Fungistatic Activity of Soil Applied with Organic Materials Against Phytophthora capsici (고추역병균(疫病菌)에 대(對)한 유기물(有機物) 시용토양(施用土壤)의 용균작용(溶菌作用))

  • We have obtained following results by executing this experiment to define effects of organic substances applied and temperature on fungistatic effect which is regarded as basic factor of inhibition against hot-pepper blight affection : 1. Fungistatic effect of soil on Phytophthora capsici is enlarged by application of organic material such as rice straw rather than matured composts. Morever, it is confirmed that fungistatic efect is more eminent at the temperature of $15^{\circ}C$ and $5^{\circ}C$ than at $30^{\circ}C$, Degree of fungistatic effect of soil tends to decrease as the goe on from the moment of using organic substance generally, in soil applied as the goes on from the moment of using organic substance generally, in soil applied rice straw it is prove that fungistatic effect is maintained stably in a long term. 2. Effect of applicating organic substances and treating by temperature on production of zoosporangium of Phytophthra capsici is reviewed. In case that sterilized soil solution is added on cultured hypae of Phytophthra capsici, increase in the production of zoosporangium is found which is statistically significant(at degree of 1%), while decrease effect statistically significant is found in case of adding soil solution used for applying bark compost. However, defferences between treatments of temperature is not found. On the other hand, when nonsterilized soil solution is added, significant defference between types of organic substance is not found, while significant difference is found between treatments of temperature, therefore formation of zoosporangium is increased at the condition of $5^{\circ}C$ comparing to that of $30^{\circ}C$. 3. Putting above result of examination concerning to composition of soil microrganisms in each treated soil and to fungistatic effect of them, it seems that fungi and bacteria in soil is mostly related to fungistatic activity of soil.

Lytic Action of Egg White Lysozyme Isolated from Ogol Fowl on Staphylococcus aureus Phage Type 29 (Staphylococcus aureus Phage Type 29에 대한 오골계 난백 Lysozyme의 용균성)

  • This experiment was carried out to investigate the bacteriolytic action of the egg white lysozyme isolated from Korean native Ogol fowl and to obtain the data for utilization of the enzyme as a food preservative. Staphyococcus aureus phage type 29 and Bacillus subtilis ATCC 6633 among the microorganisms tested were lyzed by the treatment with 0.05% lysozyme, but Staphylococcus aureus phage type 57 in addition to E. coli etc. was found to be a lysozyme- insensitive species. The lysis of S. aureus phage type 29 was maximized when incubated in nutrient broth (pH 7.0) at $37^{\circ}C$ for 24 hours and suspended it to absorbance 0.6 at 540nm in 0.05M sodium acetate but fer (pH 4.5) and then treated it with the 0.05% lysozyme for 30 min. at $30^{\circ}C$. It was found that the effect of 0.05% lysozyme in combination with 1% glycine on the growth inhibition of S. aurecus phage type 29 increased more 50% than that in the absence of glycine, but not effect with other any additeves and metal ions tested.

  • Lytic activities of the egg white lysozyme from Korea-native Ogol fowl against the alkalophilic and thermophilic Bacillus sp. TA-11 were investigated and compared. Lytic activity of the Ogol fowl lysozyme for Bacillus sp. TA-11 was the highest for the cell of post-logarithm phase and optimum concentration of the lysozyme was 0.25%, Optimum reaction pH and temperature were 4.5 and 35$^{\circ}C$, respectively. Lytic activity of egg white lysozyme from fowl for Bacillus sp. TA-11 was the highest for the cell of stationary phase and optimum concentration of the lysozyme was 0.5%. Optimum reaction pH and temperature were 5.5 and 4$0^{\circ}C$, respectively.

  • The yeast, Hansenula anomala B-7, isolated from the $Cd^{2+}$ rich soils and determined to be tolerant in the high concentration of $Cd^{2+}$ were employed in this work. Its intact cells grown in high concentration of $Cd^{2+}$ were observed to be Iysed at the early stage when transferred to a cadmium deficient broth. Its intact cells found to be not Iysed and grow well under the high concentration of $Cd^{2+}$. The Iysis of the intact cells grown at the high concentration of $Cd^{2+}$ ion was not found when the metal ions were replaced with $Cd^{2+}$ ion in the same concentration. This result indicated that Iysis of yeast cells, at least in this isolate, would be related to cell osmosis with the mineral ions added.

Enumeration of Weissella cibaria phage with cytometry, epifluorescence microscopy, and plaque assay (유세포분석기, 형광현미경, 용균반검사 분석을 이용한 Weissella cibaria 박테리오파지 정량분석 및 상관관계분석)

  • Quantitative analysis for non-host infection bacteriophage was conducted for their enumeration. Flow cytometry and epifluorescence microscopy (EPM) were selected as counting methods. Correlation analysis was performed based on the plaque assay method on the existing host infection and consisted of Pearson correlation statistical analysis, regression analysis, and difference analysis. Analyses of 12 samples with flow cytometry and plaque assay methods showed that there was a correlation of 96.7% with Pearson correlation value r=0.967, $R^2$ 0.9352, and difference value of 1.063. Analyses of 12 samples with EPM and plaque assay methods showed that there was a correlation of 99.0% with Pearson correlation value r=0.990, $R^2$ 0.9811, and difference value of 1.605. Therefore, flow cytometry and epifluorescence microscopy would be effective for enumeration of Weissella cibaria bacteriophage with plaque assay.

  • We examined some properties of yeast cell wall lytic enzyme produced by Dicyma sp. YCH-37. Several metal ions, reducing reagents, and chemical modifiers have little effects on the lytic activity, except guanidine-HCl. Yeast cells of early log phase were more susceptible to the enzyme than those of stationary phase, and heat-treated cells were more easily lysed than intact living ones. Yeast cells pretreated with organic solvents such as butanol and acetone were more susceptible to the enzyme than intact living ones. Yeast cells cultured in Yeast extract-Malt extract medium containing 0.5 M ammonium sulfate were easily lysed by the lytic enzyme, and yeast cells cultured without shaking were more easily lysed by the enzyme than those with shaking. When SDS, ${\beta}-mercaptoethanol$, Triton X-100, sodium sulfite, and KCl were added to enzyme reaction mixture each, lysis of yeast cells was more effective.
