An inclusion forming virus isolated from a fan webworm, Hyphantria cunea, in 1975 was identified as a nuclear polyhedrosis virus. With the virus isolated in Korea, it was considered that the virus would be one of the valuable microorganism in microbial control. In this connection, 1) the shape and size of the virus for identification, 2) susceptibility of the various instar larvae to the virus, 3) the effects of storage condition on the pathogenicity and the cross infection of the virus to the larvae of Bombyx mori were examined. The results are summarized as follows; 1. The polyhedron was tetrahedron or hexahedron of $2\mu$ in size and the rod-shaped virus particles consisting of $2\~14$ rods in a bundle were $330m{\mu}\times35m{\mu}$ in size. 2. The hexagonal nuclear polyhedra were found only in the nucleus of the midgut cells but were variable in size. 3. The $LD_{50}$ values for the various instar larvae of H. cunea were $8.377\times10^4\;PIBs/ml$ for the second, $4.974\times10^5\;PIBs/ml$ for the fifth instar larvae. The $LT_{50}values$ for $10^6\;PIBs/ml$ were 9.6 days for the second, 11.5 days for the third, 12.0 days for the fourth and 17 days for the fifth instar larvae. 4. The susceptibility of H. cunea to the nuclear polyhedrosis virus was greater in the first generation than in the second generation. 5. The effect of the storage conditions on the pathogenicity of the nuclear polyhedra was less in refrigerator $(5^{\circ}C)$ and in freezing $(-80^{\circ}C)$ than in room temperature $(18.5^{\circ}C)$, especially as air-dried polyhedra than as suspension. The pathogenicity of the polyhedra seemed to decrease by sunlight during storage as cadavers, since rather greater decrease in pathogenicity was found in sunny condition than in shady condition. 6. The effective spray concentration was $6.4\times10^7\;PIBs/ml$ in the field and its $LT_50$ values for the third and the fifth instar larvae were 4.8 days and 14.2 days, respectively. 7. No cross infections were found in the nuclear polyhedrosis virus between H. cunea and B. mori. larvae.
Background: Recently, many cardiac centers have been using aprotinin to reduce operative bleeding in cardiac operations using cardiopulmonary bypass. A variety of reports have confirmed the effectiveness of the drug in cardiac operations. In addition to the operations which could be considered to cause severe operative bleeding such as redo operation, long cardiopulmonary bypass operation and etc, the use of aprotinin is increasing in the field of primary cardiac operations. Varying doses of regimen have been introduced since the first report by Royston et al, and also various opinions on the effectiveness and safeness of the each regimen have been reported. We reviewed our own experience of the full dose aprotinin regimen(Hammersmith regimen) retrospectively. Material and Method: From October 1994 to February 1998, 40 cases of cardiac operative patients were randomized into two groups: aprotinin group(20 patients) which received a full dose aprotinin regimen and control group(20 patients) which did not receive aprotinin. To evaluate the degree of bleeding decrease, we analysed and compared the amount of postoperative 6 hours and 24 hours bleeding in the each group. To confirm the renal dysfunction, we measured the postoperative creatinine level. Result: In the amount of postoperative 6 hours bleeding, a statistically significant bleeding decrease was demonstrated in the aprotinin group compared to the control group(aprotinin group: 186${\pm}$40cc, control group:409${\pm}$69cc, P=0.010). Similar result was observed in the postoperative 24 hours(aprotinin group:317${\pm}$53cc, control group: 671${\pm}$133cc, P=0.024). Conclusion: We concluded that full dose regimen of aprotinin can remarkably reduce postoperative bleeding in cardiac operations without significant renal dysfunctions.
