  • Seasonal variations of nutrients (ammonium, nitrite, nitrate, phosphate and silicate), primary productivity and ammonium regeneration rate of macrozooplankton were investigate to understand the relationship between nitrogen recycling and nitrogen requirement by phytoplankton from Feburuary 1986 to November 1987 in the Kyunggi Bay, shallow estuarine water of Yellow Sea. In general, nutrients increased during the winter and depleted during the spring and the early summer except temporally sharp increase after flood in September. Ammonium was prevalently generally found in high concentration throughout the study area and it occasionally raised N/P ratio in the range of 30 to 70 as in the freshwater environment. Daily net primary productivity ranged from 30.3 to 3580.0 mgC/$m^2$/d with an average of 883.9 mgC/$m^2$/d. Annual primary productivity was determined to be 320.0 gC/$m^2$/yr. Carbon assimilation number ranged from 2.9 to 19.4 mgC/mg chl-a/h which increased in the summer and decreased in the winter. Nitrogen requirement by phytoplankton ranged from 0.4 to 45.0 mg at-N/$m^2$/d and turnover time of inorganic nitrogen ranged from 2.4 in the late summer to 122.7 days in the winter. Nitrogen regeneration rate of mixed macrozooplankton determined by bottle incubation method ranged from 0.02 to 1.34 mg at-N $m^2$/d and it could contribute from 2.8 to 38.7% with an annual average of 14.9% of total nitrogen requirement by phytoplankton in this shallow estuarine environment.

  • Schizandra chinensis has been used as a traditional Chinese medicine and is known to have various bioactive components, including schizandrin and gomisin A. In the current study, we investigated the anti-inflammatory activities and their working mechanisms of ethanol extracts of pomace of Schizandra chinensis (PSC), schizandrin (SZ), and gomisin A (GA). First, we analyzed the effects of PSC on nitric oxide (NO) production and cell viabilities in lipopolysaccharide (LPS)-stimulated RAW264.7 cells. The results indicated that PSC dramatically reduced NO production in LPS-activated RAW264.7 cells in a dose-dependent manner without affecting cell viabilities. PSC also decreased the expression of pro-inflammatory genes iNOS and COX-2, whereas the expression of TNF-${\alpha}$ was not affected by PSC. In addition, PSC inhibited phosphorylation of p38, ERK1/2, and JNK but did not change the expression of their total protein. The results indicate that PSC can regulate LPS-induced inflammatory responses by suppressing MAPK (mitogen-activated protein kinase) signaling. We also analyzed the effects of SZ and GA on NO production and cell viabilities in RAW264.7 cells. The results showed that SZ and GA also decreased NO production in a dose-dependent manner in LPS-activated RAW 264.7 cells without affecting cell viabilities. SZ reduced the expression of iNOS, whereas GA downregulated iNOS and COX-2. Overall, these findings clarify the molecular mechanisms of the anti-inflammatory effects mediated by PSC, SZ, and GA.

  • This experiment was carried out to determine the forage yield and mineral contents of silage corn(Zea mays L.), and N balance, soil chemical characteristics and nitrate-N level in infiltration water by different types and N rates of liquid manure applied. Seven treatments consisting of chemical fertilizer $200kg\;N;ha^{-1}$, liquid cattle manure 200, 320 and $440kg\;N;ha^{-1}$, liquid swine manure 200, 320 and $440kg\;N;ha^{-1}$ were arranged in a randomized complete block design with three replications. The results obtained at National Livestock Research Institute, RDA, in Suweon from May 1997 to Aug. 1997 are summarized as follows : There were no significant differences in percentage of dry matter among the treatments, but significant dry matter yield differences were found (P<0.05), and also the mean dry matter yield of liquid swine manure plot was higher than that of liquid cattle manure plot. Potassium, calcium and magnesium contents of silage corn increased with increasing liquid manure application rates (P<0.05). Nitrate-N content of silage corn in the liquid cattle manure treatments was not influenced by liquid manure application rates, but that in the liquid swine manure treatments increased with increasing the N rates of liquid manure application. pH and contents of exchangeable canons of the soil after experiment were raised by increasing the amount of liquid manure application. Available $P_2O_5$ content in topsoil(0-10cm) was highest as $340mg\;kg^{-1}$ at the plot of liquid swine manure $440kg\;N;ha^{-1}$. Concentration of nitrate-N in infiltration water increased at the plot of liquid swine manure $440kg\;N;ha^{-1}$. Amount of nitrogen balance increased with increasing liquid manure application rates. Based on the results of this experiment, it is suggested that the mean dry matter yield of liquid swine manure plot was higher than that of liquid cattle manure plot, and the amount of nitrogen balance increased with increasing liquid manure application.

