A study on the massive cultivation of cyanobacteria and strip analysis of cyanobacterial toxin (남조류의 대량배양 및 남조류 독소의 스트립분석법 연구)

  • Cyanobacterial toxins, microcystins which exist in Korean lakes show strong toxicity to fish, cattles and human. In this study, we tried to analyze cyanobacterial toxin, microcystin in the Microcystis cultivation solution using test strip, although the most common analytical methods for the detection of microcystin are HPLC and ELISA. This new anlytical method used the advantages of high specifisity and rapidness of test strip, high sensitivity of fluorescence reader. Therefore, we could analyze the trace amount of microcystin existed in various water samples without using the microcystin standards.

Systematics and Evolutionary Study of the Family Corvidae(Passeriformes) in Korea (한국산 까마귀과(참새목) 조류의 계통 진화)

  • Electrophresis methods were used to examined the degree of genetic relatedness and divergent times on the 4 genera 4 species of the family Corvidae and 1 species of the Family Cinclidae. Genetic relatedness between two families and between 4 genera of Corvidae showed similar to those reported at comparable taxonomic levels in other bird. But it appears to be considerably less than that of non avian taxa. Genetic relatedness between Garrulus glandarius brandtii and Corvus corone orientalis was the most close related(S=0.81), whereas Pica pica sericea(S=0.72) and Cyanopica cyanus koreensis(S=0.68) were showed less close relatedness respectively. The presumed divergent time between two families was 3.9 million years before present. Whereas among the 4 species of corvidae, the presumes divergent times of Cyanopica cyanus koreensis and Pica pica sericea were about 1.9 and 1.6 million years before present respectively, and between Garrulus glandarius brandtii and Corvus corone orientalis was showed recently about 1.0 million years before present

  • A survey on nation-wide distribution of fresh-water algae occurring in paddy rice fields was conducted in 1991. The algae which were collected from the whole nation were classified into a total of 54 genera including 14 genera in the blue-green algae. 29 genera in the green algae, 1 genus in the stoneworts. 3 genera in the euglenoids and 7 genera in the diatoms. The green algae and diatoms occurring in plain regions were diverse in terms of the number of genera distributed, whereas there was no regional difference in diversity of the blue-green algae and the euglenoids. Among the green algae, the suspended unicellular algae such as the genera, Chlamydomonas, Pandorina and Gonium, were widespread in plain regions, but the multicellular algae such as the genera, Spirogyra, Oedogonium, Ulothrix and Hydordictyon, were major in mountainous and attitudinal regions. The filamentous green algae such as the genera. Cladophora and Rhizoclonium, were dominant in reclaimed saline fields. The blue-green algae Oscillatoria spp. and the diatoms Navicula spp. were abundant in soil flakes.

Molecular Ecological Characterization of Wastewater Bacterial Communities in Response to Algal Growth (조류성장에 따른 하수 박테리아 군집 변화에 관한 분자생태학적 연구)

  • To deal with issues from global climate changes, renewable bioenergy has become important. Algae have been regarded as a good resource for biorefinery and bioenergy, and also have potential capability to remove nutrient and non-decompositional pollutants for wastewater advanced treatment. Although algal-bacterial ecological interaction would be a crucially important factor in using algae for wastewater advanced treatment and resource recovery from wastewater, very little is known about ecological interaction between algae and bacteria in a real wastewater environment. In this study, under a real municipal wastewater condition, we characterized wastewater pollutant treatability and bacterial communities in response to growth of Ankistrodesmus gracilis SAG278-2, which can grow in wastewater and has a high lipid contents. The growth of algal population using the wastewater was inhibited by increase in wastewater bacteria while bacterial survival and cellular decay rate were not influenced by the algal growth. Removals of recalcitrant organic matters and total nitrogen were improved in the presence of algal growth. According to T-RFLP and statistical analysis, algal growth affected time-course changes in bacterial community structures. The following 16S rRNA gene amplicon, cloning results showed that the algal growth changes in bacterial community structure, and that bacterial populations belonging to Sediminibacterium, Sphingobacterium, Mucilaginibacter genera were identified as cooperative with the algal growth in the wastewater.

