First report of three Filosporella species isolated from freshwater ecosystem in Korea (담수생태계로부터 분리된 Filosporella 3종의 국내 최초보고)

  • To investigate the diversity of aquatic fungi, we collected deposits of soil, plants, and plant litter from ponds and streams. NNIBRFG1552 was isolated from soil deposits and NNIBRFG3013 from plant deposits in Namsaengi-mot in Jeju, Korea in 2016; NNIBRFG5472 was isolated from plant litter in Bocheong-cheon, Boeun, Korea in 2018. Based on morphological characteristics and phylogenetic analysis using rDNA sequences of the internal transcribed spacer (ITS), NNIBRFG1552, NNIBRFG3013, and NNIBRFG5472 were identified as Filoporella exilis (100% similarity with KC834046), F. fistucella (99.8% with KC834047), and F. cf. annelidica (100% with KC834044), respectively. Furthermore, cultural and morphological characteristics were analyzed to confirm the molecular identification. No species of the genus Filosporella has yet been reported in Korea.

First Record of Four Yeast Strains Isolated from Freshwater in Korea (국내 담수환경에서 분리된 국내 미기록 효모 4종 보고)

  • Freshwater ecosystems are significant habitats of fungi including yeasts. This study aimed to isolate and characterize wild yeasts from freshwater environments in Korea. The yeast isolates were identified by using the D1/D2 domains of the 26S rDNA regions. We identified four strains, Candida viswanathii (NNIBRFG39781), Curvibasidium cygneicollum (NNIBRFG49003), Oberwinklerozyma silvestris (NNIBRFG39803) and Vishniacozyma foliicola (NNIBRFG6120). These yeasts had not previously been recorded in Korea. We investigated the morphological and cultural characteristics of these yeasts. All of them grew on YPD, PD and YM media. Candida viswanathii (NNIBRFG39781), O. silvestris (NNIBRFG39803) and V. foliicola (NNIBRFG6120) grew in YPD medium containing glucose and in pH range of 4-8. Curvibasidium cygneicollum (NNIBRFG49003) grew well low temperature compared to others and slowly.

Anti-inflammatory Effects of Hemistepta lyrata Bunge in LPS-stimulated RAW 264.7 Cells through Regulation of MAPK Signaling Pathway (LPS로 유도된 RAW 264.7 대식세포의 염증반응에서 MAPK 신호경로 조절을 통한 지칭개 에탄올 추출물의 항염증 효과)

  • Hemistepta lyrata Bunge (HL) has been used as a folk remedy to treat cancer, inflammation, bleeding, hemorrhoids and fever, and leaves and young shoots have been used as famine food. Nevertheless, the biological activities and underlying mechanisms of the anti-inflammatory effects remain unclear. In this study, it was undertaken to explore the functions of the aerial part of HL as a suppressor of inflammation by using RAW 264.7 cells. As immune response parameters, the productions of as nitric oxide (NO) and prostaglandin E2 (PGE2), cytokines such tumor necrotic factor (TNF)-α and interleukin (IL)-6 were evaluated. Although the release of TNF-α remained unchanged in HL-treated RAW 264.7 cells, the productions of NO, PGE2 and IL-6 were significantly increased at concentrations with no cytotoxicity. Furthermore, HL significantly attenuated the mitogen-activated protein kinases (MAPK) pathway including decreasing the phosphorylation of the extracellular signal-regulated kinase (ERK1/2) and p38 mitogen-activated protein kinases. Collectively, this study provides evidence that HL inhibits the production of major pro-inflammatory molecules in LPS-stimulated RAW 264.7 cells via suppression of ERK and P38 MAPK signaling pathways. These findings suggest that the beneficial therapeutic effects of HL may be attributed partly to its ability to modulate immune functions in macrophages.

  • An experimental investigation has been conducted to clarify roughness effects of geothermal water scale deposited onto a heating surface upon its forced convection heat transfer characteristics. Examined was a circular cylinder, on which particles of silica scale having five different sizes are uniformly distributed. The Reynolds number was varied from 13000 through 50000. Local and mean heat transfer characteristics were measured as functions of particle size and Reynolds number. Subsequently the mean fouling resistance was estimated from those results, and its characteristics are clarified. It was found that the heat transfer of cylinders greatly varies with the fouling of geothermal water scale, especially its scale height. Further, the local and average Nusselt numbers strongly depend upon the cylinder spacing and the Reynolds number.

  • This study aimed to isolate and characterize fungi from freshwater environments in South Korea and evaluate their extracellular enzyme activities. Fungal strains were collected from various freshwater sources and identified using phylogenetic analysis. The isolated fungi included known aquatic hyphomycetes and previously unreported species. Extracellular enzyme, including those of protease, amylase, lipase, cellulase, laccase, and chitinase, activities were evaluated. Among the isolated strains, several showed high enzyme activity, suggesting their potential role in organic matter decomposition in freshwater ecosystems. This research expands our knowledge of the diversity and enzyme activities of the fungi in freshwater environments, contributing to our understanding of their ecological roles.

