  • This study is a qualitative study conducted to provide basic data for therapists working in long-term care hospitals to improve their countermeasure strategies for new infectious diseases and improvement of the treatment room infection management system, and to help therapists understand their infection management work. Colaizzi's phenomenological research method was applied as the research method. Telephone interviews were conducted with nine occupational therapists and physical therapists working in nursing hospitals. The contents of the interview were recorded with the consent of the study subjects, and additional confirmation was received by email. The recorded content was analyzed after transcription, and the meaning and nature of the experience coping with COVID-19 were described. The content was organized into 6 themes, 17 main meaning and 49 meaning units. In accordance with the COVID-19 situation, the infection control system has been strengthened by reinforcing infection control education in nursing hospitals, practicing infection control, and supervising infection control outside business hours. It was found that the treatment environment was changed due to the restriction of treatment activities by practicing distancing in the treatment room, adjusting rest and meal times during working hours, and strengthening infection control. In addition, the therapist's role has been expanded and the paradigm of treatment has changed, such as considering the untact intervention, and they have experienced cohort quarantine, pre-tested for COVID-19, vaccinations, and side effects from COVID-19. However, due to the infection work, the therapist's work burden is increased, and the person is experiencing fear, depression, and work stress from the spread of COVID-19. They were also aware of the need for nursing hospital care personnel support, such as guaranteeing rest after vaccination and providing infection control tools and equipment. The results of this study are expected to be used as basic data for human and physical support for the development of infectious disease response strategy programs in nursing hospital treatment rooms and for infection control in nursing hospitals.

  • Predicting hazardous ground conditions ahead of a TBM (Tunnel Boring Machine) tunnel face is essential for efficient and stable TBM advance. Although there have been several studies on the electrical resistivity survey method for TBM tunnelling, sufficient experimental data considering TBM advance were not established yet. Therefore, in this study, the laboratory-scale model experiments for simulating TBM excavation were carried out to analyze the applicability of an electrical resistivity survey for predicting hazardous ground conditions ahead of a TBM tunnel face. The trend of electrical resistivity during TBM advance was experimentally evaluated under various hazardous ground conditions (fault zone, seawater intruded zone, soil to rock transition zone, and rock to soil transition zone) ahead of a tunnel face. In the course of the experiments, a scale-down rock ground was provided using granite blocks to simulate the rock TBM tunnelling. Based on the experimental data, the electrical resistivity tends to decrease as the tunnel approaches the fault zone. While the seawater intruded zone follows a similar trend with the fault zone, the resistivity value of the seawater intrude zone decreased significantly compared to that of the fault zone. In case of the soil-to-rock transition zone, the electrical resistivity increases as the TBM approaches the rock with relatively high electrical resistivity. Conversely, in case of the rock-to-soil transition zone, the opposite trend was observed. That is, electrical resistivity decreases as the tunnel face approaches the rock with relatively low electrical resistivity. The experiment results represent that hazardous ground conditions (fault zone, seawater intruded zone, soil-to-rock transition zone, rock-to-soil transition zone) can be efficiently predicted by utilizing an electrical resistivity survey during TBM tunnelling.

  • At the center of the Noryang waterway, the Gwangyang bay area (including the Yeosu Strait) is located at the west, and the Jinju bay area (including Gangjin bay and Sacheon bay) is located at the east. Freshwater from several rivers is flowing into the study area. In particula,r the event of flood, great quantities freshwater flow from Seomjingang (Seomjin river) into the Gwangyang bay area and from Gahwacheon (discharge from Namgang Dam) into the Jinju bay. The Gwangyang and Jinju bay are connected to the Noryang waterway. In addition, freshwater from Seomjingang and Gahwacheon also affect through the Noryang waterway. In this study, we elucidated the characteristics of the tidal exchange rate and residence time for dry season and flood season on 50 frequency, considering freshwater from 51 rivers, including Seomjingang and Gahwacheon, using a particle tracking method. We conducted additional experiments to determine the effect of freshwater from Seomjingang and Gahwacheon during flooding. In both the dry season and flood season, the result showed that the particles released from the Gwangyang bay moved to the Jinju bay through the Noryang waterway. However, comparatively small amount of particles moved from the Jinju bay to the Gwangyang bay. Each experimental case, the sea exchange rate was 44.40~67.21% in the Gwangyang bay and 50.37~73.10% in the Jinju bay, and the average residence time was 7.07~15.36days in the Gwangyang bay and 6.45~12.75days in the Jinju bay. Consequently the sea exchange rate increased and the residence time decreased during flooding. A calculation of cross-section water flux over 30 days for 7 internal and 5 external areas, indicated that the main essential flow direction of the water flux was the river outflow water from Seomjingang flow through the Yeosu strait to the outer sea and from Gahwacheon flow through Sacheon bay, Jinju bay and the Daebang waterway to the outer sea.