Isolation of Cyanobacteria Producing Microcystin from Lakes (담수 생태계에서 Microcystin을 생산하는 남조세균의 분리)

  • Four potential microcystin-producing cyanobacteria were isolated from large reservoirs which act as sources of drinking water supply in Korea. Strain DC-2, YD-l, and YD-6 were closely related to Microcystis aeruginosa based on the analysis of l6S rRNA gene and mcyA gene sequences. mcyA gene sequence of YDS2-3, isolated from Yongdam Reservoir, was closed to that of M. aeruginosa, whereas l6S rRNA gene sequence was not related to the known sequences of microcystin-producing cyanobacteria indicating this strain can be a novel cyanobacterium belonging to the genus Microcystis. When mcyA gene sequences of isolated cyanobacteria were compared with the mcyA gene sequence library of two reservoirs, the sequence of DC-2 matched with the dominant ones.

  • Lactate and malate dehydrogenase isozymes in crude extracts of various tissues from 28 avian species were investigated by means of celluose acetate electrophoresis. In a given species the presence of two types of the malate dehydrogenase isozymes could be revealed in somatic tissues; one was characterized by fast electrophoretic mobility, and the other by slow mobility. Although the bird lactate dehydrogenase isozymes existed in a variety of species-specific molecular forms, they seemed to show the same pattern within the same genus and family.

조력, 조류 및 해류발전의 기술개발 동향

  • 화석 에너지 고갈과 지구온난화의 위기로 인하여, 미개척 분야인 해양에너지 산업에 대한 전 세계적 관심이 높아지고 있다. 특히 조력발전, 조류발전 그리고 해류발전은 현 정부의 녹색성장기술 신재생에너지에 속한 산업 군으로서, 정부정책 기조와 일치하는 해양 분야의 신성장 동력 아이템이다. 조력발전의 경우 우리나라는 조석간만의 차가 큰 서해안의 지형적 이점을 바탕으로 세계 시장에서 높은 경쟁력 가지고 있으며, 국내에서도 상용화를 위한 조류발전기 개발 및 실증시험을 진행하고 있다. 그러나 해류발전은 그 개념이 국내에서 확립되어 개발 초기단계에 있는 해양발전의 새로운 영역으로서, 전 세계적으로 개발사례가 없다. 해류발전용 해류발전기는 조류발전기에 비하여 크기가 작으나 발전용량이 크며, 개발 및 제작비용이 저렴하므로 조력발전소나 방조제의 수문 근처에 추가적으로 설치하면 매우 큰 시너지 효과를 얻을 수 있을 것으로 기대된다. 본고에서는 조력, 조류 및 해류발전에 대한 개념 소개와 조력발전과 조류발전의 국내외 개발현황 분석 그리고 새로운 개념인 해류발전에 대한 기술적 가능성을 제시하고 기대효과를 간략하게 소개하였다.

Distribution and Community Structure of Phytoplankton in the Southeast Coastal Waters During Summer 2006 (2006년 여름 남해 동부 연안 식물플랑크톤 군집 변동)

  • Short-term variations of phytoplankton community structure in the southeast coastal waters of Korea from July to September in 2006 were investigated with data set of phytoplankton, chemical and physical water properties, and meterological data. A total of 11 sampling sites of 4 different depths (surface, 5 m, 10 m, and bottom) were visited on July 11-14, July 24-26, August 7-10, August 21-24 and September 5-8. We identified 151 species in 63 genera of phytoplankton in which diatoms were the most diverse group composed of 92 species in 37 genera. Dinoflagellates were the second diverse group of 52 species in 22 genera. The other groups include 7 species in 4 genera including Raphidophytes, and Euglenophyta. After rainy season, excessive nutrients from adjacent streams to the stratified water column proliferates Chaetoceros group in July. But biomass of phytoplankton and nutrient concentrations were decreased during the period of a drought in August. However, Chaetoceros was the most dominant genera in all depths of the first, second, third and fifth cruises, except the 4th cruise on August 21-24 when dominant group were dinoflagellates including Gymnodinium spp. and Cochlodinium polykrikoides. The characteristic of phytoplankton community and environment condition during summer 2006 can be summarized as: 1) low concentration of nutrients caused by a long lasting drought in August 2) no summer outbreak of C. polykrikoides because the strength of offshore waters was weak than other years, and 3) Chaetoceros spp. was the dominant species despite short period appearance of dinoflagellates.
