Experimental Studios on the Mechanism of Reproductive Cycle in the Longchin Goby Chasmichthys dolichognathus (HILGENDORF) (점망둑, Chasmichthys dolichognathus의 생식기구에 관한 실험적 연구)

  • The mechanism of reproductive cycle of longchin goby Chasmichthys dolichognathus was investigated for the annual variations of gonadosomatic index(GSI), histological changes of gonad, reproductive cycle, and initiating and terminating factors of the breeding season. The experimental fish were captured in the tide pool of Tongbaeksom, Pusan, Korea from February 1983 to September 1984. Experimental fish for the study of initiating and terminating factors of the breeding season were maintained in filtered recirculating aquariums and exposed to various temperature and photoperiod regimes. The ovary is a pair of sac-shaped organ, and the testicular structure is tubular and it is connected to the seminal vesicle which is located at the posterior end of the testis. GSI began to increase from February when the water temperature began to increase and reached peak in April. It began to decrease from August, the top water temperature season, and maintained relatively low values until January. The annual reproductive cycle includes successive six developmental stages : early growing from December to February, late growing from January to March, early mature from February to May, late mature and ripe from March to August, spawning from April to July, degeneration and resting stage from May to December. An experimental study based on the reproductive cycle of this species indicated that rising temperature under long photoperiod during spring stimulated gonadal maturation, whereas terminating factor of the breeding season with recrudescence of the gonads was demonstrated as high water temperature during summer regardless of photoperiod.

$\beta$-Mercaptoethanol이 돼지 체외수정란의 생산에 미치는 영향

  • $\beta$-Mercaptoethanol($\beta$-ME)은 일반적으로 황화합물(thiol compounds)의 일종으로, 배양액 중에서 이황화결합(disulfide bonds)을 분해하여 일정한 물질의 산화.환원반응에 관여하며, 특히 cysteine이 cystine으로 산화되는 것을 차단함으로서 cysteine의 이용능력을 증대시키고, GSH의 합성을 촉진 및 증대시키는 것으로 알려져 있고, 각종 활성산소로부터 세포를 보호하는 역할을 수행하는 것으로 보고되었다. 특히, 돼지수정란의 체외배양체계에 유의적인 영향을 미치는 것으로 보고되었다(Abebydeera 등, Theriogenol., 50:747-756, 1998). 따라서 본 실험에서는 돼지난포란의 체외성숙시 $\beta$-ME의 첨가배양이 체외수정과 배발달에 미치는 영향에 관하여 조사하였다. 돼지난포란을 10% PFF, 0.1mg/ml cysteine, 10IU/m1 PMSG, 10IU/m1 hCG 및 10ng/m1 EGF가 첨가된 NCSU23 배양액에 $\beta$-ME를 각각 0, 25, 50 및 100uM을 처리하여 22시간 동안 배양을 실시하고, 성선자극호르몬이 배제된 배양액에서 추가로 22시간을 배양하여 체외성숙을 유도하였다. 체외성숙이 유기된 난자는 난구세포를 제거하고, 2.5mM caffeine과 0.1% BSA가 첨가된 mTBM배양액에 정자를 1.25 $\times$ $10^{5}$cells/ml의 농도로 5-6시간 동안 공동배양을 실시하여 체외수정을 유도하였다. 체외수정후 일부의 수정란은 12시간에 난자 급속 염색방법으로 염색하여 다정자침입률 및 자.웅전핵형성률 등을 확인하였다. 그리고 나머지1-세포기의 수정란은 0.4mg/ml BSA가 함유된 NCSU23 배양액에 30 embryos/50ul 소적으로하여 38.8$^{\circ}C$, 5% $CO_2$의 탄산가스 배양기에서 각각 7일간 배양을 실시하였다. 조사된 결과는 SAS/STAT를 이용하여 통계분석을 실시하였다. 체외수정 12시간 후에 난자 급속 염색법으로 염색을 실시한 결과, 모든 처리구에서 핵성숙률(76.4~95.2%), 정자침투율(51.1~66.9%), 웅성전핵형성률(95.2~100%), 다정자침입률(18.2~25.6%) 및 평균침입정자수(1.2~l.4개)에서 유의적인 차이가 인정되지 않았다. 체외배양 48시간 난할률을 조사한 결과, 처리구별 차이(53.9~67.9%)는 인정되지 않았으나, 배양 7일째 배반포형성률은 각각 14.5, 25.4, 17.3 및 12.4%로서 25uM의 $\beta$-ME처리구가 유의적(P<0.05)으로 높은 배발달률을 나타내었고, 총세포수에 있어서는 대조구와 처리구간 유의적인 차이가 인정되지 않았다. 따라서 돼지 난포란을 성숙배양할 때, 25uM $\beta$-ME를 첨가배양하는 것이 양질의 돼지체외수정란을 생산하는 하나의 방법으로 조사되었다.다.

