Amount of Telomeric DNA on Pig Lymphocytes by Quantitative Fluorescence in situ Hybridization (양적형광접합보인법(Q-FISH)에 의한 돼지 백혈구 세포의 텔로미어 함량 분석)

  • The amount of telomeric DNA was quantified across different breeds(Landrace, Duroc, Yorkshire and Berksire), at different ages(90 days old and 180 days old) and among sexes(male and female) in pigs raised at the Performance Testing Station of Korea Swine Association, Jinkyo, Korea. The telomeric DNA amount was quantified by Quantitative Fluorescence In Situ Hybridization(Q-FISH) using a porcine telomeric DNA probe on interphase nuclei of lymphocytes. Analysis revealed that the amount of telomeric DNA on the pig lymphocytes was found to decrease with age. The quantity of telomeres significantly differed among breeds at 90 days of age. The colored breeds such as Berkshire and Duroc had higher amount of telomeric DNA than the Yorkshire and Landrace breed. In addition, the amount of telomeric DNA in male lymphocytes was significantly higher than that of females. In the correlation coefficients between the telomere quantity and their productive traits; average daily gain, loin percent and index value were positively correlated, whereas body length, feed efficiency and back fat thickness correlated negatively. However, the correlation coefficients were very low and not significant. Therefore, this study suggests that the amount of telomeres on lymphocytes can be considered as a physiological marker but not as a productive marker in pig.

Vegetation Structure of the Chilseon Valley in the Jirisan National Park (지리산국립공원 칠선계곡지역의 식생구조)

  • To investigate the vegetation structure of mountain valley from Cheonwangbong to Seonneotang, 23 plots($400m^2$) set up with random sampling method were surveyed. Two groups of Abies koreana community, Fraxinus mandshurica-Acer pseudo-sieboldianum community were classified by cluster analysis. Abies koreana was a major woody plant species in the valley area from Cheonwangbong to Mapogpo, and Pinus koraiensis, Acer pseudo-sieboldianum and Sorbus amurensis was partly occupied. High positive correlations was proved between Picea jezoensis and Sorb us amurensis, Taxus cuspidata, Fraxinus sieboldiana, Kalopanax pictus; Betula costata and Betula platyphylla var. japonica, Corn us controversa; Abies koreana and Pinus koraiensis; Stephanandra incisa and Betula ermani, Acer tegmentosum, Prunus sargentii; Acer pseudo-sieboldianum and Betula ermani; Fraxinus mandshurica and Deutzia prunifolia, Acer tschonoskii var. rubripes; Sorbus amurensis and Kalopanax pictus; Betula platyphylla var.japonica and Cornus controversa; Lindera erythrocarpa and Lespedeza maximowiczii, Quercus serrata, Styrax obassia; Lindera obtusi/oba and Abies holophylla; Carpinus laxiflora and Lespedeza maximowiczii; Fraxinus sieboldiana and Acer tschonoskii var. rubripes, Pinus koraiensis, Taxus cuspidata; Acer tschonoskii var. rubripes and Taxus cuspidata; Quercus mongolia and Rhododendron schlippenbachii; Quercus serrata and Styrax obassia, and relatively high negative correlations was proved between Abies koreana and Deutzia prunifolia; Lindera obtusi/oba and Pinus koraiensis. Species diversity(H') of investigated groups was ranged $2.7478{\sim}2.9573$.

Examination of Correlation between the Condition Evaluation Results of Superstructure and the Safety and Load-carrying Capacity of Bridges (노후 교량 유지관리를 위한 상부구조물의 상태평가 결과와 교량의 안전성 및 내하력과의 상관관계 분석)

  • In recent years, many infrastructure have been rapidly aging around the world, which grows interest in the maintenance of the infrastructure. Among the social infrastructure, bridge is a very important structure to transport lots of human and various products. The performance evaluation of bridge can be divided into the condition evaluation and safety evaluation, proposed by Korea Infrastructure Safety and Technology Corporation. However, there are no separate criteria for the performance evaluation of three-class bridges. In general, the performance of bridge is dominated by the results of the condition evaluation, which is lower than that of the safety evaluation. Therefore, this study assessed the correlation between the condition evaluation of superstructure and bridge and also between the condition evaluation and the safety and load-carrying capacity of bridge. The results of the study would provide a basic data for the more quantitative and higher relevant performance evaluation of the existing bridges, particularly for three-class bridges.

