Enzymatic Synthesis of Meth.yl Fructoside by Immobilized Invertase (고정화 전화당 효소에 의한 메틸 프룩토시드의 합성)

  • Methyl fructoside was synthesized from sucrose and methanol using an immobilized invertase. The enzyme was covalently bound by glutaraldehyde on porous silica coated with polyethyleneimine to give loading capacity of 120mg of invertase per one gram of dry porous silica and effective activity of 100U per one milligram of bound invertase. Polyethyleneimine coating imparted a hydrophillic character, good activity retention and high loading capacity to the surface of porous silica as well as hydrophillic microenviroment in the vicinity of bound invertase. The immobilized enzyme was formed into an alginate-enclosed silica bead to have enough activity for methyl fructoside synthesis from aqueous methanol-sucrose solution. Using the alginate-enclosed biocatalyst the yield of methyl fructoside was obtained as high as 55.9% from aqueous 30% (v/v) methanol and 0.291mo1/l sucrose with 2U/ml activity at $25^{\circ}C$, pH 4.8.

  • The quality of Injulmi, waxy rice cake is affected by milling methods. varieties. additives, steeping condition before milling and the texture properties during storage. Sinsunchalbyeo and Hwasunchalbyeo flours were made using pin-dry milling(PDM) and roller-wet milling( RWM) methods. Injulmi was made from those flours with different sugars, sucrose, maltose. oligosaccharide, and were measured textural properties of Injulmi stored at 2 $0^{\circ}C$ for 0, 12, 36 and 60 hrs, respectively. The hardness of sucrose added to injulmi made from PDM flour was the highest among any other sugars, but Injulmi from RWM flour was increased in the following order sucrose>maltose>oligosaccharide>non - added Injulmi. Adhesiveness of none added Injulmi were the lowest among sugar added Injulmi including, oligosaccharide, maltose and sucrose added Injulmi.

  • A surface plasmon resonance (SPR) sensor system for the determination of sucrose concentration was constructed with a gold thin film sensing chip. The properties of gold thin film are critical factors in exciting surface plasmon resonance phenomena. Therefore in the present paper, the fabrication conditions of gold thin film were investigated to optimize the SPR phenomena. The optimum thickness was obtained as $545{\AA}$ with $43.75^{\circ}$ resonance angle and good surface roughness limitation. about $3{\AA}$. The linear resonance angle shifts and rapid response were observed from the sucrose concentrations ranged from 0 to 40wt%.

  • Hydrogen producing bacterium, strain MeL 6-2 was isolated from the sludge of the factory areas in Anyang through the acclimation in basal salt medium (BSM) supplemented with 10 g/L of sucrose. Isolated strain MeL 6-2 was a facultative anaerobe which could grow in both aerobic and anaerobic environments. An aerobically grown pure culture isolated from enriched culture was analyzed by 16S rDNA sequencing and identified as Rhodopseudomonas sp. MeL 6-2. Effects of the concentrations of glucose and sucrose on the hydrogen production rate and the hydrogen production yield were investigated. When glucose in the range of 1~12 g/L was supplemented to the BSM, strain MeL 6-2 could grow without lag phase. An increased glucose concentration increased the specific hydrogen production rate linearly to $4.2\;mmol-H_2{\cdot}L^{-1}{\cdot}h^{-1}$ at 10 g/L, and $60\;mmol-H_2{\cdot}mg-DCW^{-1}{\cdot}h^{-1}$, but decreased slightly as the concentration increased to 12 g/L. The hydrogen production yield was maintained over a range from 2.6 to $3.1\;mol-H_2{\cdot}mol-glucose^{-1}$. When sucrose in the range of 1~12 g/L was supplemented to the BSM, strain MeL 6-2 could grow after ten hours. An increased sucrose concentration increased the specific hydrogen production rate and the hydrogen production yield to $163\;mmol-H_2{\cdot}mg-DCW^{-1}{\cdot}h^{-1}$ and to $4.5\;mol-H_2{\cdot}mol-sucrose^{-1}$, respectively.

Comparison of Free Sugar Content and Related Enzyme Activities on Different Parts of 'Changhowon Hwangdo' Peach Fruit (복숭아 '장호원황도' 과실의 부위별 유리당 함량 및 관련 효소활성 비교)

  • The free sugar content and related enzymes of four different parts, inner, outer, stylar end and stem end, of 'Changhowon Hwangdo' peach ($Prunus$ $persica$, L. Batsch) fruit were compared from August to September in 2006, i.e., from 120 to 150 days after full bloom (DAFB). The soluble solids content (SSC) of stylar end was the highest among the four fruit parts at 150 DAFB. Changes of free sugar content were similar to that of SSC in the four parts. The starch content at the stylar end was the highest at 120 DAFB, while all the other parts showed low starch contents at 150 DAFB. The free sugar composition of peach changed during fruit development. The sucrose was low at 120 DAFB and increased gradually in all parts of peach fruit. On the contrary glucose, fructose and sorbitol decreased with fruit development. The free sugar contents and related enzymes activities were investigated during fruit development. The rapid increase of sucrose contents during fruit development was more affected by sucrose synthase than sucrose phosphate synthase. Activity of SS in the four fruit parts increased continuously over the fruit development period, but activity of acid invertase showed a downward trend. This study found that the free sugar content was affected by enzyme activity for the synthesis or the cleavage. However, it was very difficult to explain sugar accumulation of peach segments with related-enzymes.

