Biological Activities of Extracts from Gamma-irradiated Aralia elata Cortex (감마선 조사한 총목피(Aralia elata Cortex) 추출물의 생리활성)

  • Gamma irradiated-treatment of natural medicinal plants can be used to improve extraction transference number and for qualitative improvement of color when applied to functional material exploration. This study investigated the biological activities of Aralia elata cortex extracts upon gamma irradiation. In addition, different physical techniques [photostimulated luminescence (PSL) and thermoluminescence (TL)] were used for irradiation identification of Aralia elata cortex. In PSL analysis, non-irradiated (0 kGy) sample showed a negative result of 400 photon counts (PCs), whereas irradiated (5, 10, and 30 kGy) samples showed positive results of 90,100.00, 312,614.33, and 321,661.67 PCs, respectively. In the TL method, growth curve showed very unusual behaviors around $200^{\circ}C$ upon natural-irradiation of the non-irradiated (0 kGy) sample and around $150{\sim}250^{\circ}C$ for the irradiated (5, 10, and 30 kGy) samples. The TL ratio was 0.1 in non-irradiated samples at 0.011, whereas the values of irradiated samples (5, 10, and 30 kGy) were 0.1 at 1.105, 1.009, and 2.206, respectively. For phenolics of gamma-irradiated Aralia elata cortex, water and 50% ethanol extracts had the highest amounts, $17.30{\pm}0.40mg/g$ and $18.87{\pm}0.46mg/g$ at 10 kGy irradiation, respectively. The inhibitory activities of angiotensin-converting enzyme and xanthin oxidase were higher in both irradiated water and 50% ethanol extracts than in non-irradiated ones. For pancreatin ${\alpha}$-amylase and ${\alpha}$-glucosidase inhibitory activities, water and 50% ethanol extracts containing $200{\mu}g/mL$ of phenolics showed high inhibitory activities of 60~100% at all irradiation doses (0~30 kGy). This result confirmed that Aralia elata cortex extracts have greater anti-diabetic effects than acabose as a diabetic remedy. Gamma-irradiated Aralia elata cortex extracts are useful as a functional material with anti-diabetic effects. Thus, Aralia elata cortex extracts can be used as a functional material with various biological activities, and gamma-irradiation can be used to amplify biological activities in plants.

Characteristics of Angiotensin Converting Enzyme Inhibitory Peptides from Aroase AP10 Hydrolysate of Octopus (Aroase AP10에 의한 문어 가수분해물의 Angiotensin Converting Enzyme 저해 Peptide의 특성)

  • The peptides from Aroase AP10 enzymatic hydrolysates of octopus proteins were isolated and tested for inhibitory activity against angiotensin converting enzyme (ACE). The Aroase AP10 hydrolysates were filtered through PM-10 membrane (M.W. cut-off 10,000) to obtain the peptides fractions with ACE inhibition activity. These fractions were applied to a Biogel P-2 column. Three active fractions (A, B, and C) were collected and applied to a SuperQ-Toyopearl 650S column chromatography, leading to the isolation of four active fractions (A-1, A-2, B-1, and C-1). Among the active fractions, C-1 had the highest ACE inhibitory activity ($IC_{50}=3.10{\mu}g$). The main composition of its amino acids is arginine, lysine, histidine and leucine, which cover about 60% of the total amino acids.

Inhibitory Effect of Nisin upon Kimchi Fermentation (김치발효에 대한 Nisin의 저해효과)

  • To examine the inhibitory effect of nisin on Kimchi, comparison of the fermentation pattern was studied between the Kimchi added nisin (100 WIg) and the control Kimchi at 15cC. The Kimchi added nisin was showed pH 4.03 at 7 days fermentation, on the other hand the control Kimchi showing pH 4.04 at 5 days. And total acidity (lactic acid%) showed 0.47,0.69,0.88 at 5,9, 14 days fermentation compared with the control Kimchi showing 0.57,0.93, 1.13 respectively. Maximum growth of lactic acid bacteria on the control Kimchi indicated 1.8, 1.6 x 108 CFU/ml at 7 days but on presence of nisin indicated 9.5,6.4 x 107 CFU/ml at 5 days. In conclusion, nisin showed inhibitory action to Kimchi fermentation from pH, total activity and lactic acid bacteria count results.

