  • 효모 (Saccharomyces Cervisiae)는 단일 세포의 형태로 존재하는 진핵 세포로써 동물세포와 유사한 기작으로 분비성 단백질을 생성한다. 따라서 박테리아와 달리 효모를 이용하면 당단백질이나 이황결합을 포함하는 분비성 단백질을 경제적으로 대량 합성할 수 있다. 효모의 필수 단백질 중 하나인 단백질 이황이성질화 효소는 소포체에 위치하며 분비성 단백질에 구조적으로 안정한 이황결합을 제공하는 효소이다. 본 연구는 단백질 이황이성질화 효소 (protein disulfide isomerase)가 지니고 있는 두 개의 활성도메인 중 분비성 단백질들의 합성 효율에 직접적으로 관여하는 부위를 찾는 연구이다. 효모 유전체로부터 단백질 이황이성질화 효소의 유전자 (PDI1)을 제거하고 효소의 변이 유전자를 주입한 후 효모의 성장 속도를 측정하였다. 또한 효모의 대표적 분비성 단백질을 각 변이 효소를 지니는 효모에 과발현시켜 합성 및 이황결합 형성 효율을 측정하였다. 단백질 이황이성질화 효소내 두 개의 활성 부위 중 아미노 말단쪽에 위치한 a 도메인에 있는 활성 부위가 분비성 단백질의 활성에 중요한 역할을 한다는 것을 알 수 있었다. 이 결과는 이황결합이나 당을 포함하는 외래 단백질의 고효율 합성을 위한 새로운 효모종 개발에 중요한 정보를 제공할 것으로 기대 된다.

Mass Spectrometry-Based Strategy for Effective Disulfide Bond Identification (질량분석기를 활용한 효과적 이황화결합 분석법 개발)

  • The determination of disulfide bonds is important for comprehensive understanding of the chemical structure of protein. So far, many strategies for the disulfide bond analysis have been suggested in terms of speed and sensitivity. However, most of these strategies have not considered free thiol residues in the target protein in the process of determining the disulfide bond. We suggested the strategy which was composed of four steps for the identification of disulfide bonds; the first step was the prediction of possible disulfide bonds, the second step was the determination of free cysteine residues, the third step was the analysis of disulfide bond using a high-resolution mass spectrometry, and the final step was the determination of disulfide bonds based on the comprehensive verification. In this study, we performed the characterization of disulfide bonds for the recombinant protein (HRPE1), where 1 and 5 inter- and intra-chain disulfide bonds were identified, respectively.

Toegye Lee Hwang's Assessment on Iljae Lee Hang's Study - Focusing on Sung Confucianism (일재(一齋) 이항(李恒)의 학문에 대한 퇴계 이황의 평가 - 성리설을 중심으로 -)

  • This article is to review academic aspects of Iljae Lee Hang through Toegye Lee Hwang's comments. Iljae Lee Hang (一齋 李恒: 1499~1576) is a representative Neo-Confucian scholar in Honam area in 16th century. His Sung Confucianism was known to Toegye Lee Hwang by Gobong Gi Dae Seung and consequentially received attention from academic world. Lee Hang's Sung Confucianism, however, has hardly drawn attention since 17th century due to Lee Hwang's negative assessment. Impeaching Lee Hang's academic attitude and methods, Lee Hwang evaluated him as having many problems. Lee Hwang criticized that Lee Hang studied Neo-Confucianism with no great effort and he was so much confident to say 'Logic of the world does not deviate from this' based on Chengzi and Zhuzi's saying which was only appealing to his ideas. Lee Hang actually cited theories of Chengzi and Zhuzi and stressed self-complacency when necessary, which therefore made him fail to exhibit consistency. Iljae partly brought Lee Hwang's criticism upon himself. Lee Hwang's negative assessment is not to be literally accepted but his assessment is helpful indeed to critically examine Lee Hang's Neo-Confucianism.

  • We see the term in Tosan 12 songs and other poems by Leehwang. The term means 'Birds fly in the sky, Fish swim in the water.' It is important I think in that it is the point of contact between his poems and thought. There is a gap between original text of , [The way of mean (중용)] and Leehwang's understanding. It is same with Buddhism and Taoism as confucianism in the idea that we cannot describe the Ultimate into the human language. is the very confucian way of that. But Leewhang has the peculiar idea that it can't pass over the Moral state even though its meaning is over the human language.

