An Antifungal Subatance, 2,4-Diacetylphloroglucinol Produced from Antagonistic Bacterium Pseudo-monas fluorescens 2112 Against Phytophthora capsici (Phytophthora capsici를 길항하는 Pseudononas fluorescens 2112가 생산하는 항진균 항생물질 2,4-diacetylphloroglucinol)

  • An antifungal substance was purified from culture broth of Pseudomonas flulorescens 2112 that showed a broad-spectrum antagonistic activity against various phytopathogenic fungi including capsici. The substance was identified as 2,4-diacetylphloro-glucinol basd on NMR analysis. The 2,4-diacetylphloroglcinol showed antibiotic activity in broad acidic range from pH 1.0 to pH 9.0. About 83% of initial activity was remained after incubation for 30min ar $60^{\circ}C$, however, the activity was dropped up to 50% after 30 min incubation in $80^{\circ}C$. When the nucleotides of P. capsici treated with 2,4-diacetylphloroglucinol were labeled with[$^{3}$ H]-Adenin, the newly synthesized and radioactive-labeled RNA was significantly reduced than those of untreated P. capsici. indicating that the 2,4-diacetylphloroglucinol inhibits RNA synthesis.

  • 포토닉스 기술에 기반한 테라헤르츠 발생/검출 소자 기술은 광대역/저잡음 테라헤르츠 시스템 기술의 기반이 된다. 이것은 주파수 대역의 특성으로 인해 소자의 설계와 제작에 있어서 반도체, 광소자 관련 기술에서 RF 회로 및 안테나에 이르는 광범위한 분야의 기술이 통합적으로 적용되어야 하는 흥미로운 분야인 동시에, 향후의 기술발전에 따라 새로운 주파수 대역이 개척됨으로써 많은 응용기술이 파생될 수 있는 중요한 분야이다. 본고는 현재 본 센터에서 개발 중인 소자들을 중심으로 포토닉스 기반 테라헤르츠 기술 전반의 현황을 요약하고, 향후의 발전방향을 예측해 보고자 한다.

Quality Changes of Salted Chinese Cabbage by Packaging Methods during Storage (포장방법에 따른 절임배추의 저장중 품질변화)

  • The effects of packaging methods on the storage properities of salted Chinese cabbage were examinined during 3-week storage. The weights of salted Chinese cabbage decreased to 69-77% by water loss and salinities decreased from 2.6% to 2.0%, respectively. The pH decreased from 6.4 to 6.2 for 1- week storage in all cases, but the pH increased to 6.6 in non packaging(Non-P), maintained at 6.2 in PVC container packaging(PVC), and decreased continuously to 5.5 and 5.7 in LDPE packaging (LDPE) and HDPE packaging(HDPE), repectively for 3-week storage. Reducing sugars increased slightly for 1-week storage for the relative concentration effect by water loss, but decreased to 1.8% for 3-week storage because of the microbial fermentation. Cutting force increased in LDPE for the continuous osmosis phenomena but decreased in Non-P for the putrefaction. As the results, LDPE was found to be the most effective packaging method.

  • Si-O-C(-H) thin film with a tow dielectric constant were deposited on a P-type Si(100) substrate by an inductively coupled plasma chemical vapor deposition (ICPCVD). Bis-trimethylsilymethane (BTMSM, H$_{9}$C$_3$-Si-CH$_2$-Si-C$_3$H$_{9}$) and oxygen gas were used as Precursor. Hybrid type Si-O-C(-H) thin films with organic material have been generated many voids after annealing. Consequently, the Si-O-C(-H) films can be made a low dielectric material by the effect of void. The surface characterization of Si-O-C(-H) thin films were performed by SEM(scanning electron microscope). The characteristic analysis of Si-O-C(-H) thin films were performed by X-ray photoelectron spectroscopy (XPS).

  • The effects of some soil conditions on the degradation rate and decomposing pattern of parathion were investigated and the obtained results are summarized as follows: Parathion degraded more rapidly in flooded soils than in non-flooded, in wet soils than in dry soils under non-flooded soils. The degradation rates in paddy and upland soils increased at high temperature than low temperature, higher pesticide concentration than low concentration and higher soil pH level. Parathion in paddy and upland soils was more persistent under soil sterilization than under non-sterilization and degraded rapidly in glucose application. Parathion was more persistent in upland soils than paddy soils under several factors described above. The metabolites identified from the paddy and upland soils by TLC include para-oxon (Rf 0.5), aminoparathion(Rf 0.27), p-nitrophenol(Rf 0.2), p-aminophenol(Rf 0.15). Soil enzyme, acid phosphatase activities decreased more at flooded soils than non-flooded, higher pesticide concentration than low concentration and higher soil pH level and the activity in glucose application was increased. Soil enzymes, urease and dehydrogenase activity decreased more at higher pesticide concentration than low concentration. Comparing with soil enzyme activity in paddy and upland soil, the former was higher than the latter.