The Properties of Extracellular Guanine Deaminase from Pseudomonas synxantha A3 (Pseudomonas synxantha A3가 생산하는 세포외 Guanine Deaminase의 성질)

  • Some properties of extracellular guanine deaminase produced by Pseudomonas synxantha A3 were studied. The enzyme was stable at pH 6.5-7.5 and generally stable when it was incubated at 4$0^{\circ}C$ for 10 minutes but inactivated gradually above 4$0^{\circ}C$. When the enzyme in 0.2M potassium phosphate (pH 8.0) was stored at room temperature, it was stable for thirty days. Alcohols and acetone were not effective for the eyzyme stability. The optimum pH and temperature for the enzyme activity were around pH 7.0-8.0 and 5$0^{\circ}C$, respectively. The enzyme was inhibited by 1mM of Hg$^{++}$, Ag$^+$ and Li$^+$ and by 0.1mM of Ag$^+$ with about 50% loss of activity. The enzyme inhibited by Li$^+$ was reactivated by EDTA. 1 mM of pentachlorophenol and p-CMB inactivated the enzyme with 50% and 40% loss of activity, respectively. The enzyme inactivated by p-CMB was reactivated by glutathione.

  • Quantum chemical quantities, enthalpy of formation(${\Delta}H_f$), HOMO and LUMO energy, and dipole moment(${\mu}_D$) were acquired by AM1, PM3, and ZINDO/1 methods for polyamines and imidazole derivatives. The investigation of the inhibitory activity on some Ni(II) complexes with polyamines and imidazole derivatives is performed by ZINDO/1 calculations. It was found that experimental inhibitory activity(IA) appeared when the value of net charge and enthalpy of formation were over 0.03 and -300 eV, respectively for Ni(II) complexes. These results showed that the Ni(II) complexes have exception on the following very unstable compounds: square pyramidal [Ni(dpt)(tn)])]$^{2+}$(dpt=3,3'-diaminodipropylamine)(tn=1,3-diaminopropane) and distorted tetrahedral [Ni(N-PropIm))$_2$(NCS))$_2$](N-PropIm=N-Propylimidazole).

Microbial Decontamination of Vegetables and Spices Using Cold Plasma Treatments (비열 플라즈마 처리를 이용한 채소와 향신료의 미생물 저감화)

  • Effects of cold plasma treatment (CPT) against Salmonella Typhimurium inoculated on cabbage and lettuce, naturally occurring bacteria in black pepper powder and red pepper powder, and Bacillus cereus inoculated onto red pepper powder were investigated. The numbers of S. Typhimurium on cabbage and lettuce were reduced by $1.5{\pm}0.2CFU/cm^2$ (900W, 5 min) and $1.1{\pm}0.1$ log $CFU/cm^2$ (900W, 10 min), respectively. The numbers of naturally occurring aerobic bacteria in both black pepper powder and red pepper powder were reduced by $2.3{\pm}0.3$ and $0.6{\pm}0.2$ log CFU/g, respectively. The numbers of B. cereus vegetative cells on red pepper powder were reduced by $1.5{\pm}0.1$ log CFU/g, but the numbers of spores remained unchanged. The inhibition of S. Typhimurium on cabbage was adequately described by Fermi's model and the Weibull model. The predicted optimum treatment power and time for S. Typhimurium inoculated onto cabbage were 746 W and 6.8 min, respectively. Our results indicate that CPT represents a useful method for microbial decontamination of vegetables and spices.

