흰쥐 해마(hippocampus)에서 acetylcholine (ACh) 유리에 미치는 $A_{1}-adenosine$ 수용체의 post-receptor 기전에 관한 지견을 얻고자 하여 $^3H-choline$으로 평형시킨 해마 slice를 사용하여 $^3H-ACh$ 유리에 미치는 여러가지 약물들의 영향을 관찰하였다. Adenosine $(0.3{\sim}300\;{\mu}M)$은 전기자극(3Hz, 2 ms, 5 $VCm^{-1}$, 구형파)에 의한 ACh 유리를 용량 의존적으로 감소 시켰으며, 이러한 효과는 $A_1-adenosine$ 수용체의 선택적 차단제인 8-cyclopentyl-1, 3-dipropylxanthine $(2\;{\mu}M)$에 의해 차단됨을 볼 수 있었다. G-단백 억제제인 N-ethylmaleimide (NEM, 10과 $30\;{\mu}M$)는 그 자체에 의하여 자극유발성 ACh 유리를 증가시켰으며, adenosine의 효과는 NEM 전처리에 의하여 완전히 소실되었다. Protein kinase C 활성화제인 $4{\beta}-phorbol$ 12, 13-dibutyrate (PDB, $1{\sim}10\;{\mu}M$)는 ACh 유리 증가를 일으켰으며 억제제인 polymyxin B (PMB, $0.03{\sim}1\;mg$)는 감소를 일으켰으나, adenosine에 의한 ACh 유리 감소효과는 PDB 및 PMB에 의해 영향을 받지 않았다. $Ca^{++}$-통로 차단제인 nifedipine $(1\;{\mu}M)$은 adenosine의 효과를 길항함을 볼 수 있었으나 $K{^+}$-통로 차단제인 glibenclamide는 adenosine의 효과에 영향을 미치지 못하였다. 8-Bromo-cAMP (100과 $300{\mu}M$) 그 자체로는 ACh 유리에 별다른 영향을 미치치 못하였으나 $300\;{\mu}M$ 8-bromo-cAMP 전처리에 의하여 $30\;{\mu}M\;adenosine$의 효과가 억제됨을 볼 수 있었다. 이상의 실험결과로 흰쥐 해마에서 $A_1-adenosine$ 수용체를 통한 adenosine의 ACh유리 감소는 G-단백에 의존적이며, 이러한 효과에 nifedipine에 예민한 $Ca^{++}$-통로와 adenylate cyclase계가 일부 관여함은 확실하나 proteinkinase C 및 glibenclamide에 예민한 $K{^+}$통로는 관여하지 않는 것으로 사료된다.
동맥경화 상태에서는 혈관이 수축물질에 예민하게 반응함으로써 혈압의 급격한 변화에 대한 회복능력이 저하한다. 한편 뽕나무 잎에는 고지방식의 섭취로 인해 발생하는 동맥경화증의 발달을 감소시킬 가능성이 있는 여러 물질이 있다고 추정된다. 뽕나무 잎을 백서에 투여하면 고지방식에 의해 유발된 혈관이 예민해지는 현상을 감소시킬 수 있는지 시험하기 위하여 고지방식 및 뽕나무잎을 8주간 투여하고 혈관수축 물질에 대한 혈압의 반응성을 시험하였다. 순환기반사에 따른 혈압반응 감수성차이를 최소화할 목적으로 동물의 중추신경계를 파괴(pithing)하고 시험하였다. 고콜레스테롤을 함유한 식이를 공급한 백서에서 교감신경계 전기자극이나 norepinephrine, phenylephrine, angiotensin II 및 vasopressin 등의 투여로 유발된 혈압상승반응이 증강되었으며, 사료에 뽕잎을 2% 또는 10% 첨가하면 정상적인 혈압상승반응을 보여주었다. 교감신경계를 파괴하지 않은 채 마취만 한 백서에서는 고콜레스테롤에 의한 반응증강이나 뽕잎에 의한 변화를 관찰할 수 없었다. 이 결과로 부터 뽕잎이 동맥경화에서 나타나는 혈관반응성 변화를 방지하는 데 유용한 것으로 판단할 수 있을 것이다.
