Restoration of Fertility by Suppression of Male Sterility- Induced Gene Using an Antisense Construct (웅성불임 유전자의 발현억제를 이용한 임성회복)

  • This study was carried out to restore the fertility by suppression of male sterility-induced gene using an antisense construct. Tobacco (cv. Petit Havana SR1) was transformed with the binary vector containing a GBAN215-6 promoter, an antisense diphtheria toxin (DTx-A) gene (pKDA215b) and a hygromycin resistant gene. Seventy-six confirmed transgenic plants regenerated from leaf disks were designated as the $R_0$ generation and selfed to produce the $R_1$ generation. From the inheritance study, five $R_1$ lines with multiple copies of the antisense construct were selected and selfed to identify homozygosity for the antisense construct. In order to restore fertility and finally to select restore lines, five $R_2$ lines with multiple copies of the antisense construct were crossed with male sterile plants. From these crosses, three different phenotypes have been observed: completely restored, partially restored, and not restored pollens, and otherwise tobacco plants were phenotypically same as normal plants. These plants were scored for the degree of restoration and selected for further study.

Seed Production Ability of Doubled Haploid Plants through Microspore Culture in Chinese Cabbage (Brassica rapa L. ssp. pekinensis) Introduced from China (중국도입 배추 소포자배양에 의한 배가반수체의 종자생산능력)

  • A total of eleven Chinese cabbage accessions were used for microspore culture and were grown to take basal data. Based on the collected data, breeding materials were chosen to develop new improved Chinese cabbage cultivars. The range of microspore-derived embryoid taken from flower buds was 1.6 to 35.4 embryoids. The embryoids from IT26110 and IT26153 among the Chinese cabbages were more than 34 per flower bud. The viability rate after cold treatment was low from 0.2 to 11.7%. The range of fertility rate was 7.7 to 58.8% in general but the IT26118, IT26122, IT26128, IT26130, and IT26164 were more than 50%. The result of their seed production ability by selfing was 11.9 seeds per siliqua in IT26128 while the others were less than 10 seeds. In the microspore culture using parents of different hereditary, the number of embryoids, the number of plants, the rate of fertility and their pure seed production ability appeared to be very different in doubled haploid lines obtained from fertile plants of Chinese cabbage.

An Increment of Crossing Efficiency with Consideration of Pollen Viability Analysis in Rose (장미 교배 효율 증대를 위한 화분 임성 검정)

  • Current rose cultivars are all composed of heterozygous genome due to long history of out crossing including interspecific hybridization. It has been adapted by artificial selection and crossing by breeders that mainly based on the crossing with fertile pollen derived from inter- or intra-specific hybridization. Pollen viability and germination ability tests provide valuable information for the designing of parentage for more successful breeding efficacy. In this study, we tested the pollen viability and germination ability in seven rose cultivars to find any relationship among several factors including pollen size, ploidy levels, and crossing compatibility. The pollen viability showed wide ranges from 39% 'Pinocchio' as minimum to 82% 'Scarlet Mimi' as maximum, whereas pollen germination rate were from 1% 'Mini Rosa' to 41% 'Scarlet Mimi' as a highest. Pollen size ranged from 41.3 to $45.4{\mu}m$ in large sized pollen and 30.7 to $37.4{\mu}m$ in small sized pollen. The mean diameter of large sized pollen is approximately 10-40% bigger than that of small sized pollen. There are positive relationships among ploidy level, total chromosome length, and pollen size. Crossing list showed that seed setting ratio and seed germination were related to pollen viability, pollen germination, and ploidy level.
