Antioxidant Activity and Whitening Effect of Cedrela sinensis A. Juss Shoots Extracts (참죽나무 새순 추출물의 항산화 활성과 미백 작용에 관한 연구)

  • In this study, the antioxidative effects, inhibitory effects on tyrosinase of Cedrela sinensis extracts were investigated. The ethyl acetate fraction of extract ($3.54\;{\mu}g/mL$) and aglycone fraction of extract ($2.15\;{\mu}g/mL$) showed more excellent free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$) than the activity of (+)-$\alpha$-tocopherol ($8.98\;{\mu}g/mL$). Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of Cedrela sinensis extracts on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system were investigated using the luminol-dependent chemiluminescence assay. The ethyl acetate fraction of extract ($0.15\;{\mu}g/mL$) and aglycone fraction of extract ($0.12\;{\mu}g/mL$) showed 10 times more excellent ROS scavenging activity than activity of L-ascorbic acid ($1.50\;{\mu}g/mL$). The protective effects of fractions of Cedrela sinensis on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The ethyl acetate fraction of extract and aglycone fraction of extracts suppressed photohemolysis in a concentration dependent manner ($5{\sim}25\;{\mu}g/mL$). The inhibitory effect of Cedrela sinensis extracts on tyrosinase was investigated to assess their whitening efficacy. Inhibitory effects ($IC_{50}$) on tyrosinase were determined with ethyl acetate fraction of Cedrela sinensis extract ($48.00\;{\mu}g/mL$) and aglycone fraction of extract ($5.88\;{\mu}g/mL$). The aglycone fraction showed 40 times more remarkable tyrosinase inhibitory effect than whitening agent, arbutin ($226.88\;{\mu}g/mL$) These results indicate that fractions of Cedrela sinensis can be used as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS. The fractions of Cedrela sinensis can be applicable to new functional cosmetics for antioxidant and whitening.

  • Vibrio cholerae non-O1 (V. cholerae non-O1) was previously called nonagglutinable or noncholera vibrios, since it fails to react with polyvalent O1 antisera. This organism is biochemically and genetically indistinguishable from V. cholerae O1 except serological difference. V. cholerae non-O1 strains are often detected in the environment including bays, estuaries, and fresh water, and also found in food. Therefore it is designated food borne bacterium in Japan. However, research papers on V. cholerae non-O1 are very rare in Korea. In order to investigate bacteriological characteristics of V. cholerae non-O1, we isolated V. cholerae non-O1 from the environmental sea water. Among the isolated V. cholerae non-O1 strains, we selected the strain which had the most strong hemolytic activity, named as V. cholerae non-O1 FM-3. The optimum growth conditions of V. cholerae non-O1 FM-3 were $37^{\circ}C$ and pH 8.5 in BHI broth (containing $0.5\%$ sodium chloride), and it grew better than V. cholerae non-O1 ATCC 25872. But both were not able to grow in BHI broth added $5.0\%$ of sodium chloride or adjusted to pH 5.0. According to the experimental results on the susceptibility test against various antibiotics, there were no significant differences between the isolated strain and reference strain (V. cholerae non-O1 ATCC 25872). Most of the antibiotics examined had bacteriostatic action against V. cholerae non-O1 FM-3 while vancomycin, oxacillin, colistin, polymyxin B, and sulfadiazine had no bacteriostatic activity.

