α-Glucosidase Inhibitory Effects for Solvent Fractions from Methanol Extracts of Sargassum fulvellum and Its Antioxidant and Alcohol-Metabolizing Activities (참모자반 메탄올 추출 분획물의 항산화 및 숙취해소능과 α-glucosidase 활성저해효과)

  • We investigated the physiological activity and solvent-partitioned fractions of methanol extracts from the green seaweed Sargassum fulvellum. The methanol extract from S. fulvellum was sequentially fractionated with n-hexane (SFMH), methanol (SFMM), buthanol (SFMB), and water (SFMA). We investigated the antioxidant activities of solvent fractions from S. fulvellum by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging capacity and an SOD activity assay. DPPH radical scavenging capacity of SFMM was 79.5% at 10 mg/ml. SOD activity of SFMM was 79.9% at 10 mg/ml. Nitrite scavenging activities of solvent fractions from S. fulvellum were investigated under different pH conditions and showed the most remarkable effect at pH 1.2. In particular, the activity of SFMB was higher than the other fractions. ADH activity and ALDH activity of SFMM were 177.0% and 167.4% at 10 mg/ml, respectively. ${\alpha}$-Glucosidase inhibitory activity of SFMH increased in a dose-dependent manner and was about 94.1% at 2 mg/ml. Elastase inhibitory activity was 93.2% at 2 mg/ml. These results revealed that S. fulvellum extracts have strong antioxidant and alcohol dehydrogenase activities and ${\alpha}$-glucosidase inhibitory activity, suggesting that S. fulvellum extracts have potential as a source of natural products for health and beauty.

Antioxidative and Antiaging Effects of Persicaria hydropiper L. Extracts (여뀌 추출물의 항산화 및 항노화에 관한 연구)

  • In this study, we investigated the antioxidative activity and inhibitory effects on elastase and tyrosinase of Persicaria hydropiper L. extracts. The free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activities ($FSC_{50}$) of ethyl acetate fractions of Persicaria hydropiper L. was $5.23\;{\mu}g/mL$. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of some Persicaria hydropiper L. extracts on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system were investigated using the luminol - dependent chemiluminescence assay. The ROS scavenging activities ($OSC_{50}$) of ethyl acetate fractions of Persicaria hydropiper L. was $0.40\;{\mu}g/mL$. The protective effects of extract / fractions of Persicaria hydropiper L. on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The Persicaria hydropiper L. extracts suppressed photohemolysis in a concentration dependent manner ($1\;{\sim}\;10\;{\mu}g/mL$). Inhibitory effects ($IC_{50}$) on tyrosinase of aglycone fraction of Persicaria hydropiper L. extracts was $8.90\;{\mu}g/mL$. Inhibitory effects ($IC_{50}$) on elastase of aglycone fraction of Persicaria hydropiper L. extracts was $2.37\;{\mu}g/mL$. These results indicate that extract / fractions of Persicaria hydropiper L. can function as antioxidants in biological systems, particularly skin exposed to UV radiation by anti-oxidative activity and protect cellular membranes against ROS. Persicaria hydropiper L. extract / fractions could be used as a new cosmeceutical for whitening and anti-wrinkle products.

Chemical Suppression of Gravitropic Bending Response in Flower Stalks of Snapdragon (Antirrhinum majus L.) (몇가지 생리활성 저해제가 금어초 절화의 굴지성 반응에 미치는 효과)

  • Numbers of chemical agents which have been shown to inhibit either auxin signal transduction pathway or ethylene formation in plant cells were applied to cut flower stems of snapdragon (Antirrhinum majus L.) and their effects on the postharvest gravitropic response were studied. The chemical treatments were done by submerging either the stem base or the top part of cut flower, which involves the gravistimulus-sensitive region, for 1 h at $25^{\circ}C$. When the chemicals were supplied from the cut stem base, the gravitropic upward bending of flower stalks kept horizontally after the treatments with 20 mM CDTA or 10 mM $CoCl_2$ was comparable to that of the untreated control, but o-vanadate showed a certain degree of effectiveness for suppressing the bending response. In contrast, the direct application of those agents to the gravitropically sensitive region of cut flowers in the presence of 0.01% Triton X-100 resulted in a substantial reduction of the gravitropic response. In the case of 20 mM $CoCl_2$ treatment, almost total elimination of gravitropism without any significant deterioration of flower quality was observed. The results indicate the possibility of preparation of a protocol involving $CoCl_2$ and a proper surfactant for commercial use to suppress the gravitropic response of cut flowers during postharvest storage and transportation.