Impacts of Temperature, Salinity and Irradiance on the Growth of Ten Harmful Algal Bloom-forming Microalgae Isolated in Korean Coastal Waters (한국연안에서 분리한 적조형성 미세조류 10종의 성장에 미치는 온도, 염분, 광도의 영향)

  • In order to understand growth characteristics of ten major species of microalgae responsible for frequent harmful algal blooms in Korean coastal waters, the growth rates of the isolates were examined in relation with the impacts of water temperature, salinity and irradiance. In addition, their bloom events since 1990 as well as monthly abundance of vegetative cells were analyzed. Heterocapsa triquetra, Eutreptiella gymnastica and Alexandrium tamarense were considered as relatively mid temperature adapted species in that growth rates were comparatively high at low water temperatures of $10{\sim}16^{\circ}C$ and drastically decreased at above $22^{\circ}C$. Prorocentrum micans and Pyramimonas sp. were categorized as relatively high temperature adapted species by showing comparatively better growths at high water temperatures above $25^{\circ}C$. Akashiwo sanguinea, Heterosigma akashiwo, Prorocentrum minimum and Scrippsiella trochoidea were eurythermal species with relative high growth rates in a broad ranges of water temperature, $16{\sim}25^{\circ}C$ were slightly halophobic, showing better growths at low salinities of $10{\sim}30$ psu than at above 35 psu. H. akashiwo, P. minimum and H. triquetra were euryhaline species with remarkable growths in a broad ranges of salinity, 15-40 psu. Frequent algal blooms by these three species at extremely low salinities below 25 psu after rainfall were attributed to their euryhaline and slightly halophobic physiological characteristics. Growth rates of H. akashiwo, P. minimum and Pyraminonas sp. increased with the increase of irradiance within the experimental ranges of $2{\sim}150\;{\mu}E{\cdot}m^{-2}{\cdot}s^{-1}$. However, A. sanguinea, A. tamarense and H. triquetra showed better growths at comparatively low irradiance of $50{\sim}100\;{\mu}E{\cdot}m^{-2}{\cdot}s^{-1}$ and drastic decreases in growth rates above $150\;{\mu}E{\cdot}m^{-2}{\cdot}s^{-1}$ of irradiance. Overall, relatively high temperature adapted species make blooms frequently in high water temperature season with strong natural irradiance, and relatively low temperature adapted species grow better at low water temperature with relatively weak natural irradiance.

  • 저 주파수에서 인덕턴스 단위(H, Henry)는 전기용량의 단위(F, Farad)로부터 Maxwell bridge 또는 LC를 이용한 resonance 방법으로써 유도가 된다. 이중 Maxwell bridge 방법은 국가의 단위표준을 유지하는 연구소 또는 기관에서 가장 많이 사용되는 "product arm" 형의 bridge로 잘 알려져 있다. 개발된 bridge는 ratio-transformer를 전압공급 쪽(source)과 신호검출 쪽(detector)에 사용하여 종래의 Maxwell bridge 평형조절 시 main balance와 Wagner balance의 2개 과정을 진행하던 것과는 달리 main balance, source balance, detector balance의 과정을 진행함으로서 기존 bridge의 불안정한 ground admittance들의 영향에 의해 나타나는 불확도를 완전히 제거하였다. 한편 개발된 bridge는 10 mH, 100 mH를 10 nF의 전기용량표준기와 $1\;k{\Omega}$, $10\;k{\Omega}$의 저항표준기로부터 유도 되도록 제작되었으며. 10 mH의 경우 $400\;Hz{\sim}5\;kHz$의 주파수 영역에서 약 $2.2{\times}10^{-6}$(k=1)의 불확도로 인덕턴스 단위를 유도할 수가 있게 되었다.

HPP, High-Pressure Process (저산성 식품에 유용한 고압처리 공정)

