The anti-hypertensive effect of ginseng in patients with mild hypertension

  • Ginseng has been traditionally used to recover vital energy from Qi deficiency in oriental countries. Recent reports suggested that ginseng could regulate blood pressure (BP), but much controversy still remain. Therefore, we intended to assess the anti-hypertensive effect of some ginseng species on Koreans and Chinese. This is a randomized, double blinded controlled clinical trial. The study subjects were recruited from the mild hypertensive patients who belonged prehypertension(120/80 to 139/89 mmHg) and stage I hypertension (140/90 to 159/99 mmHg) in Korea and China. After assigning the subjects into a Korean, a Chinese, and an American ginseng group by randomization, we prescribed ginseng with the dose of 4.5 g per a day for 4 w. To assess the anti-hypertensive effect, we compared the mean of systolic and diastolic BP between before and after ginseng medication by 24 h Ambulatory Blood Pressure Monitor (24 h ABPM). We also monitored adverse effect and laboratory findings to secure the subjects' safety. There were 64 subjects treated with Korean ginseng, 58 treated with Chinese ginseng, and 64 treated with American ginseng. All of the ginseng species reduced subjects' BP. Especially, Korean and Chinese ginseng showed more excellent effects. The secondary analysis on the subjects' nationality revealed that all of the ginseng species showed more significant anti-hypertensive effect in Chinese than in Koreans. We suggest ginseng could be useful for mild hypertension regardless of its species. And it would be safe within the dosage of 4.5 g per a day.

  • Saengmaeksan (SMS) is a well known Korean traditional herbal prescription, which consists of 3 different herbs, Radix Ophiopogonis, Radix Ginseng, and Fructus Schisandrae. SMS has been commonly used in various disease including respiratory system and cardiovascular System in Korea. The objective of this study is to find out the improving effects of SMS against Dextran Sulfate Sodium (DSS)-induced ulcerative colitis. SMS reduced clinical signs of DSS-induced colitis, including body weight loss, shorten colon length, and increased disease activity index. The results showed that SMS significantly inhibited the activation of nuclear factor-${\kappa}B$ p65 in the colon tissues of DSS-treated mice. In addition, we observed that result showed that the levels of IL-6 in plasma were increased in DSS treated group compared to those of the normal group, but these increased levels were reduced by administration with SMS. Taken together, these findings suggest that SMS has improving effects on DSS-induced ulcerative colitis, which may explain its beneficial effect in the regulation of chronic intestinal inflammation.

  • Due to the advent of the 4th Industrial Revolution, it is imperative that we aggressively continue to develop state-of-the-art, cutting edge ICT technology relative to autonomous vehicles, intelligent robots, and so forth. Especially, systems based on convergence IT are being developed in the form of CPSs (Cyber Physical Systems) that interwork with sensors and actuators. Since conventional CPS specification only expresses behavior of one system, specification for collaboration and diversity of CPS systems with characteristics of hyper-connectivity and hyper-convergence in the 4th Industrial Revolution has been insufficiently presented. Additionally, behavioral modeling of CPSs that considers more collaborative characteristics has been unachieved in real-time application domains. This study defines the non-functional requirements that should be identified in developing embedded software for real-time constrained collaborating CPSs. These requirements are derived from ISO 25010 standard and formally specified based on state-based timed process. Defined non-functional requirements may be reused to develop the requirements for new embedded software for CPS, that may lead to quality improvement of CPS.

Ecophysiological Responses and Subsequent Recovery of the Olive Flounder, Paralichthys olivaceus Exposed to Hypoxia and Iron II.   • The effects of iron on gill tissue and metabolic rate represented by oxygen consumption of olive flounder, Paralichthys olivaceus were determined. The effects were further studied by means of survival rate of the fish exposed to a serial concentrations of iron. The olive flounder exposed to iron concentrations over 0.93 mg/$\ell$ showed curvature and terminal clubbing of gill lamellae at 2 weeks post-exposure. In iron concentration 4.89 mg/$\ell$, gill of the fish were seriously damaged just after 2 weeks, showing hyperplasia of filament epithelia, deformation of lamella epithelia, chloride cell damage, and separation of lamella epithelial layer, Gills exposed to 9.78 mg/$\ell$ iron concentration resulted in fusion and necrosis of the lamellae after 2 weeks. Significant decreases of metabolic rate of the fish were observed after 4 weeks at iron concentration 0,93 mg/$\ell$ and after 2 weeks at iron concentrations over 4.89 mg/$\ell$. Survival rate of the olive flounder decreased significantly after 4 weeks at the iron concentration over 4.89 mg/$\ell$. These results lead us to conclude that, as far as the iron effects are concerned, its concentrations should not exceed at least more than 0.93 mg/$\ell$ in the fish farm and coastal waters for normal growth of the olive flounder.

