Influence of Glabrousness on the Insect Resistance and Some Agronomic Characters in Rice Plant (수도 매끄러운엽 품종과 껄끄러운엽 품종에 있어서의 벼멸구 저항성, 수량성 및 제현비율의 비교)

  • Comparisons were made between pubescent and smooth leaved cultivars or lines for the brown planthopper feeding preference, adult emergence percent, nymphal period, grain yield and brown rice recovery percent, and found that there were no significant differences of the traits tested between smooth and pubescent.

Selection on Tolerant Oak Species to Water Flooding for Flood Plain Restoration (홍수터 복원을 위한 침수 내성 참나무 수종 선발)

  • In order to select the water flooding-tolerant trees among Korean oaks, we cultivated the sapling of 6 deciduous oak species, Quercus acutissima, Q. mongolica, Q. variabilis, Q. dentata, Q. aliena and Q. serrata, under the water flooding treatment and the control, and analysed the growth difference among them. For comparing the growth between treatment and control, we measured 13 morphological and ecological traits, such as leaf Leaf number, leaf length, leaf width, leaf area, shoot length, stem length, stem diameter, root length, plant length, leaf weight, stem weight, root weight, plant weight. Q. serrata have no growth difference between water flooding treatment and control, but the rest 5 species showed the growth reduction in water flooding treatment. Tolerance of oak species to water flooding was risen in order of Quercus acutissima, Q. mongolica, Q. variabilis, Q. dentata, Q. aliena and Q. serrata. Thus, among Korean deciduous oaks, it is desirable to plant Q. serrata for flood plain restoration of river.

A taxonomic study on genus Rhynchospora Vahl in Korea (한국산 골풀아재비속 3종의 분류학적 검토)

  • Morphological and anatomical characters of selected 3 taxa of Rhynchospora were reexamined. The epidermal patterns of achene and leaf were investigated using a scanning electron microscope (SEM) and a light microscope (LM). Morphological characters such as length and width of bract, spikelet, scale, achene, stem, leaf and leaf sheath, and shape of inflorescence, spikelet, scale, apex of scale, perigynium and achene, and number of stigma and anatomical characters (transectional shape of the stem, and leaf: vascular bundles in stem and leaf epidermal patterns: shape of fundamental epidermal cell and cell wall, type of silica body, subsidiary cell shape, size and frequency of stomatal complex of leaf) were useful for the identification. Keys based on data were presented here.

  • The resistance of a non-transgenic poplar clone, 'Ogy' and three transgenic poplar lines to the cottonwood leaf beetle, Chrysomela scripta F., was evaluated by in vitro feeding. The lines were transformed with neomycin phosphotransferase II(NPT II) as a selectable marker, proteinase inhibitor II(pin2) as a resistance gene, and CaMV 35S as a promoter. An efficient method of sterilizing the beetle eggs and introducing them into plant tissue cultures was developed. The resistance of the transgenic lines was investigated in terms of effects tin leaf area consumed, insect weight, insect developmental stages, and plantlet root dry weight after feeding. Also, leaf area consumed was examined by leaf age as measured through leaf plastochron index(LPI). The leaf area consumed and insect weight were highly significant between transformants and control, and insect development in vitro was significant among the transgenic lines. Larval infestation was the most severe around LPI 4 to 5 which were young leaves. The system provided a quick, highly controlled method to screen developing transgenic plantlets directly.

Genetic transformation of Sedum erythrostichum via Agrobacterium-mediated transformation by introducing herbicide-resistant gene (아그로박테리움을 통한 제초제 저항 꿩의비름(Sedum erythrostichum) 형질전환체 개발)

  • 꿩의비름 (Sedum erythrostichum)은 매우 우수한 지피식물이며 건조에 강한 대표적 식물로 바위정원 (rock garden)을 가꾸는데 있어서 중요한 수종으로 이용되며, 유럽등지에서는 지붕에 식재하기도 하며 최근에는 빌딩옥상녹화의 대표적 수종으로 식재되고 있다. 또한 한방에서는 경천이라 불리우기도 하는데 피부상처 치유 및 미백효과가 탁월하다고 알려져 있다. 본 연구에서는 Agrobacterium을 매개로한 꿩의비름의 형질전환 시스템을 개발하고 아울러 phosphinothricin-N-acetyltransferase (PAT) 유전자를 도입하여 제초제 저항 식물을 개발하고자 수행되었다. 꿩의비름 잎을 Agrobacterium에 담근후 0.5 mg/l NAA와 2 mg/1 BA가 첨가된 MS 배지에 3일간 공동배앙 하였다. 그 후 300 mg/1 cefotaxime이 첨가된 같은 배지에 옮겨 계대하면서 Agrobacterium을 제거하였다. 약 3주후에 잎 절편으로 부터 직접적으로 부정아가 형성되기 시작 하였는데 이 시기부터 잎 절편을 25 mg/1 kanamycin이 첨가된 선발배지에 옮겨 주었다. 이 결과 배양된 잎 절편 절편 중 3.75%에서 kanamycin에 저항하는 부정아를 얻을 수 있었다. 형질전환체는 X-gluc 반응, PCR, Southern, Nothern analysis를 통하여 확인하였다. 약 94%의 형질전환 식물체는 성공적으로 토양에 옮길 수 있었으며 약 3개월후에 꽃을 피웠다. 형질전환체는 제초제인 Basta ($^{(R)}$ phosphinothricine at 200 mg/1)를 살포하여 주었을 경우 생존함을 확인 하였다.

