질병 - 분무접종법에 대한 올바른 이해

  • 양계인이라면 누구나 생독백신 접종 후에 나타나는 백신접종반응(기침 등 호흡기 증상)은 적으면서 백신접종효능(항제 형성능 및 질병방어효과)은 아주 우수한 그런 종류의 생독 백신이나 접종법을 찾게 마련이다. 국내의 경우 1일령 병아리에 대한 닭 뉴캣슬병 생독백신 접종이 의무화되어 있어 부화장에서 병아리 분양 시 분무백신기를 이용한 닭 뉴캣슬병 생독백신을 접종하게 되고, 또한 농장에서 닭 뉴캣슬병 추가백신 접종이나 닭 전염성 기관지염 백신 접종 시에도 분무접종을 실시하기도 하는 등 분무접종에 대한 경험은 누구나 가지고 있을 것이다. 그러나 음수접종법과 달리 분무접종 시 심한 백신접종반응을 경험하게 되는 양계인들도 쉽게 만나볼 수 있다. 생독백신은 음수접종법을 비롯하여 점안접종법 및 분무접종법 등 다양한 접종방법이 현재 야외농장에서 응용되고 있으며, 국내 육계사육 시에는 음수접종법과 분무접종법이 주로 사용되고 있다. 하지만 동일한 종류의 생독백신이라 할지라도 백선접종방법에 따라 백신접종 후 백신접종효능뿐만 아나라 백신접종반응에도 많은 차이를 보이게 되며 동일한 분무접종법이라 하더라도 어떠한 입자 크기의 분무기를 사용했는가에 따라서도 이러한 차이는 있다. 분무접종은 백신 바이러스가 비강,눈(harderian gland) 뿐만 아니라 상부 호흡기도에 직접적으로 작용을 하여 강력함 국소면역능을 유발하는 장점을 가지고 있으나 접종일령이나 접종 백신의 종류를 고려한 분무입자 크기의 선택이 제대로 이루어지지 않아 음수접종법에 비해 접종 부작용이 크게 나타나는 백신접종법으로 인식되어 있는 실정이다. 따라서 이번 호에서는 호흡기 질병의 예방 백신 분무접종에 대해 알아보고자 한다.

Simple Mass-screening Methods for Resistance of Tomato to Fusarium oxysporum f. sp. lycopersici (토마토 시들음병에 대한 간편 대량 저항성 검정법)

  • This study was carried out to establish the simple mass-screening methods for resistant tomato to Fusarium wilt caused by Fusarium oxysporum f. sp. lycopersici (FOL). Root dip inoculation method has been used in many studies on the resistance of tomato to disease. On the other hand, in mass-screening for resistant tomato to Fusarium wilt, the inoculation method is time-consuming and laborious procedure. Disease development of two FOL isolates on two cultivars of tomato according to inoculation method including root dip, tip and scalpel methods were investigated. In compatible interaction, tomato seedlings of each cultivar inoculated by tip method showed the lower and more variable disease severities than by root dip method. Whereas the seedlings by scalpel method represented clear resistant and susceptible responses to Fusarium wilt as root dip method. The resistance degree of each cultivar inoculated with FOL isolates by scalpel method was hardly affected by the tested incubation temperature and inoculum concentration. On the basis of the results, we suggest scalpel inoculation method as an efficient mass-screening method for resistant of tomato cultivars to Fusarium wilt. Roots of tomato seedlings at two-leaf stage grown in plastic cell tray were injured with scalpel and then spore suspension (more than $1{\times}10^7\;conidia{\cdot}mL^{-1}$) of FOL was poured directly on the roots. The infected plants were cultivated in a growth room at $25-30^{\circ}C$ for 4 weeks with 12-hours light a day.

Comparison of Efficacy of Newcastle Disease Virus $B_1$ Vaccine by Different Administration .Methods in Commercial Layer Chickens (백신접종 방법에 따른 뉴캣슬병 $B_1$생독백신의 효능비교)

