Development of Yeast Strains as Feed for Aquaculture: Possible Yeast Strains (양식을 위한 먹이사료로서의 Yeast 균주의 개발: 가능성 있는 효모 균주)

  • Possible yeast strains that could be used as feed for aquaculture were studied. It was shown that the maximum specific growth rate and the biomass yield of Kluyveromyces fragilis yeast and Candida utilis yeast under optimum pH and temperature were much higher than those of Saccharomyces cervisiae yeast which had been as established yeast diet for rotifer culture. Hence, this work was focussed on the growth characteristics of the two yeasts through flask dultures for mass production. With 5% inoculum dosage, the best values of $\mu$$_{max}$ and OD$_{max}$ were obtained with on 2.5% fructose medium and 2% YE medium for K. fragilis and C. utilis, respectively, where the values of $\mu$$_{max}$ and OD$_{max}$ were found to be 0.73 hr$^{-1}$ and 3.00 for K. fragilis and 0.59 hr$^{-1}$ and 2.80 for C. utilis. It was also found that the lag phase of the growth incresed with increasing initial zinc and NaCl concentrations and decreased with increasing inoculum dosage. Both yeasts could survive relatively well at 3.5% NaCl concentration, and only C. utilis yeast could utilize zinc.

  • In this study, the optimization of several factors for Chlorella saccharophila cultivation was investigated. The studied factors were medium type, culture type, inoculum size, sugar/nitrogen source type and concentrations. As a result, the optimized conditions for C. saccharophila cultivation were found to be the best at 3% (v/v) inoculum, 30 g/L glucose and 0.95 g/L $NaNO_3$ under mixotrophic culture. Under the optimized condition, the content of oil was high at 12 day, whereas, the amount of biomass and chlorophyll were high at 10 day.

  • Naegleria fowleri is the cause of primary amoebic meningoencephalitis in man, IL-2 levels after stimulation of T lymphocytes by PHA or N.fowleri lysates. the amounts of T lymphocyte subsets and the blastogenic responses of T lymphocytes in mice after Infected with pathogenic N. fowleri were studied comparing between two study groups, one $1{\;}{\times}{\;}10^4$ trophozoites inoculated mice and the other $1{\;}{\times}{\;}10^5$ trophozoites inoculated mice. All experimental samples were obtained on the day 7, 14 and 24 after inoculation. The mice inoculated with $1{\;}{\times}{\;}10^4$ trophozoites showed a 14.3% mortality rate, and 72.2% in the mice inoculated with $1{\;}{\times}{\;}10^5$ trophozoites. The IL-2 levels on day 14 of two experimental groups were significantly decreased as compared with the control group. Thy 1.2+T cells in the total spleen Iymphocytes of $1{\;}{\times}{\;}10^5$ trophozoites inoculated group on day 7 were significantly increased compared with the control group. There was no significant difference between $1{\;}{\times}{\;}10^4$ trophozoites inoculated group and the control group. $L3T4^{+}{\;}T$ cells and $Ly2^{+}$ T cells in the total spleen Iymphocytes of $1{\;}{\times}{\;}10^5$ trophozoites inoculated group on day 7 were sigrlificantly increased compared with the control group. The DNA S fraction of T cells in the spleen of $1{\;}{\times}{\;}10^5$ trophozoites inoculated group was significantly increased on day 7. The amount of S fractions of DNA were sequentially decreased on day 14 and 24 but they were also signiacantly increased compared with the control group. The results obtained in the experiments indicats that cell mediated immunity after N.fowleri infection acts on very important host's protection immunity around the 7th day after infection. IL-2 level was much suppressed on day 14 which resulted from the exhaustion of host immune response. It was observed that the level of IL-2 production ability and the amounts of T lymphocytes subsets and the blastogenic responses of T lymphocytes were not well correlated during the observation period.

Effect of Medium Components and Culture Methods on Prothallus Propagation of Pteridium aquilinum var. latiusculum (Desv.) Underw. ex Hell. (고사리 전엽체의 증식에 미치는 배지구성물질과 배양방법의 영향)

  • Present studies were conducted to evaluate the effects of medium strength(MS and Hyponex), carbon sources and their concentrations, agar concentrations, and inoculation amounts on prothallus propagation of Pterdium aquilinum var. latiusculum(Desv.) Underw. ex Hell in vitro. The optimum MS medium strength for prothallus propagation was 2MS concentration. Phosphate source was most effective for prothallus growth of P. aquilinum var. latisculum. The addition of 1% sucrose or glucose to MS medium promoted prothallus multiplication. Growth of prothallus was not affected by agar concentration. Propagation of homogenized prothallus was vigorous even in liquid medium. Chopped gametophytes(100 and 200 mg) were inoculated on 250 ml ${\Delta}$flask with 100 mL of 2MS concentration medium and suspension culture was done at 100 rpm for 22 days. After 20 days, prothallus multiplication slowed down, so 100 mg of chopped prothalli is recommended for initial inoculation, since initial amount of inoculum did not affect subsequent prothallus multiplication. Consequently after 20 days of suspension culture, prothallus should be subcultured or transplanted outside of growing vessels.