Gonadal Development and Sex Ratio of Artificial Seedlings of the Oblong Rockfish Sebastes oblongus (황점볼락 Sebastes oblongus 인공종묘의 생식소 발달과 성비)

  • We investigated gonadal development and sex ratio of artificial seedlings of the oblong rockfish Sebastes oblongus, based on samplings for 370 days just after parturition. The primordial germ cells and genital ridge appeared separately under the mesentery in the yolk-sac stage larva (total length: 7.10-7.77 mm) just after parturition. The primordial germ cells and genital ridge integrated to form primordial gonad in 5-day-old larvae (7.12-9.68 mm), and then proliferation of somatic cell and germ cell occurred in the gonad, which was maintained undifferentiated until 45-days after parturition (18.6-20.4 mm). The ovarian differentiation began in the larva of 50-days old (dab) after parturition (dap) (20.0-24.5 mm). The somatic tissues elongated from the both opposite end-sites of undifferentiated gonad were consequently fused and formed a complete ovarian cavity at 60-days old dap (25.5-32.0 mm). In 80-days old dap (37.3-47.2 mm), meiosis of oogonia occurred to be chromatin nucleolus stage oocyte. The perinucleolus stage oocytes appeared at in 130-days old dap (68.0-86.0 mm), and previtellogenic stage oocytes appeared in 370-days old dap (101.0-116.0 mm). Only female was observed in the artificially produced oblong rockfish in the present study. This result revealed the effect of higher temperature on the sex determination of the oblong rockfish..

TALEN Constructs and Validation for Targeting of SETDB1 Genomic DNA (SETDB1 genomic DNA 를 표적하는 TALEN construct 제작 및 분석)

  • TALEN is a newly developed gene engineering method to knock out specific genes. It contains a DNA binding domain and a Fok1 nuclease domain in the TALEN plasmid. Therefore, the engineered TALEN construct can bind to any region of genomic DNA and cut the target nucleotide, thereby inducing mutation. In this study, we constructed two TALEN constructs targeted to a protein initiation codon (DBEX2) or the 25th upstream region (DBPR25) to enable mRNA synthesis of SETDB1 HMTase. We performed the TALEN cloning in two steps. The first step was from module vectors to pFUS array vectors. We confirmed successful cloning with a colony PCR experiment and Esp31 restriction enzyme digestion, which resulted in a smear band and a 1 Kb insert band, respectively The second step of the cloning was from a pFUS array vector to a mammalian TALEN expression vector. The engineered TALEN construct was sequenced with specific primers in an expression vector. As expected, a specific array from the module vectors was shown in the sequencing analysis. The specific module sequences were regularly arrayed in every 100 bp, and SETDB1 expression totally disappeared in the TALEN-DBEX2 transfection. PCR amplification targeting of DBEX2 was performed, and the PCR product was digested with a T7E1 restriction enzyme. The expression of SETDB1 was down-regulated in the TALEN-DBPR25 transfection. Morphological changes were also observed in the two TALEN constructs with transfected HeLa cells. These results suggest that the engineered TALEN constructs in two strategic approaches are very useful to knock-out of the SETDB1 gene and to study gene function.

Different Responses to Arsenic Trioxide between NB4 and UF-1, Acute Promyelocytic Leukemia Cell Lines (급성 전골수성 백혈병 세포주간의 삼산화비소에 대한 반응)

