Volatile Compounds in the Oxidation of Unsaturated Fatty Acids Caused in Off-flavor of Porcine Liver by HS-SPME (Headspace solid-phase microextraction(HS-SPME)를 이용한 간의 비린내불쾌취에 기인하는 불포화지방산 산화생성물의 휘발성성분)

  • Headspace solid-phase microextraction(HS-SPME) was used to isolate the off-flavor volatile compounds, which are formed during the oxidation of porcine liver and unsaturated fatty acids in porcine liver induced by iron. Three fibers for HS-SPME method development were tested, and the polydimethylsiloxane/divinylbenzene(PDMS/DVB) fiber was selected as providing the best detection of analyzed compounds. In the oxidized arachidonic acid, the major compounds identified were hexanal, 1-octen-3-one(metallic), (Z)-4-heptenal, (E)-2-octenal, 1-octen-3-ol(mushroom), (Z)-2-nonenal (cardboard-like), and (E,E)-2,4-decadienal(fatty, oily). In the oxidized linolenic acid, the major ones were (E,E)-2,4-heptadienal(fishy), (E,E)-3,5-octadien-2-one and (E,Z)-2,6-nonadienal. Moreover, those of th oxidized porcine liver with iron were hexanal, 1-octen-3-one, (Z)-4-heptenal, (E,Z)-2,4-decadienal and (E,E)-2,4-decadienal. These results showed that the fishy and metallic off-flavor in porcine liver were due to the oxidation of unsaturated fatty acids in porcine liver.

The Optimal Condition for the Production and Extraction of Monacolin K from Red-Koji (홍국으로부터의 monacolin K 생성 및 추출 최적화)

  • The optimal condition for the production and extraction of monacolin K was reported. HPLC was used to determine monacolin K a kind of metabolite of Monascus from red-koji made of Monascus purpureus CBS 281.34. After culturing Monascus in solid and liquid media at $30^{\circ}C$ for 10 days, each of these were inoculated with soybean, wheat, barley, waxy rice, and rice and cultivated at $30^{\circ}C$ for 11 days. The production of monacolin K was the highest(0.35g/100g) when cultured with rice. The yield of monacolin K in red-koji increased with drying temperature and time according to the removal of water. Considering monacolin K content and the degree of death of Monascus, red-koji was dried at $80^{\circ}C$ for 60 min. Although monacolin K in red-koji was mostly extracted by 80% ethanol, there was no difference in monacolin K between shaking for 1 min and extraction for $0{\sim}24$ hr after sonication for 7 min. The extracted yield of monacolin K was the highest when the ratio of red-koji and 80% ethanol was 1:9. Moreover, the production of monacolin K appeared to be parallel with that of the pigment.

  • The fermentation process of Andong Sickhae including optimal composition of the product was investigated. Through sensory evaluation and ingredient analysis of ten samples prepared in the laboratory and the samples collected from Andong region, we found that the ratio of ingredient of the product by weigh are as follow : [glutinous rice(80) : malt(50) : radish(100) : water(500) : ginger(8) : red pepper(4)]. The level of nitrogen compound turned out to be low while that of soluble protein and salt soluble protein was high. The content of total sugar and reducing sugar was found to be considerable high and among the free sugar, maltose was the highest(80%), followed by glucose and maltotriose. Activities of acid protease and saccharogenic amylase were 1.55 unit per milliliter and $12.5D^{40}_{30}^{\circ}$ respectively. Results of sensory evaluation showed that the good Andong Sickhae turned out to have well harmonized taste of flavor, sweetness and sourness while the color looked slightly red.

  • For development of new high-value Korean traditional fermented food by using bioactive fungi and bacteria, Nuruk, Makgeolli and Cheonggukjang were prepared by mold, yeasts and bacteria from Korean traditional fermented foods and their physiological functionalities were investigated. Aspergillus oryzae N152-1 Nuruk showed the highest antihypertensive angiotensin I-converting enzyme inhibitory activity(57.2%), and Makgeolli made by Saccharomyces cerevisiae Y111-5 and commercial JS Ipguk (solid cultures of saccharifying enzyme-producing mold) was showed 42.0% of anti-obesity ${\alpha}$-glucosidase inhibitory activity. Among various Cheonggukjang, No 463 Cheonggukjang made by Brevibacterium iodinum NCDO 613(T) was showed the highest fibrinolytic activity (size of clear zone: 28 mm) and good anti-obesity ${\alpha}$-glucosidase inhibitory activity.

Elimination of Escherichia coli O157:H7 Contaminated in Frozen Beef by Electron Beam Irradiation (전자선 조사에 의한 동결육에 오염된 Escherichia coli O157:H7 의 제거)

  • Treatment with electron beam irradiation was investigated for the elimination of Escherichia coli O157:H7 which has been linked to outbreaks of foodborne illness on undercooked and raw meat. Before treatment, the maximum populations were observed at 16 hr when E. coli O157:H7 was incubated in TSB at $37^{\circ}C$. Incubation at $4^{\circ}C$ did not influence survival and growth of the strain. The numbers of E. coli O157:H7 were present about $10^{7}\;CFU/mL$ in the log $(6\;hr\;at\;37^{\circ}C)$ and stationary phase $(16\;hr\;at\;37^{\circ}C)$ of cells, respectively. Freezing $(24\;hr\;at\;-18^{\circ})$ had a more marked lethal effect. The $D_{10}$ value at $-18^{\circ}C$ of E. coli O157:H7 contaminated in frozen beef was 0.45 kGy, and inactivation factor were $6.67{\sim}11.11$ at the radiation doses of $3{\sim}5\;kGy$. Therefore, electron beam irradiation was an effective method to eleminate of E. coli O157:H7.

