The effect of temperature and light on anthocyanin synthesis in Glehnia littoralis Fr. Schm. (갯방풍의 Anthocyanin생성에 미치는 온도 및 광의 영향)

  • The experiment was carried out to clarify the effects of air temperature, light exposure and UV light on the growth and anthocyanin syntheses of Glehnia littoralis Fr. Schm. The major pigment of purfied aglycone of Glehnia littoralis Fr. Schm. was identified as cyanidin by TLC and HPLC analysis. The anthocyanin Syntheses was promoted at low temperature and in the treatment of light exposure (12hrs a day) for 4-6 days after shading. The plants grown under UV-cut film had less anthocyanin content those tose under UV-transmission film.

  • A novel UV sensor was manufactured and characterized using evanescent field coupling between photochromic dye dispersed polymer waveguide and side polished fiber. The spiroxazine (photochromic dye) dispersed polymer was used as planar waveguides. The resonant wavelength was shifted owing to refractive index variation of planar waveguide on exposed UV because of its photo-functional properties. The sensitivities are $1.21{\mu}W/mw$ and $2.75{\mu}W/mw$ when UV intensities increased after exposure times were fixed at 3 seconds and 5 seconds, respectively. Output optical power according to UV intensity increases and saturation time decreases as the intensity of UV radiations increases.

  • Amounts of DNA single strand breaks and unscheduled DNA synthesis in CHO cells exposed to MMS were increased in the presence of 3-aminobenzamide, a potent inhibitor of poly (ADP-ribose) polymerase. However, those in cells irradiated with UV-light were decreased. These results suggest that poly (ADP-ribose) polymerase acts negatively on the MMS-induced base excision repair but positively on the UV-induced nucleotide excision repair. In the combined treatment with MMS and UV-light in the presence of this inhibitor, amounts of strand breaks were just the same as those in the absence of the inhibitor. But those of unscheduled DNA synthesis were increased up to the amount induced by UV-light alone. These results may suggest that poly (ADP-ribose) polymerase affects the incision step of excision repair induced by MMS and UV-light independently, and that it may potentiate the complete cleaving of UV-induced pyrimidine dimers possibly by the repair enzymes which might have been partially inactivated by MMS.

Research and Application for Natural Extract That Contain Ultraviolet Rays Absorbent Ingredient (자외선 흡수 활성 성분을 함유한 천연추출물에 대한 연구와 응용)

  • Exessive UV radiation causes a lot of problems in our skin. In order to find out the alternative UV absorber that can safely be used in cosmetics, we have screened various natural extracts in terms of their UV absorbing effect. Some natural extracts, which possess antioxidative activities, have also been found to retard the oxidation process in our experiment. Natural compound such as 7-hydroxycymopol, baicalein, etc. could be transformed into adjuvant UV absorber by chemical modification. In cosmetics, its chemical stability against UV radiation, exposure to oxygen and other factors could be improved by using the silicone or W/S type emulsion. The values of MED (minimal erythema dose) were improved to 0.10 ${\pm}$ 0.02∼0.11 ${\pm}$ 0.02 by adding this natural extract into the cosmetic formulations. In conclusion, the results of the present study show that natural extracts could be used as an adjuvant UV absorber, if they are stabilized.

  • UV irradiation causes a variety of biologic effects on the skin. These effects can be devided to acute reactuons and chronic reacxtions by duration of UV irradiation. Acute reactions are erythema reaction, pigment reactions and changes in epidermal thickness. Among them erythema reaction is most common and conspicuous acute effects of the skin. Upon exposure to sun or artificial UV soures, a faint redness response of skin may begin. Larger exposure causes sunburn reaction which is exaggerated erythema reactionassociated with pain, swelling, vesicle and dulla. Extent and time course of erythema reaction depend upon several factors including wavelength and dose of UVR, skin conditions likeas skin type, site, color, temperature, humidity and environmental factors. Evaluation of erythema erythema induced by UV irradiation is difficult to quantify. Degree of redness of skin are usually estimated by subjective visual evaluation. The lowest exposure dose required to protuce erythema is called minimal erythema dose (mod). Repeated exposures of UVR result in photaging skin. In this condition we can see wrinkling, skin atrophy, dilated blood vessels and keratoses. In sensitive persons photocarcinogenesis is can Be developed on exposed area of skin. Recently skin canser is increasing now in our country. An effective public education and photopreventive method must be developed.

Effects of Bambusae Caulis in Taeniam Extract on the UVB-induced Cell Death, Oxidative Stress and Matrix Metalloproteinase 1 Expression in Keratinocytes (각질세포에서 자외선B가 유도한 세포 사멸, 산화적 스트레스 및 matrix metalloproteinase 1 발현에 대한 죽여추출물의 영향)

  • Ultraviolet radiation (UV) is a major cause of skin photoaging, and effective UV protecting agents are needed for the skin health and beauty. This study was undertaken to examine the effects of Bambusae caulis in Taeniam extract (BCTE) on UVB-induced cell death, oxidative stress and matrix metalloproteinase 1 (MMP1) expression in cell-based assays. HaCaT human keratinocytes were exposed to UVB in the presence of BCTE at different concentrations and resulting changes in cell viability and biochemical events were determined. The results showed that BCTE enhanced the viabilities of UVB-exposed cells, and attenuated apoptotic events such as cleavage of procaspase 3 to its active form, and the increase of Bax to Bcl-2 ratios. BCTE also attenuated the reactive oxygen generation and lipid peroxidation in cells exposed to UVB. Additionally, it attenuated the expression of matrix metalloproteinase 1 and the phosphorylation of c-Jun N-terminal kinase stimulated by UVB. Conclusively, the present study demonstrated that BCTE pro tected skin cells from the UVB-induced cell death, oxidative stress and MMP1 expression, suggesting its potential use as a cosmetic ingredient mitigating some features of the skin photoaging.

