  • In this work, a nano-sized samaria-doped ceria(SDC) was prepared by a urea-based hydrothermal method and characterized by XRD, FESEM and TEM. It was observed that the increase in synthesis time and temperature gave rise to crystallity and particles size. Moreover, the synthesised powders had a excellent ion-conductivity(0.1 S/cm at 600~$800^{\circ}C$) which is suitable for electrolyte of intermediate temperature-solid oxide fuel cell(IT-SOFC).   • Flounder is one of the major aquacultured fish and of an economically important item in Korean fisheries. Recently, there are trends of research worldwide that aim to analyze and characterize a whole genome or a whole proteome of interesting species. The data are utilized for the understanding and development of preventive and curative technologies for the serious diseases. However, there are very limited information of proteome for marine organisms, we optimized first the conditions for two-dimensional polyacrylamide gel electrophoresis (2D-PAGE) with serum form a marine fish, flounder (Paralichthys olivaceus). A pre-treatment of serum and an optimization of protein concentration analyzed were surveyed for enhancing the separation for proteins. A statistical analysis was performed on the overall 1,820 protein spots to overcome the variability among individual fishes.

  • J-chain was purified from human serum IgA by three different methods which involve preparative disc electrophoresis, ion exchange chromatography and water precipitation. It was found that the simplest and satisfactory method was precipitation in water. All the J-chain preparations obtained from the above different procedures showed similar electrophoretic mobility and band-patterns.

  • The distribution of $^{51}Cr\;and\;^{128}I$ recoil species following radiative-neutron capture in chromates, dichromates, iodates and periodates has been investigated by using paper-electrophoresis. In view of the effective recoil energy and the effect of the internal conversion, it is unlikely that an atom which has captured a neutron can remain bound in its original molecule. It is also unlikely that the energy of the recoil atom is dissipated in heating a small region of the crystal. However, the results of paper-electrophoresis separation of recoil $^{51}Cr\;and\;^{128}I$ indicated that many more of the recoil atoms were bound in the parent molecule. The disorder model for the reaction was proposed from observations of retention. In considering cations, the greater their radii, the higher is the probability of the recoil atom breaking through the secondary cage. In ammonium salt, the ammonium ion behaved as a reducing agent in the disorder zone and resulted in low retention. Crystal structures with their greater free space have shown low retention.

  • In this paper, a computer simulation is developed for isotachophoretic protein separation in a serpentine micro channel for optimal lab on a chip design using 2D Finite Element Method. This 2D ITP model is composed of 5 components such as hydrochloric acid as Leader, caproic acid as terminator, acetic acid and benzoic acid as two proteins, and histindine as background electrolyte. The computer model is based on mass conservation equation for 5 components, charge conservation equation for electric potential, and electro neutrality condition for pH calculation. For the validation of the 2D spatial ITP model, the results are compared with the Simul5 developed by Bohuslav Gas Group. The simulation results are in a good agreement in a ID planar channel. This proves the precision of our model. The 2Dproteinseparation is conducted in a 2D curved channel for Lab on a chip design and dispersions of proteins are revealed during the electrophoretic process in a curved shape.

  • Radiochemical synthesis of $N^{G}$-mono[$^{14}$ C-methyl]-L-arginine is described. The compound was synthesized from radio-active mono[$^{14}$ C]-methylamine as easily and purified by strong cation-exchange resin (NH form) liquid chromatography using a gradient of ammonium hydroxide, and crystallized as flavianate. The free amino acid was successfully prepared by strirring its flavianate and strong anion-exchange resin (OH- form), which could remove the flavianic acid from its salt in water below room temperature. Purity of the compound was tested by thin-layer chromatography, thin-layer electro-phoresis, and scintillation spectrometry.y.

  • Black phosphorus (BP) is a potential candidate for an anode in lithium ion batteries due to its high theoretical capacity and the large interlayer spacing in the monolayered phosphorene form, allowing for lithium intercalation/deintercalation. In this study, large-scale exfoliation of bulk BP was accomplished using a solution of NaOH and N-methyl-2-pyrrolidone (NMP), yielding phosphorene, which can be assembled into nanoflakes using electrophoretic deposition (EPD). Through the systematic addition of NaOH and subsequent sonication, BP nanoflakes were obtained in high yields by EPD, allowing for the integration of these nanoflakes into an anode in the film state. Anodes with a charge/discharge capacity of 172 mAh/g at a rate of 200 mA/g were obtained, which are promising for battery applications through various post-film treatments.

Purufication and Characterization of Extracellular Collagenase from Vibrio mimicus (Vibrio mimicus 가 생산하는 collagenase의 정제 및 특성)

  • A collagenase was isolated from the culture filtrate of Vibrio mimicus (ATCC 33658). The enzyme was purified to homogeneity by ammonium sulfate precipitation and DEAE-Sephadex A-50 chromatography, which an activity recovery of 22%. The molecular weight of the purified enzyme was estimated to be 42 kDa by SDS-polyacrylamide gel electrophoresis and gel filtration, indication a monomer structure. The optimum pH and temperature od the enzyme for insoluble collagen (Type I) were around 7.75 and 28$\circ$C, respectively. Some chelating agents and serine protease inhibitor inactivated the enzyme, but L-cysteine and histidine did not affect the activity. The amino acid composition indicated that the collagenase contained high amounts of amino acid residues of glycine and alanine. The K$_{m}$ and R$_{cat}$/K$_{m}$ values for the collagenase, using insoluble collagen (type I) as substrate, were 2.86 mg/ml and 972.28 U/mg-protein, respectively.

Action modes of the anti-complementary polysaccharides purified from Arecae pericarpium (대복피로부터 정제된 보체활성화 다당의 작용양식)

  • Two kinds of complement activating (anti-complementary) polysaccharides, which were expected to be immunomodulators were purified from Arecae Pericarpium (the pericarps of Areca catechu), and their action modes have been studied. The active polysaccharides, AC-2-IIIa and AC-2-IIIc from Arecae Pericarpium showed dose-dependent anti-complementary activities on $TCH_{50}$. The anti-complementary activities of AC-2-IIIa and AC-2-IIIc in metal ion-free condition were completely decreased in comparison with control whereas in case of $Ca^{2+}$-free condition, these activities were maintained, considerably. Also AC-2-IIIa and AC-2-IIIc showed relatively potent alternative complement pathway activities. Furthermore, after incubation of the normal human serum with polysaccharide of Arecae Pericarpium in the absence of $Ca^{2+}$ ion, a cleavage of C3 in the serum was found to have occurred through immunoelectrophoresis (IEP) with anti-human C3. Also, from the results of IEP using anti-human whole serum, the ratios of the height of 3rd peak to ${\alpha}2-M$ peak by AC-2-IIIa and AC-2-IIIc proved to be $1.50{\pm}0.04$ and $1.22{\pm}0.08$, respectively. These results indicate that the modes of complement activation by AC-2-IIIa and AC-2-IIIc from Arecae Pericarpium are via both the classical pathway and the alternative pathway.
