Passive Immunity by Splenocyte Transfer against Amebic Meningoeneephalitis in Mice (세포에 의한 아메바성 수막뇌염에 대한 피동면역의 전달)

  • The role of passive cell-mediated transfer of immunity against primary amoebic meningoen- cephalitis(PAME) in mice was studied. Waegleria fowleri, ITMAP 359, were cultured in CGVS medium. The ICR mice used were six week-old males of average weight of 15 g. Immunization was done by three intraperitoneal injections of $1{\times}10^6$ N. fowleri trophozoites at the interval of one week. Splenocytes were obtained from normal and immune mice spleens, and Ix107 cells were administered intraperitoneally into mice 3 days before challenge infection. Mice were infected intranasally with $7{\times}10^4$ N. fowleri trophozoites in a $3{\;}{\mu}l$ suspension under secobarbiturate anesthesia. Transplants of normal or immune splenocytes seem to alter the pattern of the PAME level- opment. The splenocytcs transferred from immune mice reduced the mortality rate in the JV. fowleri infected mice, as compared with the mice transferred with the same number of normal splenocytes or without splenocyte, The blastogenic response of the splenocytes to both lipopoly- saccharide and concanavalin A was elevated on duty 7 after infection the mice transinoculated with immune splenocytes. The serum antibody titers in the mice transferred with immune spleno- cytes were increased gradually from day 7 up to day 20 after infections by mean of ELISA. It is suggested that the transfer of splenocytes from immuniged mice conferred immunity against N. fowleri infection.

  • Ceramide rich intercellular lipid lamellae are thought to be particularly important in maintaining the structural integrity of epidermal barrier. Ceramide is synthesized de novo by serine palmitoyltransferase (SPT) phospholipid intermediates, serine and palmitic acid persist within the stratum corneum. The ceramide which is synthesized is degraded with fatty acid and sphingosine by degradative enzyme ceramidase. The depletion of ceramide in stratum corneum was reported in the atopic dermatitis. As an effort to search for the dietary source for improving the level of ceramide in epidermis, the dietary effects of various-typed silk protein were compared. Seventy male NC/Nga mice, an animal model of atopic dermatitis, were divided into seven groups: group CA as an atopic control with control diet, group S: 1% crude sericin diet, group F: 1% crude fibroin diet, group PS : peptide pattern of sericin(Mw 5000), group PF: peptide pattern of fibroin (Mw 1500), group AS: manufactured the same as amino acid profile of sericin and group AF: manufactured the same as amino acid profile of fibroin. Ten male BALB/c mice were served as group C (control group) control diet. All mice were fed on diet and water ad libitum for 10 weeks. Dry skin condition was established in group CA as ceramide content was decreased. Despite a marked decrease of mRNA and prorein expression of SPT, enzyme do novo synthesis, ceramide content of group S was dramatically increased by inhibiting the mRNA and protein expression of degradative enzyme ceramidase. However, dietary supplementation of crude silk fibroin protein (group F) and in other groups that were supplemented with either amino acid or peptide type of sericin or fibroin did not increase the level of ceramide. Together, our data demonstrate that dietary supplementation of crude sericin is more effective at improving ceramide level in epidemis of NC/Nga mice.

Expression Analysis of Diphtheria Toxin-A Gene Regulated by Lck Promoter in Transgenic Mice (형질전환생쥐에서 Lck Promoter에 의한 Diphtheria Toxin-A Gene의 발현 분석)

  • Transgenic mice containing Diphtheria Toxin-A (DT-A) gene fused to proximal lck promoter sequences was used for analysis of DT-A gene expression and thymocyte development. The diphtheria toxin gene was expressed in thymus, spleen and liver of transgenic mice confirmed by RT-PCR and Northern blotting. A FACS analysis with thymocyte cell surface antigens antibodies (CD4 and CD8) showed that the number of peripheral mature single positive thymocytes ($CD4^{+}\;and\;CD8^{+}$ cells) T-cells was severely reduced in transgenic mice compared to that in the non-transgenic littermates. A relative portion of $CD8^{+}$ single positive thymocytes was about 33.2% in transgenic peripheral T-cells while 50.6% in wild type. Reduction of $CD4^{+}$ cell numbers in transgenic mice was observed (5.9% in transgenic versus 10.3% in non-transgenic). The data from analysis of these transgenic mice indicate that the proximal lck promoter regulated the expression of DT-A gene at high level in developing thymocytes and the DT-A disrupted developing thymocytes in transgenic mice.

