  • In the 1990s, Cochlodinium polykikoides red tide has been annually occurred in the southern coast of Korea and caused the mass damage to the fisheries with a huge amount of economic loss. The present study was done to establish the biological foundation for the elucidation of the mechanism of C. polykikoiaes red tide. The growth response of C. polykikoides to physico-chemical factors such as temperature, salinity, pH, and light intensity were examined using axenic cultures to evaluate the relative importance of these factors on the dynamics of natural populations, It was found that the highest growth conditions were $25^{\circ}C,\;40\%_{\circ}$, pH 7.5, and 7,500 lux, respectively. The tolerable salinity range of growth was relatively wide at an optimum temperature and was reduced to a much narrower range at a sub-optimum temperature. These findings indicate that C. polykikoides is an eurythermal and euryhaline organism. The organism demanded higher light intensity and oceanic pH narrow in its growth. C. polykikoides utilize inorganic nutrients, such as nitrate and ammonium as N, and phosphate as P. The nutritional requirements of C. polykikoides were $40{\mu}M$ for nitrate, $50{\mu}M$ for ammonium, and $5{\mu}M$ for phosphate. The half saturation constant (Ks) for growth was $2.10{\mu}M$ for nitrate, $1.03{\mu}M$ for ammonium, and $0.57{\mu}M$ for phosphate. These values were comparatively smaller than those of other dinoflagellates reported previously. We confirmed that the organism is characterized as an eutrophic species. However, ammonium Ks value is smaller than that of other eutrophic species, This result indicates that C. polykikoides red tide may outbreak in the waters which eutrophication is in progress rather than eutrophicated waters. C. polykikoides preferred ammonium better than nitrate as a nitrogen source when in a growth stage, Therefore, our results indicate that ammonium is more important nutrient on the growth of the organism in comparison with other inorganic nutrients and C. polykikoides red tide is related with the increased ammonium concentration in the coastal waters.

  • Sunken ships cause damage to the environment due to the dispersal of fuel oil and harmful cargo goods in the hull. Since the sunken ship is mostly flooded by the seabed, it tends to be in a relatively stable condition. However, the heavy body, together with the load of remaining goods in the cargo hold, the constant contact with the seabed, and ocean currents and tidal waves, can affect dispersal of residual fuel oils out of the sunken ship. Corrosion of the sunken ship starts upon sinking, decreasing the thickness of the hull structure and sub-materials. Therefore, it is necessary to assess the structural stability against the potential breakdown of the sunken ship. Whilst evaluating the danger of the sunken ship, this result should be reflected in 'the possible discharge'. This study was undertaken to suggest a procedure for a step by step evaluation to assess the structural stability a sunken ship. The structural stability assessment to estimate the collapsibility of the hull was structure targeted at the sunken ship 'No. 7 HaeSung', which was classified as the prime example for the intensive management of sunken ships. This study was undertaken to suggest a procedure for a step by step evaluation to assess the structural stability a sunken ship and to propose a method to conduct a structural safety assessment that estimates the collapsibility of the hull by targeting the sunken ship 'No. 7 HaeSung',which was classified as the prime example for the intensive management of sunken ships. The collapsibility of the hull structure was estimated Based on the damage size of the hull structure, and the corrosion rate of the hull structure and sub-materials due to the seawater after sinking. It was confirmed that there was a low possibility of the total destruction of the hull structure at the current time. However, there is a high possibility in the potential failure of the hull structure due to increased rate of corrosion thereafter. Therefore, we believe continuous study on influence of corrosion and marine environment change to sunken ship's structural safety is necessary.

  • The heavy metal accumulations of avian eggshells were studied in order to test a feral pigeon (Columba livia)'s eggshell as an indicator for the environmental monitoring of pollutants. The reviews on the eggs of the different 19 avian species showed that it is the eggshell rather than the egg content that can better reflect the heavy metals in the environment; in most cases the CVs (coefficients of variations) of the heavy metal concentrations in the eggshells were higher than those in the egg contents. This can indicate that the heavy metal accumulations are homeostatically controlled in the egg contents, but the accumulations in the eggshells are varied according to the environmental conditions. To test the reviews, the feral pigeon eggs from the two different sites, one representing urban and the other rural environment, were analyzed for lead (Pb) and cadmium (Cd). The result showed that the eggshells of the urban pigeons (Hangang) had the higher metal concentrations than those of the rural pigeons (Hampyeong). The same difference can also be found in the internal organs (liver, bone) and blood. However, the analyses of the egg contents between the two sites did not reveal the differences. In other words, the result suggests that the feral pigeons, like the other avian species, are able to control the heavy metals into the egg contents homeostatically. Therefore, it is more useful to use the feral pigeon eggshell rather than the egg content in case of monitoring heavy metals in different habitats.

  • To assure the safety of the major edible seaweeds, we collected the 176 samples of seaweeds such as laver(Porphyra sp.), sea mustard (Undaria pinnatifida), sea tangle (Laminaria japonica) and sea lettuce (Enter-omorph sp.) from Korean coast. We investigated the contents of the trace metals in the edible seaweeds, and the values were expressed as the dry weight of sea weeds. The mean levels of the trace metals were high in the oder of Zn ($36.19{\pm}27.14 \;{\mu}g/g$), Mn ($28.75{\pm}28.03\;{\mu}g/g$), and Cu ($6.82{\pm}7.62\;{\mu}g/g$), which are necessary metals in the human body, and then followed by Cr ($1.54{\pm}2.07\;{\mu}g/g$), Ni ($1.29{\pm}2.57\;{\mu}g/g$), Pb ($0.89{\pm}1.11\;{\mu}g/g$), Cd ($0.72{\pm}0.80\;{\mu}g/g$) and Hg ($0.01{\pm}0.02\;{\mu}g/g$). The contents of Cd and Zn were high in the laver, Cr, Ni and Pb were detected highly in sea lettuce, and Hg was detected highly in sea tangle. Significant linear correlations were obtained among 28 different pairs of metals, some of them highly correlated (r>0.5, p<0.05),such as Ni-Mn, Zn-Ni, Cu-Pb and Cu-Zn in laver, Ni-Zn in sea mustard, Cu-Pb, Cu-Zn and Mn-Pb in sea tangle, and Ni-Cr in sea lettuce. And, the trace metal contents of seaweeds varied with parts and kinds of samples. The average weekly intakes of Hg, Cd and Pb from seaweeds were about $0.07{\~}3.57\%$ of PTWI (Provisional Tolerable Weekly Intakes) which has set by FAO/WHO to evaluate the safeties of trace metals in food. Therefore, it was found that they showed very safe levels in the edible seaweeds.

