Properties of Cellulase Produced from Cellulomonas sp. YE-5 (Cellulomonas sp. YE-5가 생산하는 Cellulase의 특성)

  • Enzymatic properties of avicelase, carboxymethyl cellulase (CMCase) and P-glucosidase produced by Cellulomonas sp. YE-5 were studied. Optimal temperature and pH of avicelase were 40t and 6.0, and those of CMCase and P-glucosidase were $45^{\circ}C$ and 6.5. Avicelase and CMCase were stable between pH 5.0 and 9.5, and &glucosidase was stable between pH 5.5 and 8.0. Avicelase and P-glucosidase were inactivated when incubated at $35^{\circ}C$ for 6 hrs, and CMCase was at $40^{\circ}C$ for 6 hrs. All cellulases were strongly inhibited by $Cu^{2+} \; and \; Zn^{2+}. K_m$ values of avicelase for avicel, CMCase I and CMCase II for CM-cellulose, and ($\beta$-glucosidase for p-nitrophenyl-$\beta$-D-glucoside (PNPG) were 4.76, 16.4, 16.4 $\mu g$/ml and 3.51 mM, respectively.

  • Several bacterial strains utilizing crude oil as their sole carbon and energy source were isolated from marine environment polluted by crude oil. Among them, the selected strain 4A3 showed strong degradation activity for crude oil. This strain was identified as a Provindencia rettgeri 4A3 based on the morphological, biochemical, and physiological characteristics. The optimum cultural conditions was as follows; 26$^{\circ}C$ for temperature and 7.0 for initial pH. Additionally, the optimal concentration of sodium chloride was 2.0%, indicating that this strain was derived from sea water. The emulsifying activity of 4A3 was the highest after 3 days of cultivation under the condition of 2.0% sodium chloride, pH 7.0 and 26$^{\circ}C$. This strain had one cryptic plasmid in 7.0kb size.

  • We examined some properties of yeast cell wall lytic enzyme produced by Dicyma sp. YCH-37. Several metal ions, reducing reagents, and chemical modifiers have little effects on the lytic activity, except guanidine-HCl. Yeast cells of early log phase were more susceptible to the enzyme than those of stationary phase, and heat-treated cells were more easily lysed than intact living ones. Yeast cells pretreated with organic solvents such as butanol and acetone were more susceptible to the enzyme than intact living ones. Yeast cells cultured in Yeast extract-Malt extract medium containing 0.5 M ammonium sulfate were easily lysed by the lytic enzyme, and yeast cells cultured without shaking were more easily lysed by the enzyme than those with shaking. When SDS, ${\beta}-mercaptoethanol$, Triton X-100, sodium sulfite, and KCl were added to enzyme reaction mixture each, lysis of yeast cells was more effective.

  • The possibility of substituting corn or barley for the wheat, one of the raw materials for soy sauce, was studied by measuring the amylase and proteolytic activities of koji. Also optimum conditions of koji making were determined. It was found that substitution of up to 60% of wheat content (of the total bean and wheat content) with corn, yielded good quality of soy sauce. It is also found that barley can substitute 70% of wheat content (35% of the total content).

Inhibitory Effect of Bodusan on $TNF-{\alpha}-induced$ Migration via Inhibition of Metalloproteinase Activity in Human Aortic Smooth Muscle Cells (보두산의 금속분해효소 활성 저해를 통한 사람 대동맥 평활근세포의 유주능 억제 효과)

  • Objectives : This study was evaluated to elucidate the inhibitory potential of Bodusan (BDS) and its components, Strychnos ignatii semen (SIS) and Glycyrrhizae Radix (GR), on human aortic smooth muscle cells (HASMC) migration and production of matrix metalloproteinase (MMP)-2 and MMP-9 by $TNF-{\alpha}$ treatment. Methods : Cytotoxic activity of BDS and its components on HASMC was using 5-(3-caroboxymeth- oxyphenyl)-2H-tetra-zolium inner salt (MTS) assay. Effect of BDS, SIS and GR on $TNF-{\alpha}-induced$ HASMC migration underside of matrigel filter was stained with hematoxylin-eosin. And total number of cells that migrated to the underside of the filter was counted. MMP-2 and MMP-9 activity was evaluated by gelatin zymography assay. Results : The matrigel migration assay showed that BDS effectively inhibited the $TNF-{\alpha}-induced$ migration of HASMC. Moreover, BDS significantly inhibited MMP-9 activity. Our present study demonstrates that BDS and its components inhibits $TNF-{\alpha}-induced$ HASMC migration and MMP-9 activity. The inhibitory effect of BDS extract is more potent than that of its component herb extracts. Conclusions : These results provide evidence that BDS has multiple effects in the inhibition of HASMC migration and may offer a therapeutic approach to block HASMC migration.

