  • This study was conducted to find out the optimum nitrogen fertilization for production of good quality and high yield zoysiagrass (Zoysia japonica Steud.), the changes in chemical properties of soil in pot and field experiments treated with different levels of nitrogen fertilizer. In pot experiment, the fresh and dry weights of shoots and stolons and the number of shoots increased as nitrogen levels increased, and showed no significant between 24 and $48kg\;N\;10\;a^{-1}$. In field experiment, the shoot length, fresh and dry weights of shoots, roots and stolons, the number of shoots and total stolons length linearly increased as affected by increased nitrogen application, and were not significantly different between 24 and $32kg\;N\;10\;a^{-1}$. In both experiments, pH and exchangeable cations ($Ca^{2+}$ and $Mg^{2+}$) in soil decreased as the rate of nitrogen application increased. As a results, chemical properties of soil were more deteriorated in the plots of higher nitrogen fertilizer rate. Thus, these results demonstrated that the nitrogen fertilizer rate for maximum growth of zoysiagrass was $24kg\;N\;10\;a^{-1}$ in consideration of growth and soil condition.

  • To clarify the mode of uptake of butachlor (2-chloro-2', 6'-diethyl-N-(butoxymethyl) acetanilide) by rice seedlings, its phytotoxic action to growth and physiological activities, studies were conducted with rice seedlings, at the 6th or 7th leaf-stage, which were treated with nutrient solution containing butachlor 0, 1.8, 3.6, 7.2, 10.8 or 14.4 ppm for 1, 2 or 4 days, in other case, the solutions were thereafter renewed with the untreated nutrient solution for further growth. Uptake of butachlor by rice seedlings increased linearly with increase of its concentration and duration of uptake. Butachlor inhibited root growth more than shoot growth, furthermore, the inhibitory effect on the shoot growth was greater in height than in weight or leafing rate. After 4 day-treatment, the rates of shoot growth in weight were delayed for 4 days. Butachlor inhibited water uptake rapidly and linearly with increase of its external concentration. The reduced uptake of water was followed by slow increase in the stomatal resistance of leaves. Upon completion of butachlor treatment, rate of water uptake was recovered rapidly, but the stomatal resistance with lag in time. Butachlor did not affect the uptake of cation such as ammonium, potassium and calcium, but inhibited substantially uptake of nitrate in proportion to its concentration. Especially, butachlor did not affect synthesis and degradation of nitrate reductase. In addition, butachlor has shown much greater binding to the lipidic substances from rice roots than the proteinous material. The primary mechanism of phytotoxic action of butachlor does not seem to be its effect on the protein synthesis, but great affinity to membranes. The inhibition of water uptake, and its subsequent closure of stomates is thought very important for reduced growth under mild phytotoxicity.

  • To establish an efficient herbicide screening method for purple nutsedge(Cyperus rotundus) control, its sprouting characteristics, tuber production and responses on several herbicides were investigated under greenhouse condition. The tubers stored at $4^{\circ}C$ after sterilization with the diluted prochloraz(Spotac) solution showed higher sprouting than the non-sterilized did. The harvested tubers were not dormant, and the sterilized tubers which stored at low temperature had a sprouting capability of about 80% after 6 months. If the fresh weight of purple nutsedge tubers was decreased to below 48%, they could not sprout. However, the tubers soaked in water and then stored at low temperature could sprout by 88% even 6 months later. Sprouting and initial growth of tuber were much better at $35^{\circ}C$-day/$25^{\circ}C$-night than at 30/$20^{\circ}C$ or 25/$15^{\circ}C$. The half-sected tubers, which were prepared by setting the intact tuber of above 1.2g latitudinally, were shown similar initial growth to the intact but those sected crucifically were not. These results suggest that the half-sected tuber itself can be used as a material on herbicide screening. About 1000 tubers could be harvested when 10 tubers planted in a pot($56{\times}35{\times}16cm$) filled with the artificial soil were cultivated in greenhouse of $35^{\circ}C$-day/$25^{\circ}C$-night for 3 months(April-July, 1993). Chlorimuron, Bentazon and Norflurazon were selected as the standards for the screening because of providing relatively effective control on purple nutsedge in both soil-surface and foliar spray treatment.

