Transfer of R Plasmids of Bacterial Isolates and Their Cloned R Genes in Natural Wastewater Environments (II) -Comparison of Transfer frequency- (하폐수의 자연환경에서 R Plasmid와 재조합 유전자의 전이특성(II) -전이율의 비교-)

  • Antibiotics resistance genes both in natural bacterial isolates and the genetically cloned bacteria were comparatively studied for their transfer frequencies by the method of conjugation in several different water environments. The Kmr genes in both kinds of bacteria were transferred more frequently in autoclaved wastewater of laboratory environment than in natural river water, but in Luria Bertani (LB) broth medium under the laboratory conditions the transfer frequences of the genes were much higher than in the autoclaved wastewater. The transfer frequencies at 2$0^{\circ}C$ and 3$0^{\circ}C$ were not much different in any water environments. The Km$^{${\gamma}$}$ genes of the genetically cloned bacteria and the natural isolates were transferred at the same frequency both in natural river water and in the autoclaved wastewater of laboratory environment, but in LB broth under laboratory conditions the transfer frequencies were lowered by 10$^{-3}$ to 10$^{-4}$ in the genetically cloned cells than the natural isolates. When donors of different cloned cells were conjugated with recipient of a natural isolates, the Km$^{${\gamma}$}$ genes of different donor cells were transferred at the about same frequency, but the same donor of the cloned cell were conjugated with recipients of different natural isolates, the transfer of Km$^{${\gamma}$}$ gene of the cloned cell showed some differences of 101 to 102 in frequency.

Analysis of Transfer Rate on Listeria monocytogenes Contaminated Pork Meat During Processing (돈육 가공공정 중 돈육에 오염된 Listeria monocytogenes의 전이율 분석)

  • In this study, the transfer rate of wild type Listeria mon, ytogenes (LM) was investigated to establish the standard of safety management during pork meat pr, essing for meat to meat and meat to food contact surfaces contamination at 5 and $10^{\circ}C$. The transfer rate of LM from meat to meat during the pr, essing increased from 0.02% after 30 min to 0.42% after 120 min at $5^{\circ}C$, while for conveyor belt and stainless steel, it decreased from 0.015% and 0.013% after 30 min to 0.002% and 0.0003% after 120 min at $5^{\circ}C$, respectively (p < 0.05). When temperature increased to $10^{\circ}C$, the transfer rates of LM from meat to meat, conveyor belt and stainless steel were the highest at 60 min exposure, and all decreased after 120 min. In reverse, the transfer rate from food contact surface to pork meat was significantly higher than that from pork meat to food contact surface (p < 0.01). Also, the transfer rate to conveyor belt was significantly higher than stainless steel (p < 0.05) and it was highest at 30 min exposure time in both 5 and $10^{\circ}C$. This study indicates that the transfer and adherence rates of LM are influenced by the contact time and temperature. Consequently, these results were utilized to develop a predictive model with a high level of confidence which can lead to prevent cross-contamination during pork meat processing.

Analysis of Pulmonary Metastases according to a New Staging Proposal (새로운 병기 제안에 따른 전이성 폐암의 분석)

  • The new stage of metastatic lung cancer based upon resectability, disease-free interval, and the number of pulmonary metastases was proposed in 1998 by Ginsberg, et al. We evaluated the validity of the new staging proposal for pulmonary metastases through the analysis of experiences at Severance Hospital. Material and Method: The cases of 111 patients who underwent resection of metastatic lung cancer during the eleven-year period (1990-2000) were reviewed. Of these patients, 103(92.8%) underwent compete surgical resection. The primary tumor was carcinoma in 60 cases, sarcoma in 46, and others in 5. The disease-free interval(DFI) was 0 to 35 months in 79 cases adn more than 36 months in 32 cases. Single metastasis accounted for 53 cases and multiple lesions for 58 cases. Mean follow-up was 49 months. Result: The actuarial survival after complete metastasectomy was 48.2% at 3 years and 32.6% at 5 years; the corresponding values for incomplete resection were 21.9% at 3 years. The 3-year survival rate(3-YSR) for complete resction was 40.5% and 5-year survival rate(5-YSR) was 30.4% for patients with a DFI less than 36 months, the 3-YSR, 75.8% and 5-YSR, 39.0% for those with a DFI equal or more than 36 moths; 45.8% and 30.5% for single lesions, 50.0% and 34.4% for multiple lesions. The 3-YSR and 5-YSR were 58.5% and 43.8% for stage I patients, 54.0% and 37.4% for stage II, 38.2% and 27.9% for stage III and 21.9% for stage IV. Conclusion: The result of the analysis of new stage of pulmonary metastases showed that the survival rate was different according to stage and there was no statistical significance. We need more experiences and long-term follow up to determine the prognostic factor of metastatic lung cancer surgery.

