Sperm cryopreservation methods have been improved over the last few decades. However, an optimized thawing rate has not yet been established. Therefore, we investigated the effect of thawing rate on sperm function after cryopreservation. The ejaculates collected from beagle dogs were cryopreserved and then thawed at two different thawing rates ($37^{\circ}C$ for 1 min or $70^{\circ}C$ for 15 sec). The thawed sperm were evaluated for motility, viability, morphology, plasma membrane integrity, phosphatidylserine (PS) translocation, and intracellular $H_2O_2$ level. The sperm thawed rapidly at $70^{\circ}C$ showed improved motility, viability, normal morphology, plasma-membrane integrity and non-PS translocation compared to the sperm thawed slowly at $37^{\circ}C$ (P < 0.05). However, the intracellular $H_2O_2$ levels were not significantly different between the rapid- and slow-thawed sperm (P > 0.05). In conclusion, sperm rapid thawing at $70^{\circ}C$ could improve the function of cryopreserved canine sperm, and the appropriate thawing rate would enhance the quality of the cryopreserved sperm.
본 실험은 ICR계통 생쥐의 8세포배 및 상실배를 Vitrification(VS3) 동결보존액을 이용하여 초급속동결-융해를 실시하여 배반포배까지의 체외발생한 배반포배를 이식하였을 때 수태율에 미치는 영향을 조사하여 다음과 같은 결과를 얻었다. 1. 8세포배의 평형시간(3분 및 6분)에 따른 초급속 동결-융해후 배반포배까지의 체외발생율은 57.6% 및 59.5%여TEk. 2. 상실배의 평형시간(3분 및 6분)에 따른 초급속 동결-융해후 배반포배까지의 체외발생율은 64.4% 및 68.2%였다. 3. 8세포배와 상실배를 초급속 동결-융해를 실시하여 배반포배까지 체외발생한 배를 위임신되어진 수란쥐에 19개 및 20개를 이식하여 7필(36.8%)과 10필(50.0%)의 산자를 얻었다.

Freeze-thaw tests were performed on gneiss samples collected from Terra Nova Bay, Antarctica in order to examine the engineering properties of rocks with slightly weathered (SW) and moderately weathered (MW). The tests were conducted under temperature ranging from $20{\pm}2^{\circ}C$ to $-20{\pm}2^{\circ}C$. A cycle of test consisted of 5 hours of freezing followed by another 5 hours of thawing under full saturation. In this paper, total 200 cycles of freeze-thaw test were performed with measurements of porosity, absorption, ultrasonic velocity, and shore hardness per each 20 cycle and that of uniaxial compressive strength (UCS) per each 50 cycle. The UCS of the SW rocks approximately decreased 0.07 MPa per a single cycle, while that of MW rocks decreased around 0.2 MPa per a single cycle. During the 200 cycles of SW rocks, the absorption increased from 0.23% to 0.39%, the P-wave velocity decreased from 4,054 m/s to 3,227 m/s and S-wave velocity decreased from 2,519 m/s to 2,079 m/s. Similarly, those of MW rocks changed from 0.65% to 1.6%, 3,207 m/s to 2,133 m/s and 2,028 m/s to 1,357 m/s. In conclusion, it was inferred that the properties of SW rocks experienced approximately 200-300 cycles of freeze-thaw process become close to those of MW rocks.
Variations of physical properties such as weight loss rate, wave velocity and uniaxial compressive strength after performing freeze-thaw cyclic test were measured in order to define weathering characteristics of sandstone and andesite. Weight change in specimens of the two rocks decreased with increasing the repetition number of freeze-thaw cyclic test. In particular, weight loss of andesite specimens was very irregular. P-wave velocity of sandstone specimens decreased more than 5%. On the other hand, P-wave velocity of andesite specimens do not vary up to 500 cycles and decreased more than 5% after 1000 cycles. This implies that the sandstone are easily weakened and loosened by weathering processes, while the andesite are relatively strong. In addition, the wave velocity changes of the andesite specimens coincident with the weight change. Uniaxial compressive strengths of the sandstone specimens slightly decreased at the early stage of the freezing-thawing cyclic test, then tended to be irregular after 64 cycles. In conclusion, the rock specimens showed smaller weight loss, less had lower strength reduction rate.

In this paper, the effect of freezing-thawing action on the dynamic modulus and porosity was examined by ultrasonic pulse velocity (UPV) measurement. UPV was measured every 30 cycles during the freezing-thawing test, and dynamic modulus and porosity of cement mortar were calculated by relationship among UPV, porosity and dynamic modulus. Porosity analysis was also performed to compare with calculated porosity by mercury intrusion porosimetry (MIP). From the test, it was found that dynamic modulus of cement mortar was decreased 13% after 300 cycles. The calculated porosity was increased about 30% compared with the initial porosity before freezing-thawing action. The calculated porosity showed similar increase tendency with the porosity measured by MIP. So, it can be concluded that the porosity change of cementitious materials by freezing-thawing action can be predicted by UPV measurement.

