Effect of Low Temperature and Hot-Water Treatment on Germinatino of Seeds in Dendropanax morbifera $L_{EV}$. (저온 및 온탕 침지가 황칠나무 종자의 발아에 미치는 영향)

  • This study was carried out to find out the method of increasing seed germination rate of Dendropanax morbifera LEV. was drupaceous fruit. The length and width of ovarywas 7.8 to 10.7mm, 6.9 to 9.0mm, respectively, and the shape was elliptical. The ovary had 5 loculi with one speed per loculus , but one seed among 5 weeds was exalbuminous seed. The length and width of seed was 6.3 to 7.4mm, 2.0 to2.mm, respectively, and 1,000 seed weight was 14.3 to 18.0g. Seeding after stroring at $10^{\circ}C$ for 60 to 90 days was germinated to 87% . However, fall seeding directly after harvest was increased to 85% at hot-water treatement ($40^{\circ}C$) for 90 to 120 minute.

Characteristics of Seed Germination and Promotion of Germination Rate in Pollia japonica Thunb. (나도생강의 종자 발아 특성과 발아율 향상)

  • This study was conducted to clarify the characteristics of seed germination and to promote the germination rate in Pollia japonica Thunb. that was pointed to protect by the Office of Forestry in 1997. It was better the germination rate and the growth of in the light than those of in the darkness. The seed germination of Pollia japonica showed the highest germination rate at $20^{\circ}C$ any other treated temperature. To increase the germination rate of Pollia japonica, it was conducted priming treatments and storage treatments at $4^{\circ}C$. Priming treatments using 1% NaOH, 1% KOH for 30min were effective compared to control, but soaking of 100mg/L GA3 for 24 hours, 1% NaOCl for 30min were not useful. It was higher in wet storage than in dry storage. Wet storage of seeds showed a germination rate with 95.3% in 90-day treatment, which improved 55.3% than dry storage in 90-day.

  • The study was performed to evaluate the product quality and shelf-life effect of low-fat comminuted sausages (LFS) manufactured with sodium lactate (SL, 2%), grapefruit seed extract (GSE, 0.2%) and Lentinus edodes (LE) powder (0.25%) alone or in combination. Addition of LE powder lower than 0.25% did not affect the product quality and sensory characteristics (p>0.05). However, LFS containing 2% SL in combined with 0.2% GSE inhibited the growth of inoculated Listeria monocytogenes during refrigerated storage for 8 wk. During refrigerated storage, pH decreased until 2 wk of storage and then increased thereafter. In addition, lightness decreased up to the 1st wk and then increased thereafter. However, redness value was the lowest at 8 wk, and expressible moisture (%) was decreased with increased storage time. Purge loss (%) was gradually increased over 8 wk of storage. Microbial counts of Listeria monocytogenes increased as storage time increased. Based on these results, LFS could be manufactured with LE powder (0.25%) and the addition of SL (2%) in combination with GSE (0.2%) inhibited the growth of Listeria monocytogenes during refrigerated storage of LFS.

Response of Germination Rate and Ascorbate Peroxidase Activity to Cryopreservation of Perilla (Perilla frutescens) Seeds with Variable Initial Viabilities (들깨 종자 활력 수준별 초저온 동결보존 후 발아율 및 Ascorbate Peroxidase 활성 변화)

  • BACKGROUND: Seed of perilla (Perilla frutescens var. japonica Hara) is short-lived in conventional storage conditions. For long-term conservation of plant species, cryopreservation is the method currently available. This study was performed to find out reliable methods for a long-term storage of seeds of perilla as a genetic resource. METHODS AND RESULTS: Using seeds of 9 perilla cultivars, the effects of desiccation, aging, and cryopreservation on seed germinability and ascorbate peroxidase activity in the seeds were investigated. Initial germinability of the seeds was various, and dry seeds of all cultivars survived cryopreservation without loss of viability. The highest germination was achieved at 4-5% moisture content, and stimulatory effect of cryogenic temperature on the seed germination was observed in some cultivars. Accelerated aging of perilla seeds led to reduction in germination and ascorbate peroxidase activity, and the susceptibility of seeds to aging was different among the tested cultivars. No significant difference in germination was observed for the aged seeds of control and liquid nitrogen exposed. CONCLUSION: The results of this study suggest that cryopreservation at 4-5% moisture content would be a suitable method for long-term conservation of perilla seeds without detrimental effects on germination.

