A Simple Method for Detection of Trypsin Inhibitors in Soybean (Glycine max) (대두 Trypsin Inhibitor의 간이검정법)

  • The specific reaction of trypsin inhibitors with trypsin to form stable complexes was successfully applied for detection of trypsin inhibitors in soybean. Soybean extract was treated with $Ca^{++}$ to remove globulin fraction, followed by digestion with trypsin and fractionated by chromatography on Sephadex G-50. The void volume fraction contained the trypsin-trypsin inhibitor complexes as well as trypsin. The trypsin inhibitors were then detected by their molecular weight differences on SDS-polyacrylamide gel electrophoresis, in which the complexes dissociate into trypsin and its inhibitors. With the method proposed, trypsin inhibitors were indentified by their ability forming the stable complexes with trypsin and their anti-tryptic moiety. The formation of the complexes with trypsin was further confirmed by two dimensional electrophoresis and DEAE-Sephadex A-25 chromatography. Employing the proposed method, it was found that soybean (Glycine max cv. Hill) contained 7 trypsin inhibitors.

Electrophoretic Pattern of Specific Proteins in Meat Products (육가공품(肉加工品)중 단백질의 전기영동(電氣泳動) 패턴)

  • The possibility of using sodium dodecylsulfate-polyacrylamide gel electrophoresis was studied to detect specific proteins and their content in meat products such as beef, pork, fish, soybean, fish paste, ham and fish sausage. Many complicated bands were observed in the total protein fractions of the tested samples. The number of protein bands in the low salt-soluble protein fractions was considerably lesser and showed more specific bands in comparison with total protein fractions. Actone-insoluble fractions of non-meat proteins showed different patterns from meat proteins. A heating procedure seemed to be a cause for the diminished number and quantity of resolved protein bands in sausages. The results suggest that the discgel electrophoresis can be used to detect specific proteins and their content in protein foods, if a selective extraction method is emplyed.

  • 전류를 흘렸을 때 양극과 음극에 따라 움직이는 미세한 나노입자를 이용하여 색, 글자, 그림 등을 표시하는 응용디스플레이 기술이 전기영동디스플레이(electrophoretic display)이다. 최근 전자종이 등 상품화가 진행되면서 전기영동디스플레이에 대한 관심증대와 함께 기술개발이 지속적으로 진행되고 있다. 본 연구에서는 분산중합을 이용하여 $TiO_2$ core 입자에 polystyrene을 shell로 코팅하여 마이크로캡슐형의 전기영동디스플레이에 적합한 입자를 제조하고 성능을 분석하였다. 먼저 분산제의 종류, 모노머의 농도, 개시제의 농도에 따라 제조된 대전복합입자의 크기 및 분포를 보면, 분산제의 종류를 달리 하였을때를 제외하고 대체로 균일하였다. 입경의 변화를 보면, 약 200-300nm의 $TiO_2$가 개질에 의해 400-500nm의 입경을 나타내는 것으로부터 200nm 두께의 shell층을 갖는다는 것을 확인 할 수 있었다. 또한 분산제의 종류에 따라서는 분산제를 사용하지 않는 경우가 오히려 제조된 입자의 분포가 균일함을 알 수 있었고 모노머의 농도에 따른 변화는 볼 수 없었으며 입경분포가 균일한 입자가 제조되었음을 알 수 있었다. 대전복합입자의 TGA 곡선으로부터 $300^{\circ}C$ 부근에서 polystrene shell에 의한 분해를 볼 수 있었고 $600^{\circ}C$ 이후에 잔류된 core의 $TiO_2$ 입자를 확인 할 수 있었다. 이 결과로부터 $TiO_2$ core-polystyrene shell형의 전자 종이용 대전복합입자의 제조를 확인 할 수 있었다. 또한 제조된 대전복합입자의 zeta potential을 보면, (+)전하를 띄며 64.8mV의 비교적 높은 zeta potential을 가지는 것을 확인 할 수 있었다. 그리고 $TiO_2$ 대전복합입자와 같은 방법으로 제조된 흑색 대전복합입자를 혼합하여 cell test를 측정한 결과, cell에 ${\pm}$10V의 저전압을 가했을 때에도 비교적 응답속도가 빠른 입자의 구동현상을 확인 할 수 있었다.

Changes of Protein Pattern of Mungbean Seeds, Phaseolus aureus During Germination (녹두 발아중 단백질 전기영동 패턴의 변화)

  • During the germination of mungbean seeds, the changes of water contents, total and soluble proteins, and electrophoretic pattern of the soluble proteins were examined. The moisture content of a dry mungbean was 12.7%, which was greatly increased after the soaking. Along to the germination period, the moisture contentof the mungbean sprouts was gradually increased up to 90.7%. The contents of total and soluble proteins were sharply decreased after the soaking of the mungbean and decreased gradually during the germination. PAGE of the soluble proteins showed two broad bands and three sharp bands. During the germination, two broad bands were weadened but other bands were relatively stable. SDS-PAGE showed 19 discrete bands and during the germination, the most of the bands were thinned or disappeared. But some of the protein bands were stable until the end of germination.

