Effect of Follicular Fluid on Attraction and Motility of Human Spermatozoa (사람 정자에 대한 유인능과 운동성에 미치는 난포액의 영향)

  • Follicular fluid has pivotal effects on attraction and motility of spermatozoa for successful fertilization. The effect of samples of human follicu1ar fluid(hFF) on attraction and motility of spermatozoa was investigated. Capillary tubes loaded with one of these samples, hFF sample A collected from patients with tubal factor, hFF sample B collected from patients with male factor, m-HTF and heated hFF sample were used for assessment of attraction and motility of spermatozoa following culture of 1, 2, and 4 hrs. Number and motile rate of spermatozoa in the tubes loaded with hFF sample A were significantly(P<0.05) higher than those of m-HTF, hFF sample B and heated hFF. Although the fresh hFF tended to increase the attraction of spermatozoa as compared to inactivated hFF, there was no significant difference between treatments.

Evaluation of Extended Boar Semen after Glass Wool Filtration (Glass Wool Filtration 후 돼지정액의 평가)

  • The purpose of this study was to select high-quality boar semen after the glass wool filtration of extended boar semen. After collecting boar semen, its concentration, morphology, viability, and motility were examined according the glass wool's height and time. After glass wool filtration, the sperm concentration decreased, but the proportion of normal sperms and the sperm viability increased. Nevertheless, the sperm motility showed no changes. The above results showed that the glass wool filtration of boar semen is a method of obtaining sperms with relatively low abnormal rates and high viabilities.

  • Oxidative stress is a critical factor affecting the quality and viability of sperm during boar semen storage. Oxidative stress is also a significant concern during the process of freezing semen. The process of semen storage involves exposing the sperm to various stressors, including temperature changes, cryoprotectants, and extended periods of incubation. In addition, oxidative stress can lead to the production of reactive oxygen species (ROS) within the sperm, resulting in oxidative damage to cellular components, such as lipids, proteins, and DNA. Striking a balance between ROS production and the antioxidant defense system is crucial for maintaining sperm viability and functionality during semen storage. Moreover, the prolonged storage of boar semen leads to an increase in ROS levels, which can impair sperm motility, membrane integrity, and DNA integrity. ROS-induced lipid peroxidation affects the fluidity and stability of sperm membranes, leading to decreased sperm motility. Moreover, oxidative damage to the DNA can result in DNA fragmentation, compromising the genetic integrity of the sperm. In conclusion, oxidative stress is a significant challenge in maintaining sperm quality during boar semen storage. Understanding the mechanisms underlying oxidative stress and their impacts on sperm function is crucial for developing effective strategies to minimize oxidative damage and improve sperm storage outcomes.

  • 본 연구는 개의 동결정액제조 시 동결보호 희석액 내에 첨가되는 당의 종류와 조합이 동결융해 후 정자의 생존율 및 운동성에 미치는 영향과 정자동결 시 straw size, 예비동결 시 액체질소표면으로부터 straw까지의 거리 및 융해온도가 생존율에 미치는 영향에 대하여 조사하고자 실시하였다. 포유류의 경우 부생식선의 정낭선에서 분비되는 정장내에는 정자의 체외사출 후 정자의 영양물질인 fructose 가 분비되는데, 동결정액제조 시 회석액내에 첨가되는 당 성분은 fructose 및 glucose를 일반적으로 사용하고 있다. Effect of Spring and Summer on Semen Characteristics, Frozen-Thawed Sperm Viability and Testosterone Concentration in Yorkshire Boars (요크샤 종모돈의 정액성상, 동결-융해 후 정자의 생존성 및 테스토스테론의 농도에 미치는 봄과 여름의 영향)

  • This study was carried out to investigate the effects of spring (March~May) and summer (June~August) influencing semen characteristics, frozen-thawed sperm viability and serum testosterone concentration in Yorkshire boars. Results of this study were as follows: 1. There were no significant differences in the semen volume, pH and sperm concentration of sperm-poor fraction of Yorkshire boars between spring and summer. However, sperm concentrations of sperm-rich fractions in spring were higher than those in summer (P<0.05). 2. Sperm motility and normal acrosome of raw semen in Yorkshire boars did not differ significantly between spring and summer, However, motility and normal acrosome of frozen-thawed sperm were higher in spring season than in summer season (P<0.05). 3. Serum testosterone concentrations in Yorkshire boars were 4.04 ng/$m\ell$ in spring and 2.85 ng/$m\ell$ in summer. Serum testosterone concentrations in spring were higher than those in summer (P<0.05). 4. In conclusion, when serum testosterone concentrations in Yorkshire boars were higher, frozen-thawed sperm viability was higher.