Repression of HspA2 mRNA Expression in Human Testes with Abnormal Spermatogenesis (비정상적 정자형성 환자의 정소에서 Heat Shock Protein A2 (hspA2) mRNA 발현의 감소)

  • Objective: Heat shock protein 70-2 (Hsp70-2) gene knockout mice are found to have premeiotic arrest at the primary spermatocyte stage with a complete absence of spermatids and spermatozoa. This observation led to the hypothesis that hspA2 may be disrupted in human testes with abnormal spermatogenesis. To test this hypothesis, we studied the mRNA expression of hspA2 in infertile men with azoospermia. Design: The mRNA expression were analyzed by competitive RT-PCR among testes with normal spermatogenesis, pachytene spermatocyte arrest, and sertoli-cell only syndrome. Materials and methods: Testicular biopsy was performed in men with azoospermia (n=15). Specimens were subdivided into three groups: (group 1) normal spermatogenesis (n=5), (group 2) spermatocyte arrest (n=5), (group 3) Sertoli-cell only syndrome (n=5). Total RNA was extracted by Trizol reagent. Total extracted RNA was reverse transcribed into cDNA and amplified by PCR using specific primers for hspA2 target cDNAs. A competitive cDNA fragment was constructed by deleting a defined fragment from the target cDNA sequence, and then coamplified with the target cDNA for competitive PCR. Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) gene was used as an internal control. Results: On Competitive RT-PCR analyses for hspA2 mRNA, significant amount of hspA2 expression was observed in group 1, whereas a constitutively low level of hspA2 was expressed in groups 2 and 3. Conclusion(s): The study demonstrates that the hspA2 gene expression is down-regulated in human testes with abnormal spermatogenesis, which in turn suggests that hspA2 gene may play a specific role during meiosis in human testes.

Characteristics of Sexual Maturation in the Seaweed Porphyra pseudolinearis from East Sea, Korea (동해안 긴잎돌김(Porphyra pseudolinearis)의 성성숙기 특성)

  • The seaweed Porphyra pseudolinearis as a dominant species grows at upper of the intertidal zone of the East Sea from October to March. Vegetative cells have not matured during October when observed with naked eye and microscope. In the middle of November, although it didn't distingush between female and male gametophytes by eye, it showed antheridium and carpogonium when observed vertical section under microscope. From early December, It could be distinguished female and male gametophytes clearly. From Feburary it showed shorted length of thalli by release of spermatangia and carposporangia as maturation. At early March, the colar has been decaeded and side of thalli has been melted, completed of release of spermatia. At the end of December, the average length and width of female thalli were 149.9$\pm$5.6mm and 22.2$\pm$2.3mm, respectively. The length and width of male thalli were 149.9$\pm$9.4mm and 20.7$\pm$1.8mm. At the end of January, the average length and width of female thalli were 94.6$\pm$6.4mm and 29.1mm$\pm$5.1, respcetively. The length and width of male thalli were 107.8$\pm$7.3mm and 25.9$\pm$0.9mm. From this period lengths of female and male thalli have already been shortned by the release of spermatia and carpogonia.

Cultural Landscape of Saegumjeong Area as a Recreation Place of Scholars in Joseon Dynasty (조선시대 선비들의 행락공간으로서 세검정 일대의 문화경관)

  • The area surrounding Segeomjeong, one of the famous pavilions near the capital in the Joseon Dynasty, was a favorite place of scholars to visit and enjoy the beautiful natural landscape. The recreational culture of Segeomjeong area in traditional societies representing the prototype of urban tourism in modern society indicates cultural landscape which is a combination of the surrounding landscape and human activities. At this point, the purpose of this research on Saegumjeong area as a recreation place was to examine recreational activities and landscape elements before restoration. Through the relevant historical literature, there were several recreational activities such as traveling to enjoy scenery, taking a view of stream with waterfalls, doing creative writing of reciting poems and drawing about the greatness of nature and gathering on a broad and flat rock or Tangchundae. Also, the main landscape elements-such as Saegumjeong, stream, broad and flat rocks, pine trees, Tangchundae Peak, and mountains-were extracted through paintings of Saegumjeong drawn before restoration. Consequently, in this study, it was confirmed that the Segeomjeong area was the spot that scholars visited consistently, the center for travel, and the place for communication between scholars and national banquets. This study is significant, in that it drew conclusions on the cultural landscape of the Korean people through the ages and helped to discover the meaning and value of cultural landscape of recreation places based on Korean natural characteristics and cultural climate.

