Interaction of Hydroxyethylidene bisphosphonate (HEBP) with other endodontic irrigants on tissue dissolving capacity and antimicrobial effect (근관세정제와 상호작용시 Hydroxyethylidene bisphosphonate (HEBP)의 조직용해능력, 항균효과에 대한 연구)

  • Purpose: The purpose of this study was to evaluate tissue dissolving capacity, antimicrobial effect of Hydroxyethylidene bisphosphonate (HEBP) interacting with sodium hypochlorite (NaOCl), Ethylenediaminetetraacetic acid (EDTA) as conventional endodontic irrigants and to determine tissue dissolving efficacy depended on temperature. Materials and Methods: A total of 80 bovine muscles were randomly distributed into 8 groups (n = 10). After their initial weights determined on a precision scale, the specimens in each group were immersed in the solutions for 5, 10 and 15 min and reweighted at each time period. Agar diffusion test inoculated with Enterococcus faecalis was performed for antimicrobial effect of each endodontic irrigants. Results: The ability to dissolve organic matter was greater in NaOCl group following NaOCl and HEBP mixture. Heated NaOCl ($40^{\circ}C$) and NaOCl/ HEBP mixture was greater tissue dissolving efficacy than room temperature ($25^{\circ}C$). Antimicrobial effect was greater and significant in the following order EDTA > EDTA + 1% NaOCl > $1%\;NaOCl{\geq}1%\;NaOCl$ + HEBP. Conclusion: HEBP as soft chelating agent does not disturb antimicrobial effect and less affected tissue dissolving efficacy as inherent properties of NaOCl. In the heated NaOCl/HEBP mixture analyzed, it dissolved more the organic matter than room temperature.

Quality Characteristics of Noodle Supplemented with Skate (Raja kenojei) Skin and Bone Powder (홍어 분말 첨가에 따른 국수의 품질 특성)

  • This study evaluated the quality characteristics of dried noodles made of wheat flour supplemented with different concentrations of skate powder. Cooking quality, mechanical texture properties, gelatinization temperature and viscosity were measured, after which sensory evaluation was performed with prepared noodles. Water binding capacity, solubility and swelling power of the composite skate powder and wheat flours were higher than those of pure wheat flour. Gelatinization temperature of the composite skate powder and wheat flours increased, while initial, interim, and maximum-viscosity at $95^{\circ}C$, decreased, with increasing skate powder content. In relation to color values, increasing skate flour content led to decrease in L and b values and increase in a value. For the textural characteristics, the addition of skate powder increased hardness, springiness, chewiness and adhesiveness. Overall, noodles made with 3% skate powder were preferred compared to other samples.

Generation of antibodies against N-terminus fragment of AgI/II protein from Streptococcus mutans GS-5 (연쇄상구균(Streptococcus mutans GS-5)의 항원단백질 AgI/II의 N-terminus절편에 대한 항체형성)

  • Dental caries results from localized demineralization of tooth enamel by acids of bacterial origin produced from the fermentation of dietary sugars. A group of related oral bacteria, collectively known as mutans streptococci, are implicated as the primary etiological agents of human caries. Within this group, Streptococcus mutans has been known as a causative agent for dental caries. As well as acid production yielding the demineralization of tooth enamel, adherence and colonization of S. mutans to the teeth are also important for their virulence Cell-surface fibrillar proteins, which mediate adherence to the salivary pellicle are virulence components of mutans streptococci, and primary candidates for a human caries vaccine. Here we report that the AgI/II gene from S. mutans GS-5 were cloned by PCR amplification of the bacterial chromosomal DNA and the integrity of cloned genes were confirmed by nucleotide sequencing. Sequence analyses showed the sequence alignment of 280 nucleotides between the cloned AgI/II and the reported sequence of S. mutans GS-5 showed the perfect match The cloned genes which signal nucleotide was truncated, were transferred into bacterial expression vector and the recombinant proteins were purified as His-tag fusion proteins In order to generate polyclonal antibodies against the recombinant proteins, AgI/II mr, some $100{\mu}g$ of the proteins was injected into mice three times. It can be used for an effective vaccine production to prevent dental caries caused by pathogenic S. mutans.