  • The growth inhibitory effect of Microcystis aeruginosa according to the ultrasonic irradiation time was evaluated using a large algae sample volume (10 L) for various ultrasonic irradiation times (0.5, 1, 1.5, 2, 2.5 and 3 hr) at a laboratory scale. Based on the analysis of Chl-a and cell number of M. aerginosa, algae growth inhibition was observed with the decrease in Chl-a and cell number in all experimental groups after the ultrasonic irradiation. For the experimental group (T_B, T_C, T_D) with an ultrasonic irradiation time of less than 2 hours, rapid regrowth of algae was observed after growth inhibition, but the experimental group (T_E, T_F, T_G) with an irradiation time of more than 2 hours successfully inhibited algal growth lasting one or two more days. Based on the comparison of the recovery time to initial cell number the experimental group (T_B, T_C, T_D) took less than 20 days whereas the experimental group (T_E, T_F, T_G) took about 30 days. Correspondingly, the experimental group showed a high first order decay rate (𝜅) in proportion to the ultrasonic irradiation time during the growth inhibition period. Additionally, the specific growth rates (𝜇) during regrowth in the experimental group with irradiation time of more than 2 hours were relatively low compared to those in the experimental group with less than 2 hours. Therefore, ultrasonic irradiation for more than 2 hours is required for long-term (30 days) inhibition of algal growth in stagnant waters. However, the appropriate ultrasonic irradiation time for algae growth inhibition should be determined according to various field conditions such as the volume of stagnant water, water depth, flow rate, algae concentration, etc. Finally, damages to the algal cell surface and cell membrane were clearly observed, and both destruction and disturbance of gas vesicles of M. aeruginosa in the experimental group were discovered, indicating the growth inhibitory effect of Microcystis aeruginosa according to the ultrasonic irradiation time was confirmed.

  • In this study, we analyzed the hydrological impacts of future climate change on Jeju Island using SSP-based climate change scenarios from 18 climate models and watershed modeling (SWAT-K). Despite discrepancies among climate models, we generally observed an increase in evapotranspiration due to rising future temperatures. Furthermore, a significant increase in runoff and recharge was noted due to increased precipitation. These increasing trends were particularly pronounced in the SSP5-8.5 scenario, and differences among GCM models became more significant in the late 21 century. When compared to the historical period (1981-2010), the projected changes for the far-future period (2071-2100) in the SSP5-8.5 scenario showed a 21.4% increase in precipitation, a 19.2% increase in evapotranspiration, a 40.9% increase in runoff, and a 16.6% increase in recharge on an annual average basis. On a monthly basis in the SSP5-8.5 scenario, precipitation was expected to increase by 24.5% in September, evapotranspiration by 34.1% in April, runoff by 58.1% in October, and recharge by 33.8% in September. To further assess projections based on extreme climate scenarios, we selected two models, CanESM5 and ACCESS-ESM1-5, which represented the maximum and minimum future precipitation forecasts, and compared the hydrological changes in the future scenarios. The results indicated that runoff and recharge rates were relatively higher in the CanESM5 model with the highest precipitation forecast, while evapotranspiration rates were higher in the ACCESS-ESM1-5 model with the lowest precipitation forecast. Based on the climate change scenarios used in this study, the overall available water resources on Jeju Island are more likely to increase. However, since results vary by season and region depending on the climate model and scenario, it is considered necessary to conduct a comprehensive analysis and develop response measures using various scenarios.