The Optimal Culture Conditions and Antifungal Activity of Culture Extract from Oudemansiella mucida (끈적긴뿌리버섯(Oudemansiella mucida)의 최적배양조건 및 배양 추출액의 항균작용에 관한 연구)

  • Oudemansiella mucida, an edible and medicinal mushrooms belonging to Tricholomataceae of Basidiomycota, has been known to produce antifungal substances to inhibit the mycelial growth and spore germination of the plant pathogenic fungi. To produce good amount of antifungal substances from culture media, the optimal culture conditions of O. mucida were investigated. The most favorable conditions for the mycelial growth were $25^{\circ}C$ and pH 5 in potato dextrose agar. The most favorable carbon and nitrogen sources promoting mycelial growth were maltose and calcium nitrate, respectively. The optimum C/N ratio was about 20 : 1 in case that 3% glucose was supplemented to the basal medium as a carbon source. The optimal mycelial growth of O. mucida was found in the Hennerberg medium. The crude extract from submerged culture of potato dextrose broth exhibited inhibition of mycelial growth of Colletotrichum acutatum, Botrytis cinerea and Pyricularia oryzae but, fungicidal activity is not good enough to compared with commercially available fungicides tested. Therefore, the antifungal substances extracted from submerged culture of O. mucida might have a potential to be used for biocontrol agent of fungal diseases of plants.

pH-dependence in the inhibitory effects of Zn2+ and Ni2+ on tolaasin-induced hemolytic activity (Zn2+와 Ni2+에 의한 톨라신 용혈활성 저해효과의 pH 의존성)

  • Tolaasin secreted by Pseudomonas tolaasii is a peptide toxin and causes brown blotch disease on the cultivated mushrooms by collapsing cellular and fruiting body structure. Toxicity of tolaasin was evaluated by measuring hemolytic activity because tolaasin molecules form membrane pores on the red blood cells and destroy cell membrane structure. In the previous studies, we found that tolaasin cytotoxicity was suppressed by $Zn^{2+}$ and $Ni^{2+}$. $Ni^{2+}$ inhibited the tolaasin-induced hemolysis in a dose-dependent manner and its $K_i$ value was 1.8 mM. The hemolytic activity was completely inhibited at the concentration higher than 10 mM. The inhibitory effect of $Zn^{2+}$ on tolaasin-induced hemolysis was increased in alkaline pH, while that of $Ni^{2+}$was not much dependent on pH. When the pH of buffer solution was increased from pH 7 to pH 9, the time for 50% hemolysis ($T_{50}$) was increased greatly by $100{\mu}M$ $Zn^{2+}$; however, it was slightly increased by 1 mM $Ni^{2+}$ at all pH values. When the synergistic effect of $Zn^{2+}$ and $Ni^{2+}$ on tolaasin-induced hemolysis was measured, it was not dependent on the pH of buffer solution. Molecular elucidation of the difference in pH-dependence of these two metal ions may contribute to understand the mechanism of tolaasin pore formation and cytotoxicity.

Anti-Inflammatory and Whitening Effect of the Lyophilized Powder of Oriental Plant Extracts Fermented with Streptococcus thermophilus (Streptococcus thermophilus로 발효한 한약재 발효분말의 항염증 및 미백 효과)

  • To identify new anti-inflammatory and whitening material, this study investigated the whitening and anti-inflammatory effects of the lyophilized powder from 6 oriental plant extracts (OPE; Citrus junos Tanaka, Mori cortex Radicis, Schisandra chinensis Baillon, Coix lachrymajobi var. mayuen, Angelica gigas NAKAI, and Sophora japonica L.) fermented with Streptococcus thermophilus by assessment of cytotoxicity on human dermal fibroblast, inhibitory effect of nitric oxide (NO) prodction, tyrosinase activity and melanin formation. The OPE was fermented with Streptococcus thermophilus at $37^{\circ}C$ for 2 days and the lyophilized powder was manufactured by freezing-dryer. OPE didn't show cytotoxicity at concentration of $500{\mu}g/mL$ using a cytopathic effect reduction method. OPE also exhibited inhibitory effect on nitric oxide (NO) prodction by Griess reagent system. Furthermore, OPE showed inhibitory effect on tyrosinase activity with dose dependent manner, and exhibited significant inhibition of melanin formation by measurement of melanin from culture media (p < 0.05). From these results, 6 OPE extracts showed anti-inflammatory and whitening effect and may be used as an active ingredient for cosmetics.

Antiproliferative Effect of Mistletoe Extract Added Kimchi in Human Lung Carcinoma A549 Cells (겨우살이 물추출물 첨가 김치의 A549 인체 폐암 세포 증식저해 효과)

  • The purpose of this study aimed at examining the antiproliferative effect of kimchi (kimchi B) adding mistletoe extract known as an anticancer function to improve the functions of kimchi. The study investigated the antiproliferative effect through hemocytometer counts and MTT assay, apoptosis induction through DAPI staining, and mRNA expression through RT-PCR using human lung carcinoma A549 cells. The standardized kimchi (Kimchi A) was used as a control group. As a result of hemocytometer counts and the MTT assay, it was found that kimchi samples inhibited the growth of A549 cells in a concentration-dependent manner. Kimchi B induced apoptosis in A549 cells through DAPI staining. The apoptosis induced by kimchi B was associated with the increase in the expression of pro-apoptotic Bax and with the decrease in the expression of anti-apoptotic Bcl-2 and Bcl-xL. Also, kimchi B influenced the increase in the expression of p21 mRNA, but did not have the effect on the expression of p53 mRNA. In conclusion, the antiproliferative effect of kimchi B was due to apoptosis induced by increasing Bax and decreasing Bcl-2, and increasing p21. The findings will be utilized to develop kimchi with the improved function for the patients having cancer.

