Phosphorylation as a Signal Transduction Pathway Related with N-channel Inactivation in Rat Sympathetic Neurons (N형 칼슘통로 비활성화와 연계된 세포 신호전달 체계로서의 인산화과정)

  • In N-type $Ca^{2+}$ channels, the mechanism of inactivation - decline of inward current during a depolarizing voltage step- is still controversial between voltage-dependent inactivation and $Ca^{2+}$ -dependent inactivation. In the previous paper we demonstrated that fast component of inactivation of N-type calcium channels does not involve classic $Ca^{2+}$ -dependent mechanism and the slowly inactivating component could result from a $Ca^{2+}$ -dependent process. However, there should be signal transduction pathway which enhances inactivation no matter what the inactivation mechanism is. We have investigated the effect of phosphorylation on calcium channels of rat sympathetic neurons. Intracellular dialysis with the phosphatase inhibitors okadaic acid markedly enhanced the inactivation. The rapidly inactivating component is N-type calcium current, which is blocked by $\omega$-conotoxin GVIA. Staurosporine, a nonselective protein kinase inhibitor, prevented the action of okadaic acid, suggesting that protein phosphorylation is involved. More specifically lavendustin C, inhibitor of CaM kinase II, prevented the action of okadaic acid, suggesting that calmodulin dependent pathway is involved in inactivation process. It is not certain to this point whether phosphorylation process is inactivation itself. Molecular biological research regarding binding site should be followed to address the question of how the divalent cation binding site is related to phoshorylation process.

Effect of Phosphate Types on the Strength and pH of Magnesia-Phosphate Composites (마그네시아 인산염 복합체의 강도 및 pH에 대한 인산염 종류의 영향)

  • As an elementary investigation to develop vegetation concrete with a relatively low pH value, magnesia-phosphate composites (MPC) were examined according to the phosphate types including Monoammonium, Monosodium, Monopotassium, Monocalcium, Diammonium, Disodium, Dipotassium, and Diacalcium phosphates. All of the MPC binders, the ratio of magnesia to phosphate was fixed to be 7:3. MPC mortars activated with Disodium, Dipotassium, and Diacalcium phosphates showed no compressive strength gain, even at age of 28 days. Meanwhile, MPC mortars with Monoammonium and Monosodium phosphates developed 28-day compressive strength of more than 34MPa, and showed a relatively low pH value below 9.8. Hence, Monoammonium and Monosodium phosphates have potentials as an activator for producing MPC-based vegetation concrete.

Effect of Intracellular Calcium Chelator on Phosphorylation of Spinal N-Methyl-D-Aspartate Receptor following Electroacupuncture Stimulation in Rats (칼슘 저해제가 전침자극에 의한 척수 N-Methyl-D-Aspartate 수용체 인산화에 미치는 영향)

  • Objectives : We investigated the role of intracellular calcium chelator, bis-(2-aminophenoxy)-ethane-N,N,N',N'-tetraacetic acid(BAPTA), on the modulation of phosphorylation of the spinal N-methyl-D-aspartate receptor(NMDAR) NR1 and NR2B subunits following electroacupuncture(EA). Methods : Bilateral 2 Hz EA stimulation with 1.0 mA was delivered at those acupoints corresponding to Zusanli(ST36) and Sanyinjiao(SP6) in man via needles for 30min. Results : EA analgesia was reduced by intra-peritoneal injection at a higher dose of BAPTA from termination of EA stimulation. At 60 min after EA treatment, the total number of c-fos-immunostained neurons in each regions of the dorsal horn in the $L_{4-5}$ segments was decreased by BAPTA injection, especially in nucleus proprius. The mean integrated optical density (IOD) of NR1 and NR2B subunits were increased only in superficial laminae of EA-treated rats when compared with normal rats. However, the mean IOD of pNR1 was significantly decreased by BAPTA injection in both the superficial laminae and neck region and pNR2B in the superficial laminae. Western blot analyses confirmed the decreased expression of pNR1 and pNR2B. Conclusions : We concluded that intracellular calcium may well play an important role in EA analgesia by modulating the phosphorylation state of spinal NMDAR subunits.

Monitoring on Chemical Properties of Bench Marked Paddy Soils in Korea (우리나라 논토양(土壤)의 화학적(化學的) 특성(特性) 분석(分析))

  • A survey was conducted to investigate the status of soils involving pH, electrical conductivity, total organic matter content, available phosphate and major exchangeable cations in the paddy soils sampled from 1,168 sites throughout the country. The content of soil chemical properties was lower on the average than the optimum levels for cropping. An average value showed pH 5.6, organic matter $25g\;kg^{-1}$, available phosphate $128mg\;kg^{-1}$, available silicate $72mg\;kg^{-1}$, and exchangeable potassium and the calcium and magnesium were 0.32, 4.0, $1.2cmol^+\;kg^{-1}$, respectively. Soil chemical properties was related with topography except soil pH. A soil pH and organic matter, available phosphate, exchangeable potassium increased with time while exchangeable calcium, magnesium available silicate decreased with time.