  • The immune responses of commercial layer chickens against Newcastle disease (ND) were compared among different administration methods and times of vaccination during 4 weeks of age. A total of 372 day-old chickens were devided into 4 groups of 93 birds each. Each of 3 groups was received a commercially available B$_1$ live vaccine via drinking water, eye instillation or spray method at one, 14 and 28 days of age. One group was used as an unvaccinated control. At two and 4 weeks after each time of vaccination, 15 birds from each group were challenged with virulent ND virus at the dose of 10$^{5}$ EID$_{50}$ per bird to examine the pretection rate. Ten to 15 birds from each group were bled at two weeks intervals from day old to 8 weeks of age to determine hemagglutination inhibition antibody titer. The protection rate was generally low regardless of the times of vaccination although two or more times vaccination gave higher protection than once vaccination. The low protection was considered due to low titer of the vaccine used since the vaccine titer was less than 10$^{2.5}$ EID$_{50}$ per bird. Spray method gave better protection compared to eye instillation of drinking water method which resulted in lowest response. When birds were challenged majority showed clinical signs on ND between 3 and 6 days after challenge. Death occured one or two days after onset of symptoms. Major clinical signs observed were depression (96%), drowsy(90%), anorexia (84%), diarrhoea (29%), difficult breath (15%) and torticollis (10%). Hemorrhagic lesions on post mortem were seen in duodenum (51%), trachea(36%), illeum (13%), ceacal tonsil (11%), proventriculus (10%) and some other organs. Effects of Seed Inoculation Methods on the Nodulation and the Growth of Alfalfa Seeding (근류균의 종자 접종방안의 차이가 근류형성 및 Alfalfa 유묘의 생장에 미치는 영향)

  • Alfalfa (Medicago sativa L. cv. Luna) seeded in agar was inoculated with two strains of Rhizobium meliloti isolated from root nodules of alfalfa for assessment of nodulation. The seedling growth after six weeks was remarkably increased by adding each rhizobia strains into agar media and also by nitrate application (70ug N/ml), but there was no significant difference among them. Nodulations started one week after inoculation and increased its numbers and sizes as seedling grew. Therefore, the two strains isolated from alfalfa root were concluded to be effective strains. For determining seed inoculation method the same cultivar was inoculated with both rhizobia strains using different inoculation methods such as broth-vacuum, peat-adhesive, peat & lime pelleting. They were seeded in pots of river sand and supplied with culture solution excluded nitrogen. The peat & lime pelleting was recognized the best method in both of nodulation and seedling growth after eight weeks growth. There were significant correlations between the weight of nodules and the shoot or root dry weight of alfalfa in both rhizobia strains.

  • Effective strains of cowpea bradyrhizobia JB7 $nal^rspe^r$ and CB756 $str^rrif^r$, antibiotic-resistant variants of JB7 and CB756, respectively, were used to examine changes of rhizosphere populations and nodule occupancy. Populations of each strain increased gradually in the rhizosphere, reaching a maximum of about $10^8$ cells per root system. Nodule number increased as the density of inoculum increased from $10^2$ cells to $10^8$ cells per seed. Inoculation with liquid suspension resulted in the formation of more nodules than the peat slurry or granule inoculation. When JB7 $nal^rspe^r$ and CB756 $str^rrif^r$ were introduced in equal numbers in inoculum mixtures the former consistantly occupied the majority of nodules with all three groundnut cultivars used. There was no difference in yield between nitrogen treatments and inocultation treatments.

  • The prevalence of symptomatic Hepatitis A has inclined significantly during the last several years in Korea. The average age at infection has increased, resulting in more clinically severe infections. Efforts to promote good personal hygiene have not been successful in interrupting outbreaks of hepatitis A. Vaccination is the most effective means of preventing HAV transmission among persons at risk for infection. Hepatitis A vaccination is recommended for all children at age 1 year, for persons who are at increased risk for infection, for persons who are at increased risk for complications from hepatitis A, and for any person wishing to obtain immunity.

Development of a Simple and Effective Bioassay Method to Evaluate Resistance of Watermelon Plants to Fusarium oxysporum f. sp. niveum (수박 덩굴쪼김병에 대한 간편 저항성 검정법 확립)

  • Root-dipping inoculation method has been widely used to determine the resistance of watermelon to Fusarium oxysporum f. sp. niveum causing Fusarium wilt. Although this method leads to the precise results of plant disease responses, more rapid and efficient assay methods have been still required because the root-dipping inoculation method is labor-intensive and time-consuming. In this study, we established a simple and effective bioassay method based on the comparison of various inoculation methods and growth conditions. To develop the system, the occurrence of Fusarium wilt on four resistant and susceptible cultivars was investigated by four different inoculation methods, root-dipping, scalpel, tip and soil-drenching methods. Of these inoculation methods, scalpel method resulted in clear plant disease resistance responses with the simplicity. With the use of scalpel method, we also explored the disease development of the cultivars depending on inoculum concentration, growth stage of seedlings, and incubation temperature after inoculation. Furthermore, we found that the resistance degrees of 23 cultivars derived by scalpel inoculation method were similar to the results by root-dipping method established previously.
