Pulmonary Resection Combined with Isoniazid-and Rifampin-based Drug Therapy for Patients with Multidrug-resistant Tuberculosis (다제내성 폐결핵 환자에서 폐절제술 후 일차 항결핵제 치료)

  • Background : To evaluate the clinical efficacy of pulmonary resection combined with first-line antituberculous drug therapy in patients with well-localized, cavities-containing pulmonary multidrug-resistant tuberculosis (MDR-TB). Method : From February 1998, seventeen patients with well-localized, cavities-containing pulmonary MDR-TB were enrolled and followed prospectively up to December 2004. After radical pulmonary resection, the patients were treated with antituberculous drugs comprising of isoniazid (H), rifampin (R), pyrazinamide (Z), ethambutol (E), and streptomycin (S) (3HERZS/3HERS/6HER). Results : All recovered isolates of M. tuberculosis were resistant to both isoniazid and rifampin, and to a mean of 4.8 antituberculous drugs (range, 2 to 7 drugs). Surgical procedures included lobectomy (13 patients), lobectomy plus segmentectomy (3 patients), and pneumonectomy (1 patient). The median time for postoperative sputum smear and culture conversion was 2 days (range, 1 to 23 days). Fifteen (94%) patients had durable cures (mean follow-up period, 39.0 months). One patient failed to convert her sputum and was successfully switched to second-line therapy; one patient developed active disease again almost 7 years later, likely due to re-infection with a new M. tuberculosis strain. Conclusion : Radical resection combined with administration of first-line antituberculous agents was effective in patients with well-localized, cavities-containing pulmonary MDR-TB.

Changes of Root System in Rice (Oryza sativa L.) Plant Under Salt- and Drought- Stressed Agar Medium Conditions. (Agar 배지를 이용한 건조 및 염 처리에 대한 벼 식물체의 근계 변화)

  • This study was investigated the changes of root length, tissue structure of root tip, and dry matter production of a Dongjinbyeo (DJ) cultivar subjected to 0.4 % agar medium with various concentration of NaCl (salt stress)- and polyethylene glycol 6000 (PEG, drought stress). Root length and dry weight of DJ plant were declined along the high concentration of PEG and NaCl in rice plants. To elucidate the changes of tissue structure in root tip to PEG- and NaCl-treatments, we examined the microscopic observation of root tip in NaCl- and PEG-treated rice plants using Toluidine blue O. By Toluidine blue O staining, methyl-lignin accumulation was found in the epidermis and outer cortex of the elongation zone at an early stage of PEG treatment, whereas was found only the outer cortex of the elongation zone of NaCl-treated root tip. The epidermis of NaCl-treated root tip became soften instead of methyl-lignin accumulation. TR ratio was increased along the high concentration in PEG- and NaCl-treated rice plant as a result of inhibited root elongation under PEG- and NaCl-treatment. From these morphological changes in root stimulated by drought and salt stress, we suggest that agar medium is useful to identify tolerant variety in germination stage under stressful environments.

Report on Settlement of Alien Species Red Swamp Crawfish (Procambarus clarkii) in Korea (외래생물 미국가재(Procambarus clarkii)의 국내 자연생태계 정착 보고)

  • The Red swamp crawfish Procambarus clarkii is native to the southern United States and inhibits all over the world including the United States, Asia, Africa and Europe. In Europe, it is known to disrupt ecosystems such as competition with native crayfish. In this study, habitats of red swamp crawfish were identified in Jiseok stream (15.5 km section) and Daecho stream (6.1 km section) of Youngsan river. A total 33 red swamp crawfish were captured. A female captured in Jiseok stream carried 215 newly hatched red swamp crawfish in her the burrow. As a result of the analysis of the habitat environment, the water temperature was ranged of 2.4~32.8℃, conductivity was 53.4~502.3 μS cm-1 and DO was 0.74~22.64 mg L-1. As a result of measuring HOBO, the temperature ranged from -9~48.1℃, showing a change of 57.1℃ throughout the year. The variation of the water temperature throughout the year from 2.9~33.9℃ was found to be 31℃. Red swamp crawfish have a high adaptability and a wide range of environmental resistance, and report a sharp increase in newly invaded areas, negatively affecting indigenous organisms. In addition, it can be used as a medium for the spread of diseases and parasites, so prompt investigation and research should be carried out.