HIDE, a Testis Specific Deubiquitinating Enzyme, Interacts with HSP90 (고환 특이적으로 발현되는 탈유비퀴틴효소 HIDE와 HSP90의 상호작용)

  • 연구목적: 본 연구는 아직 그 기능이 파악되지 않은 탈유비퀴틴효소 중 하나인 HIDE에 대한 기본적인 생화학적 특징과 고환에서의 발현 양상을 파악하고 있다. 연구재료 및 방법: 인간의 HIDE 유전자를 클로닝하여 효소활성이 있는지 세포 외 실험을 통해 확인하였고, 아미노산 서열을 분석하여 진화상 보존된 부분을 찾아 그 기능을 파악한 다음 HSP90과의 상호작용을 공동면역침전반응으로 확인하였다. HIDE의 조직별 발현양상을 파악하기 위해서 인간과 쥐의 RNA 블롯과 쥐의 단백질 블롯을 이용하여 각각 노던 블롯팅과 웨스턴 블롯팅을 수행하여 고환에서 많이 발현된다는 것을 알았고 이 사실을 바탕으로 쥐의 고환을 절개하여 면역조직화학반응으로써 고환 내의 HIDE 단백질의 발현양상을 파악하였다. 결 과: HIDE는 세포 외에서 유비퀴틴 잔기를 제거하는 탈유비퀴틴 활성이 있으나 세포 내에서 전체적인 유비퀴틴 복합체를 줄여주는 효과는 없었다. HIDE는 HSP90이라는 분자 샤페론과 상호작용한다. HIDE의 전사체는 고환에서 가장 많이 발현되며 다른 조직에서도 소량 발현된다. HIDE의 단백질은 웨스턴 블롯상에서 고환에서만 확인되었다. 고환 내에서의 HIDE의 발현양상은 왕성한 감수분열을 하는 정모세포에서 높았으며 지지세포나 정조세포에는 발현되지 않았다. 결 론: HIDE는 분자 샤페론 HSP90과 상호작용하며 고환 내의 감수분열 중인 세포에서 많이 발현되는 것으로 보아 감수분열이나 정자형성에 관여하는 것으로 보인다.

A Study on the Induction of Infertility of Largemouth Bass (Micropterus salmoides) by CRISPR/Cas9 System (CRISPR/Cas9 System을 활용한 배스의 불임 유도에 대한 연구)

  • A largemouth bass (Micropterus salmoides) is an ecosystem disturbance fish species at the highest rank in the aquatic ecosystem, causing a serious imbalance in freshwater ecosystems. Although various attempts have been made to eradicate and control largemouth bass, no effective measures were found. Therefore, it is necessary to find an approach to maximize the effective population reduction based on the unique characteristics of largemouth bass. This study used the transcriptome analysis to derive 182,887 unigene contigs and select 12 types of final target sequences for applying the CRISPR/Cas9 system in the genes of IZUMO1 and Zona pellucida sperm-binding protein, which are proteins involved in sperm-egg recognition. After synthesizing 12 types of sgRNA capable of recognizing each target sequence, 12 types of Cas9-sgRNA ribonucleoprotein (RNP) complexes to be used in subsequent studies were prepared. This study searched the protein-coding gene of sperm-egg through the Next Generation Sequencing (NGS) and edited genes through the CRISPR/Cas9 system to induce infertile individuals that produced reproductive cells but could not form fertilized eggs. Through such a series of processes, it successfully established a composition development process for largemouth bass. It is judged that this study contributed to securing the valuable basic data for follow-up studies to verify its effect for the management of ecological disturbances without affecting the habitat of other endemic species in the same water system with the largemouth bass.

