THE EFFECTS OF CONCENTRATION AND pH OF LACTIC ACID ON THE FORMATION OF ARTIFICIAL ROOT CARIES LESION (젖산의 농도와 pH가 인공 치근 우식 병소의 형성에 미치는 영향)

  • The purpose of this study was to evaluate the effect of concentration and pH of lactic acid on the formation of artificial root caries lesions formation in bovine teeth. The characterictics of artificially produced early root lesions were observed with polarized light microscope and the depth of lesions were mearsured with measuring microscope The results were as follows: 1. In the group of low pH and high concentration of lactic acid, the progress of lesion formation was faster than that of high pH and low concentration lactic acid group. 2. In the same group, initial lesion progress faster, but progression rate was reduced as time goes by. 3. In the development of initial root caries, cementum was act as a barrier to protect dentin from lesion progression.

Antimicrobial Activity of Garlic Extracts according to Different Cooking Methods (조리방법을 달리한 마늘 추출물의 항균활성)

  • This study was conducted to find the antimicrobial activity of garlic extracts by various processing methods(boiled, pan fried, microwave heated, pickled). Ethanol and water extracts from garlic sample were prepared and antimicrobial activities were determined against 10 microoganisms ; food borne pathogens, food poisoning microoganisms, food-related bacteria and yeasts. The ethanol extracts from the fresh and pickled garlic showed antimicrobial activities for test microoganisms, except lactic acid bacteria and yeast. However, the antimicrobial activities were decreased by heat treatment. The minimum inhibitory concentration(MIC) of the fresh garlic extracts was determined to 0.1 mg/mL against an gram positive bacterium and 0.5 mg/mL against an gram negative bacterium. The antimicrobial activities of the ethanol extracts were affected by heating methods and not by pHs.

Changes in microbial community and physicochemical characterization of Makgeolli during fermentation by yeast as a fermentation starter (효모 첨가 유무에 따른 막걸리의 발효 중 미생물 군집 및 이화학적 특성 변화)

  • This study was carried out to confirm changes in the microbial community and physicochemical characteristics of Korean traditional Makgeolli during fermentation by yeast as a fermentation starter. We demonstrate that the microbial community during fermentation affects the quality of Makgeolli. At the species level, Pediococcus pentosaceus, Weissella confusa, Pantoea vagans, and Lactobacillus graminis were dominant on fermentation mix, after 1-2 days, in the control group without yeast treatment. Acid production in the control group was higher than that in the yeast-treated group. P. pentosaceus was dominant throughout the fermentation process, and the proportion of P. vagans remarkably decreased following yeast addition. Considering quality characteristics, the alcohol content rapidly increased after yeast addition, and the lactic acid content was lower in the yeast-treated group than in the control. These results suggest that the rapid increase in alcohol at the start of fermentation inhibits the growth of lactic acid-producing bacteria. The addition of yeast may contribute to the reduction in the high amount of lactic acid, which can be one of the causes of changes in Makgeolli quality.

Influence of Anaerobic Exercise on Fatigue Material and Basic Fitness of Career Wrestlers (무산소성 운동이 경력별 레슬링 선수의 피로물질 및 기초체력에 미치는 영향)

  • This study is an experiment to look into recovering process after maximum anaerobic exercise of career wrestlers and their basic fitness. First, group time results in anaerobic power test did not show meaningful differences between groups in PP, MP, LP field. Second, blood glucose showed meaningful difference in 5-year wrestlers group according to time-wise result (p<.001). Third, time-wise results in blood lactic acid concentration showed meaningful difference all in 5-year, 10-year and 15-year career wrestlers group (p<.001). Fourth, side step and general reaction did not show meaningful differences in all the 3 groups. In conclusion, increase or decrease was seen from and also significant differences were seen from the change of anaerobic power, glucose, lactic acid, concentration, basic physical strength reaction which have influence over athletic performance of players after anaerobic exercise. It was found that 5-year career players group was good at variables of power test and glucose. Reason why recovery capability in connection with degree of fatigue was poor in 15-year career players group seemed to have correlation between career and age.

