Purpose: The human chronic myelogenous leukemia cell line, K562, expresses the chimeric bcr-abl oncoprotein, whose deregulated protein tyrosine kinase activity antagonizes via DNA damaging agents. Previous experiments have shown that nanomolar concentrations of herbimycin A (HWA) coupled with X-irradiation have a synergistic effect in inducing apoptosis in the Ph-positive K562 leukemia cell line, but genistein, a PTK inhibitor, is non selective for the radiation-induced apoptosils on $p210^{bcr/abl}$ protected K562 cells. In these experiments, the cytoplasmic signal transduction pathways, the Induction on a number of transcription factors and the differential gene expression in this model were investigated. Materials and Methids: K562 cells in the exponential growth phase were used in this study. The cells were irradiated with 0.5-12 Gy, using a 6 Mev Linac (Clinac 1800, Varian, USA). Immediately after irradiation, the cells were treated with $0.25/muM$ of HMA and $25/muM$ of genistein, and the expressions and the activities of abl kinase, MAPK family, NF- kB, c-fos, c-myc, and thymidine kinase1 (TK1) were examined. The differential gene expressions induced by PTK inhibitors were also investigated. Results: The modulating effects of herbimycin A and genistein on the radiosensitivity of K562 cells were not related to the bcr-abl kinase activity. The signaling responses through the MAPK family of proteins, were not involved either in association with the radiation-induced apoptosis, which is accelerated by HMA, the expression of c-myc was increased. The combined treatment of genistein, with irradiation, enhanced NF- kB activity and the TK1 expression and activity. Conclusion: The effects of HMA and genistein on the radiosensitivity on the K562 cells were not related to the bcr-abl kinase activity in this study, another signaling pathway, besides the WAPK family responses to radiation to K562 cells, was found. Further evaluation using this model will provide valuable information for the optional radiosensitization or radioprotection.
The purpose of this study was to evaluate the effects of fluoride releasing orthodontic sealants, light-cured (Group L1&L2) and self-cured (Group S1&S2) $FluoroBond^{\circledR}$, on enamel microhardness under artificial carious solution in vitro.112 extracted human premolar teeth were collected for experiments and divided into seven groups. A Tukon microhardness tester equipped with a Knoop diamond indenter was employed to determine microhardness. Tukon 23 program converted the number of microhardness into KHN (Knoop hardness number). The results were as follows: 1. The microhardness of enamel depth of all groups were the least at the depth of $50{\mu}m$ and that of all groups except L2 group, the greatest at the depth of $200{\mu}m$, were the greatest at the depth of $300{\mu}m$. And as the enamel depth of all groups except L2 and S2 group increased, the microhardness value also increased. 2. There was a little preventive effect in enamel decalcification both light-and self-cured orthodontic sealant groups, but had no statistical significance between the groups(p>0.05). 3. Light-cured orthodontic sealant groups had a progressive inhibiting effect in enamel decalcification at the depth of $100{\mu}m,150{\mu}m,\;and\;200{\mu}m$ (p<0.05). 4. Self-cured orthodontic sealant groups had a progressive inhibiting effect in enamel decalcification at the depth of $150{\mu}m$ (p<0.05). 5. There was no difference of the anti-enamel demineralization effect between light- and self-cured orthodontic sealant groups (p>0.05).
