The p53 gene plays a critical role in the transcriptional regulation of cellular response to stress, DNA damage, hypoxia, and tumor development. Keeping in mind the recently discovered manifold physiological functions of p53, its involvement in the regulation of cancer is not surprising. In about 50% of all human cancers, inactivation of p53's protein function occurs either through mutations in the gene itself or defects in the mechanisms that activate it. This disorder plays a crucial role in tumor evolution by allowing the evasion of a p53-dependent response. Many recent studies have focused on directly targeting p53 mutants by identifying selective, small molecular compounds to deplete them or to restore their tumor-suppressive function. These small molecules should effectively regulate various interactions while maintaining good drug-like properties. Among them, the discovery of the key p53-negative regulator, MDM2, has led to the design of new small molecule inhibitors that block the interaction between p53 and MDM2. Some of these small molecule compounds have now moved from proof-of-concept studies into clinical trials, with prospects for further, more personalized anti-carcinogenic medicines. Here, we review the structural and functional consequences of wild type and mutant p53 as well as the development of therapeutic agents that directly target this gene, and compounds that inhibit the interaction between it and MDM2.
In this study, the effect of humidity change on DNPH cartridge sampling efficiency for carbonyl compounds (CCs) has been investigated. For this analysis, gaseous standard of 6 different CCs (formaldehyde (FA), acetaldehyde (AA), propionaldehyde (PA), butyraldehyde (BA), isovaleraldehyde (IA), and valeraldehyde (VA)) was calibrated after derivatization with three types of DNPH cartridge products. Their calibration results derived at RH values between 0 and 80% were then compared against liquid phase standards. If the results of our analysis are compared by the RH values between 20 and 80%, the effect of RH can be distinguished between light and heavy CCs. For lighter CCs (like FA and AA), there was no significant change. However, for the ones heavier than PA, there was fairly noticeable increase in relative recovery ratio in RH value between 20 and 80%. Such patterns are seen consistently from all three DNPH products tested for comparison. The results of our analysis suggest that proper correction for RH change may be needed for heavier CCs by the cartridge method.
In our investigation on chemical constituents of edible mushrooms, seven compounds I-VII have been isolated from the methanolic extract of the fruit body of Sarcodon aspratus. The methanolic extract was separated by solvent partition, silica gel column chromatography and Sephadex LH-20 column chromatography, and compounds I-VII were finally purified by preparative HPLC or TLC. Their structures were assigned as 4-hydroxybenzoic acid methyl ester, 4-hydroxybenzaldehyde, cyclo(Ala-Pro), adenosine, nicotinamide, BL V, linoleic acid, respectively, on the basis of mainly $^1H\;NMR$ spectra.
The adsorption characteristics of multi-component solvent vapors including poorly-adsorbable chemicals such as toluene-xylene-MEK and toluene-MEK-IPA on the activated carbonaceous adsorbents were investigated in a stainless steel fixed bed of 10.2 cm ID and 50 cm in height in order to identify those carbons for eliminating and recovering solvent vapors from industrial emission sources. The used activated carbonaceous adsorbents were pelletized commercial activated carbons and activated carbon fiber. Breakthrough curves and adsorption capacity at atmospheric pressures were obtained. It has been found that non-polar and larger molecules have been adsorbed better than polar and smaller molecules. In special, alcohols and ketones were poorly adsorbed caused by competitive adsorbability in multi-component mixture system. However, it could be overcome by profitable employment of organization of cooperative system which was composed of different porosity activated carbonaceous adsorbents appropriately.
As the model compounds, citral and n-octanol which possess similar characteristics and structures of low molecular weight insect pheromones and $({\pm})-5-hydroxy-4-methyl-heptan-3-one$ which shows the aggregation pheromones activity of the rice weevil and the maize weevil were microencapsulated with low molecular weight polyethylene(LMPE) and urea-formaldehyde resin as wall materials. The core materials were microencapsulated as small particles in LMPE and urea-formaldehyde resin polymers and the microencapsulated polymers were white powders. And the polymer made from urea-formaldehyde resin was better than that from LMPE as wall material. The slow releasing effect and the releasing patten of the microencapsulated core materials were examined by solvent extraction method and headspace sampling method. Citral and n-octanol and $({\pm})-5-hydroxy-4-methyl-heptan-3-one$ were release more than 40 days and 15 days, respectively. The releasing pattern of urea-formaldehyde resin microcapsules showed rather smooth decrease than that of LMPE and was maintained at steady level longer.
New type dioxygen bridged complexes of palladium were prepared by using $KO_2$ as a source of superoxide ion $(O_2^-)$. The method is completely different from the traditional one which has adopted the oxidative addition of molecular oxygen to transition metal complexes in low valency. It was suggested that the reaction to prepare the dioxygen complexes proceeded via nucleophilic displacement followed by electron transfer reaction. Five new type dioxygen complexes having ${\pi}$-allyl ligand were prepared and characterized by the application of the reaction of $O_2^-$.
Equimolar aqueous solutions of D-glucose and glycine were heated at $50^{\circ}C\;and\;95^{\circ}C$ at pH 6.7. The headspace volatiles and the ether extracts from the reaction mixture were analyzed by gas chromatography and gas chromatography-mass spectrometry using a fused silica capillary column. The major components formed were identified as diacetyl, three furfurals, two pyrroles, one furanone, two pyranones and two amides. In order to elucidate the formation mechanisms of the amides formed front amino-carbonyl reaction, two model systems were adopted. N-butylacetamide were formed as major components from diacetyl-butylamine ana glyoxal-butylamine systems, respectively. The results obtained suggest that such ${\alpha}-dicarbonyls$ as 3-deoxy-D-erythro-2,3-hexodiulose and diacetyl generated in the amino-carbonyl reaction react with amino compounds, amides then being formed by cleavage of the C-C bond in the ${\alpha}-dicarbonyls$.
Growing industrialization leads to severe water pollution. Organic effluents from pharmaceuticals and textile industries released in wastewater adversely affect the environment and human health. Presence of antibiotics used for antibacterial treatment in wastewater leads to the growth of drug resistance bacteria, which is very harmful for human being. Various small organic molecules are used for the preparation of organic dye molecules in the textile industries. These molecules hardly degrade, which is present in the wastewater effluents from printing and dyeing industries. In order to address these problems, photoactive catalyst is embedded in the membrane and wastewater are passed through it. Through this process, organic molecules are photodegraded and at the same time, the degraded compounds are separated by the membrane. Titanium dioxide (TiO2) is a semiconductor which behave as excellent photocatalyst. Photocatalytic ability is enhanced by the making its composite with other transition metal oxide and incorporated into polymeric membrane. In this review, the degradation of dye and drug molecules by photocatalytic membrane are discussed.
