Simplified Procedure of Amylose Analysis by Rapid Flow Autoanalyzer RFA-300 (자동분석기 RFA-300 이용한 아밀로스 분석법)

  • Several trials and errors were repeated to develop a simplified recipe of amylose analysis using a Rapid Flow Autoanalyzer(Alpkem, RFA-300). The amylose content of rice samples analyzed by the Rapid Flow Autoanalyzer were compared with those of Williams’ and Juliano’s assay. The results by the simplified recipe of RFA amylose analysis were highly correlated with those by Williams’ and Juliano’s method(r=0.95$^{**}$-0.97$^{**}$). The relative amylose content of defatted rice starch was higher than those of non-defatted rice flour, showing very close correlations between those analyzed by three method.

  • Performance of proficiency testing and the validation of analytical method was included a scheme of quality assurance in analytical chemistry laboratory to monitor a laboratory's performance abilities and produce consistently reliable data. This study was assessed the applicability of proficiency testing scheme proposed for analytical laboratories of pesticide product in domestic. The validation of analytical methods, stability and homogeneity for formulated pesticide products (emulsifiable concentrate) of emamectin benzoate and lufenuron was confirmed for the proficiency testing. The z-score of 33 participation laboratories for emamectin benzoate were that the numbers of outlier were 2 laboratories (6.0%), z-score outside the range from -3 to 3 designated "unaccptable" were 2 laboratories and z-score in the ranges -2 to -3 and 2 to 3 designated "questionable" were 3 laboratories (9.0%). Three laboratories (9.0%) showed the z-score designated "questionable" for lufenuron. The additional proficiency testing for various product types will be needed to establish the scheme of quality control.

Improved Method for Increasing of the Oil Yields in Grape Seed (포도씨 기름의 수율증진을 위한 추출 방법 개선)

  • To establish the optimal conditions for increasing oil yields from grape seed, extraction solvent, extraction time, and temperature were examined. Using grinding, grinding and roasting, grinding and steaming, and grinding, roasting, and steaming methods. Pressing extraction method resulted in 34.0% oil yield. Grinding and roasting, grinding and steaming, and grinding, roasting, and steaming gave 64.3, 63.0, and 65.6% yield, respectively. Ether solvent treatment resulted in 77.4 and 80.9% recoveries after 24 and 48 h static, respectively. The results of sensory evaluation revealed, oils extracted under optimal condition showed the best flavor, aroma, and whor, followed by grinding, grinding and roasting, grinding, roasting, and steaming, and grinding and steaming. Taking these results together, the optimal methods for oils extraction from grape seed were as follows: grape seed ${\rightarrow}$ washing ${\rightarrow}$ drying ${\rightarrow}$ roasting ($95{\sim}100^{\circ}C$ for 20 min) ${\rightarrow}$ cooling (room temperature) grinding ${\rightarrow}$ (0.5 mm>) ${\rightarrow}$ steaming ($0.8{\sim}0.9\;kg{\cdot}f/cm^2$ for 10 min) ${\rightarrow}$ pressing ($1st\;400\;kg/cm^2\;for\;2{\sim}3\;min,\;2^{nd}\;550{\sim}600\;/cm^2$ for 10 min, $3^{rd}\;700kg/cm^2$ for 60 min) ${\rightarrow}$ oils (yield $;85{\sim}90%$).

