For the purpose of application for biomass of Pleurotus eryngii, the optimum culture condition were tested. It was found that the optimum culture condition for spot culture of pleurotus eryngii were 24$^{\circ}C$ for 18 days with PDA medium. And the optimum culture condition of bioreactor for biomass were pH 5.5, 18$^{\circ}C$ and 27 days with PDMP broth. It was possible to artificial cultivation of mycelial from Pleurotus eryngii using bioreactor for biomass under the optimum conditions, and it was also possible for Pleurotus eryngii biomass because the forming of fruiting body when Pleurotus eryngii was cultivated using mass artificial cultivated mycelial in the bioreactor.
The optimal temperature for mycelial growth the F. fomentarius was $30^{\circ}C$ and the range of the temperature for mycelial growth wsa about 10~30. The optimal pH for the growth was 4.0. The percentage of weight loss percentage wsa 17.4%. The percentage of WEC extractives wsa increased to 2.24%. The observation of micromorphological showed that the detected cell wall were erosive and thinning as typical degradation pattern of white-rot fungi.
To compare the morphological and cultural characteristics of Cordyceps militaris, forming conditions of artificial fruiting body production were investigated in this experiment. The stroma shapes of artificial fruiting body were confirmed to club and/or stick. However, both shapes were same color as a orange. Perithecia of stroma was semi-egg shape and was ranged from $30{\sim}90{\times}90{\sim}130\;{\mu}m$ and it's asci was narrowly cylindric. Ascospore of perithecia was belong to filiform and multiseptate. The conidia of C. militaris was centurally grown and shaped with globose, long clavate type, floccose, centrally raised during anamorph stage. When it was cultured between glass and PP bottle, production of artificial fruiting body, pinheading ratio and total yield were higher in PP bottle. The optimum illumination was 1,000 lx for the those production. The culture medium of barley, wheat and hulled rice showed higher artificial fruiting body compared to that of silkworm. Pinheading and yield of it's isolates was decreased at more than three subsequent transculture.

We threat sludge by heating methods with low pollution and high efficiency. Organic and inorganic components in Sludge can be almost removed through the heaging treatment process, and thefinal products are called artifical aoil or artificial media which are depended on temperature control. It can be recycled on the agricultural sites. Because it contained sort of organic matters while high heaging process with addition. To use them as agricultural materials, it need to know their characteristic transform in the nature. So we have planting bean and corn for two years with standard planting methods and to anticipate changing characteristics of artificial media and artificial soil by staying it natural condition, analyze it physical and chemical characteristics. This study will be contributed to reduce mass enviornmental problems by the treatment of Sludge and make it possible application for a agriculture use.
The embryos(6-8mm) isolated from seeds of Diospyros kaki were cultured on Murashlge-Skoog(MS), Woody Plant Medium(WPM), Campbell Durzen(CD), Lictvay's Medium(LM), Kao-Michaluk(KM), Nitsch, White, Heller, Wolter-Skoog(WS) media. The results showed that MS and WPM media were most suitable to the development of embryos into plantlets with length of $5.4{\pm}1.2 cm$ and 5-6 leaves. However, when LM and KM media were used, the addition of 1 to $2{\mu} moles/\ell GA_3$ was required for the germination of the embryos. Superoxide dismutase (SOD) activities, one of the changing factors in leaves according to physiological status displayed to be exceptionally significant in the leaves of plantlets germinated from seeds in potting sand soil contrary to those of cultured embryos specially around germination period.
In order to develop effective acclimatization methods for Aralia elata plantlets regenerated from somatic embryos, various acclimatizing conditions were compared regarding both survival rate and growth of the plantlets. The plantlets were transplanted into plastic boxes containing artificial soil in the presence of either several levels of MS liquid media, distilled water, 2% sucrose or 0.1% hyponex solution. They were then cultured by spraying of distilled water twice a week and maintained in the normal tissue culture room. Perlite was proved to be better than vermiculite on survival rate and growth of the plantlets. As the size of perlite (larger than 0.2 cm in diameter) increased, both the survival rate and growth of the plantlets improved. Among the various MS liquid media and different aqueous solutions tested, distilled water appeared to result in the best survival rate and growth. MS media were also effective in increasing survival rate and supporting growth when diluted to 1/4 and/or 1/8. The acclimatized plantlets could be transplanted directly onto the nursery bed and grown normally. The above results suggest that plantlets regenerated from somatic embryos of Aralia elata be effectively acclimatized using a plastic box containing perlite with distilled water treatment.
