Estimation of Genetic Parameters for Carcass Traits in Hanwoo Steer (거세한우의 도체형질에 대한 유전모수 추정)

  • The data were consisted of 1,262 records for carcass traits observed at Hanwoo steers from 1998 to 2001 at Namwon and Deakwanryung branch of National Livestock Research Institute, Rural Development Administration. Pedigrees of young bulls were traced back to search for magnifying inbreeding. Genetic parameters for carcass traits with Gibbs sampling in a threshold animal model were compared to estimates with REML algorithm in linear model. As the results, most of bulls were not inbred and sire pedigree group was non-inbred population. However, most of the bulls fell in some relationship with each other. Heritability estimates as fully posterior means by Gibbs samplers in threshold model were higher than those by REML in linear model. Furthermore, these estimates in threshold model using GS showed higher estimates than estimates using tested young bulls in previous study and same model. Heritability estimate by GS for marbling score was 0.74 and genetic correlation estimate between marbling score and body weight at slaughter was –0.44. Further study for correlation of breeding values between REML algorithm in linear model and Gibbs sampling algorithm in threshold model was needed.

Analysis of Diversity of Panax ginseng Collected in Korea by RAPD Technique (RAPD 방법을 이용한 국내 수집 인삼 (Panax ginseng C. A. Meyer)의 다양성 분석)

  • Genetic differences among nine land races of Korean ginseng (Panax ginseng C. A. Meyer) were examined using RAPD markers. Land races of Korean ginseng were collected from nine regions in Korea: Cheongwon, Guesan, Geumsan, Namwon, Pochun, Yangju, Yeoncheon, Yeongju. Out of 48 RAPD primers tested, 5 primers (OPA 7, OPA 13, URP 2, URP 3 and UBC 3) produced remarkable bands which showing polymorphisms among evaluated collections. Lower levels of genetic diversity were in detected same land races than among other land races. Genetic differences within and among land races indicate heterogeneity. These results indicate that cultivated ginseng in Korea is heterogeneous. Genetic similarity matrices of RAPD profiles were generated via coefficients of variation and the data were processed by the cluster analysis (UPGMA). When 90 collections were evaluated using selected 5 primers, those were clustered to 5 and 3 subgroups. These differences in genetic variation between land races of Korean ginseng implied the potential source for further breeding of Korean ginseng.

  • To obtain several foundamental information for the breeding of Korean Ginseng. 16 line selecteds in Korea were investigated and analyzed. The results obtained are as follows : 1. In all quantitative genetic characters of Korean Ginseng, the stem length showed the highest heritability(0.7050) and the leaf length the lowest(0.5674). 2. The genotypic correlation was showen the highest value (0.8649) at between the leaf length and the stem diameter and the lowest value(0.8649) at between the leaf length and the stem length, but the phnotypic correlation was shown the highest value (0.8788) at between the stem length and the stem dimeter and the lowest value (0.7950) at between the leaf length and stem length. 3. The co-heritability between the stem length and the stem diameter showed the highest value(0.6124) and that between the leaf length and the stem length showed the lowest value(0.5471). 4. The inter-class correlation among the planting location showed the highest value at the stem length and the lowest value at the stem diameter.

  • As Korean ginseng is hybrid, an individual variation is very severe, and it takes long times in new breeding because it is required 4 years to pick the seed. But, transformation technique makes the high-functional breeding in short time. The focus of these ginseng studies is to find and secure the useful gene. And it is urgent to accumulate the fundamental data for the molecular breeding and secure the useful genes. Therefore, transformation and soil acclimatization technique are necessary to molecular breeding in use of the introduction of functional genes. In this study, it add to secure of new regulation gene and useful gene as to accumulate the fundamental data for the place where it will contribute to raise the national competitive power. To analyze the useful genes in large scale, we constructed CDNA libraries with various tissues, species, and treated tissue. EST analysis of ginseng perform in large scale and build the EST database of ginseng. We perform the full length sequencing about the selected lots of clones that include the entire open reading frame of the amino acid residues and construct cDNA chip with the parental EST clones. Establishment of the transformation and a soil acclimatization system throuth the re-introduction of the selected ginseng gene that related with the secondary metabolism and anti-stress into the ginseng.

