  • In this study, yeast artificial chromosome Insert (YAC) harboring genes which related xylan metabolism was constructed by using chromosome manipulation technique. For efficient chromosome manipulation, each splitting fragment (DNA module) required for splitting process was prepared and these DNA modules were transformed into Saccharomyces cerevisiae strain YKY164. By two-rounds chromosome splitting, yeast chromosome VII (1,124 kb) was split 887 kb-YAC, 45 kb-mini YAC and 198 kb-YAC and YKY183 strain containing 18 chromosomes was constructed. Splitting efficiency for chromosome manipulation was 50- 78% and expression level of foreign genes on 45 kb-mini YAC and enzyme activity were indistinguishable from that of the YKY164 strain. Furthermore, xylan-degraded products by recombinant enzymes were confirmed and mini-yeast artificial chromosome maintained stable mitotic stability without chromosome loss during 160 generations.

  • The most problems of domestic sewage for BNR(Biological Nutrients Removal) process are deficiency of carbon source and low C/N ratio. Primary sludge fermentation is seemed to be one of the best solutions producing biodegradable organic substrates. Soluble organic materials from sludge fermentation are mainly SCFAs(Short-Chain Fatty Acids) with 2~5 carbon atoms. In this research, it was attempted to apply $A_2/O$ process with the side-stream acid fermenter to improve the nutrients removal efficiency. The result showed that proper SCFAs production is about 3.000mg/L with SRT of 4~5 days. SCFAs yield of approximately 0.10~0.16 mg SCFAs(as COD) per mg of primary sludge(as COD) were achieved. The ratio of acetic. propionic. butyric and valerie acid were 1, 0.7, 0.5 and 0.6. Significant improvements of nutrients removal over 70% in BNR process were observed. thus will reduce the demand for chemical dosing to increase nutrients removal efficiency. When the fermentate was entered $A_2/O$ process, the ratio of phosphate release to substrate uptake amounts to $0.34gPO_4-Pg^{-1}COD$.

  • Gated SPECT can evaluate the regional wall motion of the heart. We evaluated the regional wall motion of the perfusion abnormality in conventional perfusion SPECT with gated SPECT. In case of suspicious perfusion abnormalities, we tried to differentiate the artifact from true abnormality in coronary vascular disease using gated SPECT. We thought that artifacts would have normal wall motion, whereas fixed defects with decreased wall motion would probably represent coronary artery disease. A total of 275 patients who were performed coronary angiography and T1-201 rest/Tc-99m MIBI dipyridamole stress gated SPECT within 2 months were enrolled. In coronary angiography, stenosis more than 50% was considered as coronary artery disease. After injection of 111MBq T1-201 rest image was obtained on triple head SPECT system. 370MBg Tc-99m MIBI was used for the stress image. Eight-frame per-cardiac-cycle gated Tc-99m SPECT studies were done. All the images were analyzed visually. Using perfusion SPECT, the overall sensitivity and specificity were 87% and 55% respectively. Regarding artery territory, sensitivity and specificity were 68% and 73% for left anterior descending artery(LAD), 62% and 78% for right coronary artery(RCA), 42% and 90% for left ciramflex artery(LCX). Using gated SPECT, the overall sensitivity and specificity were 87% and 66% respectively. Sensitivity and specificity were 68% and 78% for LAD, 62% and 79% for RCA, 42% and 90% for LCX. Among 21 false positive cases in perfusion SPECT, 5 cases were interpreted as true negative with gated SPECT. We conclude that gated SPECT provides a valuable adjunct to perfusion SPECT in characterizing perfusion abnormalities and to improve specificity.

  • Animal glue is a traditional material used extensively as adhesive and binder in mother-of-pearl, wooden structure, traditional painting, etc. Analysis of animal glue is usually performed with IR(infrared spectroscopy) based on the IR absorption of functional group. But, it has a limitation in confirming animal glue when a sample consists of several materials because of overlapping of the absorption band. Py/GC/MS(pyrolysis/gas chromatography/mass spectrometry) is a useful tool in analyzing the constituent of polymeric materials like animal glue by identifying their pyrolysate with very small amount of sample. In this study, confirmation of animal glue in a Dancheong sample was tried with this method. Characteristic pyrolytic compounds of animal glue and tung oil used in Dancheong were identified. Dancheong sample painted with Noerok as a coloring material, animal glue and tung oil was prepared and it was possible to find characteristic peaks of animal glue after thermal degradation and artificial weathering experiment. From this, we found that animal glue can be detected using py/GC/MS in cultural heritage samples consisting of several materials and in different condition. IR was also tried to analyze Dancheong sample and the results were compared with those of py/GC/MS for the detection of animal glue.

