Studies on the Nonvolatile Organic Acids in the Extracts of Dried Squid (건오징어 추출물의 유기산 조성에 관한 연구)

  • The extracts of dried squid(Sepiella maindron) were prepared with water and 70% ethanol at boiling point and the major nonvolatile organic acids in the extracts were investigated. Eight kinds of nonvolatile organic acid were identified as lactic oxalic succinic malic citric pyroglutamic malonic and ${\beta}-ketoglutaric$ acid. Total amount of nonvolatile organic acid in the extracts prepared with water changed from 205.2mg% to 310.1mg% with extraction time while the extracts prepared with 70% ethanol were 272.4 mg% ~ 347.6 mg% The major compo-nents were in the range of 61.7% ~ 70.9% lactic acid 16.4 ~ 22.6% pyroglutamic acid 5.0 ~ 9.9% succinic acid 4.0 ~ 8.2% citric acid and total amount of nonvolatile organic acids in the extracts increased until 2 hour of extraction regardless of extraction solvent and then decreased slowly.

Infection Status of Todarodes pacificus (Mollusca: Cephalopoda) with Anisakid Larvae in the Markets from Jumungin (주문진 어시장 살오징어(Todarodes pacificus)의 아니사키스 유충 감염상)

  • A total of 248 specimens of Todarodes pacificus were purchased from the Jumungin fishery market at the East Sea area two times during the period from November 1 to December 30, 1995. Samples were examined for their infection status with larvae anisakid. Anisakid larvae were collected from muscle, viscera and omentum. One hundred sixteen larval anisakids sorted from 34 specimens of T. pacificus (13.7% of infection rate) ranged from 13.5 to 29.5 mm in their body length. They were classified based on morphological and morphometric observations as follows; Anisakis type I larvae (68 larvae, 58.6%: positive rate), Contracaecum type A (5, 4.3%), Anisakis type II (17, 14.5%), Contracaecum type D (8, 7.0%) and unknown type (18, 15.5%).

  • The paper deals the fishing condition of the stern trawlers operated in the Atlantic coast of Africa. The datas are gathered from the Korean stern trawlers operated in the area from June, 1975 to May, 1976. The obtained results are summarized as follows: 1. The mean catch per haul are calculated as, cuttle fish 14.5kg, large size squid(more than :lOOg of body weight) 28.2kg, small size squid(less than 300g) 36.4kg, octopus 47.0kg. 2. Small size cuttle fish(less than5OOg of body weight) are caught much during after spring tide in July to September, yet, large size ClIttle fish(more than 500g) are caught much during neap tide in October to January. 3. Small size squid(less than 300g of body weight) are cBught much during after spring tide in October to December, yet, large size squid(more than 3OOg) are caught much during before spring tide in October to March. 4. Octopus are caught during neap tide in July to August.

  • It was studied to the displacement and the amount of catch for migratory fishes with NOAA/AVHRR SST(Sea Surface Temperature) from 1988 to 2000 in the Korean Seas. The analyzed results from SST data showed generally the oceanic warming trend in the Korean Seas. On the increasement of SST, the distributed areas of migratory fishes which living in the warm waters were displaced gradually to the northward directions(high latitude) and then the amount of catch was increased during this studied periods against to migratory fishes which living in the cold waters.

  • This study was conducted to examine the effects of gamma irradiation on the shelf-life and sensory scores of squid Sundae under accelerated storage conditions. Squid Sundae was stored at $37^{\circ}C$ for 35 days following gamma irradiation at doses of 0, 10, and 20 kGy. For total viable cell counts, control and gamma-irradiated (GI) (10 kGy) squid Sundae were already spoiled in 4 days, whereas GI (20 kGy) squid Sundae showed complete suppression of bacterial growth during storage. There were no significant changes in pH values compared to the control. The VBN and TBARS (thiobarbituric acid reactive substance) values of GI (20 kGy) squid Sundae were significantly lower than those of the control. In addition, the induction period of GI (20 kGy) squid Sundae as measured by a Rancimat showed a higher level compared to that of the control. In the sensory evaluation, there were no significant changes between the control and GI samples. These results suggest that a dose of 20 kGy is the optimum and effective dose for preservation of squid Sundae.

Catch Variation and Fishing Period of the Set Net Fishery in Coastal Waters of Jeju Island (제주도 연안 정치망 어획량 변동과 어기)

  • The fisheries resources of the set net fishery on four fishing grounds (Pyuong Dae, Gu Eum, Du Moah, Gang Jeong) in coastal waters of Jeju Island were studied to the determine catch-variation characteristics from January, 1998 to December, 1999. A total catch of 153,862 kg was made through the survey period. The large total was related mainly to the high catch of Trachurus japonicus, which comprised 58.2% of the total. Other dominant species which account for 23.0% of the total catch were Siganus fuscescens, Todarodes pacificus, Loligo chinesis, Seriola lalandi, Sepioteuthis lessoniana, and Seriola quinqueradiata. Catch density was high in Gu Eum (more than 100,000 kg), whereas those in Pyuong Dae, Du Moah, and Gang Jeong were less than 30,000 kg in total. The fishing period of the set net fishery was from April to December, which was also closely related to the variation of the sea water temperature in the fishing grounds. The ranges of water temperature in the four fishing grounds were $18{\sim}26^{\circ}C$ in Pyuong Dae, $16{\sim}26^{\circ}C$ in Du Moah, $15{\sim}26^{\circ}C$ in Gu Eum, and $15{\sim}26^{\circ}C$ in Gang Jeong.