Probiotic Potential of Lactobacillus Isolates (Lactobacillus 분리균주의 프로바이오틱스로서의 가능성 검토)

  • The purpose of this study was to investigate the probiotic properties of lactic acid bacterial strains isolated from animal feces. BCNU 9041 and BCNU 9042 isolates were assigned to Lactobacillus brevis on the basis of their physiological properties and 16S ribosomal DNA sequences analysis. They were confirmed as safe bioresources because of their non-hemolytic activities and non-production of harmful ${\beta}$-glucosidase, ${\beta}$-glucuronidase, tryptophanase, or urease. These isolates were also highly resistant to acid (at pH 2.5) and bile acids (at concentration of 0.3%, 0.6%, and 1% oxgall). In addition, they exhibited good antibacterial activity against food-borne bacteria, especially Bacillus cereus, Listeria monocytogenes, and Shigella sonnei. Furthermore, it was demonstrated that they have the highest levels of hydrophobicity and that they showed bile salt hydrolytic and cholesterol assimilation activity. These results suggest that BCNU 9041 and 9042 have good potential for application in functional foods and health-related products.

A Study on Application for Cosmeceutical of Fagopyrum esculentum Extracts (메밀 추출물의 기능성 화장품으로서의 응용가능성에 대한 연구)

  • In this study, the antioxidative effects, inhibitory effects on tyrosinase and elastase and components of Fagopyrum esculetum extracts were investigated. Fagopyrum esculetum used for this study is hulls and dehulled seeds. The free radical(1,1-diphenyl-2-picrylhydrazyl radical, DPPH) scavenging activities($FSC_{50}$) and reactive oxygen species(ROS) scavenging activities($OSC_{50}$) of extract/fractions from Fagopyrum esculetum were measured. The aglycone($3.5{\pm}0.0{\mu}g/mL$) and ethyl acetate fractions($0.2{\pm}0.1{\mu}g/mL$) of hulls showed the most effective scavenging activities. The protective effects on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The ethyl acetate fraction exhibited the most prominent cellular protective effect(${\tau}_{50},\;771.7{\pm}9.9 min$ at 10 ${\mu}g/mL$). The inhibitory effect of aglycone fraction of hull and dehulled seeds on tyrosinase were examined ($53.6{\pm}0.5{\mu}g/mL,\;35.6{\pm}0.4{\mu}g/mL$, respectively). And The inhibitory effect of aglycone fraction of hull and dehulled seeds on elastase were investigated($3.7{\pm}0.4{\mu}g/mL\;and\;6.0{\pm}0.7{\mu}g/mL$ respectively). But 50% ethanol extract rarely exhibited the inhibitory activity on tyrosinase and elastase. (+)-Catechin and (-)-epicatechin were contained in ethyl acetate fraction of dehulled seeds. And ethyl acetate fraction of hull contains flavonoids of hyperin, quercetin, rutin and so on. These results indicate that extract/fractions of Fagopyrum esculentum can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS. Fagopyrum esculentum extract could be used as a new cosmeceutical for whitening and anti-wrinkle products.

Cellular Protective Effects of Peanut Sprout Root Extracts (땅콩나물 뿌리 추출물의 세포 보호 효과)

  • In this study, the cellular protective effect and antioxidative property of peanut sprout root extracts were investigated. Cellular protective effects of peanut sprout root extracts on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The ethyl acetate fraction of extracts exhibited a cellular protective effect in a concentration dependent manner. Particularly, the aglycone fraction of extracts showed prominent cellular protective effects in a concentration range (5~50 ${\mu}g/mL$). They are more effective than that of (+)-${\alpha}$-tocopherol, known as a lipid peroxidation chain blocker. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of peanut sprout root extracts on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system were investigated using the luminol-dependent chemiluminescence assay. The ethyl acetate fraction of extracts ($OSC_{50}$; 1.59 ${\mu}g/mL$) showed a similar ROS scavenging activity compare with that of L-ascorbic acid (1.50 ${\mu}g/mL$), known as a strong antioxidant. On the other hand, the order of free radical (1,1-diphenyl-2-picrylhydraxyl, DPPH) scavenging activity ($FSC_{50}$) was (+)-${\alpha}$-tocopherol > 80% MeOH extract > aglycone fraction > ethyl acetate fraction. These results indicate that peanut sprout root extracts can function as an antioxidant in biological systems, particularly skin exposed to solar UV radiation by scavenging $^1O_2$ and other ROS, and to protect cellular membranes against ROS.