Biological Activity of Ethanol Extracts from Amelanchier asiatica Fruits (채진목(Amelanchier asiatica) 열매 에탄올 추출물의 생리활성)

  • Amelanchier asiatica fruits have been used as a traditional medical food. This research was investigated to assess angiotensin converting enzyme, xanthine oxidase (XOase) and elastase inhibitory activity and antioxidant activities. The content of total phenolic compounds in A. asiatica fruits extracts was 17.6mg/mL. In extracts, the electron donating ability by 1,1-diphenyl-2-picrylhydrazyl free radical scavenging test of A. asiatica fruits extracts was 90.18% at $200{\mu}g/mL$. The 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid radical decolorization of A. asiatica fruits extracts was 98.81% at $200{\mu}g/mL$. The inhibition rate of the antioxidant protection factor was 1.03, and thiobarbituric acid reactive substance was 73.27% at $200{\mu}g/mL$. The XOase inhibition activity of A. asiatica fruits extracts of showed to be 13.19% at $200{\mu}g/mL$. The angiotensin converting enzyme activity was significantly inhibited by A. asiatica fruits extracts as 82.52% inhibitory rate at $200{\mu}g/mL$. Elastase inhibitory activity in the A. asiatica fruits extracts (41.48% at $200{\mu}g/mL$) was higher than vitamin C (12.8% at $200{\mu}g/mL$). These results suggests that A. asiatica fruits extracts have the greatest property as a functional food and functional cosmetic source.

Antioxidant and skin whitening effects of Inonotus obliquus methanol extract (차가버섯 메탄올 추출물의 항산화 및 미백효과)

  • This study was initiated to investigate the skin whitening activities of methanol extracts from fruiting bodies of I. obliquus. The total polyphenols and flavonoids contents of I. obliquus methanol extracts were 31.85 mg/g and 28.33 mg/g, respectively. The methanol extract of the mushroom treated on B16/F10 melanoma and NIH3T3 cell lines did not show cytotoxic activity. 2,2-diphenyl-1-picrylhydrazyl(DPPH) free radical scavenging activity and chelating activity on ferrous ions of I. obliquus methanol extract were lower than those of positive control, tocopherol and BHT. The tyrosinase and L-DOPA inhibitory activities of the extract were lower than those of positive control, kojic acid and ascorbic acid. The tyrosinase and melanin synthesis inhibitory activities of the melanoma cells treated with the extract were comparable with positive control, arbutin. The experimental results suggested that methanol extract of I. obliquus contained inhibitory activities of tyrosinase and melanin synthesis in the B16/F10 melanoma cells by dose dependent manner. High ultra-violet absorption spectra in the range of 280-350 nm showed that I. obliquus extract could protect skin from UV radiation damage. Therefore, fruiting bodies of I. obliquus can be used for developing skin whitening, anti-UV and skin care agents.

Supplementary Effects of Lentinus edodes with Different Harvest Period and Part on Neurotransmitters and Lipid Peroxide Levels in the Brain of Diabetic Mice (채취 시기 및 부위가 다른 표고버섯의 급여가 당뇨 마우스 뇌조직의 신경전달물질 및 지질과산화물 수준에 미치는 영향)

  • This study was designed to investigate the supplementary effects of Lentinus edodes which were harvested at different time period and part on acetylcholine content and its related enzyme activities in the brain of diabetic mouse model (KK mouse). We fed mice with standard diet (Control diet; CON) or 4 different kinds of experimental diets (DGC: on time harvested, cap of Dong Go; DGS: on time harvested, stipe of Dong Go; HSC: late harvested, cap of Hyang Sin: HSS: late harvested, stipe of Hyang Sin) to KK mouse for 8 weeks. Neurotransmitter such as acetylcholine contents, acetylcholinesterase activities, monoamine oxidase-B ac-tivities and lipid peroxide contents in the brain were measured. The results showed that acetylcholine content was significantly higher in DGC and HSC groups than CON group. The activities of acetylcholinesterase and monoamine oxidase-B enzyme were significantly inhibited in the brain of DGC and HSC groups compared with CON group. Lipid peroxide content was lower in DGC group than CON group. These results suggested that the cap of Lentinus edodes which were harvested on time and late time contain increased acetylcholine content and decreased acetylcholinesterase activities, monoamine oxidase-B activities and lipid peroxide contents. Thus the cap of Lentinus edodes which were harvested at different time periods may play an effective role in enhancing cognitive function.