A Study on the Location and Spatial Organization Characteristics of the Royal Tombs Uireung (의릉(懿陵) 일원(一圓)의 입지(立地)와 공간구성특성(空間構成特性)에 관(關)한 연구(硏究))

  • The purpose of this study is to examine the characteristics of the location and the spatial composition of Uireung that is located in Seokgwan-dong, Seongbuk-gu, Seoul, in order to understand the landscape architectural characteristics. The results are as follows. First, Uireung is 6.4km from Changdeokgung Palace and 5.5km from Heunginjimun Gate. It did not violate the distance standard (40km) for the royal tombs according to Joseon Dynasty Neung-won Myo-je. Second, Uireung is in harmony with the nature and shows the authoritative characteristics of the royal authority through the spatial composition and rank(Entrance Area, Ceremonial Area, Burial Area). Third, there are burial mound, stone sheep, stone tiger, stone table, stone watch pillars in the upper platform, and stone civil official, stone horse, stone lantern in the middle platform, and stone military official, stone horse in the lower platform, and T-shape shrine, worship road in the ceremonial area. There is no pond and a tomb keeper residence, but the position, size, and form can be approximated through historical research materials. There are a colony of pine trees around the burial mound and 64 species of trees such as pine tree, zelcova tree, and fir tree below the burial mound.

Microanatomical Structure of Male Reproductive Organ in the Common Squid, Todarodes pacificus (Cephalopoda: Ommastrephidae) (살오징어 Todarodes pacificus 수컷 생식기관의 미세해부학적 구조)

  • This study was performed to understand the reproductive ecology of cephalopods, described the microanatomical structure of the male reproductive organs and spermatophore in the common squid, Todarodes pacificus, a major cephalopods in Korea. The common squid was gonochorism and had sexual dimorphism, the color of the reproductive organs reflected on the mantle and the presence of the hectocotylus. Male reproductive organs were composed of testis, primary vas deferens, spermatophoric gland (seminal vesicle), spermatophoric sac (Needham's sac) and secondary vas deferens. The male has specialized reproductive organs such as the spermatophore forming organ, spermatophoric gland. Testis was histologically a seminiferous tubule type. The primary vas deferens was a thin and inverted triangular spring-like form that connected from the rear of the testis to the spermatophoric gland. Inside, it was filled with sperm of basophilic in H-E stain. The spermatophoric gland is an irregular oval connected to the primary vas deferens and spermatophoric sac, and there were a number of tubular glands. The spermatophoric sac is a tubular structure located between spermatophoric gland and secondary vas deferens, and a number of spermatophores have been identified in the lumen. The secondary vas deferens was connected to the posterior of the spermatophoric sac and had a spermatophore inside. The spermatophore was a long, transparent tube about 22 mm long, with a sperm mass inside.

Gametogenesis and Reproductive Cycle of the Cockle, Fulvia mutica (Reeve) (새조개, Fulvia mutica (Reeve)의 생식세포형성과정 및 생식주기)

  • The structure of gonads, gametogenesis and reproductive cycle of the cockle, Fulvia mutice, were studied mainly by histological observation. The materials were monthly sampled in the southern area of Yeosu from October 1980 to September 1981. F. mutica was monoecious. The gonads were situated between the liver tissues and the outer fibronuscular layers compacted by the connective tissue fibers and muscle fibers beneath the outermost layer of simple cuboidal epithelium. The gonad was composed of a number of the ovarian sacs and the testicular tubules which form the tubular structure. Testicular tubules in the mature stage sometimes contained 'testis-ova' The undifferentiated mesenchymal tissues and the eosinophilic cells were abundantly distributed on the germinal epithelium in the early development stage. With the further development of the ovary and testis, these tissues and cells gradually disapprared. The undifferentiated mesenchymal tissues and the eosinophilic cells are related to the growing of the oocytes and spermatocytes . Early multiplicating oogonium was about $10{\mu}m$ in diameter. As the oocytes grow to $27-34\times50-58{\mu}m$ by increasing cytoplasm, the oocytes connected to the basement membrane by their egg-stalks. The ripe eggs were about $60{\mu}m$ in diameter and they were surrounded by gelatinous membrane. Most male germ cells in mature stage were transformed into the spermatozoa and they formed the sperm bundles. After spawning, undischarged ripe eggs and spermatozoa remained in the ovarian sac and the testicular tubule respectively for some time, then they finally degenerated. Especially the early spent ovarian sacs in May did not contract significantly and then they took part in the secondary maturation within two or three months during the summer season. The monthly changes of the fatness well agreed with the reproductive cycle. The reproductive cycle of F. mutica could be classified into six successive stages : multiplicative, growing, mature, spent, degenerative and recovery stage. It seems that the spawning season is closely rotated to the water temperature, and the spawning occurs from May to October at about $20^{\circ}C$ in water temperature. The peak spawning seasons appeared twice a year between June and July and in September.