Production of Cellulase and Xylanase for Enzymatic Deinking of Old Newspaper (고지탈묵용 Cellulase 및 Xylanase 생산)

  • The optimal conditions for cellulase and xylanase production by Trichoderma reesei 28217 were studied for enzymatic deinking of old newspaper. The amounts of cellulase and xylanase from the strain was varied by initial medium pH, Tween 80, inoculum size of spore suspension, and carbon and nitrogen sources. The optimal conditions for cellulase production were pH 5.0-6.5, 0.02% of Tween 80, 0.5-1.0% of inoculum size of spore suspension ($1{\times}10^{7}$/ml). cottonseed meal as nitrogen source, and corn flour as carbon source. On the other hand, the optimal conditions for xylanase production were pH 6.5, 0.01% of Tween 80, corn steep liquor as nitrogen source, and disintegrated old newspaper as carbon source. The inoculum size for xylanase production was the same as for cellulase production. The concomitant production of cellulase and xylanase in shake flask culture was efficiently induced in the medium containing 0.5% cottonseed meal as nitrogen source and 1.0% old newspaper and 2.0% corn flour as carbon sources. In this case the activities of cellulase and xylanase produced were 6.11-7.22 IU/mJ and 97.7 IU/ml. respectively. However, the cellulase production in $5{\ell}$ fermentor scale was slightly decreased compared with that in flask scale. Moreover, xylanase production was severely reduced in a fermentor scale. The study for the reason of decreased enzyme production in fermentor is further needed.

Growth Stimulation of Mulberry Trees in Unsterilized Soil under Field Conditions with VA Mycorrhizal Inoculation (VA내생균근이 뽕나무의 생장에 미치는 영향)

  • Mulberry Seedlings germinated and grown under green house conditions were inoculated with Glomus mosseae, Mosse and Trappe (a Kind of Vesicular arbuscular mycorrhizae) prior to outplanting into unsterilized soil. They were grown on phosphate deficient soil for 6 months after planting. Shoot length, stem diameter and leaf yield of the inoculated plants were found to be significantly greater than uninoculated ones. It was observed in foliar mineral content that the levels of N, P$_2$O$\sub$5/, CaO of the inoculated plants were higher but the level of MgO of the inoculated plants was lower than the uninoculated ones. In the mineral content of roots, it was observed that the level of P$_2$O$\sub$5/ was higher but the level of N was lower significantly in the inoculated plants than the uninoculated ones.

  • 여러가지 난분해성 물질에 대한 생분해능을 지닌 백색부후균에 의한 매립지 침출수의 탈색을 조사하였다. 국내에서 분리한 Coriolus versicolor KR-11W와 Irpex lacteus KR-39W가 이제까지 주로 연구되어 온 Phanerochaete chrysosporium보다 높은 탈색능을 나타내었는데 I. lacteus KR-39W는 산소공급시 10%의 침출수가 함유된 YMG 배지의 진탕배양에서 85%의 색도제거율을 나타내었으며 최소배지에서도 80%의 탈색율을 보였다. P. chrysosporium에 의한 리그닌 분해능 및 분해효소 생성 보고들과 달리 진탕배양이 정치배양보다 탈색능이 높았으며 산소공급은 색도제거에 증가효과가 있었다. 균체 접종량(10-30%)과 온도(25,37.deg. C)는 탈색에 큰 차이를 보이지 않았으나 탄소원과 질소원의 농도는 상당한 영향을 나타내었다. 리그닌 분해효소군의 여러 가지 inducer와 cofactor를 C. versicolor KR-11W 배양에 첨가한 결과 많은 경우 균접종 대조군보다 2배 이상의 탈색율과 lignin peroxidase 활성의 증가를 보였으며 FeS $O_{5}$ 첨가시에는 최대 2.9배의 증가를 나타내었다. 탈색에 관여하는 효소군은 접종물에 이미 어느 정도 존재할 수 있으며 배지 및 배양조건에 따라 그 생성이 변화할 수 있으므로 이런 조건들을 잘 맞출 경우 보다 높은 탈색능이 기대된다.

Effects of mixing and reciprocal inoculation with compatible and incompatible races of Pyricularia oryzae on the enlargement of disease lesions of rice blast (친화성 및 비친화성균계의 혼합 및 전후접종이 벼 도열병의 병반확대에 미치는 영향)

  • Effect on blast lesion development was investigated in rice cultivar Milyang 23 and Jinheung using two races of Pyricularia oryzae compatible or incompatible to each of two rice cultivars. Discase lesions were measured in rice when inoculated with inoculum mixed with compatible and incompatible races, inoculated with a compatible race following with an incompatible race and vice versa. Lesions were smaller when rice was incoulated with an incompatible race and then inoculated with a compatible race than those inoculated in the reverse way both at the inoculated site and at 3cm-upper part from inoculated spot. The results suggested that induced resistance might play a role in suppression of lesion development although such an effect appeared to be influenced by heavy application of nitrogen fertilizer.

Influence of Oenococcus oeni Inoculation Time on the Quality of MBA Wine (Oenococcus oeni의 접종 시기가 국내 MBA 포도의 와인 품질에 미치는 영향)

  • This study investigated the effect of co-inoculation with Oenococcus oeni on the final quality of domestic MBA(Muscat Baily A) wine. For experiment five types of wine samples were classified for the experiment and the results are as follows; co-inoculation with Oenococcus oeni concurrently (i.e., the S+O.Co sample) reduced wine production time and facilitated the production offor high quality wine. Compared to the other samples, S+O.Co wine had a higher number of viable Oenococcus oeni cells and a rapid reduction in malic acid content although its lactic acid content increased more rapidly. Concurrent inoculation with yeast and Oenococcus oeni allowed malolactic fermentation and alcohol fermentation to be completed at the same time within 14 to 16 days. Therefore, this study showed that co-inoculation of S+O.Co wine with Oenococcus oeni had an effect through quality improvement, reduced wine making time, and increased productivity.