In order to provide basic data for efficient control of weedy rice, we analyzed emergence rate of 100 weedy rice genetic resources collected in South Korea according to growth temperature and soil burial depth. Analyzing of emergence rate using low dormancy weedy rice genetic resources, emergence was increased at 12 and $14^{\circ}C$ of mean temperature condition. When calculating the effective accumulated temperature above $10^{\circ}C$, emergence rate increased 0.6% and 0.5% in 1 cm and 3 cm of soil depth, respectively according to increase of $1^{\circ}C$. However it reduced as 0.28% in 5 cm of soil depth condition. Analyzing of emergence rate using high dormancy weedy rice, red rice showed 55%, 46%, 41% of emergence rate in 1 cm, 3 cm and 5 cm of soil depth condition, respectively. However those of white rice decreased as 10%, 8% and 5.7% in 1 cm, 3 cm and 5 cm of soil depth condition, respectively. Relative emergence, assuming the final emergence rate as 100%, reached 80%, 90% and 100% when effective accumulated temperature was $83-112^{\circ}C$, $100-123^{\circ}C$, $148-160^{\circ}C$ respectively.
This experiment was conducted to evaluate the effects of high temperature on the stem, leaf and grain of barley during the ripening period and to provide information for the development of high-temperature cultivation techniques and adaptive varieties. We used an artificial climate control facility, to provide a temperature $3^{\circ}C$ higher than the normal average temperature during the ripening stage. Although the maximum rate of starch synthesis was increased at high temperature by approximately 11%, the starch content was decreased, because the period of starch synthesis ended 4 days earlier. As in the case of starch synthesis, the expression of genes related to starch synthesis was increased at the early ripening stage in the high temperature treatment, however, the duration of expression tended to decrease rapidly. Furthermore, the partitioning rate of assimilation products in the panicle increased to a greater extent in the high temperature treatment than in the control. In contrast, for the stem and leaf, the partitioning rate of assimilation products decreased more rapidly in the high temperature treatment than in the control. On the basis of these results, it can be considered that the translocation rate of assimilation products increased to a greater extent in the high temperature treatment than in the control at the early ripening stage. These results indicate that the decrease in grain weight at high temperature during the ripening stage is attributable to an increase in the speed of starch synthesis at high temperature, but the increase in ripening speed does not compensate for the shortening of the ripening period. Finally to develop varieties and cultivation techniques suited to high temperature, we need to focus on physiological characteristics related to the duration of starch synthesis.
The increase in the frequency of occurrence of abnormal weather could include severe rainfall, which could cause rice submergence during the ripening stage. This experiment was conducted to clarify the effects of submergence during the ripening period on yield and quality of rice. The flooding treatment was conducted at 7 and 14 days after heading. Flooding conditions were created with two conditions, flag leaf exposed and overhead flooding, and each condition was divided into two conditions according to water quality-clear and muddy. Although the yield decrease was more severe at 7 days after heading because of the decrease in the ripening ratio, the head rice ratio was more affected at 14 days after heading because of the increase in the chalky kernel ratio. The maximum quantum yield (Fv/Fm), which indicates the photosynthetic efficiency, did not differ before and after the flooding treatment until flooding continued for 4 days. In addition, stem elongation occurred because of flooding as an avoidance mechanism in japonica rice. This phenomenon was expected to decrease the supply of assimilation products to the spikelet (sink). Overall, it was suggested that additional experiments should be conducted examining the change in the starch synthesis mechanism and transfer of assimilate products resulting from submergence, for development of cultivation techniques corresponding to submergence and breeding of varieties with submergence tolerance characteristics.