Effects of Temperature, Diluents, and Plastic Tubes on the Motility and Acrosome Intactness of Fresh Rooster Semen (닭 신선 정자 운동성과 첨체안정성에 보존 온도, 희석제 및 용기가 미치는 영향)

  • Chicken spermatozoa have the ability to survive in low-temperature environments; however, the effects of low temperature on sperm motility and acrosome damage have not been studied in detail. The present study investigated semen longevity following dilution of rooster semen with Beltsville Poultry Semen Extender (BPSE) and Lake extender in preservation vessels (1.5 mL e-tube and 0.5 mL straw). Spermatozoa motility in the closed-type vessel (0.5 mL straw) was higher than that in the 1.5 mL e-tube on day 3 of preservation (68.6±3.1% vs. 22.1±5.7%). The motility of rooster semen diluted with BPSE in 0.5 mL straw was also higher than that of the Lake extender on day 3 of preservation (57.7±5.6% vs. 37.7±5.4%). Furthermore, acrosome intactness was higher in 0.5 mL straw than in the 1.5 mL e-tube, and the rate of acrosome cap damage increased with preservation days. The present study demonstrates that a closed 0.5-mL straw vessel could be used for low-temperature semen preservation, with an increased motility rate and acrosome integrity in fresh rooster semen.

Enhancement of Fertilizing Ability of Frozen-Thawed Bovine and Human Spermatozoa Treated with Fertilizing Promoting Peptide or Pentoxifylline (Fertilizing Promoting Peptide와 Pentoxifylline으로 처리된 소와 사람 동결 정액의 수정능 향상)

  • This study was to examine whether the in vitro friability, motility and intact acrosome of frozen-thawed bovine and human sperm can be improved by adding Pentoxifylline (PF) or Fertilization Promoting Peptide (FPP). Human semen was frozen ultra-rapidly using Test yolk-buffer (TYB) freezing medium. Additive (PF, FPP) effects in frozen-thawed bovine and human sperm were analyzed by microscopic count for sperm motility and coomassie brilliant blue staining method f3r sperm acrosome intact. The in vitro motility of frozen-thawed bovine sperm with 5 mM PF treatment group (50.0%) was significantly higher than that of control (34.0%) (P<0.05). In the frozen-thawed bovine sperm was examined, the intact acrosome rate of 50 nM FPP treatment (49.0%) was significantly higher than those of control (30.0%) and 25 nM FPP (38.0%) treatment groups (P<0.01). In human semen, when in vitro motility of sperm with PF addition prior to freezing was examined, the result of 5 mM treatment group (51.0%) was significantly higher than those of control and 2.5 mM treatment group (39.0, 40.0%) (P<0.01). In addition, 50 nM (75.5%) FPP adding in all treatment procedures for human semen freezing (before freezing, freezing and after thawing) was significant effect on maintenance of the sperm intact acrosome percentage (control: 45.0; 25 nM: 53.0; 100 nM: 68.0%) (P<0.01). Also, the intact acrosome rate of human sperm with FPP (65.0%) was significantly higher than that with PF (43.0%) (P<0.05), although sperm motility was slightly higher in PF treatment group. These results suggest that improved sperm motility and intact acrosome of frozen thawed bovine and human sperm can be obtained by addition of PF or FPP, and that the enhanced in vitro viability, motility and intact acrosome can be obtained by addition of FPP in all semen freezing procedures.