Heat Effects for the Volatile Organic Compounds emitted from Garlic and Kimchee (열처리가 마늘과 김치에서 발생되는 휘발성 유기화합물에 미치는 영향)

  • The volatile organic compounds(VOCs) emitted from raw garlic and Kimchee were analyzed with thermal desorption or purge & trap/gas chromatography/mass selective detection method. Very offensive compounds such as methyl allyl sulfide, dimethyl sulfide, dimethyl disulfide, diallyl sulfide, methyl allyl disulfide, diallyl disulfide and dimethyl trisulfide were detected, and among them, dimethyl disulfide and dimethyl trisulfide were confirmed to be generated during the precocity of Kimchee or emitted from the stuff of Kimchee other than the garlic. Malodorous compounds emitted from the garlic or Kimchee were detected in the breath of a Korean and the refregirator keeping Kimchee. It was confirmed that the disufides emitted from the garlic or Kimchee were major components of offensive odor in the alveolar air and the refregirator. It was clarfied that heat process is very effective to reduce odorous VOCs in garlic or Kimchee.

Yu Won-Ji's Philosophical Theory and His Vindication of Toegye's Learning (졸재 유원지의 심성론과 퇴계학설 옹호)

  • This paper is intended to present the philosophical thoughts of Yu Won-Ji who was one of famous scholars in The School of Toegye. He criticized Yi I(Yulgok) and Jang Hyeon-Gwang(Yeohyeon) because they regarded Li and Ch'i as one. Yi was considered an actual founder of their theoretical and political rival party called the School of Yulgok. Jang also refused to accept Toegye's theory and had developed his own theory, which emphasized the unity of Li(principle) and Ch'i(material force). Therefore Yu might well exclude Yi I(Yulgok) and Jang Hyeon-Gwang and criticize their theories severely. First, he was convinced that Toegye's doctrine(Li-Gi-Ho-Bal-Seol) was a orthodox theory accurately succeeding to Jhu Xi's learning. Secondly, he criticised Yi I(Yulgok) for looking upon Li and Ch'i as one thing. Thirdly, he regarded Jang's doctrine as same with Yulgok's. Fourthly, he thought it is necessary to criticize Jang's theory in order to preserve Toegye's learning and establish the theoretical identity of the Toegye School.

Evaluation of Catalyst Deactivation and Regeneration Associated with Photocatalysis of Malodorous Sulfurized-Organic Compounds (악취유발 황화유기화합물질의 광촉매분해에 따른 촉매 비활성화와 재생 평가)

  • This study evaluated the degradation efficiency of malodorous sulfurized-organic compounds by utilizing N- and Sdoped titanium dioxide under visible-light irradiation, and examined the catalyst deactivation and regeneration. Catalyst surface was characterized by employing Fourier-Transform-Infrared-Red (FTIR) spectra. The visible-light-driven photocatalysis techniques were able to efficiently degrade low-level dimethyl sulfide (DMS) and dimethyl disulfide (DMDS) with degradation efficiencies exceeding 97%, whereas they were not effective regarding the removal of high-level DMS and DMDS, with degradation efficiencies of 84 and 23% within 5 hrs of photocatalytic processes. As compared with DMS, DMDS which containes one more sulfur element revealed quick catalyst deactivation. Catalyst deactivation was confirmed by the equality between input and output concentrations of DMD or DMDS, the obsevation of no $CO_2$ generation during a photocatalytic process, and the FTIR spectrum peaks related with sulfur ion compounds, which are major byproducts formed on catalyst surfaces. The mineralization efficiency of DMS at 8 ppm, which was a peak value during a photocatalytic process, was calculated as 144%, exceeding 100%. The catalyst regenerated by high-temperature calcination exhibited higher catalyst recovery efficiency (53 and 58% for DMDS and DMS, respectively) as compared with dry-air and humid-air regeneration processes. However, even the calcined method was unable to totally regenerate deactivated catalysts.

  • 셀룰로오스계 재생섬유 중 가장 먼저 개발된 Viscose rayon은 셀룰로오스 펄프를 17-18%의 가성소다 수용액에 팽윤시켜 알칼리 셀룰로스를 만든 뒤 이황화탄소(CS$_2$)에 반응시킨 후 방사하여 제조하며 흡습성, 제전성, 드레이프성 및 광택이 우수한 쾌적성 섬유이다. 그러나 탄성 회복율이 좋지 않으며 강력이 적고 특히 습윤시의 강력 저하의 단점이 있을 뿐만 아니라 제조과정에서 사용하는 이황화탄소의 환경 유해성이 큰 문제점으로 지적되고 있어 선진국들이 점차 설비를 폐쇄하거나 개도국으로 이전하고 있는 실정이다.   • 본 연구의 목적은 최인호의 "유림"에 나타난 유가사상이 실존인물을 중심으로 전개되는데 주목하여 유가사상의 본질이 무엇인지 밝히고, 이를 통해 탈근대의 단초를 제시함에 있다. 연구를 통해 알 수 있는 사실은 '예'가 동아시아인을 넘어 세계인의 사상이 될 수 있다는 사실이다. 이는 이황과 이이, 이황과 기대승의 담론을 통해 확인할 수 있다. 유가사상은 상생과 공존을 제시한 레비나스의 '타자성의 윤리'에 맞닿아 있는 것이다.