Present system of seed potato production in Korea has several weak points and consequently has difficulties in covering annual shortage of 60,000 tons of seed potatoes. The author has an opinion that this so called 'High land system' of seed potato production adopted by the Government should be replaced by the 'Coastal area system' which is proposed by the author and has many advantages over present 'High land system'(2). In coastal areas where enormous acreage of rice paddies are spread, mostly around the villages. the primary host plants of the vectors are found. Therefore, the only source of aphid vectors are limited to the villages. The farmer's houses scattered more sparsely also have minor importance. In the previous paper(2), the author reported that the aphid vector populations were lower in the coastalareas than at Taegwanryong where the Alpine Experiment Station for the production. of seed potatoes is located. However, the number to vectors at Okku showed rather high density, where the trap was placed at the distance of 200 m from a village where peach and Hibiscus trees, the primary hosts of Myzus pesrsicae and Aphis gossypii were grown. To clarify the flight distance from the source of the aphid vectors, a trial was carried out in the Kwanghwal area, Kimje-gun, Cholla-pukto. on the western coast. 13 traps were placed at four directions and the distances between the traps were 250 m. (Fig. I) The traps 'Were operated from June 21 to October 31. The results are shown in Table 1. A total of some 70 species of aphids were found, including 5 speceis of potato virus vectors. The vectors are as follows: I. Myzus persicae (Sulzer) 2. Aphis gossypii Glover 3. Aulacorthum solani (Kaltenbach) 4. Lipaphis erysimi (Kaltenbach) 5. Macrosiphoniella sanborni (Gillette) Out of a total of 12,797 aphids, 5,187$(48\%)$ vectors were found. The trap catches at the 13 locations are shown in Fig. 2 and the numbers of the vectors at each location for each vector, except Macro-siphoniella sanborni. of which only a single individual was caught, are shown in Fig. 3-6. Number of vectors at C (3,279) (Centre of the village) is considerably higher than that at Suwon (763); however, EI. SI. WI and NI. where the distanecs from Care 250 m, showed lower numbers of vectors than that at Taegwanryong (347). The number of vectors at NI was rather than at the other 3 locations at the distance of 250m from the village. This was because C was in the southern part of the village. Consequently NI was much closer to the village than the other 3 locations of the same distance from C. Numbers of catches of the most important vector. Myzus persicae, are shown in Fig.3. The distribution pattern is typical except $S-2\;and\;W_3$, where several farmer's houses were found. If only the rice paddies were found in these locations. the numbers of the vectors would be small as the distances increase. Numbers of catches of the other 3 vectors are shown in Fig. 4-6. From these results. the author has drawn the following conclusions: 1. The aphid vector sources at the rice paddy belt in the western coast are the villages. 2. The vector densities at the locations where the distances are 250 m from the centre of the village are lower than that at Taegwanryong. 3. The vector densities become gradually lower as the distances from the centre of village increase. However. depending on the host plant situation at each location, the vector densities are variable. These minor sources of aphid vectors may be eliminated so that seed potatoes can be grown. 4. Thus. under the direction of specialists, fields suitable for seed potato production can be found in the coastal areas.
Groyne to control the direction and velocity of flow in rivers is generally installed for the purpose of protecting riverbanks or embankments from erosion caused by running water. In particular, as interest in river restoration and natural river improvement increases, groynes are proposed as a key hydraulic structure for local flow control and riparian habitat establishment. Groynes are installed mainly in groups rather than as individual structures. In case of groynes installed as a group, flow around the groynes change according to spacing in between the groynes. Therefore, groyne spacing is regarded as the most important factor in groyne design. This study aimed at examining changes of flows around and within the area of groynes that take place according to the spacing of groynes installed in order to propose the optimal spacing for upward groynes. To examine flow characteristics around groynes, this study looked at flows in main flow area and recirculation flow area separately. In main flow area, it examined the impact of flow velocity increasing as a result of conveyance reduction that is exerted on river bed stability in relation to changes in the maximum flow velocity according to installation spacing. As a factor causing impacts on scouring and sedimentation within the area of groynes, recirculation flow in the groyne area can lead problems concerning flow within the area and stability of embankment. As for recirculation area, an analysis was conducted on the scale of rotational flow and the flow around embankment that exerts impacts on stability of the embankment. In addition, a comparative analysis was carried with reference to changes of the central point of rotational flow that occur within the area of groynes. As a result of compositely examining the results, the appropriate installation spacing is proposed as min. four times-max. six times considering a decrease in flow velocity according to the installation of upward groynes, river bed stability and stability of embankments against counterflow within the area of groynes.