  • As a member of ectomycorrhizal fungi, Tricholoma matsutake has a symbiotic relationship with its host, Pinus densiflora. To cultivate T. matsutake artificially, the co-cultivation of T. matsutake mycelia and bacteria from shiro was introduced. In this study, bacteria were isolated from soil samples in Bonghwa-gun, and seven bacterial isolates (B22_7_B05, B22_7_B06, B22_7_B07, B22_7_B08, B22_7_B10, B22_7_B13, and B22_7_B14) promoted the growth of T. matsutake mycelia (147.48, 232.11, 266.72, 211.43, 175.17, 154.62, and 177.92%, respectively). Sequencing of the 16S rRNA region of the isolated bacteria was performed. B22_7_B05 and B22_7_B10 were identified as Bacillus toyonensis, B22_7_B06 and B22_7_B08 as Paenibacillus taichungensis, B22_7_B07 and B22_7_B14 as P. gorilla, and B22_7_B13 as P. odorifer. These bacterial isolates were associated with the shiro community and are expected to contribute to the cultivation of T. matsutake.

  • In this study, emotion-classification gathered from users was performed, classification-experiments were then conducted using SVM(Support Vector Machine) and K-means algorithm. Total 15 numbers of channels; CP6, Cz, FC2, T7. PO4, AF3, CP1, CP2, C3, F3, FC6, C4, Oz, T8 and F8 among 32 members of the channels measured were adapted in Brain signals which indicated obvious the classification of emotions in previous researches. To extract emotion, watching DVD and IAPS(International Affective Picture System) which is a way to stimulate with photos were applied and SAM(Self-Assessment Manikin) was used in emotion-classification to users' emotional conditions. The collected users' Brain-wave signals gathered had been pre-processing using FIR filter and artifacts(eye-blink) were then deleted by ICA(independence component Analysis) using. The data pre-processing were conveyed into frequency analysis for feature extraction through FFT. At last, the experiment was conducted suing classification algorithm; Although, K-means extracted 70% of results, SVM showed better accuracy which extracted 71.85% of results. Then, the results of previous researches adapted SVM were comparatively analyzed.

  • The soft contact lens was very simple technique in handling, good sensation in fitting, good effect of a beauty, and good attachment state on the cornea in physical movement. So that, the subjects have used the correct of visual acuity. If the contact lens handling have not skill, they have acquired the side effect on eye. To analysis of a study of ultrastructure on soft contact lens surface of incubated P. aeruginosa. We have observed the soft contact lens surface by SEM. We have founded the good technical method. On the method of sample process of manufacture, the best observation of samlpe tissue was $OsO_4$ postfixation and tannic acid treatment. In this case, P. aeruginosa was a rod shape and one cilia in ultrastructure and the identification was very good. But, On the process of manufacture have not used the $OsO_4$ and tannic acid treatment, this tissue sample appeared the foreign body materials and artifacts, and the identification of the P. aeruginosa was very difficult.

  • When used for amplicon sequencing, Illumina platforms produce more than hundreds of sequence artefacts, which affects operational taxonomic units based analyses such as differential abundance and network analyses. Nevertheless it has become a major tool for fecal microbial community analysis. In addition, results from sequence-based fecal microbial community analysis vary depending on conditions of samples (i.e., freshness, time of storage and quantity). We investigated if freeze-drying samples could improve quality of sequence data. Our results showed reduced number of possible artefacts while maintaining overall microbial community structure. Therefore, freeze-drying feces prior to DNA extraction is recommended for Illumina-based microbial community analysis.

  • This study aims to elucidate the relative importance of geological characteristics, soil slope, and rainfall intensity in relation to soil erosion. To this end, indoor rainfall simulation experiments were carried out under different conditions of rainfall intensities, soil slope, and geological characteristics. The test results show that the factors affect soil erosion in the order of soil slope > rainfall intensity > organic content in the soil. Erosion rates were proportional to rainfall, and increase with increasing clay content. Therefore, the soil erosion rate increases strongly with increasing organic content and clay content. The results show that the soil erosion rate in areas of metamorphic rocks shows a marked increase compared with areas of steep slope and sedimentary rocks. These results indicate that the geological characteristics to produce soil are closely related to sedimentation before and after erosion, providing basic information for the development of models to predict soil erosion rates.

  • This study was carried out to investigate the effects of artificial light sources on growth, yield, and glucosinolate content of hydoroponically grown Peucedanum japonicum in plant factory. Treatments were given with LED Blue:White(1:1, B:W), LED Red:Blue:White(2:1:3, RBW), and LED Blue:White(1:1)+Florescent lamp(BW+FL). Number of harvested leaves and leaf weight of BW+FL were higher than BW and RBW. BW+FL in leaf length and RBW in leaf width were significant difference with other treatments. Chlorophyll content and 'L' value were not significant difference among the treatments. The 'a' and 'b' value is the lowest in BW+FL. Glucosinolate content was high in order of glucobrassicin, glucoiberin, sinigrin, gluconasturtiin, progoitrin, glucoraphamin, and epiprogoitrin in all treatments, and total glucosinolate content was the highest in RBW treatment. Moisture, crude protein, crude fat, and ash content of leaves were not different among the treatments. In conclusion, this study showed that light caused growth and secondary metabolites synthesis, and we recommend to further study between light and secondary metabolites for increasing functionality.