Antioxidative Effect and Component Analysis of Eriobotrya japonica Leaf Extracts (비파엽 추출물의 항산화 효능과 성분분석)

  • In the present study, the antioxidative properties, inhibitory activity on tyrosinase, and active components of Eriobotrya japonica (E. japonica) leaf extract were investigated. The free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$) of extract/fraction of E. japonica leaf was in the order 50 % ethanol extract ($22.625{\mu}g/mL$) < ethyl acetate fraction (6.75) < deglycosylated aglycone fraction (5.06). Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of fraction/extracton ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system using the luminol-dependent chemiluminescenceassay were investigated. $OSC_{50}$ of the ethyl acetate fraction, deglycosylated aglycone fraction, and ethanol extract were 0.75, 0.79, and $1.61{\mu}g/mL$, respectively. The cellular protective effects of E. japonica leaf extract on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The protective effects of extract/fraction of E. japonica leaf were increased in a in a concentration dependent manner ($5{\sim}50{\mu}g/mL$). Especially, ${\tau}50$ of ethyl acetate fraction at concentrations of $10{\mu}g/mL$ and $50{\mu}g/mL$ showed the most protective effects at 390.8 min and 1471.5 min. The inhibitory effect ($IC_50$) on tyrosinase of E. japonica leaf extracts was higher than arbutin, known as a skin-whitening agent. The order of inhibitory effects was acetate fraction ($75.25{\mu}g/mL$) < 50 % extract (74.1) < deglycosylated aglycone fraction (43.35). TLC of the ethyl acetate fraction showed 7 bands (EJL 1 - EJL 7). HPLC of the aglycone fraction exhibited 2 peaks, kaempferol and quercetin. The amounts of kaempferol and quercetin were 53.7 and 46.3 %. respectively. Therefore, The amounts of kaempferol and its glucoside were a little bit higher than quercetin and its glucoside in E. japonica leaf extract. Accordingly, these findings suggest that extracts/fractions of E. japonica leaf can function as antioxidants in biological systems, especially skin exposed to UV radiation, and protect cellular membranes against ROS. Thus, the extract/fraction of E. japonica leaf may be used in novel functional cosmetics as antioxidants against skin photoaging.

Antioxidant Activity of Persicaria perfoliata Extracts (며느리배꼽 추출물의 항산화 활성)

The deglycosylated fraction of extract ($12.38{\mu}g$/mL) showed the most prominent free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$). Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of P. perfoliata extracts on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system were investigated using the luminol-dependent chemiluminescence assay. The ethyl acetate fraction of extract ($0.35{\mu}g$/mL) showed the most prominent ROS scavenging activity. The protective effects of P. perfoliata extract/fractions on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The P. perfoliata extracts suppressed photohemolysis in a concentration dependent manner ($1{\sim}50{\mu}g$/mL) except the deglycosylated fraction of extract. The inhibitory effect of P. perfoliata extracts on tyrosinase was investigated to assess their whitening efficacy. Inhibitory effects ($IC_{50}$) on tyrosinase were determined with ethyl acetate fraction of P. perfoliata extract ($136.00{\mu}g$/mL) and deglycosylated fraction of extract ($68.10{\mu}g$/mL). Finally, their anti-elastase activities were measured to predict the anti-wrinkle efficacy in the human skin. Inhibitory effects ($IC_{50}$) on elastase were determined with ethyl acetate fraction of P. perfoliata extract ($67.20{\mu}g$/mL) and deglycosylated fraction of extract ($43.50{\mu}g$/mL). These results indicate that extract/fractions of P. perfoliata can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS. Extract/fractions of P. perfoliata can be applicable to new functional cosmetics for antioxidant, antiaging.