Introduction: REM sleep which shows characteristic muscle atonia and increased resistance of upper respiratory track is known to be vulnerable to sleep apnea. Previous studies reported that REM sleep-dependent (or related) obstructive sleep apnea syndrome (REM-dependent OSA) could be one of sleep disordered breathing. The present study aimed to investigate clinical findings and polysomnographic variables of REM-dependent OSA. Methods: Fifty-six patients diagnosed with mild to moderate obstructive sleep apnea by overnight polysomnography (5$53.7{\pm}16.7$ years, 42 males). REM-dependent OSA was defined as AHI-REM/AHI-NREM ratio>2. We compared clinical and polysomnographic findings between REM-dependent OSA and No REM-dependent OSA patients. Results: Among 56 patients, 37.5% (n=21, average age of $52.3{\pm}19.7$ years, 14 males) met the REM-dependent OSA criteria. There were no significant differences in age, sex and body mass index between two groups. After controlling for age, sex, body mass index and periodic leg movements index, REM-dependent OSA patients showed significantly lower AHI, lower number of oxygen desaturation events and higher stage 2 sleep proportion compared to No REM-dependent OSA patients (p=0.010, p=0.006, p=0.031, respectively). After controlling for age, sex, body mass index and periodic legs movements index, AHI-REM was positively correlated with the number of oxygen desaturation events in REM-dependent OSA group (p=0.002). Conclusion: Current results suggested that 37.5% of patients with mild to moderate severity of obstructive sleep apnea could be classified into REM-dependent OSA. REM-dependent OSA was more common in mild severity of OSA, equally prevalent in both sexes and accompanied with sleep architecture changes, i.e. increased proportion of stage 2. In addition, apneic events during REM sleep in REM-dependent OSA were related to oxygen desaturation.
The purposes of this study were to evaluate the effects of acid rain on tree growth and on the mycorrhizal formation and the effects of mycorrhizae on the host tolerance to acid rain. Simulated acid rain was applied for five months to Pinus $rigida{\times}taeda$ seedlings in pots inoculated with Pisolithus tinctorius (Pt) and Suillus luteus (Sl). Mycelial inocula of Pt and Sl were either mixed with entire pot soil (Mix) or casted as a band (Band) after soil sterilization. Three pH levels of acid rain (pH 3.0, 4.5 and 6.4 adjusted by 3 : 1 mixture of sulfuric and nitric acids) were tested. Pt-Mix was most effective in growth stimulation and resulted in 45-90% increase in tree height in sandy loam. Pt-Band was less effective than Pt-Mix in growth stimulation and mycorrhizal formation. Simulated acid rain at pH 4.5 stimulated height growth by 10-55%, while acid rain at pH 3.0 did not significantly affect the height growth. The top/root ratio was increased by pH 4.5 treatment, while pH 3.0 treatment reduced it. Mycorrhizal infection rate was not affected by acid rain. Pt inoculation reduced acid-induced leaf injury by 28-58% in both pH 3.0 and 4.5 compared with un-inoculated plants. Sl was also effective in growth enhancement, but was less effective than Pt in both mycorrhizal infection and reducing leaf injury.
This study was conducted to evaluate resistance and physiological responses of Zelkova serrata Makino seedlings to ozone in open-top chamber. One-year-old seedlings of Zelkova serrata were planted in pots in April and grown in greenhouse until August. The plants were transferred into two out-door open-top chambers with a dimension of 2.0 m in diameter and 2.0 m in height. First chamber served as a control and was supplied with ambient air. Ozone was added to the second chamber for 5 hours per day(10.00 AM-15.00 PM) for 23 consecutive days at 0.1 ppm. Each chamber housed 70 pots. Every two, three or five days after initiation of exposure, ten pots were randomly removed from the chamber and determined for the contents of chlorophyll a, b, total chlorophyll and ${\beta}$-carotene in the leaves. Photosynthesis and dark respiration were estimated by measuring $CO_2$ absorption in a gas exchange chamber and oxygen absorption by oxygen monitoring system, respectively. Superoxide dismutase(SOD) activity in the leaves was assayed by a xanthine oxidase method. First visible injury of translucent(water-soaked looking) spots appeared on the leaves 14 days after the initial exposure, and ozone accelerated senescence of old leaves. Contents of chlorophyll a and b decreased by 17%, and 31%, respectively, in ozone treatment two days after exposure. The decrease in chlorophyll b was greater than that of chlorophyll a. Content of ${\beta}$-carotene in ozone treatment decreased by 25% two days after initiation of exposure, but the reduction was recovered with time. Photosynthesis decreased by 45%, and the respiration increased by 28% in the ozone treatment. SOD activity started to increase 4 days after beginning of exposure and increased by 285% 7 days after exposure, and decreased to the level below the control treatment with the advancement of the visible injury.