  • Nephrolithiasis is the most common disorder of the urinary tract in hospitalized patients, more frequently increased in 30~50 years of age, more common in males than in females, prior right stone to left side, and than upper ureteral stone is found in cultural country, while lower ureteral stone is increased in uncultural country. Stone components are classified as calcium oxalate, calcium phosphate, magnesium ammonium phosphate, uric acid, cystine, and their mixed stone, respectively. According to the pathophysiology of urinary stones, supersaturation/crystalization of inorganic salt concentration in urine, organic matrix, inhibitor deficiency, and epitaxy theory could be based on the stone formation. Not only hypercalciuria, hyperparathyroidism, hyperoxaluria, hyperuricosuria, and cystinuria, but also renal tubular acidosis, hypervitaminosis D, and peptic ulcer, are significantly associated with nephrolithiasis. In this study upper ureteral stone component were analyzed with chemical analysis, infrared spectrum, and image analyzer from K43 patient wit erosive gastritis. As the results, mixed stone of calcium oxalate dihydrate and calcium phosphate apatite was identified, the values of clinical test in blood and urine maintained normal revels. The relapsing urinary stone from K43 have no correlation between factors for stone formation reported early, also have no evidence for risk from erosive gastritis.

Nutrition of Calcium and Phosphorus in Poultry Diets (닭에 대한 칼슘과 인의 영양)

  • Calcium and phosphorus are not only indispensable for the bone formation and body fluids equilibrium but also are major components of egg shell. It is nutritionally important, therefore, to investigate the metabolism of calcium and phosphorus and to search for optimum requirement of calcium and phosphorus and the availability of various sources of calcium an4 phosphorus by poultry. An attempt was made to review the nutrition of calcium and phosphorus in poultry diets. 1, Calcium and phosphorus have great interrelationship with vitamin D in their metabolisms. 2. Most of the plant-origin phosphorus are existing in phytic form and it leads to low availability when used in poultry rations, although calcium and phosphorus present in animal-origin or mineral supplements are highly available in general. 3. Calcium and phosphorus requirement from existing information indicated that 1.0% calcium and 0.7% phosphorus for broiler and egg-type chicks, and 3.5% calcium and 0.4% phosphorus for laying hen. 4. It has been recommended that calcium and phosphorus level should be increased when the feed intake was decreased or when the egg Production rate was higher or when the hens are old. 5. Mono-, ci-, tri-, calcium phosphate, calcium carbonate, bone meal, limestone and oyster shell u the most readily available among various sources of calcium phosphorus supplements. Soft rock phosphate, deflourinated phosphate and gypsum are somewhat inferior to the previous ones in bioavailability. 6. The effect of particle size of calcium supplements on egg shell quality and egg production rate is not yet clearly defined but recent works showed that oyster shell is more available when it was coarse and limestone is more available when it was fine in panicle. size. 7. Present data indicated that mixed feeding of oyster shell and limestone is superior to the single feeding of each on laying performance. 8. Significant interaction between phosphorus and sodium was observed, that is, excessive sodium decreased egg production in layer and body weight growth in broiler in the low phosphorus diets but increased them in the high phosphorus diets.

Effect of 35% Hydrogen Peroxide with Dicalcium Phosphate Dihydrate on the Tooth Whitening and Microhardness (35% 과산화수소에 제2인산칼슘를 함유한 치아미백제가 치아의 색과 경도에 미치는 영향)

  • The purpose of this study was to evaluate tooth whitening and microhardness after treatments with tooth bleaching agents containing dicalcium phosphate dihydrate (DCPD) and 35% hydrogen peroxide (HP) which were used in-office bleaching. Thirty enamel specimens were obtained from human premolars and randomly divided into 3 groups(n=10). Tooth bleaching agents were prepared with DCPD (0 g for controls, 0.1 g and 1 g for experimental groups) and HP solution (35% HP). All groups were applied to enamel surfaces for 60 min for 1 day. The pH of each tooth bleaching agent was measured. Tooth color, microhardness of enamel surfaces were also measured. The tooth bleaching agents containing DCPD showed a significant increase in pH compared to the ones without DCPD(p<0.05). Paired t-tests showed significant difference in color values of enamel before and after bleaching in all the groups(p<0.05). As a result, changes in color, containing DCPD group does not contain a statistically significant difference between groups was observed.(p>0.05). In all groups, tooth hardness after bleaching showed a significant decrease in microhardness (p<0.05). However, the DCPD concentration increased in the bleaching, microhardness values slightly decreased. Based on the above results, tooth bleaching agents containing DCPD and 35%HP were equally effective. Due to increases in pH and effective reduction of tooth surface decalcification, the surface characteristics are exposed to a reduced degree of negative effects, resulting in fewer constituent enamel alterations. Thus, commercial availability of the constituents of tooth whitening materials can be achieved.

