Estimation of Genetic Parameter for Carcass Traits According to MTDFREML and Gibbs Sampling in Hanwoo(Korean Cattle) (MTDFREML 방법과 Gibbs Sampling 방법에 의한 한우의 육질형질 유전모수 추정)

  • The objective of this study was to compare of genetic parameter estimates on carcass traits of Hanwoo(Korean Cattle) according to modeling with Gibbs sampler and MTDFREML. The data set consisted of 1,941 cattle records with 23,058 animals in pedigree files at Hanwoo Improvement Center. The variance and covariance among carcass traits were estimated via Gibbs sampler and MTDFREML algorithms. The carcass traits considered in this study were longissimus dorsi area, backfat thickness, and marbling score. Genetic parameter estimates using Gibbs sampler and MTDFREML from single-trait analysis were similar with those from multiple-trait analysis. The estimated heritabilities using Gibbs sampler were .52~.54, .54 ~.59, and .42~.44 for carcass traits. The estimated heritabilities using MTDFREML were .41, .52~.53, and .31~.32 for carcass traits. The estimated genetic correlation using Gibbs sampler and MTDFREML of LDA between BF and MS were negatively correlated as .34~.36, .23~.37. Otherwise, genetic correlation between BF and MS was positive genetic correlation as .36~.44. The correlations of breeding value for marbling score between via MTDFREML and via Gibbs sampler were 0.989, 0.996 and 0.985 for LDA, BF and MS respectively.

CMV-P1 Resistance Evaluation Using Enzyme-linked Immunosorbent Assay of Pepper Genetic Sources (Capsicum spp.) (Enzyme-linked Immunosorbent Assay를 이용한 CMV-P1 저항성 고추 유전자원 평가)

  • Recently, Cucumber mosaic virus (CMV)-P1 infection in pepper cultivation is very serious problem, which causes low marketability and yield. In this research, 56 domestic pepper cultivars including 20 PR (Phytophthora resistance) cultivars, 31 foreign pepper lines collected from USA and 112 genetic pepper resources form RDA Genebank were used for CMV-P1 resistance evaluation. Resistance evaluation was performed at 24 days and 51 days after artificial inoculation of peppers by enzyme-linked immunosorbent assay (ELISA). Among the 36 domestic cultivars 'Muhanjilju' were resistant to CMV-P1. All 20 PR cultivars and 29 foreign peppers except 'NuMex Twilight' and 'Chainese Giant' were susceptible. Among 112 pepper lines from RDA Genebank, nine pepper lines were resistant, and 17 pepper lines were moderately resistant, and 86 pepper lines were susceptible. Almost all domestic peppers on the market were highly susceptible to CMV-P1, whereas 17.2% of foreign pepper cultivars and genetic pepper lines from RDA Genebank were resistant or moderately resistant. Resistant pepper lines selected in this study can be used as genetic sources for breeding CMV-P1 resistant pepper.

Varietal Difference of Protein Content and Amino Acid Composition in Sesame (참깨품종의 단백질함량과 아미노산 조성)

  • To obtain the basic information an seed quality improvement in sesame, protein content of 114 varieties and amino acid composition of 12 varieties was analyzed. Protein content showed the vaietal difference ranged 20.6-30.2% and the mean was 24.72%. The highest variety in protein content was PI158066 (30.2%) originated from U.S.A. Protein content of Korean local varieties were highest among original group analyzed. Seed coat texture and seed coat color affected to protein content so, smooth type was higher than rough type in protein content, and black seeded varieties showed the hight protein content. Amino-acid composition of sesame was uneque in balance and higher than FAO reference. Total amino-acid of variety PI258372 was highest as 25.03%. Essential amino-acid (EAA) /total amino-acid(TAA) ratio of sesame was higher as 42-58.2% than soybean, corn, rice, peanut. Korean local variet 'Samcheck' showed best quality in amino-acid composition as 58.2% in EAA/TAA ratio with high tyrocin and lysine. Total amino acid content was high in order of Korean local '||'&'||'gt; introduced '||'&'||'gt; Korean bred varieties.

