Genetic Variation of Several Isoenzymes in Pinus densiflora for. multicaulis (반송(Pinus densiflora for. multicaulis)의 몇 가지 동위효소(同位酵素)의 유전변이(遺傳變異))

  • Isozyme variations in 4 enzyme systems in Pinus densiflora for. multicaulis in 31 individual trees collected throughout the country were studied by starch gel electrophoresis using haploid megagametophyte tissue to compare with those of Pinus densiflora. A minimum of 7 loci were found to code for isozymes of the enzyme systems. No variation was found at locus GOT-A; at the remaining 6 variable loci(GDH-A, GOT-B, GOT-C, IDH-A, LAP-,A, LAP-B) and 2 to 4 alleles were identified. We could not find any marker alleles to distinguish Pinus densiflora for. multicaulis from Pinus densiflora at the isozymes studied here. Allele frequency distributions at each loci were almost all the same as those of P. densiflora. The percentage of polymorphic loci(99% level), the number of alleles per locus, the observed and expected heterozygosities were 85.7, 2.3, 0.165 and 0.186%, respectively. The level of genetic diversity in Pinus densiflora for. multicaulis seemed to be less than that of P. densiflora.

Genetic Variation of Pinus densiflora Populations in South Korea Based on ESTP Markers (ESTP 표지를 이용한 국내 소나무 집단의 유전변이)

  • Genetic diversity and genetic differentiation of thirteen Pinus densiflora populations in South Korea were estimated using nine ESTP (Expressed Sequence Tag Polymorphism) markers. The numbers of allele and the effective allele were 2.2 and 1.8, respectively. The percentage of polymorphic loci (P) was 98.8%. The observed and the expected heterozygosity were 0.391 and 0.402, respectively, and the eleven populations except for Ahngang and Gangneung population were under Hardy-Weinberg equilibrium state. The level of genetic differentiation (Wright’s FST = 0.057) was higher than those of isozyme or nSSR markers. We could not find out any relationship between the genetic distance and geographic distribution among populations from cluster analysis. Also, the genetic differentiation between populations was not correlated with the geographic distance (r = 0.017 and P = 0.344 from Mantel test). From the result of FST-outlier analysis to identify a locus under selection, six loci were detected at confidence interval of 99% by the frequentist’s method. However, only three loci (sams2+AluⅠ, sams2+RsaⅠ, PtNCS_p14A9+HaeⅢ) were presumed as outliers by Bayesian method. The sams2+AluⅠ and sams2+RsaⅠlocus were originated from the sams2 gene and seemed to be the loci under balancing selection.

  • Megagametophyte tissues from seeds of 45 loblolly pine clones were subjected to horizontal starch-gel electrophoresis. The resulting gels were tested for activity of five enzyme systems (GOT, SDH, PGM, MDH, and AP). Isozymes observed were under control of 11 loci. All 45 clones could be identified with unique genotypes at above 10 loci.

  • To explore the relationships among frequencies for sets of alleles, within or between loci, is one of the first analyses in population genetic study. The general question is whether the frequency of a set of alleles is the same as the product of each of the separate allele frequencies. For two alleles of a single locus, Hardy-Weinberg equilibrium is tested and for an allele from each of two loci, linkage disequilibrium is tested. However, it is more useful if we can quantify and graphically represent this information. In this study, we suggest graphical methods to find associations between alleles. We also analyze the STR data of Korean population as an illustration.

  • Megagametophyte and embryo tissue of Pinus densiflora were subjected to study the inheritance of glutamate-oxalate transaminase(GOT) and leucine aminopeptidase(LAP), and linkage relationship among isozyme loci coding both enzymes by starch gel zone-electrophoresis. Four zones of activity were observed for GOT. No variation was found in the fastest migrating zone (GOT-A). Electrophoretic phenotypes of the other two zones (GOT-B and GOT-C) showed 1:1 segregation ration, suggesting that each zone is controlled by a single locus. Foru and three alleles were identified at both loci respectively. The isozyme pattern of the fourth zone(GOT-D), migrated cathodally, coincided precisely with that of GOT-C. Whether the two zones are controlled by the same locus or by two tightly linked loci remained unknown. In all three variant GOT zones, heterozygoes embryos produced triple band patterns, indicating that GOT isozyme in Pinus densiflora is a dimer. Two zones of activity stained for LAP were found. The segregation of the two zones (LAP-A and LAP-B) suggested that tow loci control each of both isozymes. Two and three alleles were identified at both loci. GOT-B and LAP-B were found to be tightly linked, showing an average recombination frequency of 12.5 percent. Slight deviation from independent assortment was observed between GOT-B and GOT-C, with recombination frequency of 41 percent.