The Role of $Ca^{2+}$/Calmodulin-Dependent Protein Kinase II on the Norepinephrine and GTP-Increased Myosin tight Chain Phosphorylations in Rabbit Mesenteric ${\alpha}-toxin$ Permeabilized Artery (${\alpha}$-독으로 처리한 토끼창간막동맥에서 Norepinephrine과 GTP에 의한 마이오신 인산화의 증가에 대한 $Ca^{2+}$/calmodulin-dependent Protein Kinase II의 역할)

  • The role of $Ca^{2+}$/calmodulin-dependent protein kinase II in the increase of myofilament $Ca^{2+}$ sensitivity by agonist and GTP was investigated in rabbit mesenteric ${\alpha}-toxin$ permeabilized artery. $0.3{\mu}M\;Ca^{2+}$ increased myosin light chain phosphorylations monotonically. $10\;{\mu}M$ norepinephrine and $10\;{\mu}M$ GTP potentiated increase of myosin light chain phosphorylations by $0.3{\mu}M\;Ca^{2+}$, which reaches a peak at 5 min and gradually declines to the $Ca^{2+}$ alone level at 20 min. At the early phase (1 min), $10\;{\mu}M$ KN 62, the inhibitor of $Ca^{2+}$/calmodulin-dependent protein kinase II , decreased myosin light chain phosphorylation levels by $10\;{\mu}M$ norepinephrine and $10\;{\mu}M$ GTP in the presence of $0.3{\mu}M\;Ca^{2+}.\;However\;10\;{\mu}M$ KN-62 did not affect the myosin light chain phosphorylations by $10\;{\mu}M$ norepinephrine and $10\;{\mu}M$ GTP in the presence of $0.3{\mu}M\;Ca^{2+}$ at the peak (5 min) and plateau phases (20 min). From these results, the role of $Ca^{2+}$/calmodulin-dependent protein kinase II may be different depending on time, which may play a role in increase of myofilamint $Ca^{2+}$ sensitivity by norepinephrine and GTP resulting from increase of myosin light chain phosphorylations at the early phase. However, at plateau phase, $Ca^{2+}$/calmodulin-dependent protein kinase II may not be involved in the increase of myofilament $Ca^{2+}$ sensitivity by norepinephrine and GTP in rabbit mesenteric ${\alpha}-toxin$ permeabilized artery.

Biological Availability of Various Sources of Ca and P Supplements in Young Chicks (병아리에서 인산칼슘 첨가제의 생물학적 이용성)

  • This experiment was carried out to evaluate biological availability of Ca and P in 4 different sources of tricalcium phosphate in young chicks. One hundred and twenty five-day-old male Single Comb White Leghorn chicks (10 treatments$\times$3replication$\times$4chicks) were used in trial 1 and 2, respectively, for 12 days of feeding period. Trial I was to evaluate the availability of phosphorus in the supplements, Standard purified diets were prepared to supply 0.07, 0.14 and 0.21%P using a mixture (1 : 1) of NaH$_2$PO$_4$ and KH$_2$PO$_4$ as the reference Phosphorus sources. Bone breaking strength of the tibia determeined by an Instron instrument appeared inadequate to be used as a criterion due to very high variations of the measurement within a treatment. Thus, tibia bone ash content was utilized as a criterion to evaluate th biological avilability of phosphorus in the supplements. The levels of the biological availability of the four different sources of dicalcium phoshate were 77.1, 91.0, 96.4 and 95.5%, respectively, and those of the three tricalcium phosphate sources were 94.1, 95.0 and 99.5% , respectively. Trial 2 was to determeine the levels of Ca biologically available in the supplements. Standard purified diets were made to supply 0.2, 0.3 and 0.4% Ca using CaCo$_3$ as the reference calcium source. When bone ash content was utilized as a criterion for the availability, the levels of calcium biologically available to the chicks were 78.3, 234.1, 87.6 and 244.5%, respectively, for the 4 different sources of dicalcium phosphate and 99.5, 84.0 and 101.5% , respectively, for the 3 different sources of tricalcium phosphate. The observation that two calcium sources appeared to be utilized with an unusual efficiency can hardly be explained at this moment. When they were revaluated on the basis of body weight gain, the availabilities of the four sources of dicalcium phosphate were 89.2, 58.2, 104.1 and 103.1% and of the three tricalcium phosphate were 112.6, 106.0 and 96.3% , respectively.