Development and Validation of Quick and Accurate Cephalopods Grouping System in Fishery Products by Real-time Quantitative PCR Based on Mitochondrial DNA (두족류의 진위 판별을 위한 Real-time Quantitative PCR 검사법 개발 및 검증)

  • In this study, an approach for the analysis of the five cephalopod species (octopus, long-arm octopus, squid, wet-foot octopus, beka squid) consumed in the Republic of Korea is developed. The samples were collected from the Southeast Asian countries Thailand, Indonesia, Vietnam, and China. The SYBR-green-based real-time qPCR method, based on the mitochondrial DNA genome of the five cephalopods was developed and validated. The intergroup variations in the mitochondrial DNA are evident in the bioinformatic analysis of the mitochondrial genomic DNA sequences of the five groups. Some of the highly-conserved and slightly-variated regions are identified in the mitochondrial cytochrome-c-oxidase subunit I (COI) gene, 16s ribosomal RNA (16s rRNA) gene, and 12s ribosomal RNA (12s rRNA) gene of these groups. To specify each five cephalopod groups, specific primer sets were designed from the COI, 16s rRNA and 12s rRNA regions. The specific primer sets amplified the DNA using the SYBR-green-based real-time PCR system and 11 commercially secured animal tissues: Octopus vulgaris, Octopus minor, Todarodes pacificus, Dosidicus gigas, Sepia esculenta, Amphioctopus fangsiao, Amphioctopus aegina, Amphioctopus marginatus, Loliolus beka, Loligo edulis, and Loligo chinensis. The results confirmed by a conveient way to calculate relative amplification levels between different samples in that it directly uses the threshold cycles (Ct)-value range generated by the qPCR system from these samples. This genomic DNA-based molecular technique provides a quick, accurate, and reliable method for the taxonomic classification of the animal tissues using the real-time qPCR.

Optimization of Coho Salmon Hydrolysate Using Japanese Squid Liver and Its Properties (일본산 오징어 간을 이용한 은연어 가수분해물 제조의 최적화와 가수분해물의 특성)

  • In this study, the optimal conditions for salmon hydrolysate using squid liver and compositional properties of hydrolysate were investigated. The optimal conditions were $55^{\circ}C$, pH 5.5 and 0.66~0.67% (w/w) in the ratio of squid liver to acidic and thermal treated salmon muscle. The free amino acid of hydrolysate from the acidic treated salmon muscle was higher than that of hydrolysate from the thermal treated salmon muscle, while the total amino acid and mineral were high in the acidic treated salmon muscle. Furthermore, cadmium of hydrolysate from the thermal denatured salmon muscle was below 2 ppm, and has an acceptable level as potential ingredient. The distribution of peptide molecular weight was 40.0% for 1.0~9.5 kDa, 6.7% for 0.5 kDa, and 47.4% of others in hydrolysate from the thermal treated salmon muscle. Both hydrolysates did not show any toxicity against the HepG2 cell line for up to $200{\mu}g/mL$.

  • To detect naturally occurring bioactive compounds from unused marine resources, the screening for the 5-lipoxygenase(potato lipoxygenase-II) inhibitors in Asterina pectinifera, Halocynthia roretzi skin, Nototodarus sloani ink, Anthocidaris crassispina skin, Sargassum horneri, Agarum cribrosum, Odonthalia corymbifera and Desmarestia ligulata was carried out. Water, ether, acetone and methanol fractions extracted from Sargassum horneri had strong inhibitory effect on enzymatic lipid oxidation by potato lipoxygenase-II, and their $IC_{50}$ were 320, 18, 9.5 and $100\;{\mu}g/mL$, respectively. The $IC_{50}$ of ether fraction extracted from Asterina pectinifera and acetone fraction extracted from Nototodarus sloani ink were 29.5 and $34.3\;{\mu}g/mL$, and these extracts showed relatively excellent inhibitory activity. Nonpolar solvent (ether, acetone) extracts of tested marine organisms had more inhibitory effect against 5-lipoxygenase than the polar solvent(water) extracts.

Identification of Lactic Acid Bacteria and Changes of Organic Acid during Aging of Traditional Kyungsando Squid sikhe (경상도 전통 마른 오징어 식해로부터 젖산균의 분리, 동정 및 숙성과정 중 유기산의 변화)

  • The objective of this study is to investigate identification of lactic acid bacteria and changes of organic acid during aging of traditional Kyungsando Squid sikhe. Lactobacillus brevis SH-1, Lactobacillus plantarum SH-2, and Leuconostoc mensenteroides SH-3, which were isolated from Kyungsando Squid sikhe were selected for fermentation. The viable cell and lactic acid bacteria counts of squid sikhe was increased on fermentation days, and slowly decreased after 10 days of fermentation. The viable cell and lactic acid bacteria counts of standard sikhe at 6 days fermentation was $3.7{\times}10^9$ and $8.2{\times}10^8\;CFU/g$, respectively. The organic acid of squid sikhe consisted of 7 kinds including lactic acid during fermentation. The contents of citric acid, succinic acid, and lactic acid gradually increased during fermentation, and especially, increasing of lactic acid remarkably increased than the different organic acid. The contents of acetic acid and tartaric acid slowly increased after 7 days of fermentation.