Genetic Analysis of Days to Flowering in Korean Hot Pepper (한국 건고추 개화소요일수의 유전 분석)

  • This study was carried out to calculate the genic values of days for flowering in commercial breeding lines of Korean hot pepper (Capsicum annuum L.). Two breeding lines of pepper '#2132' ($P_1$) early-flowering, and '#1308' ($P_2$) medium-late flowering, and their $F_1$ and $F_2$ generations were used in this study. By using partitioning method (Thseng and Hosokawa, 1971, 1972), it was possible to estimate, from the $F_2$ generations, the number of effective factor pairs differentiating the two parents. It was found out that the two parents were differentiated by two effective factor pairs, A:a and B:b. In the breeding lines used, the inheritance of days to flowering showed that the $F_1$ flowered a little earlier than the earlier flowering parent through the effect of over-dominance. However, $F_2$ flowered earlier or later than both parents through transgressive segregation. Conclusively, the magnitude of genic effects of A-a gene in flowering days was -13.81 days, and B-b gene was -6.73 days. The interaction between the two non-allelic factors using partitioning method was -5.26 days.

Genetic Studies of Major Agronomic Traits in Hybrid Populations of Maize Inbred Lines. (옥수수 유망자식계통들의 잡종집단에서 주요 농업형질들의 유전분석)

  • This study was conducted to find out the scientific maize breeding protocol for developing high performing single cross hybrid using introduced U.S. elite inbred lines; the pattern of inheritance, heterosis and heritabilities of six agronomic traits were studied in the progenies derived from five crosses (Mo17/B14A, Va85/B73, C103/ND203, FR35/Oh43, Wf9/A632). Among the five cross combinations, the cross combination of Mo17/B14A showed the highest heterosis for the most agronomic traits. Among 6 agronomic traits, the grain yield showed the highest heterosis effect in most cross combinations. Most of the agronomic traits in this study showed more than 50% heritability for six cross combinations, with an exception of the ear length trait. In conclusion, since Mo17/B14A showed excellent performance for most of the agronomic traits, these inbred lines were desirable combination and regarded as superior germ plasm sources for F1 hybrid development. The results of current studies will be utilized for developing high performing single cross hybrid from maize inbred lines, and will be used for the further genetic analysis of agronomic traits and maize breeding programs.

  • Residual feed intake(RFI), a linear index, is a trait derived from the difference between actual feed intake and that predicted on the basis of the requirements for maintenance of body weight and production. This study was conducted to estimate RFI genetic parameters of swine in Korea, and used 8,696 of productions data of Duroc swine species which were born from 2001 to 2014. Correlation between average daily gain and RFI breeding value has been investigated by negative correlation of -0.2(P>0.01). Estimates of heritability for RFI1(residual feed intake calculated from model average dily gain) and RFI2(residual feed intake calculated from model average dily gain and backfat) were 0.37 and 0.45. From the genetic parameter estimates found in this study, selection for low RFI in Duroc pigs has the potential to improve feed conversion ratio and reduce feed intake.

Development of PCR-based markers for selecting plastid genotypes of Solanum hjertingii (Solanum hjertingii 색소체 유전자형 선발을 위한 PCR 기반 분자마커 개발)