The purpose of this study has been conducted to reduce the lower limbs' spasticity of the patients with hemiplegia caused by cerebral stroke of apoplexy and find differences about spasticity effects among each group. The objects of this study covered 24 patients with hemiplgia who are either in the oo hospital in Daegu or under treatment from home to hospital. The objects fall into three groups which are a group of neurological development treatment, a group of functional stimulus treatment and a group of neurological development treatment and functional stimulus treatment. The result of this study were as follows : 1) The neurological development treatment has been found to reduce the lower limbs' spasticity of patients with hemiplegia caused by cerebral stroke of apoplexy and compared to before-treatment, the MAS value of spasticity has been shown to be statistically meaningful ,and gradually over the period of between 4 weeks and 8 weeks(P <.05). 2) The functional electric stimulus treatment has been shown to reduce the lower limb's spasticity of patients with hemiplegia caused by cerebral stroke of apoplexy and compared to before-treatment, the MAS value of spasticity was statistically meaningful and compared to 4 weeks, even at the time of 8 weeks, the MAS value of spasticity have shown statistical meaningness. (P <.05) 3) When neurological development treatment and functional electric stimulus treatment was applied at the same time, the lower limbs' spasticity of patients with hemiplegia was reduced meaningfully(P <.05). Compared to before-treatment at the time of 4 weeks, the MAS value of spasticity was statistically meaningful and compared to 4 weeks at the time of 8 weeks the MAS value of spasticity was also statistically meaningful(P <.05) 4) In the case of time-based MAS value of each group, functional electric stimulus treatment reduced the spasticity more meaningfully than neurological development treatment, and the group of same application of functional electric stimulus treatment and neurological development treatment showed better statistical meaningness than functional electric stimulus treatment alone(P <.05) and finally the group of same application of neurological development treatment and functional electric stimulus treatment showed more meaningful difference than neurological development treatment alone(P <.05)
Objectives : To observe physiological changes during clinical acupuncture treatment. Methods : We recruited 40 healthy volunteers who had experienced an acupuncture treatment at least once within the past three years. The experimental group was divided into four groups according to the needling site and frequency of electrical stimulation. Sites consisted of abdomen and legs. Frequencies consisted of 100 Hz and 2 Hz. The procedures of experimental treatment consisted of seven phases, Resting I phase (Resting I), Needle insertion phase (Insertion), Maintenance of needle insertion I phase (Maintain I), Electrical stimulation phase (ES), Maintenance of needle insertion II phase (Maintain II), Needle removal phase (Removal) and Resting II phase (Resting II). We measured the surface electromygraphy (SEMG) through an electrode on the frontalis muscle during all phases consecutively. Results : When SEMGs of all seven phases were analyzed, they significantly increased or decreased according to phases. SEMGs of Insertion, Maintain I, ES and Maintain II phase significantly increased more than RestingI in abdomen and legs groups. SEMGs of the abdomen group were measured as being $4.78{\pm}0.74{\mu}V$ on Resting I, $16.48{\pm}3.97{\mu}V$ on Insertion, $46.31{\pm}10.56{\mu}V$ on Maintain I, $45.88{\pm}9.72{\mu}V$ on ES, $45.56{\pm}9.69{\mu}V$ on Maintain II, $18.76{\pm}3.05{\mu}V$ on Removal, and $3.75{\pm}0.65{\mu}V$ on Resting II. SEMGs of the legs group were measured as being $3.34{\pm}0.35{\mu}V$ on Resting I, $12.11{\pm}1.76{\mu}V$ on Insertion, $36.74{\pm}6.99{\mu}V$ on Maintain I, $33.57{\pm}6.30{\mu}V$ on ES, $32.66{\pm}6.03{\mu}V$ on Maintain II, $14.08{\pm}2.15{\mu}V$ on Removal, and $2.88{\pm}0.32{\mu}V$ on Resting II. Conclusions : SEMG changed differently according to processes of acupuncture. Electrical stimulation showed different change of SEMG. Thus, acupuncture treatment may change the status of the autonomic nervous system.
Background: Peripheral nerve injury leads to neuropathic pain, including mechanical hyperalgesia (MH). Nerve discharges produced by an injury to the primary afferents cause the release of glutamate from both central and peripheral terminals. While the role of centrally released glutamate in MH has been well studied, relatively little is known about its peripheral role. This study was carried out to determine if the peripherally conducting nerve impulses and peripheral glutamate receptors contribute to the generation of neuropathic pain. Methods: Rats that had previously received a left L5 dorsal rhizotomy were subjected to a spinal nerve lesion (SNL) or brief electrical stimulation (ES, 4 Hz pulses for 5 min) of the left L5 spinal nerve. The paw withdrawal threshold (PWT) to von Frey filaments was measured. The effects of an intraplantar (i.pl.) injection of a glutamate receptor (GluR) antagonist or agonist on the changes in the SNL- or ES-produced PWT was investigated. Results: SNL produced MH, as evidenced by decrease in the PWT, which lasted for more than 42 days. ES also produced MH lasting for 7 days. MK-801 (NMDAR antagonist), DL-AP3 (group-I mGluR antagonist), and APDC (group-II mGluR agonist) delayed the onset of MH when an i.pl. injection was given before SNL. The same application blocked the onset of ES-induced MH. NBQX (AMPA receptor antagonist) had no effect on either the SNL- or ES-induced onset of MH. When drugs were given after SNL or ES, MK-801 reversed the MH, whereas NBQX, DL-AP3, and APDC had no effect. Conclusions: Peripherally conducting impulses play an important role in the generation of neuropathic pain, which is mediated by the peripheral glutamate receptors.