Silage Productivity of Korean Improved and Introduced Maize Hybrids (국내 육성 및 수입 옥수수 품종의 사일리지 생산성)

  • Silage yield Potential and some agronomic characteristics of Korean improved and introduced corn hybrids from the United States were tested for five year in Gyeongsan, Gyeongsangbug-do and for one year in Hongcheon, Gangwon-do. 1. At 20 days after emergence, plant height and dry matter of hybrids were different, while the early growth of the hybrids was not correlated to the culm length and silage yield. 2. In Cyeongsan, silage yield potential of Suwon19 was relatively high, while most susceptible to rice black-streaked dwarf virus (RBSDV) disease and leaf senescence at harvest time. In contrast, silage yield potential and resistance to RBSDV of Cwanganok were moderate, while susceptible to leaf senescence at harvest. Generally, Suwon19 showed similar or higher yield than most introduced hybrids, while some introduced hybrids showed lower silage yield than Korean improved hybrids. Most introduced hybrids were more resistant to RBSDV and leaf senescence at harvest time compared to Korean improved hybrids. 3. In Hongcheon, silage yield of three Korean improved hybrids were lower than that of NC+5514 and DK729, while similar or higher than other introduced hybrids. af senescence of all Korean hybrids was severer compared to introduced hybrids.

Breeding of King Oyster Mushroom, Pleurotus eryngii Carrying Good Traits of Cap (큰느타리버섯 갓우수 품종 육종)

  • Two strains Pleurotus eryngii KNR2312 and A8B10 (Na) which have good traits in cap quality and speedy growing were selected to breed a new strain carrying the two traits. KNR2312-2636-$10{\times}18$ (Ga) with 45.2 cap lightness and 6.5 quality was breeded by a consecutive inbreeding between KNR2312-derived monokaryons. $Ga5{\times}Na5$ came from outcrossing between Ga and Na was backcrossed with KNR2312-derived monokaryons twice to improve quality in cap color and shape. Therethrough Ga5NaKNR2312-47KNR2312-$12{\times}38$ carrying lightness of 49.5, quality of 7.3 and weight of 95.1 g was selected. The selected strain possesses good quality and dark color of cap. Especially its edge is 24 mm thick, therefore it is not likely damaged during processing and distribution.

Postharvest Nut Quality, and Changes of Soluble Solids Content and Kernel Hardness During Cold Storage in Korean Prevailing Chestnut Cultivars (밤 재배품종의 수확 후 과실품질 및 저온저장 중 밤 과실의 당도 및 경도 변화)

  • Postharvest nut quality, and changes of soluble solids content and kernel hardness during cold storage in 13 Korean prevailing chestnut cultivars were investigated to establish the chestnut grading and standardization for marketing and processing industry. Chestnut quality attributes such as nut weight, soluble solids content, kernel hardness, % with the pericarp split, and % of polyembryonic nuts were measured from 2001 to 2005. There were significant difference among cultivars in quality characteristics, and also annual variation within same cultivar, corresponded to the high genetic and environmental variability. During cold storage at $2^{\circ}C$ for 16 weeks, remarkable changes in soluble solids content were observed, and Isseumo showed the highest increase of 8% at 16 weeks of cold storage compared with postharvest. In case of most cultivars except early ripening cultivars, soluble solids content of chestnut increased until 12 weeks during cold storage, followed by decreased gradually thereafter. Kernel hardness of most cultivars except lshizuchi during cold storage increased slightly, but it was not statistically significant. This work would be a useful reference to the quality of each chestnut cultivar for the growers and breeders alike.

A Database of Simple Sequence Repeat (SSR) Marker-Based DNA Profiles of Citrus and Related Cultivars and Germplasm (SSR Marker를 이용한 감귤속 품종 및 유전자원에 대한 DNA Profile Data Base 구축)