  • Purpose: McArdle disease, glycogen storage disease type V (GSD V), is one of the most common adolescent-onset glycogen storage diseases. It is caused by recessive mutations in PYGM encoding myophosphorylase, which is critical to glycogen metabolism. Since only a few korean patients have been reported, we will observe the clinical and genetic features of three korean patients with McArdle disease. Methods: We retrospectively reviewed the medical records of three patients with genetically confirmed McArdle disease, including the results of forearm ischemic exercise test, electromyogram, nerve conduction velocity, muscle biopsy, and PYGM analysis in peripheral leukocytes. Results: All three cases were males and their age of symptom onset was 12, 5, 14 years old, respectively. A high basal level of serum creatine kinase was noted in all three patients. They experienced the recurrent episodes of rhabdomyolysis, but second wind phenomenon was not definite. In muscle biopsy, subsarcolemmal space vacuoles including periodic acid schiff stained materials were found in two patients, while no evidence of glycogen storage disease was found in the other. A total of five different mutations, $p.Arg50^*$, p.Trp798Arg, $p.Arg50^*$, p.Glu779del, $p.Asp511Thrfs^*28$ and p.Phe710del, were found in three patients. Avoidance of isometric exercise, aerobic exercise and glucose intake before each exercise were recommended for all patients. Conclusion: The three Korean patients with McArdle disease showed the typical manifestations of the condition. The most mutations were private. Therefore, identification of more cases with long-term follow-up will be required to understand the clinical and genetic features of this disease among Korean population.

  • Sodium hypochlorite and ethylene diamine tetra acetic acid are substances usually used during endodontic treatment. Several studies found that the bonding was negated with certain irrigants and some of the used irrigants have demineralizing and chealating effects, so it was advocated to omit the etching step in etch and rinse adhesive systems. The purpose of this in vitro study was to evaluate the influence of NaOCl & EDTA on the bonding strength of ethanol wet bonding. Thirty human molars were selected and mesiodistally sectioned into halves, thus providing sixty specimens. The specimens were randomly assigned to 4 groups(n=15) according to the irrigant regimen used : (1) irrigated with distilled water for 10min (control); (2) irrigated with 5.25% NaOCl(10min), flushed with 5.25% NaOCl(1min) (3) irrigated with 5.25% NaOCl, flushed with 17% EDTA (4) irrigated with 5.25% NaOCl, flushed with 17% EDTA. Each group was acid-etched with 37% phosphoric acid(except group 4) and had their dentin surfaces dehydrated with ethanol solutions : 50%, 70%, 80%, 95%, 3x100%, 30s for each application. After dehydration, a primer( 50% all bond 3 resin + 50% ethanol) was used, followed by the adhesive(ALL-BOND 3 RESIN) application. Resin composite build-ups were then prepared using an incremental technique. Specimens were sectioned into beams and submitted to a tensile load using a Micro Tensile Tester(Bisco Inc.). The data were statistically analyzed using one-way ANOVA and Tukey HSD at p<0.5 level. There was no significant difference on G1(control) and G2(irrigated with NaOCl only ). (p>0.05). G3(flushed with EDTA) showed significantly high tensile bonding strength compared to the G2 (p<0.05). G4( treated with EDTA but no acid-etching) was significantly lower value than G3. (p<0.05) Although there was no significant difference, 5.25% NaOCl seemed to have an adverse effect on the bonding strength of ethanol wet bonding. The flushing with EDTA after NaOCl irrigation prevents the decrease of bonding strength. The use of 17% EDTA as a final flush can enhance the bonding strength but EDTA flushing can't substitute for a acid-etching.

