  • We investigated the growth-inhibitory activities of cosmetics and their preservatives against pathogens and resident skin bacteria. Of the tested cosmetics, preservatives such as parabens, 1,2-hexanediol, phenoxyethanol-contained toner, emulsion, cream and baby cream exhibited potent antibacterial effects against Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa. Parabens, 1,2-hexanediol and phenoxyethanol inhibited the growth of pathogens, as well as skin-resident bacteria such as Staphilococcus epidermidis, Shigella flexneri, Enterobacter aerogenes and so on. The application of a basic cream containing phenoxyethanol to human skin was shown to disturb the skin microbiota: at the phylum level, Proteobacteria increased and at species level, 4P004125_s increased and Propionibacterium humerusii decreased. Based on these findings, parabens, 1,2-hexanediol and phenoxyethanol have antimicrobial activity and cosmetics containing phenoxyethanol may disturb skin microbiota.

  • Survival and stability of 19 bifidobacterial strains included 13 isolates from Korean infants against acid, bile acid and oxygen exposure were examined. Acid resistance of selected strains at pH values of 4.0, 3.0 and 2.0 was tested. Among the bifidobacterial strains tested, B. bifidum B3, B. longum D6, and B. adolescentis F1 exhibited higher viable cell counts exposed to acid whereas other strains had various results. The abilities of the strains to grow in the MRS broth containing 0.2% thioglycolic acid and 0.2% oxgall were tested and the tolerance of B. bifidum B3 and B. longum D6 to bile acid were higher than that of others. Even though in same species, the tolerance of tested strains to bile acid were variable. Stabilities of tested strains to oxygen exposure were variable and B. bifidum and B. longum strains showed relatively higher viable cell counts after 48 hours exposure to aerobic incubation. These results demonstrated that the survival and stability of bifidobacterial strains to acid, bile acid, and oxygen exposure were variable and strain-dependent. Due to their tolerant ability to environmental factors like acid, bile acid, and oxygen, B. bifidum B3 and B. longum D6 had good potential properties as probiotic cultures and may be useful for industrial application.

  • Stable cell preservation is an essential factor in the regenerative medicine for cell therapies and transplantation of biologic materials. In this study, we studied to provide more stable hypothermic preservation by protection of cell damage during the preservation at $4^{\circ}C$. The result of searching for key components that have excellent efficacy in hypothermic preservation of cells, we have identified the fact that the hypothermic preservation adding protein hydrolysates such as yeast hydrolysate is far superior to others. All protein hydrolysates that are derived from animal, plant and microbe sources have superior efficacy, especially the peptides which have molecular weights under 10 kDa have the best efficacy among the components of protein hydrolysate. The protein hydrolysates prevented the decrease of ATP level in the cells caused by hypothermic environment and they inhibited the generation of ROS. Adding antioxidants and control agents of osmotic pressure were showed to have more superior efficacy in hypothermic preservation. Finally, KUL261 solution (DMEM/F12 1 : 1 medium, yeastolate 1%, $\alpha$-tocopherol $100{\mu}M$, dextran 2.5%), the preservation solution developed in this study, showed the best efficacy in both cell viability and cell growth more than other conventional preservation solutions. In conclusion, the improved hypothermic preservation solution that contains the protein hydrolysates as a key component provide the best preservation efficacy. It provides better efficacy than other preservation solutions and will contribute to both the development of regenerative medicine and global commercialization in this therapeutic field.