Effects of Hesperidine, Naringin and Their Aglycones on the In Vitro Activity of Phosphatidate Phosphohydrolase, and on the Proliferation and Growth in Cultured Human Hepatocytes HepG2 Cells (In Vitro 에서 Phosphatidate Phosphohydrolase 활성과 HepG2 세포증식에 미치는 Hesperidine, Naringin 및 그 Aglycone Flavonoid의 영향)

  • Effects of four citrus flavonoids, hesperidin, naringin and their aglycones on phosphatidate phosphohydrolase(PAP, EC 3.1.3.3) activity were examined using isolated rat microsomes as an enzyme source. In addition, these flavonoids were tested to see whether they exert any influence on the proliferation and growth in cultured human hepatocytes HepG2 cells. Flavonoids at concentration up to $10{-4}M$ had no significant effect on the number of cells and cell proliferation by MTT cell growth assay method, whereas aglycone flavonoids, hesperetin and narigenin, at concentration of $10{-3}M$ significantly inhibited cell proliferation. Hesperetin inhibited PAP activity in a dose-dependent manner starting at concentration of $10{-5}M$. Narigenin at concentration of $10{-2}M$ inhibited PAP activity markedly, while the other flavonoids did not show any significant effect. The present study, therefore, demonstrated that aglycone flavonoids exerted portent effects on PAP activity and on cell proliferation.

Inhibitory Effects of Grapefuit Seed Extract on Growth and Aflatoxin Production of Aspergillus parasiticus (Grapefruit 종자추출물을 이용한 Aspergillus parasiticus의 생육 및 Aflatoxin 생성억제 효과)

  • This study was conducted to determine the potential of grapefruit seed extract to support Aspergillus parasilicus growth and aflatoxin production. The grapefruit seed extract inhibited the growth and aflatoxin production of the fungi in the level of more than 4,000 ppm and 3,000 ppm in the medium, respectively. Grapefruit seed extract appears to block the conversion of acetate, averufin and versiconal acetate into aflatoxin in vitro experiments. The addition of grapefruit seed extract to the feeding experiment systems did not inhibit the incorporation of 14C-labeled versicolorin A, versicolorin A hemiacetal and sterigmatocystin into aflatoxin. In the electron microscopic examination the biocidal action of grapefruit seed extract was related to the disturbance of cell menbrane funtion, inhibiting cellular respiration.

Differential Inhibitory Effect of Artemisia Extract between Staphylococcus aureus and vaginal Lactobacillus spp (쑥 추출물의 포도상구균과 질 유산균에 대한 선택적 저해효과)

  • In this study, Artemisia mongolica fischer extract was examined for its possible differential inhibitory activity against pathogenic bacteria including Staphylococcus aureus and Lactobacillus spp. isolated from women's vagina. First, seven lactobacillus spp. were selected based on their in vitro high anti-staphylococcal activity. Various samples extracted using different concentrations of organic solvents (acetone, ethanol, methanol) were examined for optimal anti-staphylococal activity. When the Artemisia extract obtained using $100\%$ solvents was added to the cell suspension at $17\%$ (vol/vol), Lactobacillus sp. KLB 224 maintained its viability for 48 hr, whereas S. aureus was completely inactivated showing differential antimicrobial activity of the extract. Using scanning electron microscopy the effect of the extract on the cell morphology was observed: S. aureus showed markedly distorted cell morphology while Lactobacillus sp. KLB 224 appeared to remain intact.

Tyrosinase Reaction in AOT/Isooctane/Reverse Micelles (AOT/이소옥탄/역미셀계에서의 Tyrosinase 반응)

  • It is difficult to accurately evaluate the effect of lipophilic compounds in aqueous reaction system of enzymes because they are immiscible with water. To screen lipophilic inhibitors of tyrosinase which catalyzes the synthesis of melanin in vivo, an optically clear organic system composed of organic solvent, surfactant, and water, often called reverse micelles(RM), was introduced. Optimal RM to let tyrosinase act normally was composed of isooctane as an organic solvent and dioctyl sulfosuccinate(AOT) of 100 mM as a surfactant. When a molar ratio of water to surfactant was 15, tyrosinase(105.3 units) in RM showed a similar reactivity toward 3,4-dihydroxyphenylalanine(0.18 mM) as in the aqueous assay system. In the presence of cinnamic acid, the product formation of tyrosinase reaction was proportional to the reaction time. This indicates that the inhibitory effect of lipophilic compounds could be analyzed in RM.