  • The production of distilled soju by fermenting nonsteamed rice was evaluated using commercial enzyme products. White koji and modified nuruk had alpha-amylase activities of 31.90 U/g and 3,532.71 U/g, respectively, and gluco-amylase activities of 698.32 U/g and 4,899.58 U/g, respectively. The enzyme products had activities of 5,604.15-225,182.00 U/g and 13,517.41-120,822.41 U/g, respectively. At enzyme concentrations of >800 mg/L, the Chung-moo-purified enzyme had an alcohol productivity of ≥19%. Nurukzyme R400, Sanferm Yied, and Diazyme X4 exhibited alcohol productivities of >19% at concentrations of >600 mg/L. The alcohol content of the vacuum distillates was 41.31%-44.86%. The volatile component with the alcohol content adjusted to 25% was analyzed and principal component analysis was performed. The volatile components in white koji, Diazyme X4, and Sanferm Yield were similar. The modified nuruk treatment group had a relatively high ethyl lactate content compared to the white koji treatment group. The Nurukzyme R400 treatment group had high contents of butyric acid and ethyl butyrate. The Chung-moo-purified enzyme was characterized by a low component content. Thus, when enzyme products were used in nonsteamed rice fermentation, no effect on the alcohol productivity and quality of vacuum distilled soju was observed, suggesting that it can replace white koji and modified nuruk.

  • Purpose : We describe the changes of rat glomerular epithelial cells when exposed to high levels of glucose and advanced glycosylation endproducts(AGE) in the in vitro diabetic condition. We expect morphological alteration of glomerular epithelial cells and permeability changes experimentally and we may correlate the results with a mechanism of proteinuria in DM. Methods : We made 0.2 M glucose-6-phsphate solution mixed with PBS(pH 7.4) containing 50 mg/mL BSA and pretense inhibitor for preparation of AGE. As control, we used BSA. We manufactured and symbolized five culture dishes as follows; B5 - normal glucose(5 mM) + BSA, B30 - high glucose(30 mM) + BSA, A5 - normal glucose(5 mM) + AGE, A30 - high glucose(30 mM) + AGE, A/B 25 - normal glucose(5 mM) + 25 mM of mannitol(osmotic control). After the incubation period of both two days and seven days, we measured the amount of heparan sulfate proteoglycan(HSPG) in each dish by ELISA and compared them with the B5 dish at 2nd and 7th incubation days. We observed the morphological changes of epithelial cells in each culture dish using scanning electron microscopy(SEM). We tried the permeability assay of glomerular epithelial cells using cellulose semi-permeable membrane measuring the amount of filtered BSA through the apical chamber for 2 hours by sandwich ELISA. Results : On the 2nd incubation day, there was no significant difference in the amount of HSPG between the 5 culture dishes. But on the 7th incubation day, the amount of HSPG increased by 10% compared with the B5 dish on the 2nd day except the A30 dish(P<0.05). Compared with the B5 dish on the 7th day the amount of HSPG in A30 and B30 dish decreased to 77.8% and 95.3% of baseline, respectively(P>0.05). In the osmotic control group (A/B 25) no significant correlation was observed. On the SEM, we could see the separated intercellular junction and fused microvilli of glomerular epithelial cells in the culture dishes where AGE was added. The permeability of BSA increased by 19% only in the A30 dish on the 7th day compared with B5 dish on the 7th day in the permeability assay(P<0.05). Conclusion: We observed not only the role of a high level of glucose and AGE in decreasing the production of HSPG of glomerular epithelial cells in vitro, but also their additive effect. However, the role of AGE is greater than that of glucose. These results seems to correlate with the defects in charge selective barrier. Morphological changes of the disruption of intercellular junction and fused microvilli of glomerular epithelial cells seem to correlate with the defects in size-selective barrier. Therefore, we can explain the increased permeability of glomerular epithelial units in the in vitro diabetic condition.