  • The production of bacterial protease and its characteristics were investigated with Bacillus subtilis globigii CCKS-118 which was isolated from Korean traditional soy sauce. The optimum culture condition of the strain for the production of alkaline protease was as follow : 2% soluble starch, 0.2% yeast extract, 0.1% $(NH_4)_2SO_4$, 0.2% $MgSO_4$, pH 7.5, $35^{\circ}C$ and 20h rs. The optimum pH and temperature for the enzyme action of alkaline protease producing Bacillus subtilis globigii CCKS-118 were pH 9.0 and $50^{\circ}C$, respectively. The enzyme was relatively stable at $pH\;6.0{\sim}9.0$ and at temperature below $40^{\circ}C$. The activity of the enzyme was inhibited by $Hg^{2+}$ whereas $Cu^{2+}$ gave rather activating effects on the enzyme activity. The enzyme was inhibited by phenylmethane-sulfonyl fluoride indicating serine pretense metal ion group are required for the enzyme activity. Km value was $1.242{\times}10^{-4}M$, $V_{max}$ value was $25.99\;{\mu}g/min$. This enzyme hydrolyzed casein more rapidly than the hemoglobin.

  • Rotating Biological Contactor (RBC) was tested for the removal of total ammonia nitrogen (TAN) by using simulated aquaculture system. RBC performance was evaluated by controlling revolution rate of disk and hydraulic residence tile (HRT). The optimum revolution rate of disk was 4 rpm, As HRT of RBC was increased, TAN removal efficiency of RBC and TAN concentration of rearing water were increased. HRT for maintaining lowest TAN concentration of rearing water was 9.5 minutes and at that condition TAN concentration of rearing tank was $1.03 g/m^3$

  • Rotating Biological Contactor (RBC) was tested for the treatment of artificial rearing water in n simulated aquaculture system. Performance of RBC on the removal of TAN and COD was evaluated by controlling hydraulic residence time (HRT). As HRT of RBC was increased, TAN removal rate ana removal efficiency of RBC and TAN concentration of rearing water were increased, but COD removal rate was decreased. Total alkalinity consumption rate was increased by increasing HRT of RBC. Ratio between total alkalinity consumption rate and TAN removal rate was 7.73. HRT for maintaining lowest TAN and COD concentration of artificial rearing water was 14,6 minutes and at that condition TAN and COD concentration of the water was 1.28 and $5.59 g/m^3$, respectively.

  • To investigate the response on feeder cell cultures, protoplasts isolated from cell suspensions initiated from mature seed scutellum-derived callus of the Japonica rice variety Taipei 309, were cultured on filter membranes under various conditions. The effects of various factors, such as gelling agents, feeder cell and protoplast densities, species of feeder cells and heat shock treatment, have been investigated to improve protoplast plating efficiencies on filter membranes. Maximum protoplast plating efficiencies were obtained when protoplasts were cultured on KPR medium semi-solidified with Sea Plaque agarose at a density of $5\;\times\;10^{5}\;ml^{-1}$ protoplasts in the presence of Lolium multiflorum as feeder cells (0.5 ml pcv per 10 ml of protoplast culture medium). Pre-culture heat shock treatments for 1 min. and 5 min. to the protoplasts did not give any appreciable increase on the plating efficiency of protoplasts in the presence of feeder cells. Maltose-supplemented medium was superior for plant regeneration from protoplast-derived colonies compared with medium containing only sucrose. The plants were transferred to the glasshouse, flowered and were fertile.

  • Pretense production and its characteristics were investigated for Rhizopus stolonifer, Rhizopus oryzae and Absidia corymbifera which were isolated from Korean traditional meju. The optimum culture conditions of the strains for the production of protease in basic medium [wheat bran : 1% glucose solution=1 : 1 (w/v)] were 3$0^{\circ}C$ and 4 days. The optimum pH and temperature for the enzyme activity of crude enzymes produced by Rhizopus sto-lonifer, Rhizopus oryzae and Absidia corymbifera were pH 6.0 and 5$0^{\circ}C$, respectively. The enzymes were relatively stable at pH 4.0~7.0, at temperature below 4$0^{\circ}C$, and at NaCl concentration lower than 16%. The $K_{m}$ value for Hammastein casein was 3.3$\times$10$^{-4}$ , 0.75$\times$10$^{-4}$ and 1.3$\times$10$^{-4}$ M, and $V_{max}$ value was 17.2$\mu\textrm{g}$/min, 9.4$\mu\textrm{g}$/min and 7.8$\mu\textrm{g}$/min, respectively.y.