  • Selection of pepper (Capsicum sp.) cultivars tolerant to acetolactate synthase (ALS)-inhibiting herbicides {imazethapyr, 2-[4,5-dihydro-4-methyl-4-(1-methylethyl)-5-oxo-1H-imidazol-2-yl]-5-ethyl-3=pyridine-carboxylic acid, and primisulfuron methyl 2-[[[[[4,6-bis(difluoromethoxy)-2-pyrimidinyl]amino] carbonyl]amino]sulfonyl]benzoate} was investigated. Pepper cultivars such as Red Top, Happy Dry, Golden Tower, and Hagyeorae showed relatively tolerant response to imazethapyr, while cultivars; Korea, Cheongyang, Oriental Glory, and Hanam were susceptible. Red Horn, Jopoong, Kwangbok, and Wangcho cultivars were tolerant to primisulfuron whereas Korea, Dahhong, Chamjoah, and Poongchon cultivars were susceptible. Determination of growth inhibition by ALS-inhibiting herbicides showed that the $I_{50}$ estimates of growth from the susceptible- and tolerant-cultivars were 0.075 and 0.20kg ai/ha for imazethapyr; 0.06 and 0.16kg ai/ha for primisulfuron, respectively. Furthermore, the $GR_{50}$ estimates of growth from the susceptible and tolerant cultivars were 0.05 and 0.20kg ai/ha for imazethapyr; 0.07 and 0.16kg ai/ha for primisulfuron, respectively. This result, based on the $GR_{50}$ and $I_{50}$ values, indicates that responses of pepper to ALS-inhibiting herbicides between tolerant- and susceptible-cultivars were different about 3- to 4-fold to imazethapyr, and 2- to 3-fold to primisulfuron.

  • In this study, germination characteristics and herbicidal response of green kyllinga(Kyllinga brevifolia var. leiolepsis H.) were investigated. The storage method desirable for a rapid dormancy release was to keep the seed under low temp. and wetting condition for one to two months, or high temp($40^{\circ}C$) and drying condition for three months. The dormancy of rhizome was hardly observed. The optimum temperature for germination of seed and rhizome was around $30^{\circ}C$ and 16-$20^{\circ}C$, repectively. The germination of dormancy-breaked seed was completely dependent on light. Shoot emergence ratio(%) was decreased with increase of planting depth ; for example, only 18% of rhizome segments planted in the depth of 4cm under soil surface emerged above soil surface. Flooding at earlier growth stage resulted in significant decrease in shoot emergence as well as in dry weight. The germinablity of rhizome was almost lost as a decreased in fresh weight reached to 50%. Usually, green kyllinga was sensitive to herbicides such as bentazone, bensulfuron and benfuresate etc. which were known to be effective in Cyperaceae weeds, indicating that green kyllinga can be used as a representative plant in the screening of herbicides for Cyperus weeds.

  • Greenhouse study was undertaken to find tank-mix feasibility of quinclorac with molinate and propanil, selective post-emergence herbicides in controlling barnyardgrass, for reducing the application rate of quinclorac. Following foliar application in combination of quinclorac at 0.038, 0.075, 0.150, and 0.300kg ai/ha with molinate at 0.190, 0.380, 0.750 and 1.500kg ai/ha, and propanil at 0.263, 0.525, 1.050, and 2.100kg ai/ha at 3.5-leaf stage of barnyardgrass, fresh weight and weeding efficacy and their interaction by Colby's efficacy method were evaluated. Percent inhibition of barnyardgrass growth by quinclorac, molinate and propanil at recommended rate were 78.1, 26.1, and 61.7%, respectively. The dose combination shown above 85% in weeding efficacy were from 0.300kg of quinclorac with 0.75kg of molinate and 0.150kg of quinclorac with all rates of propanil. Therefore, combination of quinclorac with molinate tended to additive interaction and that of quinclorac with propanil appeared partially synergistic interaction. Conclusively, for reducing the application rate of quinclorac, the combination of quinclorac with propanil was more synergistic than that of quinclorac with molinate.