It is well recognized that damage resulting from freeze-thaw cycles is a serious problem causing deterioration and degradation of concrete. In general, freeze-thaw cycles change the microstructure of the concrete ultimately leading to internal stresses and cracking. In this study, a new method for one-sided stress wave velocity measurement has been applied to evaluate freeze-thaw damage in concrete by monitoring the velocity change of longitudinal and surface waves. The freeze-thaw damage was induced in a $400{\times}350{\times}100mm$ concrete specimen in accordance with ASTM C666 using s commercial testing apparatus. A cycle consisted of a variation of the temperature from -14 to 4 degrees Celsius. A cycle takes 4-5 hours with approximately equal times devoted to freezing-thawing. Measurement of longitudinal and surface wave velocities based on one-sided stress wave velocity measurement technique was made every 5 freeze-thaw cycle. The variation of longitudinal and surface wave velocities due to increasing freeze-thaw damage is demonstrated and compared to determine which one is more effective to monitor freeze-thaw cyclic damage progress. The variation in longitudinal wave velocity measured by one-sided technique is also compared with that measured by the conventional through transmission technique.

The aim of this study was to overcome some of the limiting factors that the maxi cryopreservation straw of 5 ml presents in processing boar semen. Cryopreservation of semen samples was conducted in 0.5 ml and 5.0 ml straws at two freezing rates: -140℃ in 8 minutes and 30 seconds (FR-1) and -140℃ in 14 minutes (FR-2). The straws were then thawed and the semen parameters were compared by Computer Assisted Sperm Analysis, and sperm morphology and acrosome status were examined by Coomassie blue staining. The effects of different thawing temperatures and durations were also compared, namely 37℃ for 115 sec, 50℃ for 45 sec, or 70℃ for 25 sec. In general, the FR-1 group showed higher (p<0.05) sperm viability and motility than the FR-2 group in the 5.0 ml straws. Compared to other ranges, thawing at 50℃ for 45 sec showed the highest sperm viability and motility (68.4±3.6% and 69.5±2.2%, p<0.05), suggesting that thawing temperature should be adjusted concurrently with freezing rate. Sperm morphology and acrosome integrity did not significantly differ among the groups (p>0.05). The data obtained in this study suggest that improving the freezing-thawing protocol for one artificial insemination dose straws (5.0 ml) retains the sperm's parameters from 0.5 ml cryopreservation, and is more convenient to handle, which could result in enhanced reproductive performance.

본 연구에서는 저온의 순수물 속의 등온 수직원기둥에 의해 야기되는 열전달 특성을 구명함에 있어서 짧은 원기둥 범주에 속하는 종횡비 0.5인 얼음 원기둥을 이용 하여 실험적으로 열전달 특성을 구명하였다. 그리고 전 유동장을 가시화 하였으며, 얼음의 융해율로써 누셀트(Nusselt)수를 측정하여 기존의 결과들과 비교검토하였다.
We have collected shale specimens from the Pungam Basin in Kangwon province and investigated change of physical properties by freezing and thawing in water as well as in acidic fluid. The temperature range was $-20{\pm}2^{\circ}C\~15{\pm}2^{\circ}C$. Specimens were frozen for 12 hours and thawed in water for 8 hours. Then, they were saturated in the vacuum chamber for 4 hours to make specimens fully saturated. This procedure was 1 cycle. We have measured absorption, ultrasonic velocity, shore hardness, slake durability and uniaxial compressive strength at every 5th cycles. The physical properties increased or decreased as freezing and thawing cycles increased. Uniaxial compressive strength decreased by 0.40MPa per cycle in water and by 0.48MPa in acidic fluid. Elastic constant also decreased by 0.21GPa per cycle in water and by 0.30GPa in acidic fluid. Absorption increased by $0.29\%$ and $0.37\%$ per cycle in water and acidic fluid, respectively. These results indicate that decrease in uniaxial compressive strength, elastic constant and absorption by freezing and thawing in acidic fluid is more rapid than in water. Ultrasonic velocities, shore hardness and slake durability show no differences in water and acidic fluid. When we compared our results with the temperatures in the Hongchon during the winter season, $6\~12$ cycles may be equivalent to 1 year.
고로슬래그 미분말을 사용한 콘크리트\ulcorner 수화속도가 느려 어린 재령시 동해의 영향을받기 쉽다. 본 연구에서는 고로슬래그 미분말을 사용한 콘크리트의 동결융해 저항성을 알아보기 위해 고로슬래그 미분말의 치환율과 물-결합재비를 변화시켜 제조한 콘크리트에 대해 동결융해시험을 실시하였다. 또한 동일한 치환율, 물-결합재비의 콘크리트에 AE제를 첨가시켜 동결융해 저항성의 개선효과를 알아보았다. 시험결과 고로슬래그 미분말의 치환율이 증가할수록 동결융해 저항성은 작게 나왔다. 또한 non-AE 콘크리트의 경우 물-결합재비가 51%, 45%일 때 내구성지수는 각각 2.4%, 40.0%이하로 매우 나쁘게 나타났으나, AE콘크리트의 경우 물 -결합재비가 45%와 51%인 콘크리트의 내구성지수는 각각 90.2% 80.9%이상으로 동결융해 저항성이 매우 우수하게 나타났다.