Transfer Rate of Cross Contamination of Listeria monocytogenes between Pork Meat and Workers' Hands during Pork Meat Processing (포장돈육 가공공정 중 돈육과 작업자 손과의 Listeria monocytogenes의 교차오염 전이율)

  • This study was performed to determine the transfer rates of each foodborne pathogen from pork meat packaging during the processing. We analyzed the transfer rate of Listeria monocytogenes from contaminated pork meat to worker's hands (wearing polyethylene gloves, PEG; cotton gloves, CG; and bare hands), cutting boards and knives, and vice versa. Transfer rate of CG 100.00% was higher than that of bare hands 2.513% and PEG 1.511%. In particular, when wearing Co, the transfer rate from the CG to bare hands with CG was 0.08%. Also, the range of transfer rates from contaminated pork meat to cutting board and knife was 0.352-3.791%. In contrast, transfer rates from the workers' hands (with PEG/CG and bare hands) to cutting board, knife, and pork meat ranged from 0.001 to 0.141%. There was a lower transfer rate from workers' hands than from pork meat. These findings indicate that use of PEG could effectively reduce or prevent the cross-contamination compared to CG and provide important information concerning the consecutive transfer of L. monocytogenes during food processing.

Stable Transmission and Continuous Expression of Human Interleukin-10 Transgene in the Offspring of Transgenic Mice (형질전환 생쥐의 후대에서 인간 Interleukin-10 유전자의 안정적 전이와 지속적인 발현)

  • The transgenic mice carrying human Interleukin-10 (hIL-10) gene in conjunction with bovine (3 -casein promoter express hIL-10 in milk during lactation. In this study, stability of germ line transmission and expression of hIL-10 transgene integrated into host chromosome were monitored up to generation F8 of transgenic mice. When male mouse of generation F8 was crossbred with normal females, approximately half of offspring (50.9±5.8%) were identified as transgenic mice. Generation F9 to F15 mice also showed similar transmission rates (66.0±20.1%, 61.5±16.7%, 41.1±8.4%, 40.7±20.3%, 61.3±10.8%, 49.2±18.8% and 43.8±25.9%, respectively), implying that hIL-10 transgene can be transmitted stably up to long term generation in the transgenic mice. Expression levels of human IL-10 from milk of generation F9 to F14 mice were 3.6± 1.2 mg/ml, 4.2±0.9 mg/ml, 5.7±1.5 mg/ml, 6.3±3.5 mg/ml, 6.8±4.5 mg/ml and 6.8±3.1 mg/ml, respectively, which was showed high-level expression compared with that of generation F1 (1.6 mg/ml) mice. In conclusion, our results suggest that transgenic mice can be continuously passed their transgenes to the progeny through the breeding program with the same productivity of human IL-10 protein in their milk.

Optimization of Transglucosylation Reaction of Stevioside (스테비오시드 당전이 반응의 최적화)

  • Stevioside was transglucosylated to improve its sweetness. Eighteen conditions were tested using the tables of orthogonal arrays of $L_{18}\;(2^{1}{\times}3^{7})$. Statistical analysis showed that the transglucosylation rate was significantly affected by temperature, the ratio of co-substrate (maltodextrin) to stevioside, pH, DE of maltodextrin and concentration of stevioside, in their order. Optimum conditions selected for temperature, ratio of co-substrate to stevioside, pH and DE of maltodextrin were $80^{\circ}C$, 1.0, 6.0 and 15, respectively. Glycosyl-stevioside with 68% yield of transglucosylation was produced at the optimum condition and found to have better quality in sweetness than stevioside and rebaudioside A.