  • An inhibitor of squalene synthase, a key enzyme in the cholesterol biosynthetic pathways and a target for improved agents to lower plasma levels of low-density lipoprotein, was sequentially purified from Curcuma longa by acetone extraction, silica gel column chromatography, and sephadex LH-20 column chromatography. Active compound, YUF-01, was successfully purified and analyzed as $C_{20}H_{21}O_6$ by electron ionization mass spectrum. Through $^1H-NMR$ and $^{13}C-NMR$ analyses, YUF-01 was identified as curcumin, which showed strong inhibition of squalene synthase.

  • Microorganisms producing milk-clotting enzyme were isolated from 1,506 strains which were collected from soil on the various places of Korea, and from strains which were already identified. Dothiorella ribis was taken as a good strain producing milk-clotting enzyme. When it is cultured on wheat bran, the optimum experimental conditions for the production of milk-clotting enzyme were consequently obtained as follows: 1) $30{\sim}35^{\circ}C$ of temperature and 4.0 of pH. 2) $60{\sim]80%$ of cultivating water to the weight of wheat bran. 3) addition of $(NH_4)_2SO_4$ as a nitrogen source, $NaCl\;and\;KH_2PO_4$ as an inorganic salt, and 3% of sucrose as a carbon source. 4) four days for a period of cultivation.

  • The extraction mechanism and extraction conditions of cellular lipid of freeze-dried cells of Rhodotorula rubra YUFE 1526, which was reported as coenzyme Q producing microorganism, were studied. 1. Methyl alcohol was the most appropriate solvent for extraction of cellular lipids and the resulting total lipid was 19.17 weight %. 2. When 1 % (w/v) of freeze-dried cell was extracted by methyl alcohol, the extraction yield was 90.49 % at $50^{\circ}C$ for 2 hr. 3. The diffusivity varied with extraction temperature and the empirical equation was derived as follows : $2.2e^{0-02907T}{\times}10^{-9}\;mm^2/min.$ 4. The diffusivity of cellular lipid of Rhodotorula rubra YUFE 1526 was $9.4{\times}10^{-9}\;mm^{2}/min.$ 5. The extraction model was well fitted on the extraction of cellular lipid.

  • The amylase was0 incubated in a 9mm hole on the starch agar gel plate bored with a cork borer. When 0.1N-iodine solution sprayed on the plate, the formed uncolored zone were observed. An activity of amylase has been determined by measurement of diameter of the uncolored zone. We named this method 'cork borer method'. When amylase was incubated on the starch agar gel plate, the following points were obtained. 1) The optimum pH for the formation of the zone in case of amylases(Biotex, Spitase) which produced by Bacillus is neutrality and alkali, while that for Glucoamylase, Biodiase, and Mucorrennin which produced by Rhizopus and Mucor is from 5 to 7. 2) The diameter of the zone was increased by the incubation time and amylase activity. 3) The zone was easily formed at low level of starch concentration and was formed much bigger than at $50^{\circ}C\;than\;at\;10^{\circ}C$. From the above results, after malting the starch agar gel plate, keeping constant concentration of the starch, the measurement of amylase activity is in efficiency upon the constant reaction time and temperature.

  • Lactobacillus helveticus YM-1and Streptococcus lactis Ml$_3$ were inoculated together in reconstituted non-fat skim milk medium, and then their proteolytic activity and stimulatory compound for acid production were investigated. Significant difference between Lactobacillus helveticus YM-1 and Streptococcus lactis Ml$_3$was observed in the proteolytic activities. The proteolytic activity of Lactobacillus helveticus YM-1 and Streptococcus lactis Ml$_3$ was 105 $\mu\textrm{g}$/$m{\ell}$ and 30 $\mu\textrm{g}$/$m{\ell}$ when converted the amounts of hydrolysates of milk protein determined by Folin Ciocaleau phenol method into their tyrosine equivalent Stimulatory compounds in cell-free filtrate of Lactobacillus helveticus YM-1were identified as peptide with a molecular weight of approximately 4, 300 for the acid production by Streptococcus lactis Ml$_3$. Some kinds of amino acids, such as histidine, lysine, arginine and glutamic acid, were rich in acid hydrolysates of peptide. Among amino acids, histidine, glutamic acid and phenylalanine stimulated acid production, on the contrary isoleucine inhibited.