This study was carried out to obtain the basic data on artificial culture of Lampteromyces japonicus. Favorable media for mycerial growth were glucose peptone medium, malt yeast extract, yeast malt peptone, potato dextrose medium. The optimum conditions for the mycelial growth were $30^{\circ}C$ and pH 6.0. Dextrose as a carbon source was favorable to mycelial growth. The optimal dextrose concentration was 1.2%. As nitrogen sources, yeast extract, $(NH_4)_2HPO_4$ and 0.2% for glutamine. The mineral salts of $Al_2(SO_4)_3{\cdot}14H_2O$ were effective and the optimal concentration was 0.1 M.
A Gram (-), rod-shaped bacterium in size of 1.3∼$1.8{\times}0.35{\mu}m$ inhibiting the growth of cyanobacterium (Ana-baena cylindrica) was isolated and designated NG-2 in this manuscript. This isolate showed positive reactions for catalase and oxidase, and optimal growth conditions of 35∼TEX>$40<^{\circ}C$ and pH 9.0. In a mixed-culture of A. cylindrica and the isolate, each microorganism grew inverse-proportionally, and the cyanobacterial vegetative cells almost completely disappeared within 24 hours. NG-2 lysed A. cylindrica only under light, which means that lytic activity of NG-2 was dependent on the photosynthetic activity of host. When observed under phase contrast microscope, the isolate lysed vegetative cells of A. cylindrica in scattered state in a liquid medium, whereas het-erocysts have not been lysed. When cyanobacterial cell walls have been lysed partly, NG-2 attatched around A. cylindrica filament and formed colony, then encouraged complete lysis of cyanobacterial cells. The isolate showed similar lytic activity in natural water as in an artificial medium. And lytic activity of NG-2 was enhanced when attached on expandable polystyrene bead.
Isoates with changed pathogenicity were selected among iprodione-resistant Alternaria mali to investigate any relationship between their pectolytie enzyme activity and pathogenicity. In artificial medium, strongly pathogenic isolates $S_1$, and $R_3$ showed higher enzyme activity than weakly pathogenic isolate $R_8$.Activity of endo-polymethylgalacturonase and end-o-polygalacturonase was more than 3 times. But activity of pectin methylesterase and pectin Iyase by isolaste $S_1$ was higher than those by $R_3$ and $R_8$ isolates. In apple medium dialyzed against distilled water, activity of enzyme by each isolate was increased but growth of each isolate was reduced. When iprodione was added to the medium, enzyme activity and growth of isolate $S_1$, were reduced but strongly pathogenic isolate .$R_3$ among iprodione-resistant ones showed increased enzyme activity except for exo-polygalacturonase in dialyzed apple medium.
This experiment was carried out to find the method for mass production by artifical cultivition and to analyze the components of Paecilomyces japonica according to several media. Time of inoculation of the Paecilomyces japonica using silkworm was on first day of five molting and infection rate was 72.0%. Optium medium for mass production of the Paecilomyces japonica was known effective for increasing dry weight and fruitbody at brown rice 80g plus pupa powder 20g. Dry weight of Paecilomyces japonica using fungus of silkworm was 1.2g including pupa and length of fruitbody was appeared 3.0cm to 3.5cm. Content of $\beta$ - glucan was very high as 40.5% at inoculation on the first day of the five molting while 16.4% at brown rice, 20.7% at pupa, 23.1% at brown rice plus pupa powder, and 28.7% at pine sawdust plus wheat bran. Mycelium was poor and pinkly conidiospore was formed on media of centipede, maggot and powder of silkworm.
