  • The histochemical characteristics of the cutaneous mucous glands and its fine structure of the Korean fire bellied toad, Bombina orientolis Boulenger, were studied with light and electron microscopes. The mucous glands of the toad are simple alveolar gland, and are composed of inner glandular epithelial cells and outermost myoepithelial cells. Histochemical studies suggest that main secretory materials released from the mucous glands might be acid mucopolysaccharides which is more abundant in the ventral skin than in the dorsal skin. The scanning electron microscopic observation showed that the morphology of the opening sites of the gland duct is similar to the morphology of stomata on the plant leaves. Glandular epithelial layers of the mucous glands are composed of two cell types, the mucous secreting cells and the mitochondria rich cells. The mucous secreting cells could be classified according to the morphology and inner textures of cytoplasm and those characteristics were seemed to be related to the degree of maturation of the cells. Furthermore the mucous within the mucous secreting cells appears to be synthesized in the rough endoplasmic reticulum.

  • Histological investigation of seasonal changes of the mucus cells of ricefield-dwelling mud loach Misgurnus mizolepis was carried out on the skin in three regions of the fish: dorsal, lateral, and occipital. Although there were no significant differences in general morphology, the mucus cells of the epidermis showed a severe change in size and number by skin region. The density of the mucus cells was generally the highest in winter and the lowest in summer in all three skin regions. In particular, during two distinct periods, March to April when water is supplied to the ricefields, and August to September when the ricefields become dry after harvest, showed that the change in density and distribution of the mucus cells in both lateral and occipital regions is greater than during any other period. Moreover, the mucus cells for the dorsal and occipital regions temporally increased in size in June during the period of the highest water temperature of the year. Therefore, we may consider these seasonal changes as part of a protective adaptation to overcome extreme environmental conditions and to increase uptake of oxygen by cutaneous respiration.

  • The authors observed the ultrastructure of the mucous glandular epithelial cells in the amphibian skin by mean of electron microscope. The specimens from the experimental animals were fixed in 2.5% glutaraldehyde-oaraformaldehyde fixative in phosphate buffer at pH 7.2 prior to fixation in 1% osmium tetroxide, dehydrated in graded ethanol and acetone, embedded in Epon 812 mixture, and sectioned with Sorvall MT-2 ultramicrotome. The ultrasections were contrasted with uranyl acetate and lead citrate and observed with a JEOL-100B electron microscope. The results were as follows: 1. The cutaneous mucous glands in amphibia consisted of the glandular epithelial and the myoepithelial cells. 2. Several different cells in ultrastructure were observed in the mucous glandular epithelium of the adult amphibian skin. a. The dark and the light cells were observed in Hynobius leechi. b. The mitochondria-rich and the round secretory granule-containing cells were observed in Bombina orientalis. c. The round secretory granule-containing and the foam-like granule mass-containing cells were observed in Kaloula tornieri. d. The cutaneous mucous gland of Rana nigromaculata were divided into two types: A and B-type glands. In the A-type mucous gland, the mitochondria-rich and the round secretory granule-containing cells and in the B-type mucous gland, the mitochondria-rich, the secretory granule-containing and the ER-rich cells were observed. 3. Based upon the above findings, the authors infer that the mucous granular epithelium of the amphibian skin consists of the mitochondria-rich undifferentiated, the secetory granule-containing and mature, and the ER-rich evacuated cells.

  • The species of the slug used in this experiment is the Korean terrestrial slug (Incilaria fruhstorferi), which is examined for the cytochemical and ultrastructural research on the mucous granule-producing cells and the epithelial cells. I. Epidermal tissue According to the part of the epidermal tissue of this slug, the epidermal tissue is divided into the mantle, the foot and the dorsal epidermis. These epidennal tissue are composed of the irregular simple columnar epithelium, which are formed into the sensory epithelial cells, the supporting epithelial cells, the mucous granule-producing cells, and the clear epithelial cells are similar to the sensory epithelial cells. Both the sensory epithelial cells and the supporting epithelial cells are observed between the mantle and the foot epidermis, but the clear epithelial cells are only seen in the dorsal epidermis. II. Mucous granule-producing cell The acid mucous granule-producing cells and the neutral mucous granule producing cells are observed between the irregular simple columnar epithelium of the mantle, the foot and the dorsal epidermis. According to the part of the epidermal tissue, the number of these mucous granule-producing epithelial cells are differently distributed between the epidermis respectively.