EXPRESSION OF DSPP AND BSP MRNAS DURING ODONTOBLAST DIFFERENTIATION IN THE NFI-C (-/-) MICE (NFI-C 결손 생쥐의 상아모세포 분화과정에서 DSPP와 BSP mRNA의 발현)

  • Nuclear factor I (NFI) exists in the odontoblast and osteoblast. NFI-C null mice demonstrated aberrant odontoblast differentiation, abnormal dentin formation, and molar lacking roots. The purpose of this study was to examine phenotype of the aberrant odontoblast in NFI-C null mice and to evaluate the expression of DSPP and BSP mRNAs in NFI-C null mice with in-situ hybridization. The results were as follows: 1. In the NFI-C (-/-) mice, the crown dentin of molar showed normally formation, but there was no root dentin. 2. In the NFI-C (-/-) mice, the labial dentin of mandibular incisors showed relatively a lot of dentin formation, but the lingual dentin showed defect. 3. In the NFI-C (-/-) mice, the odontoblast of mandibular incisors revealed abnormal shape and trapped in osteodentin-like mineralized tissue. 4. In the NFI-C (-/-) mice, the odontoblast in the crown dentin of molars showed strong expression of DSPP, the odontoblast in the root dentin of molars was not expression of DSPP. In the NFI-C (-/-) mice the odontoblast in the mandibular incisors showed weekly expression of DSPP 5. In the wild mice, the odontoblasts of mandibular incisors were not expression of BSP, but in the NFI-C (-/ -) mice the odontoblast of mandibular incisors showed strong expression of BSP These results suggest that odontoblast in the NFI-C (-/-) mice changes the phenotype into osteoblast.

Friend Virus 를 이용한 Reverse Transcriptase 억제제의 생체제의 약효검색

  • HIV와 유사하게 retro virus의 일종인 Friend virus of Anemia strain (FVA)을 BALB/c mice 에게 감염시 reverse transcriptase 의 활성에 의해 비장의 erythroid progenitor cell 에서 증식되므로서 비장의 비대 및 빈혈을 초래하게 된다. 이를 지표로 하여 reverse transcriptase 억제작용을 지닌 항 virus 약물을 천연성분으로 부터 검색하고자 하였다. 우선, 대조약물로 사용한 기존의 항 AIDS 약물인 zidovudine (AZT) 을 FVA 가 감염된 BAB/c mice 에게 18일간 투여시 (약 100 mg/kg/day, p.o. ) 대조군에 비해 비장의 비대 및 reverse transcriptase 활성이 90% 이상 억제되었으며, 이들의 혈청을 정상 BALB/c mice 에게 재투여시에도 유사한 결과를 나타내어 reverse transcriptase 를 억제하므로서 항 virus 작용을 나타낸다는 것을 입증하였다. 그러나, 혈액중 Hemoglobin등 빈혈의 지수는 대조군과 유사한 수치를 나타내므로서 정상으로 회복되지를 못하였다. 이것은 zidovudine 자체가 지니는 골수억제에 의한 부작용 때문인 것으로 고려된다.

Effects of Walgookwhanhab-Bojoongikgitang administration on immune-function in Balb/c mice stressed by electric footshock (월국환합보중익기탕(越鞠丸合補中益氣湯)이 전기자극(電氣刺戟)스트레스를 받은 mouse의 면역기능(免疫機能)에 미치는 영향(影響))

  • The present experiments were designed to study the influence of WalgookwhanhabBojoongikgitang on immune functions of Balb/c mice under stress condition. WalgookwhanhabBojoongikgitang was orally administered to the mice for 15 days. On the 10th day the mice were immunized with sheep red blood cells (SRBC) and then subjected to electric footshock for 5 days (2 sessions a day, 11 footshocks a 31 min-session). The immune responses to SRBC were determined by means of hemagglutination and BIT cell populations in the spleen were studied by FACS analysis on the 16th day. The results were as follows. 1. After electric footshock, IDlce became sluggish and crowded to one side of the cage. Increased antibody titer for SRBC, increased B cell population, and decreased T cell population in the spleen were also observed. These results confirm that electric footshock caused stress inducing immunological and behavioral changes in Balblc mice. 2. Walgookwhanhab-Bojoongikgitang administration significantly antagonized the effect of electric footshock on the antibody titer for SRBC. As a result, antibody titers for SRBC in the mice treated with Walgookwhanga-Bojoongikgitang were maintained at the similar levels as those of control group mice even after the electric foots hock. 3. Walgookwhanhab-Bojoongikgitang administration also antagonized the effect of electric footshock on the BIT populations in spleen. As a result, Band T populations in the mice treated with Walgookwhanhab-Bojoongikgitang were maintained at the similar levels as in the control group mice even after the electric footshock. Taken together, Walgookwhanhab-Bojoongikgitang seem to help Balb/c mIce to maintain their humoral immune response and immune cell populations at a normal range under the stress conditions, suggesting its possible therapeutic use as a immune function modulator.

  • Effects of royal jelly(RJ) on the immune system in normal and cyclophosphamide(CY)-treated mice were investigated. The results were as following: 1. Body weight, spleen weight, thymus weight, WBC, cell-mediated immunity (CMI, contact hypersensitivity to DNFB), humoral immunity (HI, Hemagglutinin-, Hemolysin-titer) were increased or decreased dependent on the day of administration of RJ in normal mice. But it showed no effect on liver weight and RBC. 2. Combined treatment with RJ in CY-treated mice on the day which RJ showed the increasing activities in normal mice inhibited the decrease of survival rate, body weight, spleen weight, WBC and CMI caused by CY, but no effect on the decrease of thymus weight and HI induced by CY.

