Effect of Gamma Irradiation on the Protein Solubility, Purge Loss and Shear Force of Beef (감마선 조사가 우육의 단백질 용해성, 수분손실 및 전단력에 미치는 영향)

  • Post mortem bovine muscle. semitendinosus was used to investigate the effects of gamma irradiation on physicochemical properties. The muscle was cut into pieces of 2 cm in thickness and packaged with 2 different methods; vacuum-packaged and air-packaged. The packaged samples were irradiated at designated doses of 0, 1, 3 and 5 kGy by Cobalt-60 irradiator. Muscle protein solubility, purge loss, composition and contents of free amino acids, and shear force were observed during storage at $4^{\circ}C$. We found no significant differences in the purge loss and the contents of free amino acids. Muscle protein solubility slightly increased depending upon the increase of the dose. The decrease of shear force by gamma irradiation was observed. Therefore, it is considered that meat quality can be improved by gamma irradiation.

A Study on the Utilization with the Protein Forthification Material of Skip-jack Dark Meat Protein by Enzymatic Hydrolysis (효소 분해에 의한 가다랭이 혈합육 단백질 농축물의 단백질 보강제로서의 이용에 관한 연구)

  • For the effective utilization of dark meat separated as by-product from skip-jack canning, the dark meat concentrate(DPC) was prepared by removal of extractable materials with ethanol from dried dark meat. Dark meat protein hydrolysate(DPH) was prepared by the hydrolysis of DPC with ${\alpha}-chymotrypsin$. ${\alpha}-Chymotrypsin$ hydrolysed DPC to the extend of 79% during 10hr. The solubility over a pH range 1~12 showed similar trend on the both of DPH and DPC. The highest solubility was 81% on the DPH and was 36% on the DPC at pH 3. The lowest solubility was 65% on the DPH and was 22% on the DPC at pH 7. The content of total free amino acid was higher in the DPC than in the DPH, but the content of total essential free amino acid was higher in the DPH. Especially, the contents of taurine in the DPC and DPH were much higher than those of other amino acids. The result of sensory evaluation on the fish sauce analogue showed good taste, color and odor at the supplemented level of 8g DPH per 100ml of raw solution of fish sauce analogue and didn't show signifcaint difference compared with market fish sauce(p<0.05). On the preparation of surimi gel, 2% substitution of DPH for the supplemented starch was the most appropriate level.

The Synergistic Action of the AL-Protease from Arthrobacter luteus on the Lysis of Yeast Cell Walls (Arthrobacter luteus가 생산하는 AL-Protease의 효모세포벽 용해 촉진작용)

  • The yeast cell wall lytic action of the alkaline AL-protease, which was found out of the crude Zymolyase that a kind of yeast cell wall lytic $endo-{\beta}-1$, 3-glucanase produced from Arthrobacter luteus, was investigated with the viable cells of S. sake and it's cell wall preparation. AL-protease on the lysis of the viable yeast cells showed very low activities with the alone, but the lytic activities were highly increased with the combination of AL-protease and Zymolyase. On the stepwise treatment of the viable yeast cells with AL-protease and Zymolyase, the cells were lysed highly only by the course having a treatment with Zymolyase after pretreatment with AL-protease. Thus synergistic action of AL-protease was not observed with any some commercial enzymes, known as a type of alkaline and serine protease such as AL-protease, and was also found to be affected greatly by the culture conditions and species of the yeast tested. AL-protease caused the release of some peptide and a lot of sugar from the cell wall preparation, but could not lysed the cell wall more than 66%. Whereas Zymolyase could lysed the cell walls almost completely with alone. On the basis of these results, the synergistic action of AL-protease on the lysis of S. sake cells is hypothesized that at first AL-protease bind to the yeast cell surface layer consisting of mannan and protein, and then changes their conformation to facilitate the penetration of Zymolyase from the outside to the inside framework layer constituted of alkali insoluble ${\beta}-1,\;3-glucan$.

새로운quinolone계 항균제의 경구용제제화 연구

  • 열분석 (DSC, TG/DTA)을 통해 Q-35는 A, B, C의 세가지 다형이 존재함을 알수 있었으며 이중 상대습도에 따른 흡습성의 변화가 적고, 가습에 의해 다형전환이 일어나지 않는 C형이 제제에 유리하다고 사료된다. 또한C형은 분쇄, 연합 및 타정에 의한 다형의 전환이 없었다. Q-35의 용해도는 물, 메탄올, 에탄올에 각각 0.30.3.55,6.31mg/m1 이었으며 PH5이상에서는 급격히 용해도가 감소하고 산성에서는 용해도가 크게 증가하는 양상을 나타냈다. 적층시험, 배합시험을 통해 선정한부형제의 표준 처방 혼합물로 정제를 저조한 결과Q-35의 성형성은 양호하였고 Q-35의 함량이 증가함에 따라 붕해시간이 다소 지 연됨을 알수 있었다. 그러나 Q-35원료는 유동성이 적어 직타법이 적합하지 않았으며, 습식 과립압축법에 따라 저조한 정제의 붕해시간은 5-9분, 15분 후 용출은 91.$\pm$5.0% 이었다. Q-35정제를 4$0^{\circ}C$, 4$0^{\circ}C$ ㆍ 75%RH에서 6개월, 6$0^{\circ}C$에서 3개월 보존후 함량을 측정한 결과 각각 100.0, 98.7, 98.9%이었으며 그밖의 항목에서도 안정한 결과를 얻어 Q-35정제는 온도 및 습도어 안정한 것으로 사료된다. 대량생산 연구결과 Q-35의 결정수가 이탈되지 않도록 건조, 코팅 공정 중 정제의 온도를 5$0^{\circ}C$ 이하로 유지시켰다.