The Effect of Extract of Cervi pantotrichum Cornu on Human Sperm Motility (녹용(Cervi pantotrichum cornu) 추출물이 인간 정자 운동성에 미치는 영향)

  • This study was conducted to determine the effect of extract of Cervi pantotrichum cornu on human sperm motility, Four different types of media were prepared such as plain Ham's F-10 medium(control medium), control medium containing 0.3% bovine serum albumin(BSA)(medium A), control medium containing the extract of Cervi pantotrichum cornu aqua-acupuncture medium(medium B) and medium B containing 0.3% BSA(medium C). Human semen were washed and divided into 4 fractions and sperm were cultured in those medium for up to 72 hours at 37$^{\circ}C$ in a humidified atmosphere of 5% $CO_2$ in air. A total twenty eight semen samples including 14 normozoospermia and 14 asthenospermia were used for this study. In normozoospermia group, motility of control medium and medium A, B and C were 4.1%, 1.3%, 64.5%, and 77.1%, respectively after 24 hours of incubation, and were 0.0%, 0.0%. 8.8% and 44.9%, respectively after 48 hours of incubation. In asthenospermia group, motility of control medium and medium A, B and C were 2.0%, 2.2%, 58.3% and 85.1%, respectively after 24 hours of incubation, and decreased to 0.0%, 0.2%, 5.8% and 29.6%, respectively after 48 hours of incubation. In both groups, highest sperm motility was observed in medium C group when compared with other media. Furthermore motile sperm were found in medium C after 72 hours of incubation while no motile sperm was observed in the other media. Therefore it could be concluded that the extract of Cervi pantotrichum rornu affects on the human sperm motility.

Effects of Osmolality and $Ca^{2+}$ on Sperm Motility in Marbled Sole, Limanda yokohamae (삼투질농도와 $Ca^{2+}$이 문치가자미 (Limanda yokohamae) 정자의 운동성에 미치는 영향)

  • A series of experiments were conducted to study effects of osmolality and $Ca^{2+}$ on sperm motility in marbled sole, Limanda yokohamae. Spermatozoa were immotile when the milt was mixed with solutions (electrolyte or non-electrolyte) of lower osmolality than the average seminal plasma osmolality (351 mOsm/kg), but became motile after mixing milt with an hyperosmotic solution. However, with the osmolality above 1,639 mOsm/kg spermatozoa ceased their movement. When the milt was suspended in the $Ca^{2+}$ free artificial seawater (ASW) containing ethylenglycoltriamine (EGTA), the percentage of motile spermatozoa decreased in proportion to the increase of EGTA. Most spermatozoa were quiescent in a medium containing 3 mM EGTA. The motility of spermatozoa prevented by 3 mM EGTA was recovered by the subsequent addition of $Ca^{2+}$ over the concentration of 0.25 mM. Effects of calmodulin antagonist, trifluoperazin (TFP) on spermatozoa were examined to elucidate the possible functions of calmodulin in sperm motility. TFP immobilized spermatozoa 5n ASW at the concentration of 0.5 M. These findings suggest that calcium is an effective external factor in the initiation process of sperm motility.

Properties of Semen and Sperm Motility of Black Porgy Acanthopagrus schlegelii Acclimated in Freshwater (담수순화 감성돔 Acanthopagrus schlegelii의 정액특성 및 정자운동성)

  • The comparison of the chemical properties of semen of black porgy Acanthopagrus schlegelii long-term acclimated reared in freshwater (BFW) and seawater (BSW) with sperm activity of salinity and ion composition. The chemical properties of seminal plasma on BFW of the factors that most there was not significant difference in the BSW. However, osmolality in seminal plasma of BFW and BSW was $307.0{\pm}4.6$ and $337.3{\pm}10.1$ mOsm/kg, respectively, where BFW showed significant lower concentration in contrast to BSW. Salinity effect on sperm motility of BFW and BSW in 0 psu solution, no sperm motility was observed, whereas in 10 psu solution, both BFW and BSW sperms showed low motility and short time post sperm activation. However, diluted in 20 and 32 psu solutions, highest motility and long time post sperm activation were observed in BFW and BSW sperm. SAI of BFW and BSW varied in depend on the osmolality regardless of ion kind and it showed the highest value in the similar osmolality of artificial seawater (956 mOsm/kg). Accordingly, even in sperm released from BFW, factors initiating sperm motility are determined by osmolality.