  • 고압처리로 멸균된(high-pressure- sterilized) 저산성 식품은 아직 본격적으로 시장에 나오지는 않고 있다. 그러나 고압처리로 살균시켜(high-pressure-pasteurized) 제품의 부가가치를 높인 상품들은 일부 시판되고 있다. 미국시장에는 구아카몰(guacamole),굴 등이 있고, 일본과 유럽에는 잼, 젤리, 생선제품, 육제품, 슬라이스 햄, 샐러드 드레싱, 쌀떡, 주스, 요구르트 등이 판매되고 있다. 기술이 발전되어 처리비용이 떨어짐에따라 우유나 오렌지 주스 같은 저부가가치의 제품(high-volume commodity products)에도 고압처리 기술이 적용되는 예를 보게 될 것이다.미국 FDA의 "열처리된 저산성식품은 용접밀봉된 용기(캔, hermetically sealed container)에 포장되어야 한다." 는 규정(Title 21, Part 113 of the Code Federal Regulations)은 고압처리 식품을 염두에 두고 만들어진 규정은 아니지만, 저산성식품의 고압처리 공정(high-pressure processing)은 강력한 살균력을 제공하고 있다. 따라서 위의 FDA 규정(21 CFR 113)의 모든 조항은 고압처리 저산성 식품이 상업적인 생산에 들어가기 전에 시행되어야만 하는 것이다. 그 규정의 주요 조항중의 하나는 고압처리 공정에 대한 전문적인 지식을 가진 전문가나 위원회 등에 의하여 세부적인 고압처리공정 절차를 확립하는 것이다. 공정의 전문가들은 안전성을 증명하기 위해서 적합한 과학적인 방법을 사용하여 고압처리 공정을 확립해야만 한다. 저산성 식품에 대한 고압처리 공정은 주의깊게 설정된 공정조건하에서 재현성 있게 안전한 식품을 생산할 수 있어야 한다. 살균에 대한 안전성과 재현성이 입증되면 고압에 의한 살균처리공정 설계는 일반적인 대량생산 공정으로 활용될 수 있게 된다. PP(고압처리공정)의 유효성이 보다 확실하게 입증되고 널리 활용되려면, 공정의 살균력과 일관성(uniformity)을 증명하는 미생물적, 물리적, 화학적, 공학적인 여러 분야가 통합된 검증방법이 필요하다. 전통적인 열처리 공정은 스팀열의 가열시간으로 설명이 충분하다. 물론 HPP살균에서도 시간과 온도는 주요 변수이지만, 압력에 의한 미생물살균도 고려되어야만 한다. 또한 점도, 밀도, 구성성분, pH, 수분활성도 같은 해당제품의 특성도 제품의 고압처리 살균공정, 특히 미생물의 사멸에 영향을 미칠 수도 있다.

Quality Characteristics of Low Salt Kalopanax pictus Shoot Jangajji Using Soybean Sauce (저염 개두릅 장아찌의 품질 특성)

  • In order to develop new Kalopanax pictus shoot Jangajji (KJ) with low salt concentration, physicochemical changes of KJ soaked at different concentration of soybean sauce (10~50%) were investigated for 50 days. The pH of KJ increased significantly with increasing concentration of soybean sauce in soaking solution (P<0.05). Titratable acidity and NaCl concentration of all KJ increased during storage for 50 days at $10^{\circ}C$. The total bacterial count slowly increased during early storage time and reached its highest level (4.74 log CFU/g) after storage for 30 days. KJ-20 (KJ added with 20% soy sauce) showed the highest scores for taste, flavor, texture, and overall acceptability. The total polyphenol and flavonoid contents of KJ extract increased with increasing concentration of soybean sauce. DPPH and ABTS radical scavenging activities of KJ extract were 27.22% and 38.37% (control), 40.74% and 43.65% (KJ-10), 41.79% and 50.37% (KJ-20), 43.09% and 52.60% (KJ-30), 48.62% and 53.45% (KJ-40), and 50.85% and 60.79% (KJ-50), respectively.

A comparison of the Effects of Intravenous Fluid Warming and Skin Surface Warming on Peri-operative Body Temperature and Acid Base Balance of Elderly Patients with Abdominal Surgery (수액가온요법과 피부가온요법이 개복술 환자의 저체온 예방에 미치는 효과 비교)

  • Purpose: The purpose of this study was to compare the effects of intravenous fluid warming and skin surface warming on peri-operative body temperature and acid base balance of abdominal surgical patients under general anesthesia. Method: Data collection was performed from January 4th, to May 31, 2004. The intravenous fluid warming(IFW) group(30 elderly patients) was warmed through an IV line by an Animec set to $37^{\circ}C$. The skin surface warming(SSW) group(30 elderly patients) was warmed by a circulating-water blanket set to $38^{\circ}C$ under the back and a 60W heating lamp 40 cm above the chest. The warming continued from induction of general anesthesia to two hours after completion of surgery. Collected data was analyzed using Repeated Measures ANOVA, and Bonferroni methods. Results: SSW was more effective than IFW in preventing hypothermia(p= .043), preventing a decrease of $HCO_3{^-}$(p= .000) and preventing base excess(p= .000) respectively. However, there was no difference in pH between the SSW and IFW(p= .401) groups. Conclusion: We conclude that skin surface warming is more effective in preventing hypothermia, and $HCO_3{^-}$ and base excess during general anesthesia, and returning to normal body temperature after surgery than intravenous fluid warming; however, skin surface warming wasn't able to sustain a normal body temperature in elderly patients undergoing abdominal surgery under general anesthesia.