Development of Biosensor for Simultaneous Determination of Glucose, Lactic Acid and Ethanol (포도당, 젖산 및 에탄올의 동시 측정용 바이오센서의 개발)

  • The purpose of this study is to develop biosensor for determination of glucose, lactate, and ethanol in foods and food-stuffs simultaneously. The multiple cathode system was prepared with an oxygen electrode having one anode and hexagonal cathode. Glucose oxidase, mutarotase, lactate oxidase, alcohol oxidase and catalase were used for immobilization to determine glucose, lactate, and ethanol. These components including ethanol were simultaneously determined by the immobilized enzymes in the multiple cathode system. The determination of the components by enzyme sensor was based on the maximum slope of oxygen consumption from enzyme reaction of each sensor part. The response time for analysis was 1 min. The optimum condition for glucose, lactate and ethanol sensor was found to be 0.1 M potassium phosphate buffer, pH 7.0 at $40^{\circ}C$. Interferences of various sugars and organic acids were investigated. Less than 10% of error was found in determination of the components except organic acids. This difference was compensated by the modified equation. This system was confirmed by conventional methods. It was concluded that the multiple cathode system of this study is for an effective method to determine sugar, organic acid, ethanol simultaneously in foods.

The Effects of Sugar Addition in Yogurt Prepared from Egg White Powder and Casein (난백분말과 카제인으로 만든 요구르트에서 당의 첨가 효과)

  • A curd yogurt was prepared from egg white powder (EWP) and casein added with sugars (glucose, fructose, lactose). The effects of sugar addition on acid production and growth of Lactobacillus were studied. The effects of sugar addition on sensory property and volatile aroma compounds were also studied. Acid production by L. acidophilus in EWP 2% (W/V), casein 3% (w/v) and sugar 0.5,1 or 2% W/V) was lower than that of L. acidophilus in milk (control). Acid production in sample added with glucose or fructose of 1% or 2% (W/V) was higher than that of 0.5% (W/V), while acid production in lactose added sample was not affected with the concentration of lactose. Number of viable cells of L. acidophilus at 24 hr in milk, glucose added sample, fructose added sample and lactose added sample was 3.6${\times}$10/Sup 9/, 5.6${\times}$10$\^$8/, 6.0${\times}$10$\^$8/,and 3.2${\times}$10$\^$7/, respectively. Through 30hr fermentation, acid production and number of viable cells of L. acidophilus in milk were higher than those of sugar added samples. Sensory property of fructose added sample was slightly better than that of milk yogurt (reference), while that of lactose added sample was significantly inferior. Though the composition of volatile aroma compounds was slightly different according to sample, gas chromatographic analysis detected acetone, ethanol, diacetyl and acetoin in samples fermented by L. acidophilus.

Effect of Whey Brew Cultured by Lactobacillus helveticus ATCC 55163 and Propionibacterium acidipropionici 5020 on Quality Characteristics of Bread (Lactobacillus helveticus ATCC 55163과 Propionibacterium acidipropionici 5020로 배양한 유청발효물이 빵의 품질특성에 미치는 영향)

  • This study evaluated the effect of whey brew cultured by Lactobacillus helveticus ATCC 55163 and Propionibacterium acidipropionici 5020 on bread quality characteristics. Ten and 15% whey brew were added to flour-based bread, after which bread volume, pH, total titratable acidity (TTA), moisture content, water activity, texture, organic acid content, and sensory evaluation were analyzed. The bread volume and TTA of control were the largest among the samples, whereas pH was the lowest. Moisture content did not significantly differ depending on the amount of whey brew added, though water activity was highest in the bread with 10% whey brew. However, hardness was the lowest in bread with 10% whey brew. Propionic acid was not detected while succinic acid, lactic acid, and acetic acid were detected in small amounts in the control compared to the test samples. Succinic acid, acetic acid, and lactic acid content was high in bread with 15% whey brew, with propionic acid present at a very high amount. In terms of sensory evaluation, bread with 10% whey brew had the highest score. As a result, high quality characteristics were associated with the bread with 10% whey brew, whereas long preservation was a characteristic of the bread with 15% whey brew.