  • This study was conducted to investigate the effect of silicate mixed with nitrogen fertilizer on improving the growth and reducing nitrogen input of zoysiagrass. Plant height, fresh and dry weight of shoots, roots, and stolons, the number of shoots and total of stolons length were increased with highest in silicate mixed with nitrogen 24 kg/10a than nitrogen 24 kg/10a, and it showed no significance in silicate mixed with nitrogen 18 kg/10a. Nitrogen use efficiency in mixed silicate fertilizer was increased by 25-30% than single nitrogen fertilization. Moreover, the contents of available $SiO_2$, and organic matters of silicate fertilization on soil was higher than not silicate fertilization on soil. The silicate enhanced the growth and density of zoysiagrass, while it was a crucial factor to affect the chemical property of the soil.

  • This experiment was performed to identify and isolate a growth inhibiting compound from Aralia continentalis. In order to isolate the growth inhibiting compound from Aralia continentalis the bioassay test of lettuce seed germination and rice seedling growth were used. Through these bioassays the growth inhibiting compound which was spotted at $R_f$ 0.51 on Tlc was isolated. This compound inhibited the lettuce growth by 79% at the concentration of 1000ppm. When sprayed with $FeCl_3$ reagent, it developed a bule spot. It had UV-absorbance at 217 nm and 342 nm, and $OH^-$ of $3600cm^{-1}$, C=O of $1700cm^{-1}$, C=C of $1600cm^{-1}$, and C-O of $1200cm^{-1}$ on IR spectrum. Through HPLC analysis this compound was identified as a ferulic acid ($C_{10}H_{10}O_4$) having 25 min. retention time.

  • We investigated the levels of resistance and accumulation of terapyrroles, reactive oxygen species, lipid peroxidation, and antioxidative enzymes for reasons of growth reduction in herbicide-transgenic rice overexpressing Myxococcus xanthus, Arabidopsis thaliana, and human protoporphyrinogen oxidase (Protox) genes. The transgenic rice overexpressing M. xanthus (MX, MX1, PX), A. thaliana (AP31, AP36, AP37), and human (H45, H48, H49) Protox genes showed 43~65, 41~72 and 17~70-fold more resistance to oxyfluorfen, respectively, than the wild type. Among transgenic rice lines overexpressing Protox genes, several lines showed normal growth compared with the wild type, but several lines showed in reduction of plant height and shoot fresh weight under different light conditions. However, reduction of plant height of AP37 was much higher than other lines for the experimental period. On the other hand, the reduction of plant height and shoot fresh weight in the transgenic rice was higher in high light condition than in low light condition. Enhanced levels of Proto IX were observed in transgenic lines AP31, AP37, and H48 at 7 days after seeding (DAS) and transgenic lines PX, AP37, and H48 at 14 DAS relative to wild type. There were no differences in Mg-Proto IX of transgenic lines except for H41 and H48 and Mg-Proto IX monomethyl ester of transgenic lines except for MX, MX1, and PX. Although accumulation of tetrapyrrole intermediates was observed in transgenic lines, their tetrapyrrole accumulation levels were not enough to inhibit growth of transgenic rice. There were no differences in reactive oxygen species, MDA, ALA synthesizing capacity, and chlorophyll between transgenic lines and wild type indicating that accumulated tetrapyrrole intermediate were apparently not high enough to inhibit growth of transgenic rice. Therefore, the growth reduction in certain transgenic lines may not be caused by a single factor such as Proto IX, but by interaction of many other factors.

  • Growth and anatomical responses of rice and barnyardgrass(Echinochloa crus-galli) to oxyfluorfen were examined under various growing conditions in a greenhouse. With foliar application at 0.08kg ai/ha 7 days after seeding or transplanting, oxyfluorfen completely reduced the shoot and root growth of barnyardgrass under all growing conditions. However, oxyfluorfen reduced the growth of direct seeded and transplanted rice by 0~64% and 0~12%, respectively. Phytotoxic effect on rice growth, with respect to plant height and shoot fresh weight, was more pronounced under water(53~63%) than under dry-condition(16~33%), and in broadcast-seeded(34~62%) than in drill-seeded rice(16~33%). Anatomical changes of epidermal and vascular bundle sheath cells of rice leaves were not induced by oxyfluorfen, but mesophyll cells were partially ruptured and total leaf thickness was reduced. Oxyfluorfen, however, constricted and ruptured mesophyll and vascular bundle sheath cells and severely reduced leaf thickness of barnyardgrass under all growing conditions examined. These anatomical changes were more severe under dry- than under water-seeded condition.