  • 2020년대에는, 7조개의 스마트 디바이스의 급증, 70억의 인구, 새로운 고품질 매체에 대한 요구가 급증하면서, 새로운 이동통신 환경 및 비즈니스 모델의 변화에 대한 선제대응이 필요하고, 현재 셀률라의 1000배의 용량, 사용자당 Gbps 서비스, 그리고 사용자당 1000분의 1의 에너지효율을 높이면서도, 사용자와 인터액션과 감성을 만족시키는 서비스에 대한 선도적인 연구개발이 필요하다. 이를 통해서, 현재 이동 통신 사업자들의 투자 대비 수익 감소에 따른 불합리한 투자 및 수익 구조를 개선할 수 있을 것으로 예상한다. 이러한 기술을 바탕으로 5G 이동통신 네트워크는 Early Adpater 시장뿐 아니라, 보편적 모바일 핵심 인프라기반으로 미래의 사회 문제를 해결하고 지식의 양극화를 적은 비용으로 해소할 수 있어야 하며, 또한 창조지식 국가 실현을 위한 성장엔진으로, 새로운 혁신 산업 및 지식산업의 생태계를 창조함으로서, 산업 육성 패러다임의 역할을 할 수 있어야 한다. 본 고에서는 이동통신 환경 및 서비스 변화를 예측하면서, 5G 이동통신에서 실현해야 할 이동통신 기술의 비젼과 그 추진 전략에 대해서 살펴보고자 한다. 그 구체적인 추진 전략으로서 국가 정책 및 연구 개발 활성화 방안을 수립하고, 그 실행을 위한 민관 연합의 협력 기구인 5G 포럼의 구성과 역할에 대해서 살펴본다.

  • 본 연구는 N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) 를 처리한 CHO-K1세포에서 DNA 복제억제와 그 회복과정의 분자론적 기작을 규명할 목적으로 방사선 이중표지에 의한 DNA합성율의 측정, 알카리 자당농도구배 초원심분리법에 의한 DNA 분자량과 후복제 회복율을 측정하여 다음과 같은 결과를 얻었다. DNA 합성율은 2nM 이하의 낮은 농도의 MNNG 처리군에서는 급격히 감소하였으나, 5nM 이상의 농도에서는 그 감소양상이 둔화되었다. 억제되었던 DNA 합성율은 시간경과에 따라 회복되어 처리 후 4시간 째에는 대조군 수준 또는 그 이상으로 회복되었다. MNNG 처리 후 DNA 분자 크기의 분포와 새로 합성된 DNA 분자의 생장양상을 알카리 자당농도구배 초원심분리법으로 조사한 결과 MNNG 처리 후 시간 경과에 따라 새로합성된 DNA 분자들의 크기분포는 1*107 달톤 이하의 DNA 분자들의 합성양이 특이하게 증가하였다가 감소함을 보였다.

  • TRS 무선망은 셀룰라 시스템과 동일한 네트워크 교환 기술로서 다수의 기지국으로 배치된 광역의 서비스 시스템에 의해 그 내에서는 어디를 가더라도 교신이 가능하도록 되어 있다. TRS 시스템 설계에서 고려해야 하는 우선사항은 용량에 관한 설계이다. 예상된 가입자로부터 지역별 통화량을 조사와 가입자당 통화량을 조사한 후, 각 세부 구역별 통화량 값을 구한 다음 가용 주파수 대역과 가입자당 수용 가입자의 값으로부터 FA당 통화 용량 및 기지국 유형별 가입자 수용용량을 계산한다. 더불어 통화품질 수준을 고려하는 엔지니어링이 필요하게 된다. 본 논문은 TRS 무선망에서의 용량설계 연구내용의 일부로, 최종적인 채널용량을 산정함으로써 최종적 목표인 기지국 수/유형/용량을 결정하는 것을 목표로 한다.

  • The ISFET glucose and sucrose sensors containing platinum electrode and photopolymeric enzyme membrane were fabricated. The platinum working electrode was used for the electrolysis of hydrogen peroxide, which was the other product of the enzyme reaction, to improve sensing characteristics of the sensors. In order to improve response time, photo-crosslinkable polymer(PVA-SbQ) was used to the matrix for the enzyme immobilized membrane. The characteristics of glucose and sucrose sensors were investigated according to the variation of platinum electrode area. The response time was about $3{\sim}5$ minutes and determinations of glucose and sucrose in the range of about $30{\sim}300mg/dl$ could be possible.