  • 주물작업이란 용융된 금속을 미리 준비한 주형에 부어 원하는 형태의 주물을 만드는 금속성형작업으로서, 생산 공정에서 고열을 이용하여 금속을 용해, 주조하는 등의 작업 특성상 부유분진의 발생이 필연적이며, 일산화탄소, 금속 흄, 휘발성 코울타르 피치, 소음 및 진동, 고열, 전리방사선 등의 유해인자로 인한 피해가 극심하다. 특히 분진은 주물의 생산공정 중 용해, 조형, 탈사 및 후처리 등 거의 전 공정에서 발생되며, 결정형 유리규산(SiO$_2$)을 다량 함유하고 있어 인체에 흡입되었을 때, 건강의 악화 및 직업병의 원인으로서 작업 능률을 떨어뜨리는 요인으로 작용하고 있다(Burgess, 1995).   • A conventional advanced control algorithm is proposed in this paper for improved temperature regulation of a TV-glass melting furnace. The TV-Glass melting furnace is a typical MIMO(Multi-Input Multi Output) system which is subject to various thermal disturbances. Because of its complexity, a detailed mathematical model of the furnace is hard to establish. To design a temperature control control system of the furnace, major input-output variables are selected first, and simple FOPDT(First Order Plus Dead Time) models are established based on the physical meaning and experimental process data. Based on the FOPDT models, a multi-loop control system composed of cascade and single loops are designed for effective control of the MIMO system. Practical implementation on the 150 ton/day furnace using the DCS(Distributed Control System) showed that the proposed control technique performs better than manual control.

  • This paper presents a practical application of fuzzy logic control to temperature control of glass melting furnace. Due to the characteristics of glass melting furnace, a hybrid algorithm of conventional PI controller and fuzzy logic controller is proposed and discussed. Practical implementation results of the production furnace showed the effectiveness of the proposed control algorithm.

타이타늄 주조 기술

  • 타이타늄은 경량성, 고강도성, 내화학성, 인체친화성, 내환경성 등의 물성을 이용하여 다양한 산업분야에 활용되고 있다. 다양한 산업분야에 정형부품으로 타이타늄을 가공하는 기술 중에서 가장 경제성이 높은 분야로 주조공정을 들 수 있다. 주조공정으로 부품을 제조하면 복잡한 형상을 기계가공하지 않고 거의 정형으로 제조할 수 있으며, 기계적 물성 또한 단조품에 버금가는 제품을 생산할 수 있다. 또한 다량의 제품을 생산할 수 있기 때문에 경제성에서도 유리하다. 이와 같은 주조품을 제조하는데는 진공에서 불순물의 유입을 최소화 할 수 있는 특수용해공정과 주형과의 반응을 최소화 할 수 있는 주형재료와 주조방안이 필수적이다. 본 고에서는 반응성이 높은 타이타늄을 용해하는 기술과 주형과의 반응이 최소활 될 수 있는 주형재료의 선정, 그리고 실 제품에 활용하기 위한 다양한 후처리 기술에 대하여 요약하였다.

Pretreatment of Feedstock with High Free Fatty Acid (고농도 유리지방산을 함유한 원료유지의 전처리)

  • Fatty acid methyl esters, also referred to as biodiesel, have been determined to have a great deal of potential as substitutes for petro-diesel. In order to enhance productivity in the biodiesel production process, feedstocks were previously recommended to be anhydrous, with a free fatty acid content of less than 0.5%. In this study, the effects of several catalysts, methanol molar ratio, catalyst amount, and reaction time on the reduction of free fatty acid level were studied with a simulated feedstock consisting of 20% oleic acid in rapeseed oil. Ferric sulfate was selected as the best catalyst. Increasing the catalyst amount and methanol molar ratio is very effective in decreasing the acid value of the simulated mixture. Our results may provide useful information with regard to the development of more economic and efficient free fatty acid removal system.

Changes of Silk Protein Compositions by Solubility Condition (용해조건에 따른 견 단백질의 조성 변화)

  • Changes of silk protein compositions of average molecular weight (Mw) and free amino acid composition to different solubility conditions were studied by SDS-polyacrylamide electropholesis, gel permeation chromatography (GPC), and free amino acid analysis method. We can not detected average molecular weight distribution of different hydrochloric acid (HCl) conditions as SDS-polyacrylamide method, but as using GPC method, molecular weight distribution of 2N-HCl, 1N-HCl and 0.5N-HCl (3 hrs at $110^{\circ}C$ treated) are confirmed Mw 800, 1,500 and 3,700, respectively. The average molecular weight of calcium chroride and calcium chloride-enzyme treated samples are shown Mw 46,800 and 12,500, respectively. The degree of hydrolysis and the composition of the free amino acid in the fibroin hydrolysates effected significantly composition of free amino acids of the fibroin powder. The increase of the degree of hydrolysis and ratio of free amino acids and oligopeptides were found to be directly related to the concentration of hydrochloric acid and treatment of enzyme, resulting in the increase of water solubility.