Antibacterial, Antioxidative and Antiaging Effects of Allium cepa Peel Extracts (양파껍질 추출물의 항균, 항산화 및 항노화 효과에 관한 연구)

  • In this study, the antibacterial, antioxidative and inhibitory effects of Allium cepa peel extracts on tyrosinase and elastase were investigated. MIC values of the ethyl acetate fraction of Allium cepa peel on especially, S. aureus among the skin resident flora (Staphylococcus aureus, S. aureus; Propionibacterium acnes, P. acnes; Pityrosporum ovale, P. ovale; Escherichia coli, E. coli) were 0.06%. The aglycone fraction showed more excellent free radical (1,1-diphenyl-2-picrylhydrazyl radical, DPPH) scavenging activity ($FSC_{50}=5.05{\mu}g/mL$). Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of the ethyl acetate fraction and aglycone fraction in the luminol-dependent $Fe^{3+}-EDTA/H_2O_2$ system were 0.05 and $0.03{\mu}g/mL$, respectively. The cellular protective effect of the aglycone fraction on the rose-bengal sensitized photohemolysis of human erythrocytes exhibited more prominent (${\tau}_{50}$, 480 min at $25{\mu}g/mL$). The inhibitory effects ($IC_{50}$) of the ethyl acetate fraction and aglycone fraction on tyrosinase were 9.16 and $8.68{\mu}g/mL$, the inhibitory effect ($IC_{50}$) of the aglycone fraction on elastase was $14.12{\mu}g/mL$ The transepidermal water loss of the cream containing 0.1% ethyl acetate fraction was decreased from $8.3g/m^2h$ in control to $6.8g/m^2h$ in the subjects applied with cream containing the ethyl acetate fraction. These results indicate that extract/fractions of Allium cepa peel can function as antioxidant in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS, and possibly as antiaging agents. Allium cepa peel extract could be used as a new cosmeceutical for whitening and anti-wrinkle products.

A Case of Gilbert's Syndrome with Severe Neonatal Hyperbilirubinemia

  • Gilbert's syndrome is caused by a reduction in the activity of uridine diphosphate glucuronosyltransferase (UGT) and induces chronic, non-hemolytic unconjugated hyperbilirubinemia. It has been suggested that 3-10% of the population has Gilbert's syndrome. Commonly, Gilbert's syndrome causes mild symptoms. However, a case of Gilbert's syndrome with severe neonatal hyperbilirubinemia is presented here. The patient developed jaundice three days after birth. Five days after birth, the patient's total serum bilirubin level was 34 mg/dL. The patient received intensive phototherapy and was given oral phenobarbital. Hemolytic hyperbilirubinemia was excluded on the basis of laboratory tests. Heterozygote polymorphisms of the promoter region (-3279T>G) and exon 1 (211G>A) were found in UGT1A1 gene. After discharge, the patient did not require any further treatment. This is the first case of proven Gilbert's syndrome with severe neonatal hyperbilirubinemia in Korea.

Effects of the microbiological properties and pathogenicity of Photobacterium damselae subsp. damselae under different culture conditions (배양 조건이 Photobacterium damselae subsp. damselae의 미생물학적 성상 및 병원성에 미치는 영향)

  • The effects of microbiological properties and pathogenicity of Photobacterium damselae subsp. damselae were investigated under different culture conditions, temperature, pH, NaCl and iron concentration on culture media. Favorable conditions for bacterial growth were between 15-30${^{\circ}C}$, pH 5-9, 0-4% NaCl concentration and iron contents of over 10 mM, whereas the bacterial growth was inhibited under iron chelator existence. When P. damselae was cultured in iron-limited tryptic soy broth, total protein concentration of extracellular products, cytotoxic ability of ECPs on cell line, bacterial viability in flounder serum, phospholipase and siderophore activities of ECPs were significantly increased. On the other hand, the activities of P. damselae cultured under iron-added conditions were decreased. In this study, the iron-limited conditions were similar to the host in which iron concentration is low. During infection caused by P. damselae, the conditions could be related to the pathogenesis of the pathogen.