  • The tetraploid Solanum hjertingii, a wild tuber-bearing species from Mexico is a relative of potato, S. tuberosum. The species has been identified as a potential source of resistance to blackening for potato breeding. It does not exhibit enzymatic browning nor blackspot which are physiological disorders. However, due to their sexual incompatibility, somatic hybridization between S. hjertingii and S. tuberosum must be used to introduce various traits from this wild species into potato. After somatic hybridization, molecular markers are essential for selecting fusion products. In this study, the chloroplast genome of S. hjertingii was sequenced by next-generation sequencing technology and compared with those of other Solanum species to develop specific markers for S. hjertingii. The chloroplast genome has a total sequence length of 155,545 bp, and its size, gene content, order and orientation are similar to those of the other Solanum species. Phylogenic analysis including 15 other Solanaceae species grouped S. hjertingii with S. demissum, S. hougasii, and S. stoloniferum. After detailed comparisons of the chloroplast genome sequence with eight other Solanum species, we identified one InDel and seven SNPs specific to S. hjertingii. Based on these, five PCR-based markers were developed for discriminating S. hjertingii from other Solanum species. The results obtained in this study will aid in exploring the evolutionary aspects of Solanum species and accelerating breeding using S. hjertingii.

Genetic Diversity of Korean Native Chicken Populations in DAD-IS Database Using 25 Microsatellite Markers (초위성체 마커를 활용한 가축다양성정보시스템(DAD-IS) 등재 재래닭 집단의 유전적 다양성 분석)

  • A number of Korean native chicken(KNC) populations were registered in FAO (Food and Agriculture Organization) DAD-IS (Domestic Animal Diversity Information Systems, http://www.fao.org/dad-is). But there is a lack of scientific basis to prove that they are unique population of Korea. For this reason, this study was conducted to prove KNC's uniqueness using 25 Microsatellite markers. A total of 548 chickens from 11 KNC populations (KNG, KNB, KNR, KNW, KNY, KNO, HIC, HYD, HBC, JJC, LTC) and 7 introduced populations (ARA: Araucana, RRC and RRD: Rhode Island Red C and D, LGF and LGK: White Leghorn F and K, COS and COH: Cornish brown and Cornish black) were used. Allele size per locus was decided using GeneMapper Software (v 5.0). A total of 195 alleles were observed and the range was 3 to 14 per locus. The MNA, $H_{\exp}$, $H_{obs}$, PIC value within population were the highest in KNY (4.60, 0.627, 0.648, 0.563 respectively) and the lowest in HYD (1.84, 0.297, 0.286, 0.236 respectively). The results of genetic uniformity analysis suggested 15 cluster (${\Delta}K=66.22$). Excluding JJC, the others were grouped in certain cluster with high genetic uniformity. JJC was not grouped in certain cluster but grouped in cluster 2 (44.3%), cluster 3 (17.7%) and cluster8 (19.1%). As a results of this study, we can secure a scientific basis about KNC's uniqueness and these results can be use to basic data for the genetic evaluation and management of KNC breeds.

Phylogenetic Study of Genus Haliotis in Korea by Cytochrome c Oxidase Subunit 1 and RAPD Analysis (Cytochrome c oxidase subunit 1과 RAPD 분석에 의한 한국 전복속의 계통 연구)

  • Abalones are gastropod mollusks belonging to the genus Haliotis. Pacific abalones are regarded as a very important marine gastropod mollusk in Korea, Japan, China, and also in food industries around the world. In Korea, 6 species of abalone have been reported to occur along the coasts: Haliotis discus hannai, Haliotis discus discus, Haliotis madaka, Haliotis gigantea, Haliotis diversicolor supertexta, and Haliotis diversicolor diversicolor. This study was performed to discriminate the genetic variances by the partial sequences of the mitochondrial cytochrome c oxidase subunit I (COI) genes and random amplified polymorphic DNA (RAPD) analysis against four species of Pacific abalone (H. discus hannai, H. discus, H. madaka, H. gigantea). COI gene is reasonably well conserved and has been sequenced in various invertebrate taxa. The RAPD analysis technique is a relatively simple and low cost method that allows differentiation of taxa without the need to know their genomes. In this study, we investigated the genetic diversity, phylogenetic relationships within each species. The COI and RAPD analysis were able to distinguish between H. gigantea and the other three species. However, these analysis methods were inadequate to distinguish between H. discus and H. madaka. These results are believed to be able to provide a basis data for future hybrid breeding research by defining the genetically closely related four species of abalone, which is to develop new hybrid abalone for export using hybrid breeding.