  • The water quality and phytoplankton presence in the ballast water (BW) of 37 vessels originating from the international ports of China were investigated to facilitate negotiations for exemptions to the Ballast Water Management Convention (BWM Convention). The shortest duration given BW spent in a vessel was $3.91{\pm}4.61days$ in area "A", which included the Bohai Sea. Total suspended solids, dissolved organic carbon, and particulate organic carbon ranged from 1.80 to $266mg\;L^{-1}$, from 1.09 to $5.79mg\;L^{-1}$, and from 0.17 to $3.65mg\;L^{-1}$, respectively. A low average concentration of nutrients was measured in BW from area "C", but the concentration of nutrients in BW from area "B" (around the Changjiang estuary) was high, which may be related to the relevant supply of freshwater. A high chlorophyll-a concentration (> $1{\mu}g\;L^{-1}$) was measured in six vessels, three of which carried BW in the area "A". High abundance of phytoplankton (> $10,000cells\;L^{-1}$) was measured in four vessels, two of which carried BW in the area "C". Vessel No. 37, originating from Hong Kong Bay in area "C", showed a high density of dinoflagellates. The results suggest that BWM Convention exemption negotiations with China should be performed cautiously.

  • In this study, we have investigated the distributions and killing effects of marine bacteria that tend to kill the red tide microalgae, C. polykikoides in the area of Masan bay from June to October, 1996. To summarize, C. polykikoides killing bacteria were detected at $10^2$ to $10^3$ cells/ml of seawater samples during the survey period, and the bloom was observed in September by containing $4.8\times10^3$cells/ml. It appears however that the number of these bacteria is decreased ($2.0\times10^2$cells/ml) in October, A total of 110 strains were isolated from seawater samples and seawater filtrate (pore size, 0.8 $\mu$m)-containing mixed culture of C. polykikoides in which the mixed culture was grown in f/2 medium. As results we have successfully isolated Micrococcus sp. LG-1 which decreased to less than 10cells/ml within 6days and 5days sfter inoculation of Micrococcus sp. LG-1 into the la9 and logarithmic growth phases of C. polykrikoides respectively. Therefore, it appears that inoculation of Micrococcus sp. LG-1 against the logarithmic C. polykrikoides is more effective than the lag growth phase, (n addition, the killing effects were increased in accordance with bacterial cell densities inoculated in a dose dependent manner. Especially, the filtrate of kitling bacterium culture (nore size, 0.2 $\mu$m) revealed a dramatic effect in which C. polykrikoides were decreased to less than 10 cells/mf of culture within 1 hr, 1,5 hrs, 1,5 hrs, 3.5 hrs. and 5,5 hrs after inoculations of the culture filtrate with concentration of $30\%,\;20\%,\;10\%,\;5\%$ and $2.5\%$, respectively. Moreover Micrococcus sp. LG-1 showed a selective specificity against C. polykrikoides and any other killing effects of Micrococcus sp. LG-1 were not observed against Alexandrium tamarense, Prorocentrum micans, Scrippsiella trochoidea. ana Gymnodinium sanguineum.

  • Paralytic shellfish poisoning (PSP) including Saxitoxin (STX) is caused by harmful algae, and poisoning occurs when the contaminated seafood is consumed. The mouse bioassay (MBA), a standard test method for detecting PSP, is being sanctioned in many countries due to its low detection limit and the animal concerns. An alternative to the MBA is the Neuro-2a cell-based assay. This study aimed to establish various test conditions for Neuro-2a assay, including cell density, culture conditions, and STX treatment conditions, to suit the domestic laboratory environment. As a result, the initial cell density was set to 40,000 cells/well and the incubation time to 24 hours. Additionally, the concentration of Ouabain and Veratridine (O/V) was set to 500/50 μM, at which most cells died. In this study, we identified eight concentrations of STX, ranging from 368 to 47,056 fg/μl, which produced an S-shaped dose-response curve when treated with O/V. Through inter-laboratory variability comparison of the Neuro-2a assay, we established five Quality Control Criteria to verify the appropriateness of the experiments and six Data Criteria (Top and Bottom OD, EC50, EC20, Hill slop, and R2 of graph) to determine the reliability of the experimental data. The Neuro-2a assay conducted under the established conditions showed an EC50 value of approximately 1,800~3,500 fg/μl. The intra- & inter-lab variability comparison results showed that the coefficients of variation (CVs) for the Quality Control and Data values ranged from 1.98% to 29.15%, confirming the reproducibility of the experiments. This study presented Quality Control Criteria and Data Criteria to assess the appropriateness of the experiments and confirmed the excellent repeatability and reproducibility of the Neuro-2a assay. To apply the Neuro-2a assay as an alternative method for detecting PSP in domestic seafood, it is essential to establish a toxin extraction method from seafood and toxin quantification methods, and perform correlation analysis with MBA and instrumental analysis methods.