Habitats Environmental Characteristics of Polypodium vulgare L. in Ulleung-do (울릉도 미역고사리(Polypodium vulgare L.) 자생지의 입지환경특성)

  • The habitats characteristics of Polypodium vulgare L. in Ulleung-do were investigated to compile basic data for conservation and restoration. Natural habitats were located at altitudes of 410~748m with inclinations of $12{\sim}80^{\circ}$. Sixty six vascular plants were identified from 10 quadrats in 4 habitats. Dominant species among the woody plants, based on importance value, were Acer pictum subsp. mono(49.52%) in the tree (T1) layer, Sorbus amurensis(28.99%) in the subtree (T2) and Schizophragma hydrangeoides(51.99%), Ligustrum foliosum(8.82%), Fagus engleriana(7.25%) in the shrub (S) layer. Importance value for members of the herb (H) layer were as follows: Polypodium vulgare 23.23%; Maianthemum dilatatum 9.65%; Phryma leptostachya var. asiatica 9.23%; Dryopteris crassirhizoma 8.40%; Carex shimidzensis 6.75% and Dystaenia takesimana 5.42%. The importance value of the last five species were high, so they were at affinity with Polypodium vulgare in their habitats. Species diversity was 1.18, and dominance and evenness were found to be 0.11 and 0.84, respectively. The soil types were sandy loam. Average field capacity was 30.42%, and the organic matter and pH were 17.95%, and 4.70. Correlation coefficients based on environmental factors, vegetation and soil analysis were showed that positive correlations between species diversity and species richness, whereas between species diversity and dominance, coverage of Polypodium vulgare and species richness were showed negative correlations.

Vegetation Structure of Mountain Ridge from Songchu to Dobong in the Bukhansan National Park, Korea (북한산국립공원 도봉산 송추-도봉 구간의 능선부 식생구조)

  • To investigate the vegetation structure of the mountain ridges ranging from Songchu to Dobong, this research set up 22 plots$(400m^2)$ as survey target areas. As a result of the analysis of woody plant cluster, it was classified as two groups-Quercus mongolica community and Quercus mongolica-Pinus densiflora community. Quercus mongolica was found as a mostly dominant woody plant species in the ridge areas from Songchu to Dobong, while Pinus densiflora, Quercus serrata and Quercus acutissima were mixed up partly in low altitudes. High negative correlations were shown between Quercus mongolica and Quercus serrata, Quercus serrata and Acer pseudosieboldianum, Pinus densiflora and Acer pseudosieboldianum, and relatively high positive correlations were found to exist between Quercus acutissima and Quercus serrata; Quercus mongolica and Rhododendron mucronulatum; Pinus rigid a and Rhododendron schlippenbachii; Sorbus alnifolia and Rhododendron mucronulatum; Pinus rigida and Pinus densiflora; Pinus densiflora and Robinia pseudoacacia; Acer pseudosieboldianum and Styrax obassia; Magnolia sieboldii and Symplocos chinensis. Species diversity index(H') by community at the surveyed areas stayed in the scope of $0.997\sim1.160$, which indexes showed nothing different from the vegetation structure of other national parks.

Seasonal Variation of Phytoplankton Community in the Hantan River (한탄강의 식물플랑크톤 군집의 계절적 변화)

  • This study was conducted to investigate a seasonal variation of phytoplankton communities at 6 stations in the Hantan River from November, 2001 to August, 2002. Electric conductivity gradually increased from the upper reaches to the down reaches. DO differed not much from the upper reaches to down reaches, but it decreased at the station where the pollutants inflowed. T-N and T-P concentrations were high at station 4${\sim}$6. Chlorophyll-a concentration gradually increased at the mid and down reaches. It was high in May but decreased in August by the heavy rains. Phytoplankton communities were identified a total 354 taxa composed of 99 genus, 320species, 19 varieties, 3 forma and 12 unidentified species. There were 135 taxa of Chlorophyceae, 134 taxa of Bacillariophyceae, and the other taxa were Euglenophyceae, Cyanophyceae, Xanthophycaea, Chrysophyceae and Dinophyceae. Standing crops ranged from 8,600 to 337,100 cells/mL and it was lower in August than May by the rains. Dominant species included Achnanthes alteragracillima, Achnanthes convergens, Achnanthes minutissima and Cymbella minuta var. silesiaca which were the saproxenous species at the reaches, and were Cyclotella meneghiniana, Chlamydomonas pseudopertyi and Nitzschia which were saprophilous and eutrophic species at the down reaches. The correlation coefficients between phytoplankton standing crops and EC, BOD and T-N, T-P was 0.68, 0.60, 0.60 and 0.70, respectively. All correlation coefficients between Chlorophyceae and EC, BOD and T-P were higher than 0.6. A. convergens were negative correlations with EC and BOD, but N. palea were positive with EC, BOD, T-N and T-P.