Anti-obesity Effects of Capsule-filled Cheonggukjang Added with Arrowroot (Pueraria thunbergiana) Extracts in ob/ob Mice (칡(Pueraria thunbergiana)추출물 청국장 캡슐의 ob/ob Mice에 대한 항비만 효과)

  • This study was designed to evaluate the anti-metabolic syndrome effects of capsule-filled cheonggukjang (CGJ) added with arrowroot (Pueraria thunbergiana) extracts on body weight, adiposity and lipid metabolism in ob/ob mice. Experimental groups were normal control group (NC: basal diet), positive control group (PC: 2% CGJ), CGJ added with arrowroot extracts group (AR: 2% arrowroot in CGJ), and capsule-filled CGJ added with arrowroot extracts group (ARC: 2% arrowroot CGJ capsule). Each group was fed experimental diet for 10 weeks. Final body weight gain and atherogenic index were significantly lower in the ARC than NC group. Serum levels of total cholesterol, LDL-cholesterol, and triglycerides, blood glucose and atherogenic index were significantly lower in the ARC than NC group. Furthermore, fatty liver and regional lipid accumultion in ob/ob mice were inhibited in the ARC group. The hepatic activities of superoxide dismutase, catalase and glutathione S-transferase were significantly higher in the ARC than NC group. Therefore, the anti-matabolic syndrome effects of the ARC group were higher than the AR group. In conclusion, these results indicated that CGJ added with arrowroot mediates its anti-obesity effects in ob/ob mice by improving lipid metabolism and antioxidant enzyme.

Effects of Mucilage from Yam (Dioscorea batatas DECENE) on Blood Glucose and Lipid Composition in Alloxan-induced Diabetic Mice (마(Dioscorea batatas DECENE) 점질물이 Alloxan 유발 당뇨 마우스의 혈당 및 지질 성분에 미치는 영향)

  • Effects of Mucilage fraction from Yam (Dioscorea batatas DECENE), which has long been used as a wild vegetable and folk medicine, on blood glucose and lipid composition in alloxan-induced diabetic mice were investigated. Diabetes mellitus was induced in male ICR mice by the injection of alloxan into the tail vein at a dose of 75 mg/kgBW. Alloxan-induced diabetic mice were administered the yam mucilage fraction1 orally ; y500F1 group (500 mg/kgBW/day fraction1) and y750F1 group (750 mg/kgBW/day fraction1); and the normal and alloxan-control group were orally administered with saline for 10 days. The body weight gain and food intake were monitored every day. The concentrations of glucose, triglyceride, total cholesterol, and LDL-cholesterol of serum and liver levels of glucose, triglyceride, and cholesterol were determined. Also weight of liver, heart, spleen and kidney were measured. The fraction 1 of yam mucilage lowered body weight gain significantly (p<0.05) and decreased serum glucose levels in alloxan-induced diatetic mice compared to that of alloxan-control group. In alloxan induced diabetic mice serum triglyceride level was lowered and liver HDL-cholesterol level increased significantly (p<0.05). In conclusion, it was assumed that yam mucilage fraction 1 has anti-hyperglycemic and anti-obesitic effects by reducing body weight gain and decreasing serum glucose and triglyceride level.

Resistance to Naegleria fowleri infection passively acquired from immunized splenocyte, serum or milk (면역시킨 마우스의 비장세포, 혈청 또는 모유를 통해 얻을 수 있는 Naegleria fowleri 감염에 대한 방어 능력)

  • A pathogenic free-living amoeba, Naegleria fowleri, causes primary amoebic meningoencephalitis to human and experimental animals. This infection is rare, but the mortality is very high. Nowadays, drug treatment or active immunization of human or mice are being tried with partial effectiveness. This study shows passive immunization effect by transfer of immunized spleen cells, serum, or milk from immunized mother in mouse experimental model. Young BALB/c mice were immunized intraperitoneally with $2~3{\times}10^{6}$ trophozoites of N. fowleri, and spleen cells and sera were collected for injection to recipient mice. There were seven transfer groups, i.e., immunized mouse serum, spleen cells, serum and spleen cells, normal mouse serum, spleen cells, serum and spleen cells, and control group. Three days later, BALB/c mice were inoculated with $1{\times}10^{4}$ trophozoites of N. fowleri intranasally. After infection, decreased mortality ana prolonged survival time of mice were noted in immunized Bloops compared with non.immuniBed control group. The groups Injected with immunized spleen cells or normal serum shewed lower moltality than that of controls bult showed no changes of Serum IgG level. The groups injected with immunized serum or normal spleen cells showed increased serum IgG level after immunization but hundred percent mortality was observed. Mother mice were ifnfnunised increperitqneeliy with $2~3{\times}10^{6}$ trephozoites of N. fowleri at the end of pregnancy and weaning Period. Soon after the delivery, Jitters born of non-immunszed mother were matched with immunized mother for feeding immune milk. After three weeks, the litters were infected with $1{\times}10^{4}$ trophozeites of N. fowleri or sacrificed for serum collection to measure the IgG levels. The results show that anti-JV. fowleri IgG from mother was transferred to litter through milk but this IgG did not inauence the mortality or survival time of the infected mice.