Evaluation of leaf morphology for distinguishing Prunus (Rosaceae) from Jeju, Korea (제주도산 벚나무속 잎 형질의 분류학적 검토)

  • Leaf morphology was examined for Prunus species from Jeju Island. Analyzed were the leaf blade length, petiole length, width of blade, number of veins and angle of the base to the mid-vein as quantitative characters and the distribution of trichome, position of glandsand leaf features qualitative characters. A total of 25 OTU were phenetically analyzed by UPGMA. The resuling phenograms slightly differ from the currently recognized taxonomic system in two points. Prunus mume was clustered with P. padus and P. buergeriana. Prunu spendula and P. yedoensis were separated from the cluster of P. jaamasakura, P. speciosa, P. sargentii and P. jamasakura var. quelpaertensis. Except for the members of subgenus Cerasus, subgenera Padus (P. buergeriana and P. padus), Microcerasus (P. japonica) and Amygdalus were well defined. Some morphological characters of leaves such as the ratio of blade length to width, the length of blade to petiole, number of veins, the distance between the gland and base, the angle of base to mid-vein, and the distribution of trichome were useful as diagnostic features for Prunus from Jeju Island.

Numerical Taxonomy of Cirsium japonicum Complex(Asteraceae) in Korea (한국산 엉겅퀴군(국화과) 식물의 수리분류학적 연구)

  • Patterns of morphological variation in Cirsium japonicum complex were examined using numerical analysis, and the delimitation of taxa was evaluated. Principal components analysis of taxa using 24 morphological characters and 11 leaf characters revealed the presence of two major groups; C. japonicum var. japonicum and C. japonicum var. spinosissimum. Circium japonicum f. nakaianum could be included in the category of C. japonicum var. japonicum. In conclusion, Korean C. japonicum complex was composed of two varieties and one form, C. Japonicum var. japonicum, C. japonicum var. spinosissimum, and C. japonicum var. spinosissimum f. alba.

A taxonomic study on the Fuirena, Kobresia, and Scleria of Korea(Cyperaceae) (한국산 사초과 3속(검정방동산이속, 좀바늘사초속, 너도고랭이속)의 분류학적 검토)

  • Morphological characters of three taxa belonging to three genera (Fuirena, Kobresia, and Scleria) in Korean Cyperaceae were reexamined. The anatomical patterns of stems and leaves, and epidermal patterns of achenes and leaves were investigated by a scanning electron microscope (SEM) and a light microscope (LM). Based upon the results, the scientific and Korean names, which have not been clearly established, were reviewed. Morphological characters such as length and width of stem, leaf, bract, scale, achene, and shape of achene, spikelet, style base, scale, perianth, and anatomical characters such as shape of stem and leaf, shape of achene epidermal cell and silica body of leaf, size and frequency of stomatal complex, presence of bulliform cell were useful for an identification of the three taxa.

Inheritance of Several Qualitative Characters in Cyclamen persicum (시클라멘의 몇 가지 형질의 유전)

  • This experiment was conducted to characterize the inheritance of petal color, flower-eye color, flower shape, leaf variegation pattern, and leaf-end shape in Cyclamen persicum. The segregation of $F_2$ and backcross populations from crosses between inbred lines was tested for Mendelian inheritance mode. From four different combinations of crosses, it was found that the petal color was controlled by a single incomplete dominant gene. The other characters, flower-eye color, flower shape, leaf variegation pattern and leaf-end shape were confirmed to be controlled by single complete dominant genes.

The taxonomic implication of leaf anatomy in tribe Sorbarieae (Spiraeoideae: Rosaceae) (쉬땅나무족(Sorbarieae Rydb., 장미과) 잎의 해부학적 형질 및 분류학적 유용성)

  • A comparative study of leaf anatomy in tribe Sorbarieae (Adenostoma, Chamaebatiaria, Sorbaria, Spiraeanthus) including one related genus Lyonothamnus was carried out using light microscopy. Anatomical characteristics of the leaf blade and midrib were described and taxonomically evaluated. The anatomical characters which described in this study are as follows: thickness of leaf midrib, blade in cross section, cuticle, epidermal cell, stoma, trichome, mesophyll, crystal, main vasculature type. All features were compared and the vascular patterns of midrib were distinguished two types. - Type 1: Trace tripartite (Adenostoma), Type 2: Trace continuous, subtype 2A: flat arc (Chamaebatiaria, Spiraeanthus), subtype 2B: U-shape arc (Lyonothamnus, Sorbaria). In conclusion, some of leaf anatomical characters (e.g., cuticle, epidermal cell, trichome, mesophyll, main vasculature type) can be useful for diagnostic features. Hypostomatic type, dorsiventral mesophyll, Ushape vasculature type would constitute a major characters for genus Sorbaria in Sorbarieae. The detailed anatomical description of studied taxa is provided, and its systematic importance is also briefly discussed.