  • Acute promyelocytic leukemia (APL) is a myeloid leukemia caused by over-expression of fusion protein, PML/RAR$({\alpha})$, which was the result of chromosomal translocation and induces the blockage of differentiation of affected promyelocytes. Pharmacological dose of retinoic acid induces the activation of and subsequent degradation of PML/RAR$({\alpha})$ fusion protein, and then APL cells undergo through the normal differentiation pathway. Arsenic trioxide has proved effective in causing remission of acute promyelocytic leukemia by inducing apoptosis of this tumor cells, whereas the heterogeneity of cellular susceptibility to this cytotoxic agent limited its usage on more types of tumors in clinic. This work showed that arsenic trioxide could induce apoptosis of a panel of acute promyelocytic leukemic cell lines, all-trans-retinoic acid (ATRA) sensitive NB4 cells and ATRA resistant UF-1 cell. They were investigated with regard to the correlation between the inherent or intrinsic cellular level of GSH and the apoptotic susceptibility of the cells to arsenic trioxide. We manifested, in two cell types, the inherently existed difference in intracellular GSH level reactive to the arsenic trioxide, and a positive correlation between the GSH level and their apoptotic sensitivity to arsenic trioxide. And it showed that arsenic trioxide could differentiate promyelocytic cancer cells to the cells possessed of dendritic cell surface markers. Unravelling the cause of the different susceptibility between leukemic cells and proving that promyelocyte could be differentiated to dendritic cells by arsenic trioxide will help not only to understand the mechanism underlying the complete remission of acute promyelocytic leukemia induced by arsenic trioxide, but also to expand its clinical usage.

A Characteristics on the Ancient Glass Beads Excavated from the Site of Hapgang-ri in Sejong, Korea (세종 합강리 유적 출토 고대 유리구슬의 특성 연구)

  • A convergence research was conducted on glass beads excavated at the ruins of Hapgang-ri, Sejong, using the archeology and conservation approach. The period of appearance of Jugu Towangmyo in the ruins of Hapgang-ri can be estimated to begin in the late 2nd century; for Jugu Towangmyo No. 15, the period spans from the late 2nd to early 3rd century. Form, color, and manufacturing techniques of complete glass beads were examined, and the cross-sections and chemical composition of 16 samples were observed. Based on these analyses, the glass beads were divided into blue, purple, and red colors. The blue-colored beads could be further sub-divided based on their gloss and brightness; in contrast, the red-colored beads were highly uniform with regard to these parameters. Based on the stripe and bubble arrangement on the surface of the glass beads, their drawing technique was identified. Traces of heat treatment or polishing were observed at the ends of the beads. According to their chemical composition, the 16 samples were classified into 3 potash glass and 13 soda glass groups; in the latter, the properties of the stabilizers were divided according to the blue and red bead colors. The stabilizers of the red beads are unique in that they allow the distinction among beads excavated in other areas in South Korea owing to their compositional differences. Colorants in blue- and red-colored beads are cobalt (containing MnO), and copper and iron, respectively.

An Exploratory Study on Luminescent Properties and the Relevant Applications of POF-based Flexible Textile Display for Mountaineer Wear with Safe-guard Function (안전보호 기능의 산악복을 위한 유연 광섬유 직물 디스플레이의 발광특성 및 적용에 관한 탐색적 연구)

  • Recent years have witnessed that IT-convergence technology has become the most important issue in the global market. Along with this trend, demand for PSS(i.e., Product-Service Systems) design has been rapidly increased in the smart clothing market. A case of the PSS design research, this study aimed to identify optimum conditions for weaving of POF-based flexible textile display(abbreviated as "PFTD") for mountaineer wear with safe-guard function regarding luminescent properties. Based on the findings regarding the optimum weaving condition of PFTD, several designs of mountaineer wear were suggested in this study. A total of 15 PFTD samples were prepared under various weaving conditions of weave structures and density of POF, and the luminance values of each sample were measured. As the results, the types of PFTD with structures and density of 'satin 3:1', 'satin 2:1', 'twill 3:1' and 'twill 2:1' indicated relatively higher luminescence. And based on the results and recent sports fashion trends, two suited mountaineering wears applying PFTD were illustrated in this study.

  • Lots of interdisciplinary studies for fusion of high technologes and the other areas of research had been tried in these days. In sports training area, high technologies like a vital sign sensor or an accelerometer were adopted as training tools to improve the performance of the sports players. The purpose of this study is finding the proper locations on the human body for attaching the motion sensors in order to develop a smart sportswear which could be helpful in training players. The rowing was selected as a subject sport as lots of movements of the joint on human body could be seen in rowing motions. The players of rowing could be devided into two weight divisions, the lightweight and the heavyweight. In this study, the change rates of distance between markers on human skin as the players moved were took on the back, the elbow, the hip and the knee area on human body by 3D motion capturing system. The distances between markers and the differences between the lightweight and heavyweight were analyzed. Finally, this study provided the guide lines for designing a motion sensing smart sportswear.