UV-cured Pressure Sensitive Adhesive for Protective Film Application (보호필름용 자외선경화형 점착제)

  • In this study, we made UV cured acrylic pressure sensitive adhesive containing silicone-urethane-acrylate (SUA) oligomer for a coating on protective film and investigated the effect of SUA oligomer content and UV-dose on adhesion properties. The results illustrated that peel strength decreases with increasing oligomer content and UV-dose, while holding power increases. The gel fractions sharply increased after UV irradiation and then remain constant with prolonged UV exposure. From peel-off test, sample S70 (70% oligomer content) shows the best peelability and removability without remaining any mark or adhesive material on the test substrate than S50 (50% oligomer content) and S60 (60% oligomer content). Sample S70 also showed a surface energy lower than $26mJ/m^2$ and a transmittance higher than 95% at UV-dose 1857 and $2270mJ/cm^2$ which met the required properties for protective film application.

  • UV-vis spectroscopy is one of the powerful tools for measuring the concentrations of reactant and products during a chemical reaction. However, there is an limitation of using the technique when the reaction undergoes in high concentration and high temperature. Color analysis using camera images can provide the identical results with UV-vis analysis with regardless of the sample concentration and temperature. The catalytic reduction reaction of resazurin to resorufin was investigated using the color analysis with the color spaces such as CIE L*a*b*. Moreover, the color analysis enabled the independent analysis of two different material's concentrations without the deconvolution of overlapped wavelengths unlike the case of using UV-vis spectroscopy.

UVB-Shielding Effects of para-Coumaric Acid (파라-쿠마린산의 자외선B 차단 효과)

  • Recent studies have uncovered attractive properties of para-coumaric acid (PCA) as a potential skin hywhitening agent. The purpose of the current study was to examine its UVB-shielding effects. Effects of PCA on the viability of HaCaT cells exposed to UVB were assessed in vitro in comparison with other aromatic amino acid metabolites that have similar UV absorption spectra. For in vivo test, PCA cream (1.5 %) and cream base were topically applied to the dorsal skin of SKH-1 hairless mice and the inflammatory responses due to UVB exposure were monitored by changes in skin color (erythema) and thickness (edema). The cream application-UVB exposure regimen was repeated every other day for a total of 12 sessions. When HaCaT cells were irradiated with UVB, there was a dose-dependent decline in cell viability. The cell viability decline due to UVB exposure (10 mJ $cm^{-2}$) was significantly prevented by 100 ${\mu}M$ PCA, cinnamic acid, urocanic acid, or indole acrylic acid by 39, 27, 39, or 31 %, respectively. Topical application of PCA cream onto the dorsal skin of hairless mice (10 ${\mu}g\;cm^{-2}$) attenuated the changes of color parameters, $L^*$, $a^*$, $b^*$ values, and thickness of the UVB (150 mJ $cm^{-2}$)-exposed skin by 59, 50, 58, and 53 %, respectively. The current study, together with the previous studies that demonstrated the antimelanogenic effects of PCA, suggested that PCA may prevent not only dyspigmentation but also inflammatory reactions in the UVB-exposed skin.

  • 국내의 등산용 아웃도어 제품의 경우, 기능성인 투습도와 내수압만을 증대시키려는 연구가 주류를 이루고 있었다. 하지만, 실제 등산용 아웃도어의 경우 산악지형인 고지대에서 사용시간이 많으므로, 이에 따른 장시간의 직접적인 태양광 노출로 인한 인체에 치명적인 영향을 야기시키고 있지만, 인체 보호용 헬스케어 아웃도어 제품에 대한 연구는 미흡한 실정이다. 태양광은 자외선 2.5%, 가시광선 51.5%, 적외선 46.0%의 광량 비율을 가지고 있으며, 이 중 자외선은 광량은 적지만 에너지적으로 높아 유기물 분해 및 열화를 일으킨다. 이러한 자외선을 차단하기위해 아웃도어 의류에서는 유무기하이브리드 소재를 표면에 코팅시키게 되며, 기능성 코팅액내에 함유되어 있는 나노분말의 경우 이산화티타늄($TiO_2$), 산화세륨(CeO), 산화아연(ZnO), 삼산화텅스텐($WO_3$), 산화마그네슘(MgO) 등이 주로 사용되어 진다. 본 연구에서는 자외선 흡수소재로 나노산화아연분말을 이용하여, 그 입도 및 코팅용 희석 용매내의 분산성을 확인하고, 함유량을 달리한 코팅 수지를 제조하여, 코팅시편 제조 후 그 특성을 비교/분석하여 자외선 차단 효과를 확인하고자 한다.