Major Microbial Composition and Its Correlation to the Taste of Jeonju Kimchi (전주 김치의 적숙기 주요 미생물 조성과 맛의 상관성)

  • The 21 Kimchi samples collected by a contest for the best Kimchi homemaker in Jeonju were analysed to find major microbial composition and the correlation between sensory scores and the microbial properties, which include total microbial numbers, dominant strain number, subdominant strain number, yeast strain numbers, and floral diversity. The most common microbial composition of the Kimchi samples was Leuconostoc mesenteroides as dominant strain, and Lactobacillus sakei as subdominant strain, the pattern of which was also observed in all three Kimchi samples with the highest sensory scores, but not in all three Kimchi samples with the lowest sensory scores. However none of the other examined properties showed any significant correlation to sensory scores. This result means that the taste of fermented Kimchi depends mainly on the composition of bacterial species and that of Leuconostoc mesenteroides as dominant strain and Lactobacillus sakei as subdominant strain is the best for good taste. Yeast strains were found in the range of $3.8{\sim}4.6log\;CFU/g$ from 8 Kimchi samples and all the strains were identified to be Saccharomyces servazzii. However, fungi were not detected from any of the Kimchi. samples.

Effect of Crude Carbohydrate Content in Livestock Manure Compost on Organic Matter Decomposition Rate in Upland Soil (가축분 퇴비 조섬유 조성이 밭 토양에서 유기물 분해율에 미치는 영향)

  • The objective of this study was to characterize organic mater decomposition with application of livestock manure compost in upland soil. Six different composts, which were chicken (CHM), pig (PIM), and cow (COM) manure compost added and chicken (CHMS), pig (PIMS), cow (COMS) manure compost with sawdust added, were prepared for this study. These composts have different composition of crude carbohydrate (hemicellulose, cellulose, and lignin). The buried-bag method was used to determine the rate of organic mater decomposition and the changes of crude carbohydrate content during 36 months in the field. In all treatment, hemicellulose content was sharply decreased within 8 months, but considerable amount of lignin was remained after 36 months. After 40 months, the rates of carbon decreasing were 81, 80, 72, 69, 67, and 64 % for CHM, PIM, COM, CHMS, PIMS, and COMS, respectively. The estimated equation of carbon decreasing rate (D), $D=aT^b$, was fit to the carbon decreasing rate vs. elapsed time (T) using a non-linear regression procedure. After 40 months, significant difference of carbon decreasing rate between observed and estimated was not found. The relationship between constant a, b and hemicellulose content in the compost was not observed in this experiment. The cellulose and lignin content in the compost were positively correlated to the constant b and negatively correlated to the constant a.

Effect of nitrogen content in media on yield of Pleurotus ostreatus in bag culture (느타리 봉지재배시 배지질소함량이 수량에 미치는 영향)

  • We analyzed the relationship between the main chemical properties (pH, total nitrogen content, total carbon content, and CN ratio) of sterilized medium and mushroom yields in 1 kg bag culture of Pleurotus ostreatus. The mushroom yields appeared to be highly correlated with the pH, total nitrogen content, and CN ratio, and were the highest under a total nitrogen content of 2.0-2.2% and a the CN ratio of 20-22.5. However, of the three parameters, total nitrogen content showed the highest correlation with the mushroom yield. The coefficient of determination(r2) between the total nitrogen content and the sum of the yield was 0.931, while that of the pH and CN ratio was relatively low. The nitrogen content of the medium was the most important factor in determining the yield of oyster mushrooms.

Study on analysis of PAHs in consumer products (공산품 함유 PAHs 분석법에 관한 연구)

  • Polycyclic aromatic hydrocarbons (PAHs) that are carcinogenic and persistent will be restricted in consumer products from December 27, 2015 by EU REACH regulation. Pretreatment using Soxhlet extraction and quantitative analysis by GC-MS were studied to develop the method for analyzing 18 PAHs in consumer products as well as to detect the amounts and the kinds of PAHs in consumer products such as grips of a bag and a hammer, a cable and a plastic sandal. Linearity and precisions were evaluated by analyses of the standard PAH solutions ranging from 0.3125 mg/L to 5.00 mg of each of 18 PAHs. Linearity of resulting standard curves for all 18 PAHs were obtained with $R^2$ above 0.999. Precisions of the retention times and the peak areas were found to be 0.00%~0.05% and 1.16%~3.69% of relative standard deviations, respectively. The recoveries for spiked samples were all around 95%~105% after Soxhlet extration using three different solvents such as dichloromethane, hexane and toluene. The limits of quantitation for 18 PAHs in solutions and polymer samples by GC-MS were evaluated to be 0.327 mg/L (Benzo[ghi]perylene)~0.464 mg/L (Acenaphthylene) and 1.635 mg/kg (Benzo[ghi]perylene)~2.32 mg/kg (Acenaphthylene) based upon dilution factor of 5, respectively. Under the developed analytical method, only trace amounts of phenanthrene were detected in three samples while 15 kinds of PAHs including phenanthrene were detected in a grip of hammer with concentrations of maximum 83.4 mg/kg of Phenanthrene and minimum 8.5 mg/kg of Acenaphthylene. Further studies are needed to decrease the quantitation limit and to check the feasibility of decreasing Soxhlet time as well as to demonstrate cases that the clean up is required.