  • This study was performed in order to know the histochemical changes of mucosubstances in mucous glands under normal and dry conditions. The frogs, Rana nigromaculata Hallowell, were collected near Seoul area in mid-August. The experimental group was retained under dry conditions for 12, 24, 30 and 36 hrs to make a comparison to normal group. The results obtained were as follows: 1. Mucosubstances in mucous glands of the frog skin contain neutral mucin and sulfated and non-sulfated mucins. Mucosubstances in the cells of the gland neck are neutral mucin, while the gland body is composed of neutral, sulfated and non-sulfated mucins. 2. Mucosubstances in mucous gland of the frog skin decreased distinctively under dry conditions, as is evident from the hitochemical properties, te neutral mucins were influenced more clearly than acid mucins. 3. Histochemical compositions of the mucosubstances in the cell of the mucous gland (the gland neck and the gland body) were not changed. 4. The phenomena were considered as a general adaptation syndrome to keep homeostasis.

  • We studied any variations of skin mucus cells of Misgurnus mizolepis caused by inducing a great change of water temperature including high temperature- and low temperature-adapted groups and then compared them with a control group of normal water condition. The high temperature-adapted group showed no significant different in size and number of the mucus cell (P>0.01), whereas in the low temperature-adapted group, the surface area of mucus cell layer and shape of its mucus cell, and the number of mucus cell remarkably increased in all the skin regions of dorsum, lateral region and occiput (P<0.01). Returned to the same condition as the control group, the low temperature-adapted groups showed the same features that appear in the normal water condition (P<0.01). Based on these results, the skin mucus cells of M. mizolepis seems to be very sensitive to cold water temperature and therefore they may play a key role in assessment of its environmental conditions.

  • This study describes the light microscopical cell types and histochemical characteristics as a preliminary study for the research on integument of the walleye pollock Gadus chalcogrammus in accordance with the physiological and environmental changes. The lateral line of the integument surface showed a curve in the anterior part and was straight from the middle to the posterior part. Integument is composed of outer epidermis and inner dermis. The epidermis is a stratified layer composed of epithelial cells, mucous cells, and club cells. Epithelial cells are classified into squamous superficial cell, cuboidal intermediated cell and columnar basal cell. The thickness of epidermis was 122.9 ㎛, and the ratio of epidermis thickness to body length was 0.03%. The mucous cell and club cell of unicellular gland were mainly distributed in the apical and middle layer of epidermis. The mucous cell contained mucosal materials of acidic glycoprotein. The proportion of mucous cells and club cells were 21.3 (± 7.0)% and 4.0 (± 1.0)% of epidermal area, respectively. The dermis was dense connective tissue layer and composed of mainly collagen fibers. It also contained fibrocytes, blood vessels, melanophores and scales.

  • The mucosubstances in the gastrointestinal mucous secreting cells of Rana rugosa were detected histochmeically during pre-hibernating, hibernating, post-hibernating and active phases. The results of the observation were as follows: 1. The mucosubstances in the gastrointestinal mucous secreting cells of active frog were strongly PAS-active in stomach, PAS-active and alcianophilic at pH 2.5 in small intestine and alcianophilic at pH 2.5 in large intestine. 2. The PAS-active mucosubstances in the gastric surface epithelial cells were increased remarkably during hibernation. 3. The alcianophilic mucosubstances at both pH 2.5 and pH 1.0 were decreased remarkably in the goblet cells of small intestine during hibernation, but a little PAS-active ones were increased. 4. The alcianophilic mucosubstances at pH 2.5 were decreased remarkably and a lttle PAS-active ones also were done in the goblet cells of large intestine during hibernation. 5. The increases of the contained quantity of mucosubstances in the gastric surface epithelial cells during hibernationi may have theeffects of preventing cohesions of gastric mucosae and suppressing activities of gastric acid and enzymes. The mucosubstances neutral acidity in the intestine during hibernation may be secreted, because of acidity being done near neutrality in its lumen, due to remarkable decrease of intestinal juices and gastric acids.