Studies on the Optimum Fermenting Conditions of Dongchimi for Production of Ion Beverage (이온음료 제조를 위한 동치미의 최적 담금 조건에 관한 연구)

  • The study was conducted for optimum fermenting conditions of Dongchimi(pony tailed chinese radish kimchi) in production of ion beverage. The changes of pH and total acidity were increased as the temperature increased. Non-volatile organic acids, such as lactic acid, citric acid, malic acid and succinic acid were produced in Dongchimi fermentation. The amount of lactic acid was increased higher, followed by citric acid and malic acid. However succininc acid was produced a little of amount at $0^{\circ}C$. Lactic acid producing bacteria number increased in initial period and then decreased in last period of fermentation. During lactic acid producing bacteria was increased, the amouont of lactic acid was increased. The flavor components were tentatively identified as methyl pentane, ethyl thioethene 2, 3-diazaindolizine, dimethyl disulfide. The optimum fermenting conditions of Dongchimi for production of ion beverage were 24~29 days at $0^{\circ}C$, 9~12dyas at $5^{\circ}C$ and 16~22days at $10^{\circ}C$, respectively.

Fermentation of Momordica charantia Extract using Leuconostoc mesenteroidesies and Physiological Activity of Product (Leuconostoc mesenteroidesies 균주를 이용한 여주 추출물 발효 및 생산물의 생리활성 특성)

  • In this study, Momordica charantia (MC) fermented with Leuconostoc mesenteroides (MC-LM) were assessed for the antioxidant and the antidiabetic activities. Antioxidant activities of MC and MC-LM were evaluated using 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)(ABTS) radical. Although MC-treated groups showed little activity, 47% of activity was observed at $500{\mu}g/mL$ concentration for MC-LM and increased significantly(p<0.05) as MC-LM concentration increased. MC-LM more effectively inhibited the oxidative damage of DNA by peroxyl radical than MC and the inhibition of the strand breakage increased significantly as MC-LM concentration increased(p<0.05). Measuring the inhibition of ${\alpha}-glucosidase$ activity, which is closely related to the regulation of blood sugar, resulted in MC reduced the activity of ${\alpha}-glucosidase$ by 30% at 8 mg/mL and MC-LM at the same concentration by 60%. In addition, the effect of MC-LM on the cell viability of alloxan-treated RIN-m5F resulted in a significant increase in cell survival(p<0.05) in the group treated with MC-LM and a 20% increase in the concentration of $1000{\mu}g/mL$. As a result of insulin secretion by alloxan-treated RIN-m5F cell, the level of insulin secretion tended to increase in all group treated with MC-LM. At the concentration of $1000{\mu}g/mL$, the insulin secretion was increased by 15% in MC-LM group than in MC group. In conclusion, the results of this study suggest that fermented bitter gourd has antioxidant and antidiabetic effects.

Characteristics of Whelk Internal Organ Jeotgal with the Addition of Bromelain (Bromelain 첨가에 따른 골뱅이 내장 젓갈의 특성)

  • To use the by-products of whelk processing, whelk internal organ with the addition of bromelain were used to make jeotgal, Korean traditional salted and fermented seafood sauce. The products were prepared at different salt concentrations of 10, 15 and 20% with various bromelain contents 0, 0.01, 0.1 and 0.5%. The samples were stored at $10^{\circ}C$ and the chemical and microbiological analyses were evaluated for four weeks. The initial pH (near 6.1) in all conditions decreased in the beginning stage of aging, then gradually increased until three weeks and finally decreased to $5.6{\sim}6.0$. The sample with lower salt concentration showed less pH change For two weeks. The samples treated with higher bromelain in 10% and 15% salt concentration showed higher pH values after 4 weeks compared to the nontreated control. Amino nitrogen in the samples increased at lower salt concentration and at higher bromelain content. The amino nitrogen showed maximum value, 780 mg%, at 10% salt and 0.5% bromelain treatment. Total nitrogen contents increased with aging periods, especially increased rapidly in high concentrations of bromelain treated samples at early stages of aging. The higher salt concentrations in the product decreased the total microbial number and lactic acid bacterial number.