  • The present study investigated identification of cultivars through phylogenetic analysis of 108 Citrus varieties and related cultivars using simple sequence repeat (SSR) markers. Two hundred three SSR primer pairs were used to detect polymorphic markers among 8 Citrus cultivars consisting of 4 mandarins, 1 orange, 1 tangor, 1 tangelo, and 1 pumelo. Eighteen SSR primer pairs were reproducible and showed highly polymorphic alleles. These markers were applied to assess genetic variations of the 108 varieties. Each marker detected 5-14 alleles, with an average of 9.28. The polymorphism information content varied from 0.417 to 0.791 with an average of 0.706. Cluster analysis with SSR markers resulted in 13 major groups reflecting cultivar types and pedigree information. Twelve orange cultivars in the $I-1^{st}$ sub-cluster and 23 mandarin cultivars in the $II-1^{st}$ sub-cluster, respectively, were not discriminated using the SSR markers. This could be due to narrow genetic backgrounds originated through bud mutation or nucellars seedlings. The SSR profile database of Citrus cultivars will be useful as a tool for protection of plant breeders' intellectual property rights in addition to assessing genetic diversity in Citrus cultivars and germplasms.

Breeding of a New Yellow Gerbera hybrida "Golden Time" with High Yield for Cut Flower (황색 대륜 절화용 다수성 거베라 신품종 "골든타임" 육성)

  • A new yellow gerbera (Gerbera hybrida Hort.) cultivar "Golden Time" was developed from a cross between '99-ga-2' and '99-na-7-6', followed by seedling and line selections at the Flower Research Institute, Gyeongnam Agricultural Research and Extension Services (ARES) in 2007. Detailed characteristics of the new cultivar were evaluated from 2004 to 2007. "Golden Time" has semi-double type large flowers in a good harmony with yellow (RHS, 17-B) ray floret and brown center. It has good, stable flower shape and strong peduncles, and its vase life was 12.3 days. The average flower yield of "Golden Time" was about 49.5 stems per plant/year in greenhouse trails during 2004 and 2007. This cultivar was registered to the Korea Seed and Variety Service (KSVS) for commercialization in 2009. Year-round production of this cultivar is possible in the greenhouse condition throughout the Korean climate.

A Mid-Maturing Apple Cultivar "Hongso", High Density Cultivation Type having a Good Taste (밀식재배형 식미우수 중생종 사과 "홍소(紅笑)")

  • "Hongso" was originated from a cross between "Yoko" and "Hongro" in NIHHS, RDA, in 1989. "Hongso" was preliminarily selected in 2002 for its high fruit quality. After regional adaptability test at five sites from 2003 to 2006 as "Wonkyo Ga-30" it was finally selected in 2006. Optimum harvest time is early September which is similar to that of "Hongro". Fruit shape is globose conical and skin color is light red. Mean fruit weight is 295g and soluble solids content is 14.1oBrix. Fruits acidity is 0.34%, which is higher than that of "Hongro"(0.23%). It has a good taste for harmony of sugar and acidity. Storability is 3 weeks in room temperature. It is apt to russet on fruit skin. It is susceptible to Bitter rot. "Hongso" has a good cross compatibility with major cultivars such as "Fuji", "Hongro" and "Tsugaru". Tree habit is semi-spreading and tree vigor is weak. "Hongso" is high-density cultivation type cultivar, because it has a precocity, spur-type.

Semi-dwarf Grain Sorghum (Sorghum bicolor L. Moench) Variety 'Sodamchal' with Waxy Endosperm (기계화 수확 적합 단간 찰수수 "소담찰")

  • The sorghum (Sorghum bicolor (L.) Moench) variety 'Sodamchal' was developed and registered by Department of Sothern Area Crop Science, National Institute of Crop Science (NICS) in 2013. This variety was developed from the cross between 'Hwanggeumchal' with brown grain and 'Jungmo4001' with semi-dwarf trait. 'Sodamchal' is the first semi-dwarf stature with waxy endosperm ever developed in Korea. The yield performance of 'Sodamchal' was 2.95 MT/ha in local adaptability test for 2 years. It had 91 cm culm length, which was 58 cm shorter than that of 'Hwanggeumchal', 26.3 cm panicle length, 22.0 mm thickness of culm, 21.1 g in 1,000 grain weight. It could be reduce the lodging problem and also adapted to mechanized cultivation. 'Sodamchal' produces high quantities of polyphenol and radical scavenging activitiy with diphenyl-picrylhydrazyl (DPPH), 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) in pigmented testa. Above all 'Sodamchal' is suitable for use as a parental source for the development of improved pollinator parent lines for developing semi-dwarf sorghum with waxy endosperm and antioxidant activity.