Limonium tetragonum is a halophyte grown naturally in the coastal region in South Korea. This study was conducted to investigate the effects of salt concentrations on seed germination, seedling growth, and antioxidant capacity of L. tetragonum. Seeds were collected from naturally grown plants of L. tetragonum and those at full maturity were used in this experiment. All experiments were performed at 0%, 0.5%, 1.0%, or 2.0% of salt concentrations. Seed germination rate was highest as 86% at $20^{\circ}C$ and followed as higher in order of $25^{\circ}C$, $30^{\circ}C$ and $15^{\circ}C$. The germination rate was about 80% at 0% or 0.5% of salt concentration, but it was very low at the salt concentrations higher than 1%. Growth of L. tetragonum seedlings showed no difference in Hoagland solution containing NaCl in the range of 0% to 1.0% and seedlings survived at 2.0% of NaCl concentration. As the salt concentration increased, the content of $Na^+$ in the shoot increased, but that of $K^+$, $Ca^{{+}{+}}$, or $Mg^{{+}{+}}$ decreased. The antioxidant activity and the content of total polyphenol and total flavonoid in the shoot were similar at 0% and 0.5% of NaCl and were highest at 2.0% of NaCl concentration. In conclusion, performance of seed germination and plant growth of L. tetragonum was highest at 0% and 0.5% of NaCl concentration, and showed no difference in antioxidant activity, total polyphenol contents, and total flavonoid contents at the same salt concentrations.
To investigate the effect of tillage methods on the silage barely growth and the soil environment in paddy field, liquid pig manure(LPM) was applied after harvesting rice at Jisan series soil for 2 years. Five plots, a LPM applied rate as N%; 0, 100, 150, 200(basal dressing) and 100(basal dressing)+50(additional fertilizer) were divided by tillage methods; non-tillage, non-tillage+rice straw and rotary tillage method. Emission amounts of $NH_3$ gas highly decreased in the rotary tillage and the non-tillage+rice straw plot compared to non-tillage plot. The contents of soil organic matter and exchangeable cation were increased in the applied LPM plot. $NH_4-N$ and $NO_3-N$ contents in soil were the highest in the non-tillage+rice straw plot and followed by the rotary tillage and highly decreased along with the growth of plant. Run-off rate of mineral components were higher in order of the rotary tillage plot£æthe non-tillage plot£æthe non-tillage+rice straw plot and then leached to $SO_4$, $NO_3-N$, K plentifully. The yield of silage barley in dry weight was higher in order of the non-tillage+rice straw plot>the rotary tillage plot>the non-tillage plot. To estimate the feed value of silage barley, crude protein, acid detergent fiber(ADF) and neutral detergent fiber(NDF) contents were analyzed. Crude protein and ADF contents were the highest at rotary tillage N150% plot as 9.7 and 29.4%, respectively. NDF contents was the highest at non-tillage+rice straw N150% plot as 56.7%. In conclusion, we recommend not to incinerate rice straw and to apply LPM at non-tillage status in cultivating the silage barley. This may prevent water pollution and increase barley yields.
Development of transgenic plant increasing crop yield or disease resistance is good way to solve the world food shortage. However, the persistence of marker genes in crops leads to serious public concerns about the safety of transgenic crops. In the present paper, we developed marker-free transgenic rice inserted high molecular-weight glutenin subunit (HMW-GS) gene ($D{\times}5$) from the Korean wheat cultivar 'Jokyeong' using Agrobacterium-mediated co-transformation method. Two expression cassettes comprised of separate DNA fragments containing only the $D{\times}5$ and hygromycin resistance (HPTII) genes were introduced separately into Agrobacterium tumefaciens EHA105 strain for co-infection. Each EHA105 strain harboring $D{\times}5$ or HPTII was infected into rice calli at a 3: 1 ratio of EHA105 with $D{\times}5$ gene and EHA105 with HPTII gene expressing cassette. Then, among 66 hygromycin-resistant transformants, we obtained two transgenic lines inserted with both the $D{\times}5$ and HPTII genes into the rice genome. We reconfirmed integration of the $D{\times}5$ and HPTII genes into the rice genome by Southern blot analysis. Wheat $D{\times}5$ transcripts in $T_1$ rice seeds were examined with semi-quantitative RT-PCR. Finally, the marker-free plants containing only the $D{\times}5$ gene were successfully screened at the $T_1$ generation. These results show that a co-infection system with two expression cassettes could be an efficient strategy to generate marker-free transgenic rice plants.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