  • BACKGROUND: Nitroxoline is an antibiotic agent. It is used for the treatment of the second bacterial infection by the colibacillosis, salmonellosis and viral disease of the poultry. When the nitroxoline is indiscreetly used, the problem about the abuse of the antibiotics can occur. Therefore, this study presented the residue analytical method of nitroxoline in food for the safety management of animal farming products. METHODS AND RESULTS: A simple, sensitive and specific method for nitroxoline in chicken muscle by high performance liquid chromatograph with PDA was developed. Sample extraction with acetonitrile, purification with SPE cartridge (MCX) were applied, then quantitation by HPLC with C18 column under the gradient condition with 0.1 % tetrabutylammonium hydroxide-phosphoric acid and methanol was performed. Standard calibration curve presented linearity with the correlation coefficient ($r^2$) > 0.999, analysed from 0.02 to 0.5 mg/L concentration. Limit of quantitation in chicken muscle showed 0.02 mg/kg, and average recoveries ranged from 72.9 to 88.1 % in chicken muscle. The repeatability of measurements expressed as coefficient of variation (CV %) was less than 12 % in 0.02 and 0.04 mg/kg. CONCLUSION(S): Newly developed method for nitroxoline in chicken muscle was applicable to food inspection with the acceptable level of sensitivity, repeatability and reproducibility.

  • This study was conducted to establish the standard method for the contents of biotin in milk formulas. To optimize the method, we compared several conditions for liquid extraction, purification and instrumental measurement using spiked samples and certified reference material (NIST SRM 1849a) as test materials. LC-MS/MS method for biotin was established using $C_{18}$ column and binary gradient 0.1% formic acid/acetonitrile, 0.1% formic acid/water mobile phase is applied for biotin. Product-ion traces at m/z 245.1 ${\rightarrow}$ 227.1, 166.1 are used for quantitative analysis of biotin. The linearity was over $R^2=0.999$ in range of $5{\sim}60{\mu}g/L$. For purification, chloroform was used as a solvent for eliminating lipids in milk formula. The linearity was over 0.999 in range of 5~60 ng/mL. The detection limit and quantification limit were 0.10, 0.31 ng/mL. The accuracy and precision of LC-MS/MS method using CRM were 103%, 2.5% respectively. Optimized methods were applied in sample analysis to verify the reliability. All the tested milk formulas were acceptable contents of biotin compared with component specification and standards for nutrition labeling. The standard operating procedures were prepared for biotin to provide experimental information and to strengthen the management of nutrient in milk formula.

  • Purpose : Inhaled corticosteroids (ICS) are used as first-line agents for the treatment of persistent asthma; however, their use is accompanied by apprehension of potential systemic adverse effects. This study aimed to assess the effects of ICS on bone mineral density (BMD) and bone metabolism in children with asthma. Methods : From February 2008 to September 2008, 26 asthmatic children treated with ICS (ICS group), 15 asthmatic children treated with leukotriene receptor antagonist (LTRA) (LTRA group), and 30 healthy children (Control group) were selected from the Korea University Anam Hospital. BMD and serum bone-specific alkaline phosphatase (BALP) levels were measured. The asthmatic children underwent spirometry and methacholine bronchial challenge test. Results : There were no significant differences in BMD in the lumbar spine (P=0.254) and proximal femur (P=0.297) among the 3 groups. The serum BALP levels were significantly higher in both the ICS (P=0.017) and LTRA (P=0.025) groups than in the Control group. None of the parameters pertaining to ICS use, such as the mean daily dose during the last 6 months, the total cumulative dose, duration of use, and age of commencement of use, showed significant correlations with BMD (P>0.05 for all parameters). Conclusions : We demonstrated that a low dose of ICS does not exert any significant adverse effect on bone metabolism in asthmatic children. These findings support the current recommendations with regard to the use of ICS for asthmatic children.

  • In order to compare the effectiveness of different phosphorus forms (pulverized as regranulized) and formulars (compound fertilizers) of fused phosphate and to compare the effectiveness of boron from different forms (straight borax, granulized with fused phosphate and included in compound fertilizer), a field experiment was conducted on a relatively fertile upland soil using soybean as a test crop. Results are summarized as following. 1. Yield of soybean was significantly higher in the plots fertilized with P when compared to check plot (no p). However, no significant differences were observed among the different forms or formulars of fused phosphate, indicating that the regranulization or formulation of compound fertilizer did not influence the effectiveness of phosphorus. 2. The application of boron, regardless of its formular, brought about higher B content in plant tissue and higher grain yield. There was a tendency that boron contained in ragranulized fused phosphate was more effective than straight borax. The results also indicated that when boron is to be included in NPK compound fertilizer, its content should be at the least 1.5% as $B_2O_3$. 3. The effectiveness of boron appeared as increased number of pod, higher B, and Ca contents in plant and the amounts of uptake of these elements by soybean. 4. The analysis of soil after the experiment indicated that different forms or formulars of fused phosphate were similarly effective in increasing the available P, Ca, and Mg contents in the soil. 5. Slight increase in available B contents in the experimental plot soils applied with boron fertilizer was observed after the trial.