효율적인 Follicle Stimulating Hormone의 생산을 위한 Retrovirus Vector System의 확립

  • 본 연구에서는 vesicular stomatitis virus G glycoprotein (VSV-G)를 envelope로 가지는 pantropic retrovirus vector system을 이용하여 재조합 human FSH 유전자가 전이된 형질전환 닭을 생산하고자 하였다. Human FSH $\alpha$$\beta$ 유전자와 CTP linker는 human pituitary gland cDNA library에서 RT-PCR 방법을 이용하여 cloning하였으며, 각각의 fragment는 FSH$\beta$-CTP-FSH$\alpha$ 순서의 단일사슬로 연결하였다. 연결된 FSH$\beta$-CTP-FSH$\alpha$는 retroviral vector 내의 $\beta$-actin promoter의 조절 하에 도입한 후, PT67 packaging cell line에 transfection하여 virus를 생산하였으며 생산된 virus는 pantropic한 virus producing cell인 GP293에 infection하여 FSH 유전자가 도입된 virus를 생산하였다. FSH 유전자의 발현을 in vitro에서 확인하기 위하여 CHO (chinese hamster ovary) 세포에 virus를 감염시킨 후, 세포의 배양액을 취하여 electrochemilumine-scence immunoassay 방법으로 정량하였다. In vitro에서 전이 후 발현이 확인된 FSH 외래유전자의 retroviral vector virus를 초원심분리로 고농축하여 stageX의 계란의 배반엽 층에 주입하였으며, 그 결과 18%의 부화율과 91%의 부화한 닭의 유전자 전이율을 확인할 수 있었다. 전이된 유전자의 확인은 FSH$\beta$와 Neo 유전자에 대한 primer를 이용한 RT-PCR의 방법을 이용하였다. In vitro에서와는 달리 in vivo에서는 FSH 유전자의 전이는 확인되었으나 발현을 확인하지는 못하였는데, 이는 적은 수의 실험군이 형질전환율에 비해 상대적이지 못하였거나, 외래 유전자인 FSH의 발현에 의한 생리적인 부작용이 유발되어 해당개체가 부화되지 못한 것으로 추정된다. 본 문제점을 해결하기 위하여 실험군의 수를 늘리고 외래 유전자에 대한 controllable expression system이 보완될 필요성이 요구되며, 이러한 점이 해결된다면 높은 유전자 전이율에 기인하여 retrovirus를 이용한 형질전환 방법은 형질전환 가금의 생산에 있어서 매우 효율적이고 주목할 만한 방법으로 사료된다.

Comparison of Cross-contamination of Salmonella spp. on Pork Meat and Workers' Hands during Pork Cutting Processing (돈육 가공공정에서 돈육과 작업자 손 간의 Salmonella spp.의 교차오염 비교)

  • This study describes cross-contamination events that occur in animal food processing. We analyzed the number of Salmonella spp. contamination transferred from pork meat to workers' hands (wearing polyethylene gloves; PG, cotton gloves; CG, and bare hands), cutting boards and knives, and vice versa. Transfer rate of CG 38.80% was higher than that of PG 3.11% and bare hands 1.35%. In particular, when wearing CG, the transfer rate from the CG to bare hands with CG was 0.07%. Also, the range of transfer rates from the contaminated pork meat to cutting board and knife was 0.20-1.99%. In contrast, the transfer rates from the worker's hands (with PG/CG and bare hands) to cutting board, knife, and pork meat ranged from 0.0015-0.21%. There was a lower transfer rate from workers' hands than from pork meat. These findings indicate that the use of PG compared with CG could effectively reduce or prevent the cross-contamination and provide important information concerning the consecutive transfer of Salmonella spp. during food processing.

편광분석법을 이용한 GaN 유전율 함수의 온도 변화에 대한 연구

  • III-V 족 반도체 물질 중, GaN는 넓은 밴드갭을 가지고 있어 발광 다이오드나 레이저 다이오드, 트랜지스터, 스핀트로닉스 등의 응용에 유용한 물질이다 [1]. 실시간 성장 제어 및 최적화된 특정 소자 응용을 위해서는 GaN의 다양한 온도에 대한 유전율 함수 정보가 필수적이다. 편광분석법을 이용한 상온에서의 hexagonal GaN 유전율 함수는 이미 여러 연구에서 보고되었고, 80~650 K 사이의 온도 범위에 대한 언구도 수행되었다 [2,3]. 그러나, 온도변화에 대한 GaN 유전율 함수와 $E_0$ 전이점에 대한 해석은 부정확하다. 따라서 본 연구에서는 사파이어 기판 위에 분자살박막증착장치를 이용하여 c-축 방향 (0001)으로 성장 시킨 hexagonal GaN를 0.74~6.42 eV 에너지 구간에서 보다 확장된 온도 영역(26~693 K)의 유전율 함수를 편광분석법을 이용하여 측정하였다. 측정된 GaN의 유전율 함수를 회기분석법을 통한 2차 미분 표준해석법을 이용해 분석 하였고, 그 결과 $E_0$와 excitonic $E_0$ 전이점을 명확히 얻을 수 있었다. 온도가 감소함에 따라 격자상수 및 전자-포논 상호작용이 감소하여 전자 전이점이 청색천이 하고, 그 구조가 명확해 지는 결과를 얻었다. 본 연구의 결과는 GaN 유전율 함수의 온도 의존성에 대한 데이터베이스를 제공함은 물론, 실시간 모니터링과 GaN를 기반으로 하는 광소자 제작 등에 유용할 것이다.