최근 첨단 과학의 발달과 함께 인체조직에 대한 적용도가 뛰어난 소재가 개발되어 초소형의 이식형 장치가 개선되어감에 따라 전력공급방법의 다양한 연구가 이루어져 유도코일을 이용하여 무선으로 전력을 전송하는 장치가 연구되어오고 있다. 이에 저자는 이론적으로 효율이 100%인 E급 전력증폭기를 사용하여 $2{\sim}30mm$의 공극거리에서 가장 이상적인 주파수를 1MHz로 설정하여 제작하였고, 직경 46mm의 송수신 코일을 이용하여 코일의 비정렬에 대한 전송율이 이격거리가 10mm일 때, 20% 감소되었다. PLL을 사용하여 주파수추적동조법으로 공극거리 15 mm 이내에서는 완만하게 20% 정도의 에너지 전송효율을 얻을 수 있었다. 또한 최적의 공진거리에서 50 mA 정도의 출력전류가 얻을 수 있다는 것은 초소형 전기 자극기와 같은 이식형 장치를 동작시키는데 구동 전력으로 가능하다는 것을 알 수 있었다.
청신경의 이상으로 발생하는 감각신경성 난청의 경우, 달팽이관이나 청각신경에 전극을 이식하여 전기자극을 가함으로써 청지각을 살릴 수 있다. 이를 위해 우선적으로, 각 청각신경들이 담당하여 인지할 수 있는 소리의 주파수 분포를 표시한 음계소지도를 파악해야 한다. 본 논문에서는 청각신경신호 검출 장치용 다중채널 아나로그 프론트엔드 회로를 제안한다. 제안된 아나로그 프론트엔드의 각 채널은 AC 커플링 회로, 저 전력 4차 Gm-C LPF와 단일 기울기 ADC로 이루어진다. AC 커플링 회로는 청각신호의 불확실한 DC 전압 레벨을 제거하고 AC 신호만 전달한다. Gm-C LPF는 청각신호의 대역폭을 고려하여 설계 되었으며, 플로팅-게이트 기법이 적용된 OTA를 사용하였다. 채널별 ADC를 구현하기 위해서, 최소의 면적으로 구현할 수 있는 단일 기울기 ADC 구조를 사용하였다. 측정 결과, AC 커플링 회로와 4차 Gm-C LPF는 100 Hz - 6.95 kHz의 대역폭을 가지며, 단일 기울기 ADC는 7.7 비트의 유효 해상도를 가진다. 그리고, 채널 당 $12\;{\mu}W$의 전력이 소모 되었다. 전원 전압은 3.0 V가 공급되었고, 코어는 $2.6\;mm\;{\times}\;3.7\;mm$의 실리콘 면적을 차지한다. 제안된 아나로그 프론트엔드는 1-poly 4-metal $0.35-{\mu}m$ CMOS 공정에서 제작 되었다.