Experimental Study on the Natural Convective Heat Transfer Characteristics of Ferrofluid for Concentric Annuli under Rotating Magnetic Field (회전수 및 자기장강도 변화에 따른 이중원관내 자성유체의 자연대류 열전달 특성에 관한 실험적 연구)

  • The objective of this study is experimentally to investigate natural convective heat transfer characteristics of the ferrofluid for a concentric annuli under rotating magnetic field with variations of the revolution and the magnetic field strength. The rotating magnetic field was provided by induction motor with 6 poles and 3 phases and the revolution and the magnetic field strength were controlled by an inverter driver and a voltage meter, respectively. Temperatures of the inner pipe and the outer pipe in the tested concentric annuli were maintained at $30^{\circ}C$ and $25^{\circ}C$, respectively, during the test and the direction of the rotating magnetic field was a counterclockwise. As a result, the natural convective heat transfer characteristics of the ferrofluid for a concentric annuli were increased with the rise of the revolution and magnetic field strength due to the increased heat dissipation between hot side and cold side of the concentric annuli.

The Mechanism of Whole Plant Extract of Viola verecunda on the Proliferation of Dermal Papilla Cells (콩제비꽃 전초 추출물의 모유두세포 증식 기전)

  • Proliferation and maintain of dermal papilla during progression of hair-cycle are crucial to the duration of anagen and regulated by diverse signaling pathway such as PI3K/Akt/Wnt/β-catenin pathway. In this study, we investigated the effects and mechanisms of Viola verecunda on dermal papilla cells. Treatment of dermal papilla cells with whole plant extract of V. verecunda resulted in cell proliferation, which was accompanied by up-regulation of cyclin D1, phospho (ser780)-pRB and cdc2 p34, and down-regulation of p27kip1. V. verecunda extract also promoted the levels of phospho (ser473)-Akt and phospho (ser780)-pRB in a time-dependent manner. Inhibition of PI3K/Akt by Wortmannin suppressed progression of cell-cycle, thereby attenuated the increases in proliferation of dermal papilla cells by V. verecunda extract. We further investigated Wnt/β-catenin pathway with respect to the effects of V. verecunda extract on the proliferation of dermal papilla cells. Treatment with V. verecunda extract results in up-regulation of Wnt/β-catenin proteins such as phospho (ser9)-GSKβ, phospho (ser552)-β-catenin and phospho (ser675)-β-catenin. In addition, Wortmannin abrogated V. verecunda extract mediated up-regulation of cdc2 p34 and down-regulation of p27kip1. These finding reveal that the proliferative effect of V. verecunda mediated by alteration of cell-cycle via activating PI3K/Akt/Wnt pathway in dermal papilla cells.

Evaluation Antioxidant and Anti-inflammatory Activity of Ethanolic Extracts of Myriophyllum spicatum L. in Lipopolysaccharide-stimulated RAW 264.7 Cells (이삭물수세미(Myriophyllum spicatum L.) 에탄올 추출물의 항산화와 항염증 효과)

  • Myriophyllum spicatum L. has been used as an ornamental in ponds and aquariums, and as a folk remedy for inflammation and pus. Nevertheless, the biological activity and underlying mechanisms of anti-inflammatory effects are unclear. This study is aimed at investigating the antioxidative and anti-inflammatory activities of ethanol extract of Myriophyllum spicatum L. (EMS) in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. Antioxidant activity of EMS was assessed by radical-scavenging effects on ferric reducing antioxidant power (FRAP) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radicals. As inflammatory response parameters produced by LPS-stimulated RAW 264.7 cells were quantified to assess the anti-inflammatory activity of EMS. Our results showed that EMS increased FRAP and DPPH radical-scavenging activity. In EMS-treated RAW 264.7 cells, the production of NO, PGE2, TNF-α and IL-1β was significantly inhibited at the non-cytotoxic concentration. In addition, EMS significantly attenuated LPS-stimulated the toll-like receptor (TLR) 4/myeloid differentiation protein (MyD) 88 signaling pathway, and inhibited nuclear translocation of nuclear factor-kappa B(NF-κB). Positive correlations were noted between anti-inflammatory activity and antioxidant activity. In conclusion, it was indicated that EMS suppresses the transcription of inflammatory factors by inhibiting the TLR4/MyD88/NF-κB signaling pathway, thereby suppressing LPS-stimulated inflammation in RAW 264.7 cells. This study highlights the potential role of EMS against inflammation and associated diseases.

A Study on the Ultrasonic Effect for Turbulence Enhancement in the Flow Field of a Coaxial Circular Pipe (동심원관 유동장에서 난류증진을 위한 초음파 영향에 관한 연구)

  • A study on the ultrasonic effect for turbulence enhancement is carried out in the horizontal flow field of a coaxial circular pipe. A large transparent acryl tank is made to perform several experiments for the above research. The front flow field from jet exit is divided as 4 measuring regions to observe characteristics of the above flow field according to those with and without ultrasonic. An ultrasonic transducer with 2MHz high frequency is used to give them the ultrasonic forcing. Characteristics such as the velocity distribution, the kinetic energy and the turbulence intensity are visualized, observed, examined and considered at Re No. 2000. In results, it is clarified that the ultrasonic increases the turbulence enhancement. And the optimum and harmonious intensity suited to the power of flow is needed to maximize the turbulence enhancement.
