Comparison of Pollination Efficiency on Different Pollination Methods in Yellow poplar (Liriodendron tulipifera) (백합나무의 인공교배 방법에 따른 교배 효율성 비교)

  • Yellow poplar (Liriodendron tulipifera L.) is an insect-pollinated tree species with large, perfect flower, and its seed sets average only about 10 percent naturally. In its controlled pollination, pollination bags are usually taken to prevent unwanted pollination, but bagging is an expensive and time-consuming process. Therefore, this study was conducted to determine the need of pollination bag by estimating how much unintended pollination would occur when different cross methods were applied. Five different pollination methods were applied as follows: 1) natural open pollination (i.e. insect pollination) as a reference, 2) self-pollination; no removing reproductive organs with bagging, 3) open pollination; emasculated(removing sepal, petal and stamen) without bagging, 4) controlled pollination; emasculated with bagging and 5) controlled pollination; emasculated without bagging. Very low value of full seed rate (0.2%) was observed in method 3, it was suggested that removing stamen and petal restrict the activity of pollen vectors like bee. Difference in the full seed rate between method 4 and method 5 was not significant (27.9% versus 24.0%, respectively). Consequently, controlled pollination without bagging might be an alternative method for extensive breeding and mass production of seeds in yellow poplar.

Development of the Artificial Insemination Instrument of Bumblebee Queens (뒤영벌 인공수정기 개발)

  • An artificial insemination instrument of bumblebee queens was firstly developed. This instrument consists of location tube, transfer tube, holding tube, head product, and probe apparatus for reproductive tract etc. This instrument was designed to minimize stress and damage of reproductive tract of bumblebee queens. The regulator handle apparatus in artificial insemination instrument was used the principle of lever, that manipulates easy, accurate and rapid insemination of bumblebee queens. By using this instrument, the insemination rate was over 90%. This instrument will be useful for the breeding and conservation of excellent character of bumblebees.

  • 진주조개(Pinctada fucata martensii)는 일본의 남부지역에만 분포하는 종으로 양질의 진주를 생산하는 주된 양식 대상종이다. 우리나라에서는 1960년대 일본으로부터 이식하여 양식하고 있으나, 자연채묘에 의한 종묘 수급에 어려움을 겪고 있다. 이러한 어려움을 해결하기 위해서는 인공 종묘생산이 이루어져야 하며, 이의 확립을 위해서는 건강하고 우량형질을 지닌 어미와 함께 원활한 유생의 공급이 필요하다. 그 한가지 방법으로서 발생배의 대량 냉동보존을 들 수 있는데, 만일 진주조개 배우자의 냉동보존 기술이 확립된다면, 연중 인공 종묘생산과 선발육종이 가능해지고 우량종의 유전자를 보존할 수 있게 된다.   • 가무락조개, Cyclina sinensi(Gmelin)는 백합과에 속하는 식용 이매패로서 한국, 중국, 일본의 연안에 분포한다. 특히, 우리나라에서는 서ㆍ남해안 해역의 니질에 서식하며(Yoo, 1976), 지금까지 가무락조개에 관한 연구ㆍ조사는 일본 히로시마산 가무락조개의 초기발생(Haragaki, 1966)과 한국산 가무락조개의 인공산란유발ㆍ인공수정 및 성장(Choi, 1971; Choi and Song, 1973; Choi, 1975), 생식주기(Lee and Cho, 1985) 등이 정성분석으로 보고되어 있을 뿐 정량적 분석에 의한 정확한 산란기 등이 제대로 밝혀져 있지 않은 실정이다. Effect of Zebularine on Chromosomal Association between Meiotic Homoeologous Chromosomes in Wheat Genetic Background (Triticum aestivum L.) (제부라린이 생식세포분열 동안 동조 염색체 사이의 염색체 접합에 미치는 영향)

  • The objective of this study was to identify the effect of zebularine, a DNA methylation inhibitor, on the chromosomal association between homoeologous chromosomes in the wheat genetic background. Zebularine at a final concentration of 10 µM was used to treat the spikes of the double monosomic wheat addition line (DMA) with one Leymus mollis chromosome and one Leymus racemosus chromosome, both of which were in a homoeologous relationship. In late prophase, zebularine led to chromosome breakage in the Leymus homoeologous chromosomes. Chromosome breakage caused an increase in the frequency of chromosomal associations between the Leymus homoeologous chromosomes. Ordinary DMA showed 65 cells (35.3%) with chromosomal associations and 119 cells (64.7%) with no association, whereas treated DMA showed 102 cells (60.0%) with chromosomal associations and 67 cells (39.4%) with no association. In diakinesis, the Leymus bivalent showed a chromosomal association in the whole euchromatic region. In metaphase, the Leymus bivalent showed association in the whole chromosomal region, unlike other Leymus bivalents with partial chromosomal association. Chromosomal association by chromosome breakage occurred not only between Leymus chromosomes but also between Leymus and wheat chromosomes. The frequency of other chromosomal association (such as fusion and insert) was increased. Chromosome breakage by zebularine treatment is a useful method at the chromosome level as the spores with others are hereditary stable, although the homologous index (h) was not significantly different between ordinary DMA and treated DMA. It is necessary to study how to control zebularine treatment with a more stable concentration for chromosome breakage during meiosis.

Tolerance of Rice, Soybean, and Hot Pepper to Simulated Acid Rain at Different Growth Stages (벼, 콩, 고추의 생육시간별 인공 산성비에 대한 내성)

  • To compare the tolerance of crops to acid rain at different growth stages a simulated acid rain (SAR) of pH 2.7 was applied to rice, soybean, and hot pepper from vegetative growth stage to harvest (Veget.-Harvest) and from reproductive growth stage to harvest (Reprod.-Harvest). Visual damages of crops by SAR were greater in the order of rice < hot pepper < soybean and greater at Veget.-Harvest than at Reprod.-Harvest treatment. Chlorophyll content of all crops was greater in the order of Veget.-Harvest < Reprod.-Harvest treatment < control, but photosynthetic activity was not affected by SAR treatments. Nitrogen concentration and uptake of rice plants at harvest were similar among SAR treatments, but those of soybean and hot pepper were greater at Veget.-Harvest treatment than at Reprod.-Harvest treatment or control. Sulfur concentration of all crops was not affected by SAR treatments, but total sulfur uptake of soybean was greater in SAR treatments than untreated control. Grain yield of rice and soybean was not affected by SAR although grain fertility, percent ripened grains, and 1,000-grain weight of rice at Veget.-Harvest treatment were lower compared with Reprod.-Harvest treatment or control. Fruit dry weight of hot pepper was greater in the order of Veget.-Harvest < Reprod.-Harvest < control due to decreased fruit number per plant and average fruit weight. At one time application of SAR at flowering stage, brown spots were observed on the spikelets of rice at below pH 2.3. Petals of soybean and hot pepper were wilted at pH below 1. 7 and 2.0, respectively, but fruit setting was not affected by the pH of the SRA.