망막에서 나오는 활동전위와 같이 복잡한 신경망을 거쳐 처리되는 전기신호를 분석하기 위해서는 기존의 단일 전극 기록법으로는 어렵다. 단일 전극을 통한 활동전위의 기록은 개개의 신경세포 특성을 알아내는 데에는 유용한 방법이나 신경세포 간의 시간적, 공간적인 관계는 알아낼 수 없다는 한계를 가지고 있으므로 이같은 한계를 극복하기 위하여 다채널 전극을 이용한 신경신호 기록방법이 최근에 개발되어 널리 이용되고 있다. 다채널전극 기록 방식인 MEA60 시스템은 세포 밖에 위치한 60개의 전극이 생체신호를 동시에 기록한다. 세포 fi에 위치한 각각의 전극이 포착한 신경 신호는 하나의 망막신경절세포 반응이라기보다는 여러 세포의 반응이 동시에 기록될 가능성이 높다. 그러므로 여러 세포의 반응이 함께 기록된 신호로부터 각각의 세포로부터 나오는 파형을 구분하는 작업이 반드시 필요하다. 본 연구에서는 다채널 전극으로 기록한 망막 신경절세포 신호로부터 MATLAB을 이용하여 활동전위 파형을 검출하고 분류하는 과정을 구현하여 보았다. 이러한 분류과정은 추후 진행되는 신호분석방법인 자극 후 시간 히스토그램(poststimulus time histogram, PSTH), 자기상관관계(autocorrelogram), 상호상관관계(cross-correlogram)를 보기 위하여 반드시 거쳐야 하는 전처리(preprocess) 과정이다. 본 연구에서는 MATLAB을 이용한 파형 구분 프로토콜을 확립하였을 뿐만 아니라 이러한 프로토콜이 신경절 세포의 활동전위 파형을 검출하는 데 유용한 방법임을 입증하였다
아스코크로린(Ascochlorin, ASC)은 Ascochyta viciae로부터 추출된 프레닐페놀 물질로, 혈청 콜레스테롤과 트리글리세라이드 수치를 감소시키고 종양 성장을 억제한다는 연구 결과가 보고되어 있다. 본 논문에서는 아스코크로린이 생리학적 혹은 병리학적인 작용과 염증반응에서 약리학적으로 유도되는 반응을 어떻게 조절하며, 이러한 메커니즘에 대해 이해하기 위해 mouse macrophage Raw264.7 세포에 아스코크로린을 처리하여 이에 대한 프로테옴의 특이적인 발현에 대해 분석하였다. 따라서 본 연구는 LPS를 처리한 mouse macrophage Raw264.7 세포에 아스코크로린을 처리하여 염증과정에 관련된 단백질의 발현 양상을 확인하기 위해 프로테오믹스를 시행하였다. Mouse macrophage RAW264.7 세포에 아스코크로린을 처리한 조건과 무처리한 조건으로 나누어 two-dimensional electrophoresis (2-D SDS-PAGE), matrix-assisted laser desorption/ionization mass spectrometry (MALDI-TOF-MS)와 bioinformatics 방법으로 아스코크로린을 처리한 mouse macrophage Raw264.6 세포의 프로테옴을 분석하였다. 그 결과 mouse macrophage Raw264.7 세포에 아스코크로린 처리 시 Calreticulin이 4배 감소, ${\beta}-actin$도 4배 감소 그리고 vimentin이 1.5배 감소함을 확인 할 수 있었다. 그러나 rabaptin 아스코크로린 처리에 의해 3배 증가함을 확인 할 수 있었다. 이러한 단백질 발현은 RT-PCR을 수행하여 결과에 대해 재확인 하였으며, 프로테오믹스와 동일한 결과를 얻을 수 있었다. 따라서 본 연구를 통해 LPS 처리에 의해 활성화된 mouse macrophage RAW264.7 세포에 ASC를 처리한 후 이차원 전기영동법을 이용하여, 단백질의 발현 변화 및 양상을 규명하고 단백질 지도를 확립 하였으며, RAW264.7 세포를 이용한 면역세포 모델에서 ASC의 항염증 작용을 중심으로 생리활성 조절기능을 확인 할 수 있었다. 향후 분자 기능 조절 연구와 전 임상 연구를 통해 ASC의 생리활성 조절 기능을 규명한다면 ASC는 항염증 및 항암활성을 갖는 약물로 개발될 것으로 기대된다.

본 연구는 핵이 제거된 소난자 내에 소의 태아 섬유아세포를 이식한 후 난자의 발달능력을 조사하였다. 소 태아 섬유아세포는 45일된 응성 태아로부터 분리한후 MitoTracker로 염색하고 세포 주기를 동기화시키지 않은 세포를 핵이 제거된 소난자의 위란강 내에 이식하였다. 섬유아세포와 소난자의 복합체는 전기 자극을 주어 융합시키는 방법을 이용하였으며, 융합된 난자는 Calcium ionophore와 6-DMAP를 이용하여 난활성을 유도시킨 다음 CR1aa 에서 배양하였다. 핵치환 된 난자의 핵은 핵질 응축과정, 팽대과정, 전핵 형성과정이 일어났으며, 이러한 과정에서 재구성된 난자는 분열 과정을 거쳐 18∼26시간 사이에는 2-세포기로 발달하였다. 섬유아세포의 미토콘드리아는 핵치환시 난자 내로 이전되었는데 이것들은 난자 내에서 빠르게 사라지는 것으로 관찰되었으며, 핵융합 후 8시간째에는 섬유아세포의 미토콘드리아가 전혀 관찰되지 않았다. 2-세포기로 분열된 난자의 21% 가 이식 가능한 단계인 배반포 단계까지 발달하였다. 이러한 결과는 소의 태아 섬유아세포가 탈핵된 소난자 내에서 성공적으로 재분화과정을 거치며, 이렇게 재조합 된 수정란은 배반포 단계까지 발달이 가능하다는 것을 보여주고 있다.

③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