Hypoglycemic Effect of the Functional Food Manufactured by Fermented Soybean as Main Materials in Streptozotosin - Induced Diabetic Rats (Streptozotocin 유발 당뇨쥐에 있어서 발효콩을 주원료로 한 기능성 식품의 혈당강하 효과)

  • The present study was undertaken to evaluate the effect of functional food, which was processed with fermented soybean as main ingredient (FS), on the body weight, organ weight, plasma glucose, and plasma lipid in diabetic rats caused by streptozotocin (STZ). The body weight was decreased more slowly in the FS group than in the diabetic, and the food intake increased significantly in all diabetic groups. The food efficiency was very low in all diabetic groups, but increased significantly in the FS groups than diabetic control (p<0.05). In comparing the weight of organ, the weight of liver and kidney were increased in all diabetic groups than in the control, and decreased slightly in FS groups. The weight of heart and spleen were not different among all test groups. In the oral glucose tolerance test, the blood glucose in the diabetic group was the highest in 60 minutes. And the blood glucose in the FS group was the highest in 30 minutes, and decreased significantly after 120 minutes to the level of fasting glucose. The glucose in serum was decreased significantly in the FS groups fed the functional food for 4 weeks, compared to the diabetic control (p<0.05). Total cholesterol, triglyceride and atherogenic index (AI) in serum were significantly higher in diabetic control, compared to the normal (p<0.05), and decreased by 16.4%, 15.4% and 48.3%, respectively, in the FS fed 400 mg/kg of functional food. HDL-cholesterol was increased significantly in the FS-400, compared to the diabetic control. These results support that functional food using fermented soybean improve glucose and lipid metabolism in diabetic rats.

Ecotoxicological Study of Gammarus sobaegensis by pH Depression in Artificial Channels - Drift behavior - (인공수로에서 산성화 영향에 따른 소백옆새우(Gammarus sobaegensis)의 생태독성학적 연구 - 표류행동을 중심으로 -)

  • This study was carried out to examine the drift of Gammarus sobaegensis by acid stress as pH depression in Oweol creek from April 1996 to October 1996. The behavioral drifting was revealed to the characteristics of G. sobaegensis that is tend to increase as acid stress. And, tolerance level of G. sobaegensis to pH depression was different among the size classes. Individuals belong to small to medium size classes were weak in lower pH. Pattern of response in G. sobaegensis has a lower tolerance to acid stress at below pH 4.0 than above pH 5.0 in the artificial channel and show the possibility as an effective aquatic ecotoxicity test organism. The result of analysis of variance, water temperature (F-ratio : 66.596, p< 0.0005) and the size classes (F-ratio : 71.386, p< 0.0005) except pH level (F-ratio : 353.415, p<0.0005) were showed to the major factor for drift behavior by acid depression. Evaluation of the Colonization of Lactobacillus plantarum in Mouse Gut by Terminal Restriction Fragment Length Polymorphism Analysis (Terminal Restriction Fragment Length Polymorphism 분석을 이용한 Lactobacillus plantarum의 생쥐 장관 정착 평가)

  • T-RFLP (terminal restriction fragment length polymorphism) analysis, one of the most highly adopted culture-independent microbial community analysis methods, was applied to evaluate the colonization of probiotics in experimental animal gut. Lactic acid bacteria that exhibited cinnamoyl esterase activity were isolated from Korean fermented vegetables and identified by 16S ribosomal RNA sequence analysis. Lactobacillus plantarum KK3, which demonstrated high chlorogenic acid hydrolysis by cinnamoyl esterase activity, and acid/bile salt resistances, was cultured, freeze-dried, and fed to mice and the microbiota in their feces were monitored by T-RFLP analysis. The T-RF of L. plantarum was detected in the feces of mice after the start of administration and lasted at least 31 days after the initial 7 day feeding. T-RFLP analysis was considered a useful tool to evaluate the gut colonization of probiotic L. plantarum. In order to prove that L. plantarum was from viable cells, we reisolated L. plantarum in the feces using cinnamoyl esterase activity media as the screening step. The colonization of L. plantarum KK3 in the mouse gut was confirmed by this research.

Isolation and Characterization of Zymomonas mobilis DNA Fragments Showing Promoter Activity in Escherichia coli (Escherichia coli에서 Promoter 활성을 보이는 Zymomonas mobilis DNA 조각의 분리와 분석)

  • For the purpose of isolation of the Zymomonas mobilis DNA fragments showing promoter activity in Escherichia coli, a promoter screening vector, PCMT215 was constructed by transferring a promoterless chloramphenicol acetyltransferase (CAT) gene of pYEJ001 into pMT21 which contains $\beta$-lactamase gene and multiple cloning sites. A library of Z, mobilis Sau3AI DNA fragments was constructed in E. coli using the newly constructed pCMT215. Fourteen clones showing resistance to chloramphenicol ranging in concentration from 30 to 750 $\mu$g/$m\ell$ were selected. From five clones of them, the Z. mobilis DNA fragments expressing CAT gene of the recombinant plasmids were sequenced and then sites of transcriptional initiation were identified. The nucleotide sequences of the cloned DNA shared AT rich regions, poly A's or T's stretches and palindromic regions. The positions of transcriptional initiation for CAT gene occurred at more than one site spaced over by 4 to 190 base pairs on the cloned fragments in E. coli.