  • 용수 다소비 산업으로 분류되는 제지공업의 특성 상, 용수사용의 억제 및 용수의 무방류화에 대한 압력이 증가되고 있는 반면, 초지환경은 날로 열악해지고 있는 실정이다. 한 예로써, 용지의 재활용율 및 재사용 비율이 증가함에 따라 원료의 저급화가 심 각한 수준에 이르고 있으며, 이러한 초지환경의 변화는 각질화되고 극도로 미세화된 다량의 미세섬유 발생 및 잡고지로부터 유입되는 점착성 이물질과 무기 충진제의 도입 등을 유발함으로서 제품의 물리적 성질 저하, 탈수부하 증가로 인한 생산성 저하, 약품사용량 증가 등을 초래하여, 결과적으로는 폐수처리장의 부하를 증대시킴과 동시에 청수사용량 및 폐수방류량을 증가시키는 악순환을 되풀이하게 된다. 국내 제지산업이 국제적 경쟁력을 갖추기 위해서는 제품의 증대가 필수적이나, 원료와 설비의존도를 감안할 때, 제품특성과 공정 분을 결정하는 습부공정을 정확히 파악하여 최적 운전조건을 확립하는 것이 가장 경제적이고 용이한 방법일 뿐만 아니라 전체 제지공정의 정정화에도 기여할 수 있을 것으로 판단된다. 일정규모이상의 종이를 생산하는 제지공장은 거의 대부분 컴퓨터로 처리되는 온라인 공정조절 시스템(On-line process control system)을 구비하고 료의 도입에서부터 설비운전 및 제품의 기본적인 품질관리까지 실시간의 자동화되고 안정적인 운전과 관리시스템을 유지하고 있다. 그러나 전체 제지공정 중, 제품 물성과 운전조건의 대부분을 결정하는 습부공정만큼은 아직도 주기적인 분석이 행해지고 있지 않을 뿐만 아니라, 슬러리의 농도나 보류도 정도를 제이하면 분석항목 조차도 변변히 확립되어 있지 않을 실정이다. 이는 다량의 물 속에 존재하게 되는 용전물질(Dissolved solids, DS), 부유물질(Suspended solids, SS), 섬 유(Fibers), 무기 미 세분(Inorganic f fines), 그리고 투입되는 약품간에 발생하는 계면동전현상 및 이러한 현상과 최종 지제 품의 물성간의 상관관계에 대한 이해 정도나 경험 부족에서 기인하는 것으로 생각된다. 1 1983년 미국제지기술연합회(T APpI)의 제지용 첨가제 분과위원회(Papermaking A Additive Committee)에서는 습부공정의 적절한 조절을 위해 어떤 항목들이 필요하며 그 중요도는 어떠한가를 조사하였으며, 그 결과로 전하밀도, 제타전위, 보류도 및 여수 도, 무기 미세분 함량 등이 중요하다고 보고하였다. 그러나 지료의 전기적 특성을 실시 간으로 측정하는 기자재류가 최근에 이르러서야 도입되고 있는 실정이기 때문에 충분 한 현장적용 사례가 보고되지 못하고 있으며, 결과적으로 얻어진 정보와 최종 지제품의 물성 및 초지기 운전조건과의 상관관계를 확립하는 작업결과는 더욱 찾아보기 어렵다. 따라서, 본 연구에서는, 라이너지 제조공장의 습부공정에 투입되는 약품이 지료 의 전기적 성질, 즉 전하밀도와 제타전위에 미치는 영향을 모니터링함으로서 지료의 전기적 성질들과 습부 공정상태와의 상관관계를 살펴보고자 하였다. 본 연구는 다음과 같은 순서로 진행하였다.

  • Lodging of rice plant is the most important damage by unfavourable weather conditions in rice cultivation. High levels of nitrogen application and growing of Japonica rice variety is one factor to increase the lodging damage. Lodging of rice plant decreasing grain yield; 34% at milky. 21% at dough. 20% at yellow stage. decreasing grain Quality. increasing green rice. and increasing labor cost to harvest. To decrease lodging damage. the lodging resistant varieties will be selected and cultural practices such as amount and time of nitrogen application. planting density. water management. and disease and pest control methods have to be study for the short and strong culm. and good rooting system. Also. application methods such amount and time of plant growth regulators and new types of chemicals will be develop for the reducing lodging of rice plant. To decrease the lodging damage in direct seeding cultivation. first identifying the differences of lodging mechanisms between hand transplanting and direct seeding, second establish the suitable direct seeding methods such as seedling establishement. fertilization. and water menagement.

The Experiment for the Technical Balance Work on the Automatic Silk Reeling Machine ( I ) (자동조사기 기술체계확립을 위한 시험 (I))