  • We observed the structure of phenylalanine ammonia-lyase gene (PAL) which is one of the best studied plant defense-related genes responding to pathogen infection by producing suberin, lignin, and phytoalexins. In tomato, at least 5 different genetic loci have been identified by genomic southern blot hybridization and nucleotide sequence analyses of partially cloned gene fragments (Lee et al. 1992). However, our results suggest that two other isoforms designated as PAL X1 and PAL X2 are located on the chromosome in tomato plant. Furthermore, the preliminary results obtained from southern blot hybridization analyses of subcloned fragment digested with several restriction endonuclease indicated that PAL X1 and PAL X2 clones contain at least two copies of PAL gene and partial nucleotide sequence analyses of each subcloned fragment with the same primer taken from known nucleotide sequence of PAL5 gene indicated that they are located side by side on the same chromosome.

  • A linkage map for Japanese red pine (Pinus densiflora) was constructed on the basis of two DNA marker systems of random amplified polymorphic DNAs (RAPDs) and inter-simple sequence repeats (I-SSR). Haploid genomic DNAs were extracted from megagametophyte tissues of 96 individual seeds in a single tree. A total of 98 DNA markers including 52 RAPD markers amplified by 25 primers and 46 I-SSR markers amplified by 18 primers were verified as Mendelian loci showing 1 : 1 segregation in 96 megagametophytes which were ${\chi}^2$-tested at 5% significance level. Of them, 63 segregating loci turned out to be linked into 20 linkage groups by the two-point analysis. However, 35 loci (17 RAPD and 18 I-SSR) of the 98 segregating loci did not coalesced into any linkage groups at a LOD of 3.0. The linked 63 loci were separated by an average distance of about 25.5 cM, which were spanned 1097.8 cM as a whole. The minimum and maximum map distances of the linkage groups were 4.3 cM and 54.9 cM, respectively. Incorporation of I-SSR loi into linkage map of RAPD loci resulted in extended and partially more saturated linkage blocks.

  • The genetic structures at one leucine aminopeptidase locus (LAP-A) of acorns, seedling raised in greenhouse and forest form the two beech provenances. West Germany and Rumania, were investigated and compared with each other. In many pair wise comparisons significant differences in genotypic structure as well as genic structure were ascertained between different developmental stages. In both the provenances, the allele $A_2$ seems to have advantage at both seedling stages raised under two different conditions. Homozygous carriers of $A_2$ allele survived best in greenhouse, while heterozygous carriers especially with $A_2$ allele possessed great viability under more variable environmental conditions. Since a distinct different genetic background was present in two base populations, the identical effect of the allele $A_2$ confirms the adaptiveness of this locus. With aid of some measures such as viability parameter and genetic distance, the character of occurred viability selection of further explained. The possible significance of this locus at this early stage is discussed in relation to adaptation of this long lived tree species to heterogeneous environment.

  • Soybean [Glycine max(L.) Merr.] is an important crop, accounting for 48% of the world market in oil crops. Improvements in economic traits, such as quality and oil constituents, arethe most important objectives in soybean breeding. The objective of this study was to identify quantitative trait loci (QTLs) that control seed size and fatty acid contents in soybean. 115 $F_{2:10}$ recombinant inbred lines (RIL) developed from a cross of 'Keunolkong' and 'Iksan10' were used. Narrow-sense heritability estimates based on a plot mean on 100 seed weight, saturated fatty acid (palmitic acid + stearic acid), and oleic, linoleic, and linolenic acid content were 0.72, 0.60, 0.83, 0.77 and 0.81, respectively. The 100 seeds weight was related to seven QTLs located on chromosomes 1, 3, 8, 9, 16 and 17. Two independent QTLs for saturated fatty acid content were identified on chromosomes 17 and 19. Five independent QTLs for oleic acid content wereidentified on chromosomes7, 11, 14, 16 and 19. Five QTLs for linoleic acid content were located on chromosomes 2, 11, 14, 16 and 19. Three QTLs for linolenic acid content were located on chromosomes 8, 10 and 19. Oleic, linoleic, and linolenic acid had one major common QTL on chromosome 19. Thus, linoleic and linolenic acid content were identified as common QTLs.

  • The estimates of genetic diversity based on 7 polymorphic loci coding for 6 isozymes were compared between the tolerant and the sensitive trees in an air polluted Prunus sargentii stand located on Ulsan industrial area. Although we could not observe statistically significant differences in allele and genotype frequencies between the two groups except the gene locus Got-2, the unique alleles and genotypes were observed in the tolerant group. All the genetic parameters such as genetic multiplicity, genetic diversity, and heterozygosity revealed that a greater amount of genetic variations existed in the tolerant group.