The Combined Effect of Fast Neutron and Hyperthermia according to the Sequence and Interval in MKN-45 Cells (MKN-45 세포에서 속중성자와 온열치료의 순서 및 간격에 따른 병용효과)

  • Purpose : It has been well established that the response of cells and tissues to low LET radiations (X- or gamma-ray) can be enhanced by combining with hyperthermia. However, there has been relatively little work of hyperthermia on the possible modification of either cellular or tissue responses to other types of radiation. So, we investigated the combined effect of fast neutron irradiation and hyperthermia according to the sequence and time interval of the two. Materials and Methods : In MKN-45 cells, a human stomach cancer ceil line, suwiving fractions were measured according to the sequential treatment of 0, 4, 2, 0 hour-intewal for fast neutron irradiation (1.5 Gy) combined with hyperthermia (41 $^{\circ}C$ for 30 min or 43$^{\circ}C$ for 30 min). Results : D$_{0}$ and n of MKN-45 for neutron were 0.8 Gy and 2.5, respectively. The surviving fraction by 1.5 Gy of neutron was 0.36$\pm$0.34. Interacting powers were mostly ranged between 1 and 2, but they were 3.0 and 2.7, respectively for hyperthermia (41 $^{\circ}C$ for 30 min) fellowed by neutron irradiation 6 and 4 hours later. Conclusion : The combined effect of fast neutron (1.5 Gy) and hyperthermia (41 $^{\circ}C$ or 43$^{\circ}C$ for 30min) is largely independently additive. Preceding mild hyperthermia (41 $^{\circ}C$ for 30 min) 4 or 6 hours before neutron may cause decreased sensitivity to subsequent neutron irradiation.

The Influence of the Sympathetic Nervous System on the Development and Progression of Cancer (교감신경계가 암의 발전과 진행에 미치는 영향)

  • Living creatures possess long-conserved mechanisms to maintain homeostasis in response to various stresses. However, chronic and continuous exposure to stress can result in the excessive production of stress hormones, including catecholamines, which have harmful effects on health. Studies on the relationship between the sympathetic nervous system (SNS) and cancer have been conducted based on the traditional hypothesis that stress can promote cancer progression. Many preclinical and epidemiological studies have suggested that the regulation of ${\beta}$-adrenergic signaling, which mediates SNS activity, can suppress the progression of solid tumors. SNS activation has highly pleiotropic effects on tumor biology, as it stimulates oncogenes, survival pathways, the epithelial - mesenchymal transition, and invasion. Moreover, it inhibits DNA repair and programmed cell death and regulates the tumor microenvironment, including immune cells, endothelial cells, the extracellular matrix, mesenchymal cells, and adipocytes. Although targeted therapies on the molecular basis of tumor proliferation are currently receiving increased attention, they have clinical limitations, such as the compensatory activation of other signaling pathways, emergence of drug resistance, and various side effects, which raise the need for pleiotropic cancer regulation. This review summarizes the effects of the SNS on the development and progression of cancer and discusses the clinical perspectives of ${\beta}$-blockade as a novel therapeutic strategy for this disease.

Deubiquitinase Otubain 1 as a Cancer Therapeutic Target (암 치료 표적으로써 OTUB1)

  • The ubiquitin system uses ligases and deubiquitinases (DUBs) to regulate ubiquitin position on protein substrates and is involved in many biological processes which determine stability, activity, and interaction of the target substrate. DUBs are classified in six groups according to catalytic domain, namely ubiquitin-specific proteases (USPs); ubiquitin C-terminal hydrolases (UCHs); ovarian tumor proteases (OTUs); Machado Joseph Disease proteases (MJDs); motif interacting with Ub (MIU)-containing novel DUB family (MINDY); and Jab1/MPN/MOV34 metalloenzymes (JAMMs). Otubain 1 (OTUB1) is a DUB in the OTU family which possesses both canonical and non-canonical activity and can regulate multiple cellular signaling pathways. In this review, we describe the function of OTUB1 through regulation of its canonical and non-canonical activities in multiple specifically cancer-associated pathways. The canonical activity of OTUB1 inhibits protein ubiquitination by cleaving Lys48 linkages while its non-canonical activity prevents ubiquitin transfer onto target proteins through binding to E2-conjugating enzymes, resulting in the induction of protein deubiquitination. OTUB1 can therefore canonically and non-canonically promote tumor cell proliferation, invasion, and drug resistance through regulating FOXM1, ERα, KRAS, p53, and mTORC1. Moreover, clinical research has demonstrated that OTUB1 overexpresses with high metastasis in many tumor types including breast, ovarian, esophageal squamous, and glioma. Therefore, OTUB1 has been suggested as a diagnosis marker and potential therapeutic target for oncotherapy.