  • Panax ginseng C.A. Meyer is a well-known Korean traditional medicine. Until now, even though major research of ginseng has been focused on the pharmacological effect, clinical application and chemical analysis of extracted secondary metabolite for several years, the physiology and gene functions of ginseng were not well known. In this research, we have developed the protein extraction methods of ginseng root and hairy root for proteome analysis in order to elucidate the gene(s) function of ginseng. Using the liquid nitrogen (equation omitted) TCA method as protein extraction method, about 660 protein spots were detected on the 2-DE gel of hairy root. Additionally, comparative analysis result of 2-DEs of ginseng root (equation omitted) hairy root suggested that proteomes of same organism could be changeable according to the culture condition, growth stages and other stimulus.

  • Bowman-Birk type protease inhibitors of soybeans were purified with gel filtration on Sephadex G-75. Significant differences were found in the content and in the electrophoretic patterns of the purified protease inhibitors. Ten among fourteen electrophoretic bands appeared as protease inhibitors. The chymotrypsin and trypsin inhibiting activities in soybeans showed three and four fold variation respectively. And the cultivars with higher chymotrypsin inhibiting activities seemed to have higher cysteine contents.

  • With naked barley varieties which are different in their spring habits, variations in seed proteins during ripening were checked by acrylamide gel electrophoresis. In case of 7% acrylamide gel among protein bands ⅰ-band was observed only in both Sedohadaka (intermediate type) and Wanju (spring type) throughout the ripening period, and j-band was detected in all the three varieties until 33 days after heading, but not in Nonsangwa (winter type) thereafter. In case of 6M urea-7% acrylamide gel z-band was traced only in Nonsangwa, contrary to i-band, throughout the ripening period. U-band was observed in all the three varieties until 23 days after heading, but not in Nonsangwa thereafter. X-band showed opposite from u-band. Throughout the ripening period Sedohadaka was significantly more similar in the electrophoretic patterns to Wanju than to Nonsangwa.

Iontophoretic Transdermal Delivery of Alendronate in Hairless Mouse Skin (In-vitro에서 전기이온영동법을 이용한 알렌드로네이트의 경피약물전달)

  • This study examined the transdermal delivery of alendronate across hairless mouse skin. The effects of iontophoresis, perforation with a microneedle, and a combination of a microneedle pretreatment and iontophoresis were evaluated in vitro test. Hydrogel patches were polymerized by UN polymerization to supply a hydrogel patch to the iontophoretic transdermal drug delivery system. The alendronate content in the iontophoretic delivery patch was $5.0\;mg/cm^3$. The amounts of alendronate that permeated across the hairless mouse skin when current densities of 0.25 and $0.50\;mA/cm^2$ were supplied to the iontophoretic alendronate patch were $0.80{\pm}0.03$ and $2.00{\pm}0.02{\mu}g$, respectively. After pretreatment with a microneedle, the amounts of alendronate that permeated across the hairless mouse skin increased to $70.65{\pm}0.37$ and $162.23{\pm}0.40{\mu}g$, respectively. The biocompatibility of the iontophoretic alendronate patch was examined according to the international standardization organization 10993.

Solubility and Electrophoretic pattern of Korea Ginseng Protein (한국산 인삼 단백질의 용해성 및 전기영동 패턴)

  • For the systematic investigation of biochemical characteristics of Korean ginseng protein, protein fractions were analyzed by the techniques of sodium dodecyl sulfate (SDS) polyacrylamide gel electrophoresis. The effect of pH and various salts on extractibility of ginseng protein were determined while the amino acid composition was studied by amino acid autoanalyzer. The protein was consisted of 66.08% of albumin and 20.51% of glutelin. Extractability of ginseng protein was the lowest in pH 3.0 and the highest in $pH\;6.0{\sim}8.0$. Among the neutral salts solution, $0.4M\;Na_2CO_3$ showed maximum extractability while $1.0M\;MgSO_4$ solution showed the least extractability. Resonable precipitation was obtained by 40% of acetone and ammonium sulfate. It has been shown by SDS polyacrylamide gel electrophoresis that the soluble protein had 11 bands. The molecular weight for the main protein of the soluble protein wasestimated to be 43,000. In amino acid composition of water extracted protein, arginine content was the highest 47.17% while on the contray, proline and cystine contents were very low.

  • Protein profiles of beef loin were constructed by comparing two-dimensional gel electrophoresis patterns of the proteins from different growth stages of Hanwoo, Korean cattle. Proteins from the lean muscle of 0, 6, 12 and 24 months old Hanwoo were separated by isoelectric focusing (IEF) on 16 cm tube gels, and further processed second dimensionally by 12% SDS-polyacrylamide gel electrophoresis (PAGE) using $18{\times}20$ cm gel. Proteins with pI values ranging 3.0 to 9.0 and molecular weight of 15 to 100 kDa could be clearly detected on gel by silver staining. Interestingly, many of the proteins significantly increased and decreased during growth happened to be low molecular ones. To isolate the increased proteins, the soluble proteins were obtained from the tissue by 1% Triton X-100 extraction, then, fractionated by 30% and 50% ammonium sulfate. The isolation condition of each particular protein was determined. According to the conditions, two of the increased proteins were isolated, and transferred to PVDF membrane and microsequenced.