  • This experiment was carried out to investigate the cocoon reeling condiditions required to technical balance work on the automatic silk reeling machine. The results obtained were as follows. 1) Reasonable reeling velocity with regard to raw silk yield and to reeling efficiency varied according to cocoon reelability. It was observed to be about 150m/min on good reelability cocoon (850m of nonbreaking bave length), about 120m/min on medium reelability cocoon (650m of nonbreaking bave length) and from 90m/min to 120m/min on bad reelability cocoon (500m of nonbreaking bave length) 2) Low temperature drying of cocoon (T$_3$) improved the reelability of cocoon, but increased the reeling accidance with deteriorating the quality of raw silk more than the control(T$_1$) or high temperature drying of cocoon (T$_2$). No significant difference was observed in its raw silk yield and silk by-product with regard to cocoon drying temperature. 3) Incomplete drying of cocoon (T$_3$) improved the reelability of cocoon, but increased the reeling accidance with deteriorating the quality of raw silk more than the control(T$_1$) or over drying of cocoon(T$_2$). No significant difference was observed in its raw silk yield and silk by-product with regard to cocoon drying degree. It was cocoon drying condition of cocoon was the control(T$_1$) or high temperature(T$_2$) rather than low temperature (T$_3$) in cocoon drying process. Control (T$_1$) or over drying of cocoon (T$_2$) was adequate rather than incomplete drying of cocoon (T$_3$) for the improvement of the quality of raw silk on the automatic silk reeling process. 4) It was found that the reasonable cooking condition of cocoon was optimum cooking with 4$0^{\circ}C$ of reeling temperature or incomplete cooking with 45$^{\circ}C$ of the reeling temperature in the automatic silk reeling of the domestic cocoon.

Changes of Crude Components and Essential Oil Content by Shading Treatment in Codonopsis lanceolata Trautv (차광처리에 따른 더덕의 조성분과 정유성분 변화)

  • This experiment was conducted to increase essential oil and aromatic contents on roots of Codonopsis lanceolata Trautv with different shading treatments. Vine length was prolonged by shading treatment compared with non-shading. Fresh root weight in 75% shading treatment was highest with 13g per plant. Crude components such as crude protein, fat and fiber contents decreased, while crude ash increased as shading degree increased. Forty-eight volatile aromatic compounds in the root were identified by GC/MS. Major aromatic compounds were 1-hexanol, cis-3-hexanol, and trans-2-hexanol. In particular, trans-2-hexanol was highest in the 75% shading treatment attaining the 160.32 % area. Conclusively, it was suggested that the 75% shading treatment was to some extent effective to improve the content of essential oil and aromatics in the roots of Codonopsis lanceolata Trautv.

The Study of Milling Properties for Optimization of Treatment and Recycling of Converter Slag (제강슬래그 처리 및 재활용의 최적화를 위한 분쇄 특성에 관한 연구)

  • To treat and recycle a large quantity of converter slag. the milling properties of -14/ +24 mesh-sized slag has been considered. The optimal conditions in milling process were investigated for producing powder-type slag and the required consumption was derived for the economical grinding. The characteristics of milling processes were studied in the variation of the rotational speed, milling time, filling ratio of ball, and size and amount of feed. The grinding efficiency was also examined. The optimal rotational speed in this experimental condition was observed to be the value of 79% of critical speed. The extent of grinding was increased with increasing the grinding time. but the efficiency of milling was decreased with the time. 50% ball filling was shown to have the optimal grinding effect, and less amount and small-sized feed made the milling efficiency high. As the result, using Bond's equation, power required for efficient milling was considered and the highest value was observed in the condition of high grinding time and optimal rotational speed.

  • There has been a great deal of controversy in the views on the impact of the pesticide pollution from the golf courses. The stream and reservoir associated with a existing golf course were investigated. The parameters include the ones related with eutrophication, heavy metals, pesticides. and aquatic ecological conditions. It was found that the impact on the aquatic ecosystem by the pollution from the golf course was not so serious. An aquatic ecological food chain was well established in the reservoir which belongs to the golf course. In contradiction to the general recognition, the livestock farming and agricultural farming gave much more adverse impacts on the stream than the golf course.

Analysis of Anti-Allergic Activities by Chaff Vinegar Liquor (왕겨초액의 추출물에 의한 아토피 완화 효과 비교)

  • To evaluate whether chaff vinegar liquor (CVL) has potential in ameliorating allergic symptoms in mice, we tested the anti-allergic activities with a DNFB (dinitrofluorobenzene)-induced allergic mouse model. The DNFB-induced allergic symptoms were reduced by 20% compared to the control by chaff vinegar liquor supplemented with Perilla frutescens var. acuta. Allergic symptoms were examined by measuring the width of epidermis swelling. The hematoxyline & eosin (H&E) staining also revealed that the chaff vinegar liquor dramatically decreased the allergic symptoms in the epidermis of the ear. These results collectively suggest that the chaff vinegar liquor supplemented with P. frutescens var. acuta has potential in ameliorating allergic symptoms, showing that the liquor could be a useful biomaterial(s) for the nutraceutical or cosmetic industries.