This study was conducted to determine the toxic effects of air pollutants on landscaping trees, Pinus densiflora, Pinus koraierasis, Ginkgo biloba, Liriodeytdron tulipifera, Platanus occidentalis and their resistance to the pollutant toxicity in urban and industrial regions of Seoul and Taejon, Korea. Total sulfur content and superoxide dismutase activity were analysed in tree foliage of Pinus densiflora, Pinzes koraiensis, Ginkgo biloba, Liriodendron tulipifera, Platanus occidentalis. In addition, SOD activity was analyzed in the foliage of tree seedlings, i.e. Pinus densijlora, Pinus koraiensis, Ginkgo biloba, Liriodendron tulipifera, with the lurnigation of $SO_2$ in gas chamber 4 hours a day for six days. In all species total sulfur content and SOD activity had a positive correlation. Air pollutants accumulated in tree tissues were supposed to enhance the enzyme activity like SOD providing with the resistance mechanisms. Trees under the air pollution stress increased enzyme activity to develop internal self-resistance against pollutants, but after a critical point enzyme-activity decreased gradually and resulting in injury after all, Deciduous trees had greater filtration capacity than conifers and coniferous trees showed greater resistance against air pollutants than deciduous species. Foliage SOD activity was higher in polluted area than in unpolluted area for most species. Coniferous species and mature trees had higher SOD activity than deciduous seedlings. Especially Pinus koraiensis, Ginkgo biloba and Plcatanus occidentalis had higher SOD activity than other species. The tree species with the high SOD activity showed strong resistance against air pollutants. In 2nd-year needles of Pinus densiflora seedlings and current and 2nd-year needles of Pinus koraiensis seedlings containing high native SOD activity, SOD activity increased with the increase of $SO_2$ level. But in seedlings containing low native SOD activity, SOD activity increased at 0.5ppm $SO_2$ level while it decreased at 1.5 and 2.5ppm $SO_2$. Changes of SOD activity was different between species and in most species SOD seemed to participate in resistance mechanism.
Purpose : To evaluate the usefulness of functional MRI (fMRI) of visual cortex in patients with ischemic infarction in the occipital lobe. Materials and Methods : Four patients with the symptoms and signs of visual cortical ischemia were included. Functional MRI was performed by 2D-FLASH technique with the parameter of 90/56msec TR/TE, $40^{\circ}$ flip angle, $240{\times}240{\;}FOV,{\;}64{\times}128$ matrix number, 8.32 seconds acquisition time, 8mm slice thickness. An axial slice including both visual cortices was selected and alternative activation and resting of the visual cortex was performed using red color photostimulator. all patients undertook visual field test, and vascular abnormality was examined by MRA (n=4) and DSA (n=2). fMRI results were compared with the results of a visual field test, conventional MRI and cerebral angiography. Results : On fMRI, decreased activity of the visual cortex was found in the occipital lobe corresponding to stenosis of the posterior cerebral artery or its branch noted on angiogram. However, 2 of 4 patients showed no abnormal findings on conventional MRI. Visual field defect was noted in 3 patients, one and of whom showed no abnormality on conventional MRI and diffusion-weighted image, but revealed decreased activity in the corresponding visual cortex on fMRI. Conclusion : fMRI may be a sensitive method for detection of the status of decreased blood flow or vascular reserve which other methods can not.
