Evaluation of Tomato Genetic Resources for the Development of Resistance Breeding Lines against Late Blight (잎마름역병 저항성 육종을 위한 토마토 유전자원의 저항성 평가)

  • Occurrence of tomato late blight (Phytophthora infestans) has caused significant losses in tomato yield in all over the world. Evaluation of the level of resistance in tomato gene resources for main breeding and initiation of the resistance breeding program are important for control of this disease. Resistant assay of 78 tomato cultivars/lines to late blight in pots and field experiment was carried out under controlled and natural conditions in 2009. All commercial cultivars including 'Legend' were susceptible. However, 10 lines including KNU-2, KNU-6-1, KNU-11, KNU-13, KNU-14-1 lines distributed from University of California, Riverside and L3708, $AV107-4{\times}L3708$, $07-15{\times}L3708$, $BS67{\times}L3708$ lines which have resistant gene Ph-3 and $06-9-62A{\times}06-9-62A$ were highly resistant to late blight. These highly resistant lines can be used as resources of resistance to late blight in a tomato breeding program in future.

Resistance Evaluation of Radish (Raphanus sativus L.) Inbred Lines against Turnip mosaic virus (순무모자이크바이러스에 대한 무 육종 계통 저항성 평가)

  • Leaves of twenties radish (Raphanus sativus L.) inbred lines were mechanically inoculated with Turnip mosaic virus (TuMV) strain HY to evaluate TuMV resistance of the radish inbred lines. The inoculated radish plants were incubated at $22^{\circ}C{\pm}3^{\circ}C$ and resistance assessment was examined using symptom development for 4 weeks. Based on the reactions of differential radish inbred lines, 16 radish lines were produced mild mosaic, mottling, mosaic and severe mosaic symptoms by TuMV infection. These results were confirmed by RT-PCR analysis of TuMV coat protein gene, suggesting that TuMV is responsible for the disease symptoms. Four resistant radish lines did not induce systemic mosaic symptoms on upper leaves and chlorosis in stem tissues for 4 weeks, showing they were symptomless by 8 weeks. Further examination of TuMV infection in the 4 radish lines showed no TuMV infection in all systemic leaves. These results suggest that the 4 radish lines are highly resistant to TuMV.

Evaluation of Pepper Genetic Sources (Capsicum spp.) for Disease Resistance Breeding (병저항성 육종을 위한 고추 유전자원의 저항성 평가)

  • For initiation of resistance breeding program of the red pepper, 21 PR ($Phytophthora$ resistance) cultivars, 36 cultivars collected from USA and 'Supermanitta' which is a susceptible cultivar against phytophthora blight were assayed against phytophthora blight, powdery mildew, and anthracnose. For seedling assay of phytophthora blight, three different mating type strains of $Phytophthora$ $capsici$ were used (A1, A2, Sterile). The result showed that most of the pepper of PR cultivars were resistance or moderately resistance at each mating type. 'Yeokganghongjanggun' was resistant to all three $P.$ $capsici$ strains and 'PR-Datta' and 'PR-Manitta' were resistant or moderately resistant at each type. In case of the collected cultivars, 'NuMex J.E.Parker', 'Omni Color', and 'SCM334' were resistant to all the three types and some cultivars including 'Sweet Banana' and 'Tabasco' were moderately resistant to each type fungi. 'Orange Habanero' and 'Black Cuban' were resistant to powdery mildew and 'Supermanitta' and 'PR Keumdong' were moderately resistant, while 'Santa Fe Grande', 'NuMex Pinata' and 'Puya' were very susceptible. In the case of anthracnose, 'Aji Limon' and 'Capsicum baccatum var. pendulum 3-4' were resistant and 'Pobalno', 'Omni Color', 'Negro', 'Mesilla', 'Mulato', 'Bhut Jolokia', 'Big Dipper', 'Black Cuban', 'NuMex Pinata', and 'NuMex Big Jim' were moderately resistant. The most PR cultivars except 'Taesan' were susceptible or very susceptible. These resistant individuals identified through this experiment can be used as sources of resistance to pepper pathogens in the future breeding programs.