Germination Rate and Microbial Safety during Cultivation of Disinfected Seeds (새싹 종자 소독 여부에 따른 발아율과 재배기간별 미생물 오염도)

  • This study investigated the microbial safeties and germination rates of five domestic sprout species(alfalfa, broccoli, clover, red cabbage, and red radish) grown from disinfected seeds. The 48 h germination rates of all seeds were over 90%, regardless of treatment. Seed total plate count(TPC) and coliform levels were reduced significantly(p<0.05) by treatment with 20,000 ppm calcium hypochlorite solution at $25^{\circ}C$ for 15 min, following FDA recommendations. However, after germination, all sprouts regardless of treatment exhibited bacterial counts of $10^7-10^8CFU/g$. Listeria monocytogenes was detected at $10^3-10^4CFU/g$ on germinated non-disinfected clover seeds at days 1, 2, and 5. In conclusion, although sprout germination from disinfected seeds potentially permits the growth of sprouts with lower pathogen counts, there were no significant differences in TPC or coliform levels between sprouts grown from disinfected seeds and control sprouts. Further work is needed to improve the microbial safety of cultivated sprouts and to find optimal conditions for seed germination.

Germination Behaviors and Seed Longevities of Three Ulmus Species in Korea (한국산(韓國産) 느릅나무속 3수종(樹種)의 종자저장성(種子貯藏性) 및 발아특성(發芽特性))

  • This study was conducted to investigate temperature effect on seed germination behaviors and seed viability changes by drying periods of three Ulmus species (Ulmus davidiana var. japonica (Rehder) Nakai, U. pumila L., and U. parvifolia Jacq.) distributed in Korea. Statistical analysis showed that temperature had a significant effect on mean germination time in three Ulmus species, but its effect on germination percentage was only shown in U. parvifolia (p<0.01). U. davidiana var. japonica and U. pumila germinated relatively consistent in all temperature conditions, especially the latter showed high germination behaviors in low temperatures (10-15$^{\circ}C$). U. parvifolia germinated well in high temperature (25$^{\circ}C$), while this species rarely germinated in low temperature conditions (10-20$^{\circ}C$). The results confirm that seeds of U. pumila and U. parvifolia are orthodox seeds which can be stored under 4-8% moisture condition, and U. davidiana var. japonica seed is recalcitrant seed which is difficult to store for a long time. In conclusion, these differences of the germination behaviors for three Ulmus species seem to be adaptive regimes of their natural habitats, respectively.

Effects of Light and Storage Condition on the Germination of Oenothera lamarckiana Seeds (큰달맞이꽃 종자(種子)의 발아(發芽)에 미치는 광(光) 및 저장조건(貯藏條件)의 영향)

  • The effects of light and storage conditions on the germination of Oenothera lamarckiana were investigated. Germination was induced by red light, but inhibited by green, blue or infrared. No germination occurred in the dark. Radicle was more photosensitive than other parts of the seed. The rate of germination was proportional to increase in Light intensity ranging from 10 to 3000 Lux. Photosensitivity occurred 6 hrs after water imbibition of air-dried seeds. Maximum photosensitivity was detected 1-2 days after seeding under the light condition at 300 Lux. Germination was almost completed 3 days after seeding. When seeds stored in the chilling and wetting condition were kept at constant temperature in the dark, they germinated well. Air-dried seeds, however, didn't germinate. Whereas, the seeds which kept at alternating temperature had the tendency that seeds stored at room temperature (25$^{\circ}C$) germinated better than those stored at low temperature (4$^{\circ}C$).