Purpose: The purpose of this study was to search activated genes that could be related to radiation adaptive response (RAR) induced by low-level radiation from $^{99m}Tc$ in human cell lines. Methods: We used gene discovery array (GDA) and representational difference analysis (RDA) methods. $^{99m}Tc$-pertechnetate was added to $2{\times}106/mL$ NC-37 cells (human lymphoblastic cells) to make concentrations ranging from 148 MBq/mL to 148 Bq/mL by serial 10 fold dilutions. After 44 hours, 2 Gy gamma irradiation was given to them using a Cs-137 cell irradiator. Results: As compared to the control (Con) group to which no $^{99m}Tc$ was added, those cells to which 148 and 14.8 KBq of $^{99m}Tc$ were added showed significantly lower damage to chromosomes, which was evaluated by metaphase analysis. Cells with 148 KBq $^{99m}Tc$ (T148 group) showed most significant protection. Activated genes in the T148 group as compared to Con group were evaluated by GDA and GDA methods. GDA revealed genes of casein kinase 2 (CK2) beta chain, immunoglobulins (lg), human leukocyte antigen (HLA)-B, and two novel genes. Twenty RAR related clones were selected by RDA method. The size of those genes was from 234 to 603 base pairs. Conclusions: RAR was induced by low dose irradiation from $^{99m}Tc$ in NC-37 cell lines. Genes related to the response included CK2, lg, HLA-B in human lymphoblastic cell lines.

BACKGROUND: Rabbiteye blueberry(Vaccinium ashei) is one of the most widely grown blueberry types in the world, together with Northern and Southern highbush blueberry(Vaccinium corymbosum). Rabbiteye blueberry have higher soil adaptability and fruit productivity but less cold tolerance to low temperature than highbush blueberry. The objective of this study is to investigate freezing tolerance of floral buds and establish a cultivation zone for rabbiteye blueberry cultivars. METHODS AND RESULTS: Bearing branches which have similar thickness and same number of floral buds were collected in the early January at the blueberry germplasm preservation plot located in Namhae Sub-station, National Institute of Horticultural and Herbal Science in Gyeongsangnamdo. Cold response of bearing branches were investigated by electrolyte leakage and freezing tolerance of floral buds were determined by ovary browning ratio of 50%($LT_{50}$). Cultivation zone was established based on mean annual extreme minimum temperature for 30 years, from 1981 to 2010. The electrolyte leakage of bearing branches in rabbiteye blueberry increased as temperature decreased and was lowest in 'Brightwell' but highest in 'Bluegem' when they were kept in $-5^{\circ}C$. Besides, the electrolyte leakage increased in 'Brightblue', 'Brightwell', 'Climax', 'Delite', 'Gardenblue', 'Southland' and 'Woodard' in $-20^{\circ}C$. Freezing tolerance($LT_{50}$) was lowest in 'Bluegem' and 'Homebell'($-13.3^{\circ}C$), and highest in 'Tifblue'($-25^{\circ}C$) among different rabbiteye blueberry cultivars. $LT_{50}$ of 'Southland' was from -15.0 to $-16.7^{\circ}C$, that of 'Delite', 'Brightwell',' Austin' and 'Climax' was $-18.3^{\circ}C$, and that of 'Bluebelle', 'Woodard' and 'Powderblue' was $-20^{\circ}C$. CONCLUSION: This study indicate that The hardiness zones of rabbiteye blueberry were classified into Six cultivation zones and cultivation zones of most cultivars were the south of Jeollanam-do and Gyeongdangnam-do, except for 'Tifblue.'

Vegetative plants in Yakutia are naturally frozen when they are covered with snow in the fall, and they function as green cryo-fodder that is a source of biologically active substances and nutrients for herbivorous animals. We observed a considerable increase in the total fatty acid content in the leaves of Avena sativa, Elytrigia rеpens, Equisetum variegatum and Equisetum scirpoides during the fall period. However, the degree of unsaturation of fatty acids was not higher in the frozen plants covered with snow than in the summer plants, with the exception of E. scirpoides, a dwarf horsetail found in the Pole of Cold in the northern hemisphere. In the internal adipose tissue of the Yakut horse (young horse meat), 18 fatty acids were found, including 10 saturated ones. Monounsaturated oleic С18:1 (n-9) acid and polyunsaturated α-linolenic С18:3 (n-3) acid were equally prevalent among the unsaturated fatty acids, accounting for 70% of the total unsaturated fatty acids. This composition of polyenoic fatty acids in the internal adipose tissue indicates that the Yakut horse actively feeds on the fall vegetation and the wintergreen sedge-grass. We believe that the high plant-specific free fatty acid content in the tissue of Yakut horses may play an important role in the regulation of their resistance to long-term low-temperature stress.
